    Immunization against Hepatitis B Surface Antigen (HBsAg) in a Cohort of Nursing Students Two Decades after Vaccination: Surprising Feedback

    Health-care students can be exposed to biological risks during university training. The persistence of long-term immunogenicity against hepatitis B virus (HBV) was analyzed in a cohort of nursing students two decades after primary vaccination. A total of 520 students were enrolled at the University of Palermo and were evaluated for levels of anti-HBsAg antibodies. ThestudentswereexaminedduringtheïŹrstyearoftheirDegreeCourseandwerecheckedtwoyears later. All students with anti-HBsAg <10 mIU/mL during their ïŹrst or third year were boosted within onemonth. Theproportionofstudentsthatwerevaccinatedduringadolescenceshowinganti-HBsAg ≄10 mIU/mL was higher than that observed in students who were vaccinated during infancy (69% versus31.7%;p-value<0.001). ReceivingHBVvaccinationatadolescencewassigniïŹcantlyassociated with a fourfold increased possibility of having anti-HBsAg titers≄10 mIU/mL (adj-OR = 4.21, 95% CI: 2.43–7.30). Among the students who were checked at the third year and boosted after the ïŹrst year (n = 279), those who were vaccinated during infancy showed a higher percentage of antibody titers <10 mIU/mL (20.3% versus 8.7% among vaccinated during adolescence; p < 0.01). This study conïŹrms that HBV vaccination at adolescence might determine a higher long-term persistence of anti-HBsAg titers≄10 mIU/mL and that anti-HBV booster could increase levels of anti-HBsAg over a relatively short period, especially in subjects who were vaccinated during infancy

    The Logic of Probability: A Trip through Uncertainty

    In real life we have to deal with uncertainty, imprecision and vagueness. Many ideas were introduced and studied in detail to manage with these problems. Now we briefly expose the main formal concepts which describe non-ideal situations, i.e. Probability, Statistics and Fuzzy Logic. Probability has recent origins with respect to other branches of mathematics which have deep roots in the past, like geometry or algebra.We may say all this started with Antoine Gombaud, Chevalier de MĂ©rĂ© (1607–1684), who asked Blaise Pascal (1623–1662) about gambling with dice. The correspondence between Pierre de Fermat and Blaise Pascal, which began in 1654, initially on these questions, led to the introduction of basic concepts, i.e. probability and expectation. Only in 1657, Christian Huygens in "De Ratiociniis in ludo aleae" proposed a first systematic study of the new branch of mathematics. However, the need of an axiomatic construction of the theory of probability arose to analyze more general and complex situations than gambling. A strong formalization was supplied by the monograph "Foundations of the theory of probability" (1933) by Andrey Nikolaevich Kolmogorov.Statistics represent the most popular application of probability theory, providing research tools in several areas, including physical and natural sciences, technology, psychology, economics and medicine. Statistics are the bridge that connects experimental data to the mathematical theory behind itself.Fuzzy logic, sometime confused with probability, wants to express and formalize all the sentences which are not true or false at all; the philosophical idea is that "everything is a matter of degree" (Zadeh). La logica della probabilitĂ : un viaggio attraverso l’incertezzaNella vita reale ci si trova di fronte a molte situazioni caratterizzate da incertezza, imprecisione, vaghezza. Sono state introdotte diverse modellizzazioni per il trattamento di tali concetti e problemi. Ci proponiamo di esporre sinteticamente alcuni lineamenti fondamentali di ProbabilitĂ , Statistica e Fuzzy Logic.La probabilitĂ  ha origini recenti rispetto alle altre branche della matematica che hanno profonde radici nel passato, come la geometria o l’algebra.Possiamo dire che un passaggio iniziale importante si Ăš avuto con Antoine Gombaud, Chevalier de MĂ©rĂ© (1607-1684), che pose a Blaise Pascal (1623-1662) una questione riguardante il gioco dei dadi. La corrispondenza tra Pierre de Fermat e Blaise Pascal, che ha avuto inizio nel 1654, su questioni simili, ha portato all'introduzione di concetti di base, come probabilitĂ  e aspettativa. Successivamente Christian Huygens, in "De ludo Ratiociniis in aleae", ha proposto un primo studio sistematico della nuova branca della matematica. Tuttavia, la necessitĂ  di una costruzione assiomatica della teoria della probabilitĂ  sorse per l’esigenza di analizzare situazioni piĂč generali e complesse rispetto al gioco d'azzardo.     GeneBase 1.1: a tool to summarize data from NCBI gene datasets and its application to an update of human gene statistics

