Differences in the microbial profiles of early stage endometrial cancers between Black and White women
Objective: Black women suffer a higher mortality from endometrial cancer (EC) than White women. Potential biological causes for this disparity include a higher prevalence of obesity and more lethal histologic/molecular subtypes. We hypothesize that another biological factor driving this racial disparity could be the EC microbiome. Methods: Banked tumor specimens of postmenopausal, Black and White women undergoing hysterectomy for early stage endometrioid EC were identified. The microbiota of the tumors were characterized by bacterial 16S rRNA sequencing. The microbial component of endometrioid ECs in The Cancer Genome Atlas (TCGA) database were assessed for comparison. Results: 95 early stage ECs were evaluated: 23 Black (24%) and 72 White (76%). Microbial diversity was increased (p < 0.001), and Firmicutes, Cyanobacteria and OD1 phyla abundance was higher in tumors from Black versus White women (p < 0.001). Genus level abundance of Dietzia and Geobacillus were found to be lower in tumors of obese Black versus obese White women (p < 0.001). Analysis of early stage ECs in TCGA found that microbial diversity was higher in ECs from Black versus White women (p < 0.05). When comparing ECs from obese Black versus obese White women, 5 bacteria distributions were distinct, with higher abundance of Lactobacillus acidophilus in ECs from Black women being the most striking difference. Similarly in TCGA, Dietzia and Geobacillus were more common in ECs from White women compared to Black. Conclusion: Increased microbial diversity and the distinct microbial profiles between ECs of obese Black versus obese White women suggests that intra-tumoral bacteria may contribute to EC disparities and pathogenesis
Hypertensive conditions of pregnancy, preterm birth, and premenopausal breast cancer risk: a premenopausal breast cancer collaborative group analysis
Purpose: Women with preeclampsia are more likely to deliver preterm. Reports of inverse associations between preeclampsia and breast cancer risk, and positive associations between preterm birth and breast cancer risk are difficult to reconcile. We investigated the co-occurrence of preeclampsia/gestational hypertension with preterm birth and breast cancer risk using data from the Premenopausal Breast Cancer Collaborative Group. Methods: Across 6 cohorts, 3096 premenopausal breast cancers were diagnosed among 184,866 parous women. We estimated multivariable hazard ratios (HR) and 95% confidence intervals (CI) for premenopausal breast cancer risk using Cox proportional hazards regression. Results: Overall, preterm birth was not associated (HR 1.02, 95% CI 0.92, 1.14), and preeclampsia was inversely associated (HR 0.86, 95% CI 0.76, 0.99), with premenopausal breast cancer risk. In stratified analyses using data from 3 cohorts, preterm birth associations with breast cancer risk were modified by hypertensive conditions in first pregnancies (P-interaction = 0.09). Preterm birth was positively associated with premenopausal breast cancer in strata of women with preeclampsia or gestational hypertension (HR 1.52, 95% CI: 1.06, 2.18), but not among women with normotensive pregnancy (HR = 1.09, 95% CI: 0.93, 1.28). When stratified by preterm birth, the inverse association with preeclampsia was more apparent, but not statistically different (P-interaction = 0.2), among women who did not deliver preterm (HR = 0.82, 95% CI 0.68, 1.00) than those who did (HR = 1.07, 95% CI 0.73, 1.56). Conclusion: Findings support an overall inverse association of preeclampsia history with premenopausal breast cancer risk. Estimates for preterm birth and breast cancer may vary according to other conditions of pregnancy
Geographical and temporal distribution of SARS-CoV-2 clades in the WHO European Region, January to June 2020
We show the distribution of SARS-CoV-2 genetic clades over time and between countries and outline potential genomic surveillance objectives. We applied three available genomic nomenclature systems for SARS-CoV-2 to all sequence data from the WHO European Region available during the COVID-19 pandemic until 10 July 2020. We highlight the importance of real-time sequencing and data dissemination in a pandemic situation. We provide a comparison of the nomenclatures and lay a foundation for future European genomic surveillance of SARS-CoV-2.Peer reviewe
Inhibitors of COP-mediated Transport and Cholera Toxin Action Inhibit Simian Virus 40 Infection
Simian virus 40 (SV40) is a nonenveloped virus that has been shown to pass from surface caveolae to the endoplasmic reticulum in an apparently novel infectious entry pathway. We now show that the initial entry step is blocked by brefeldin A and by incubation at 20degreesC. Subsequent to the entry step, the virus reaches a domain of the rough endoplasmic reticulum by an unknown pathway. This intracellular trafficking pathway is also brefeldin A sensitive. Infection is strongly inhibited by expression of GTP-restricted ADP-ribosylation factor 1 (Arf1) and Sar1 mutants and by microinjection of antibodies to betaCOP. In addition, we demonstrate a potent inhibition of SV40 infection by the dipeptide N-benzoyl-oxycarbonyl-Gly-Phe-amide, which also inhibits late events in cholera toxin action. Our results identify novel inhibitors of SV40 infection and show that SV40 requires COPI- and COPII-dependent transport steps for successful infection

Study of hydrogen-powered versus battery-powered automobiles
A study has been conducted of two future candidate automobile propulsion systems that do not rely upon petroleum or natural gas as an energy source. Potential vehicle characteristics for each system have been identified. The first vehicle system employs a gaseous, hydrogen-fueled, internal combustion engine and either a liquid or metal hydride energy storage system. The second vehicle system employs an electronically controlled, electric motor powertrain and a battery energy storage system. Major tasks included in this study were the technical and economic assessments of the state of the art and future alternatives in hydrogen production and delivery, the hydrogen vehicle assessment, the battery-electric vehicle assessment, and the comparison of the principal vehicle alternative in 1985, 1990, and 2000. The comparison includes weight, size, cost, energy, and design range relationships and the implications on expenditure of all major energy sources. The study is summarized, results are presented, and conclusions are drawn. Comments are made on the future roles of hydrogen and electricity in automobile propulsion
Study of hydrogen-powered versus battery-powered automobiles
A study conducted to compare the technological status and the resultant potential vehicle characteristics for hydrogen- and battery-powered automobiles that could be produced from 1985 to 2000 is documented in 3 volumes. The primary objectives of the study were: the assessments of applicable energy storage and propulsion technology for the two basic vehicle types (applied to four-passenger cars); a rigorous comparison of vehicle weight, size, and usefulness versus design range; and an investigation of the relative efficiencies of expending energy from various primary sources to power the subject vehicle. Another important objective, unique to hydrogen powered vehicles, was the assessment of the technology, logistics, and cost implications of a hydrogen production and delivery capability. This volume, Volume III, contains three major sections: the assessment of battery electric vehicle technology for energy storage and the drivetrain system; the technical and economic comparison of hydrogen- and battery-powered vehicles derived primarily from data in the previous vehicle technology assessments, with consideration of alternative energy sources; and a series of appendices that support the vehicle definitions and comparisons
