    Centrosome fragments and microtubules are transported asymmetrically away from division plane in anaphase

    Spinning disc confocal microscopy of LLCPK1 cells expressing GFP-tubulin was used to demonstrate that microtubules (MTs) rapidly elongate to the cell cortex after anaphase onset. Concurrently, individual MTs are released from the centrosome and the centrosome fragments into clusters of MTs. Using cells expressing photoactivatable GFP-tubulin to mark centrosomal MT minus ends, a sevenfold increase in MT release in anaphase is documented as compared with metaphase. Transport of both individually released MTs and clusters of MTs is directionally biased: motion is directed away from the equatorial region. Clusters of MTs retain centrosomal components at their focus and the capacity to nucleate MTs. Injection of mRNA encoding nondegradable cyclin B blocked centrosome fragmentation and the stimulation of MT release in anaphase despite allowing anaphase-like chromosome segregation. Biased MT release may provide a mechanism for MT-dependent positioning of components necessary for specifying the site of contractile ring formation

    The parish churches, cathedral, and corporation of Salisbury: continuity and change c.1480-c.1650

    An analysis of the accounts of the churches of Salisbury, in conjunction with the ledgers of the civic authority, provides evidence of change and continuity in the worship of the citizens and the social role of religion for a single city in the sixteenth and seventeenth centuries. This occurred during a period when the jurisdiction and control of the bishop was reduced from the whole city to just the cathedral and its surrounding close. At the same time, the civic authorities became more involved in the business of the church, and by the time of the Civil War it was employing lecturers and providing housing for the ministers of the city churches. Individual chapters explore lay devotion in the form of the trade guilds, and the cults of St Osmund and the Holy Name; illumination including the provision of window glass and the use of artificial illumination in which tallow became more commonly used; the interior and exterior soundscape of bells, organs and other music, and an examination of the Caroline church through the trial of the iconoclast Henry Sherfield. Both change and continuity in all of these aspects can be seen during the period of the long Reformation, along with the sensory experiences of the laity, and the construction of artefacts including windows and organs. This study has revealed some atypical findings in the city of Salisbury: firstly, the continuation of trade guild celebrations during the long Reformation, including church worship and the memorialization of members. Secondly, through experimentation with beeswax and tallow, where evidence is provided that tallow candles may not have produced such a poor light or smoky environment as previously thought. Thirdly, it is shown that church bells continued to be rung in the city, their use evolving over time, as the number of religious services reduced, whilst ringing to announce local and national events increased. Finally, music continued to be sung and played during the sixteenth and seventeenth centuries, with the purchase of replacement organs and their continued maintenance, including the instrument in the church of St Edmund, despite the congregation being known for its puritan tendencies

    AbstractPrP has a central role in the Transmissible Spongiform Encephalopathies (TSEs), and mutations and polymorphisms in host PrP can profoundly alter the host's susceptibility to a TSE agent. However, precisely how host PrP influences the outcome of disease has not been established. To investigate this we have produced by gene targeting a series of inbred lines of transgenic mice expressing different PrP genes. This allows us to study directly the influence of the host PrP gene in TSEs. We have examined the role of glycosylation, point mutations, polymorphisms and PrP from different species on host susceptibility and the disease process both within the murine species and across species barriers

    Textural and geochemical constraints on andesitic plug emplacement prior to the 2004–2010 vulcanian explosions at Galeras volcano, Colombia

    Hazardous sequences of vulcanian explosions are thought to result from the repeated emplacement and destruction of degassed, highly crystalline magma plugs in the shallow conduit of arc volcanoes. The processes governing the timing and magnitude of these explosions are thought to be related to magma ascent rate and efficiency of degassing and crystallisation. We study a rare suite of time-constrained ballistic bombs from the 2004–2010 period of activity of Galeras volcano to reconstruct magma plug architecture prior to six individual vulcanian explosions. We find that each plug was vertically stratified with respect to crystallinity, vesicularity and melt volatile content, melt composition and viscosity. We interpret this structure as resulting from multiple bubble nucleation events and degassing-driven crystallisation during multi-step ascent of the magma forming the plug, followed by spatially variable crystallisation within the plug under contrasting conditions of effective undercooling created by degassing. We propose that the shallow conduit evolved from more open degassing conditions during 2004–2008 to more closed conditions during 2009–2010. This resulted in explosions becoming smaller and less frequent over time during 2004–2008, then larger and more frequent over time during 2009–2010. This evolution was controlled by changing average ascent rates and is recorded by systematic changes in plagioclase microlite textures. Our results suggest that small volume vulcanian explosions (~ 105 m3) should generally be associated with longer repose times (hundreds of days) and produce ballistics characterised by small numbers of large, prismatic plagioclase microlites. Larger volume vulcanian explosions (~ 106 m3) should be associated with shorter repose times (tens of days) and produce ballistics characterised by high numbers of small, more tabular plagioclase microlites

    The responses of an anaerobic microorganism, Yersinia intermedia MASE-LG-1 to individual and combined simulated Martian stresses

    The limits of life of aerobic microorganisms are well understood, but the responses of anaerobic microorganisms to individual and combined extreme stressors are less well known. Motivated by an interest in understanding the survivability of anaerobic microorganisms under Martian conditions, we investigated the responses of a new isolate, Yersinia intermedia MASE-LG-1 to individual and combined stresses associated with the Martian surface. This organism belongs to an adaptable and persistent genus of anaerobic microorganisms found in many environments worldwide. The effects of desiccation, low pressure, ionizing radiation, varying temperature, osmotic pressure, and oxidizing chemical compounds were investigated. The strain showed a high tolerance to desiccation, with a decline of survivability by four orders of magnitude during a storage time of 85 days. Exposure to X-rays resulted in dose-dependent inactivation for exposure up to 600 Gy while applied doses above 750 Gy led to complete inactivation. The effects of the combination of desiccation and irradiation were additive and the survivability was influenced by the order in which they were imposed. Ionizing irradiation and subsequent desiccation was more deleterious than vice versa. By contrast, the presence of perchlorates was not found to significantly affect the survival of the Yersinia strain after ionizing radiation. These data show that the organism has the capacity to survive and grow in physical and chemical stresses, imposed individually or in combination that are associated with Martian environment. Eventually it lost its viability showing that many of the most adaptable anaerobic organisms on Earth would be killed on Mars today

    Controls on explosive-effusive volcanic eruption styles

    One of the biggest challenges in volcanic hazard assessment is to understand how and why eruptive style changes within the same eruptive period or even from one eruption to the next at a given volcano. This review evaluates the competing processes that lead to explosive and effusive eruptions of silicic magmas. Eruptive style depends on a set of feedbacks involving interrelated magmatic properties and processes. Foremost of these are magma viscosity, gas loss, and external properties such as conduit geometry. Ultimately, these parameters control the speed at which magmas ascend, decompress and outgas en route to the surface, and thus determine eruptive style and evolution