    We release GeneBase 1.1, a local tool with a graphical interface useful for parsing, structuring and indexing data from the National Center for Biotechnology Information (NCBI) Gene data bank. Compared to its predecessor GeneBase (1.0), GeneBase 1.1 now allows dynamic calculation and summarization in terms of median, mean, standard deviation and total for many quantitative parameters associated with genes, gene transcripts and gene features (exons, introns, coding sequences, untranslated regions). GeneBase 1.1 thus offers the opportunity to perform analyses of the main gene structure parameters also following the search for any set of genes with the desired characteristics, allowing unique functionalities not provided by the NCBI Gene itself. In order to show the potential of our tool for local parsing, structuring and dynamic summarizing of publicly available databases for data retrieval, analysis and testing of biological hypotheses, we provide as a sample application a revised set of statistics for human nuclear genes, gene transcripts and gene features. In contrast with previous estimations strongly underestimating the length of human genes, a 'mean' human protein-coding gene is 67 kbp long, has eleven 309\u2009bp long exons and ten 6355\u2009bp long introns. Median, mean and extreme values are provided for many other features offering an updated reference source for human genome studies, data useful to set parameters for bioinformatic tools and interesting clues to the biomedical meaning of the gene features themselves.Database URL: http://apollo11.isto.unibo.it/software/

    Le complexe GIT-PIX : Une plate-forme de régulation des GTPases ARF et Rac/Cdc42

    Les protĂ©ines G sont des commutateurs molĂ©culaires contrĂŽlant divers aspects de la vie cellulaire tels que la transduction du signal, la rĂ©organisation du cytosquelette et les mĂ©canismes de transport vĂ©siculaire. Ces protĂ©ines fixent alternativement les nuclĂ©otides GDP et GTP, ce qui leur permet de transiter entre deux conformations structurelles, respectivement inactive et active. L’activation des protĂ©ines G rĂ©sulte d’une interaction avec un facteur d’échange qui stimule la dissociation du GDP pour le remplacer par du GTP. C’est sous cette forme liĂ©e au GTP que les protĂ©ines G vont pouvoir interagir avec leurs effecteurs et les activer. La stimulation de l’activitĂ© intrinsĂšque d’hydrolyse des protĂ©ines G par des protĂ©ines GAP (GTPase activating protein) va conduire Ă  un retour vers la forme inactive liĂ©e au GDP. De nombreuses Ă©tudes ont montrĂ© l’existence d’interconnexions entre les voies de signalisation impliquant les GTPases ARF (ADP ribosylation factor) et Rac/Cdc42. La dĂ©couverte d’un complexe protĂ©ique comprenant PIX, un facteur d’échange pour Rac/Cdc42 et GIT1, une protĂ©ine GAP pour les ARF, a rĂ©cemment donnĂ© un nouvel Ă©clairage sur les moyens mis en oeuvre par la cellule pour rĂ©guler de façon Ă©troite l’agencement du cytosquelette et la dynamique des membranes. De plus, la plate-forme PIX-GIT1 est associĂ©e Ă  des rĂ©seaux de signalisation comprenant des suppresseurs de tumeur. Ces donnĂ©es rĂ©centes incitent Ă  prendre ce complexe en considĂ©ration dans le contexte des pathologies cancĂ©reuses.We recently described that the tumor suppressor factor Scribble anchors the PIX exchange factor for Rac/Cdc42 and the ARF-GAP GIT proteins at the plasma membrane. Because it has been postulated that the GIT-PIX proteins dimerize and tightly self-assemble to form a high molecular weight complex, this nexus may be capable of linking together important signalling molecules to control cytosqueleton polymerization and membrane dynamics. To date, most studies that have tempted to unravel the function of these proteins have found their implication in a great variety of cellular functions (receptor recycling, endo-exocytosis, cell migration, synapse formation
) but have mostly neglected to consider the multimeric organization of this hub. There is no doubt that our comprehension of physiopathological disorders such as cancers will be improved when the nature of the complex pathways integrated by the GIT-PIX nodule will be understood

    A partire da un articolo di Bernard Adamy pubblicato nello «Schopenhauer Jahrbuch» del 1985 intitolato Schopenhauer Rezeption bei Karl Kraus, il presente contributo intende mettere in evidenza gli aspetti che avvicinano il pensiero Kraus a Schopenhauer. Infatti, ispirandosi alle tesi schopenhaueriane sullo stile, con una pungente satira, nella sua «Die Fackel» Kraus attacca le istituzioni politiche e culturali della Vienna asburgica, denunciando soprattutto gli orrori della prima guerra mondiale e la corruzione della stampa, e contemporaneamente offre il suo punto di vista sullo stile e la teoria linguistica

    Origanum vulgare subsp. hirtum essential oil prevented biofilm formation and showed antibacterial activity against planktonic and sessile bacterial cells.

    Essential oils from six different populations of Origanum vulgare subsp. hirtum were compared for their antibiofilm properties. The six essential oils (A to F) were characterized by a combination of gas chromatography with flame ionization detector and gas chromatography with mass spectrometer detector analyses. All oils showed weak activity against the planktonic form of a group of Staphylococcus aureus strains and against a Pseudomonas aeruginosa ATCC 15442 reference strain. The ability to inhibit biofilm formation was investigated at sub-MIC levels of 200, 100, and 50 m g/ml by staining sessile cells with safranin. Sample E showed the highest average effectiveness against all tested strains at 50 m g/ml and had inhibition percentages ranging from 30 to 52%. In the screening that used preformed biofilm from the reference strain P. aeruginosa, essential oils A through E were inactive at 200 m g/ml; F was active with a percentage of inhibition equal to 53.2%.     Oncolytic Adenoviruses for Cancer Therapy

    Many immuno-therapeutic strategies are currently being developed to fight cancer. In this scenario, oncolytic adenoviruses (Onc.Ads) have an interesting role for their peculiar tumor selectivity, safety, and transgene-delivery capability. The major strength of the Onc.Ads is the extraordinary immunogenicity that leads to a strong T-cell response, which, together with the possibility of the delivery of a therapeutic transgene, could be more effective than current strategies. In this review, we travel in the adenovirus (Ads) and Onc.Ads world, focusing on a variety of strategies that can enhance Onc.Ads antitumoral efficacy, passing through tumor microenvironment modulation. Onc.Ads-based therapeutic strategies constitute additional weapons in the fight against cancer and appear to potentiate conventional and immune checkpoint inhibitors (ICIs)-based therapies leading to a promising scenario.Peer reviewe

    Set of Classical PCRs for Detection of Mutations in Candida glabrata FKS Genes Linked with Echinocandin Resistance

    Clinical echinocandin resistance among Candida glabrata strains is increasing, especially in the United States. Antifungal susceptibility testing is considered mandatory to guide therapeutic decisions. However, these methodologies are not routinely performed in the hospital setting due to their complexity and the time needed to obtain reliable results. Echinocandin failure in C. glabrata is linked exclusively to Fks1p and Fks2p amino acid substitutions, and detection of such substitutions would serve as a surrogate marker to identify resistant isolates. In this work, we report an inexpensive, simple, and quick classical PCR set able to objectively detect the most common mechanisms of echinocandin resistance in C. glabrata within 4 h.
