Insights into Campylobacter jejuni Desulforubrerythrin catalytic mechanism

Dissertation presented to obtain the Master Degree in Molecular Genetics and BiomedicineThe following work aims to contribute to a better understanding of systems involved in resistance to oxidative stress species, namely hydrogen peroxide. The work is focused in one protein from the pathogen Campylobacter jejuni: desulforubrerythrin. Desulforubrerythrin is a non-heme iron protein in which the catalytic centre harbours a diiron cluster. Besides, the protein has a desulforedoxin domain at the N-terminal and a rubredoxin domain at the C-terminal. With the objective of understanding the protein catalytic mechanism three site-directed mutants, as well the wild type protein, were over expressed in Escherichia coli, purified and studied through biochemical and spectroscopic techniques. The amino acid residues selected for mutations are two tyrosines near the catalytic centre (residues 59 and 127). These residues are strictly conserved in rubrerythrins; moreover in diiron centres containing proteins tyrosines play a role in dissipating oxidizing species of iron (IV) by forming a tyrosil radical. The selected residues were replaced by a phenalanine residue which gave rise to three mutants: Y59F, Y127F and Y59F Y127F. These were characterized having as reference the wild type protein. All proteins have a molecular mass of 24 kDa and are tetramers in solution. The EPR and UV-visible techniques confirmed the presence of the three metallic domains in the wild type and Y59F mutant. The Y127F mutant was successfully used to test a protocol for diiron centre reconstitution in desulforubrerythrin. Finally, crystals of the wild type and, for the first time, of the Y59F and double mutants were obtained. The X-ray data for the mutants were collected with a resolution of 1.9 Å and its structure will be determined

O caráter problemático dos "trabalhos de casa": Estudo de caso numa turma de 3.º ano do Ensino Básico

Este trabalho parte da constatação de divergências de opinião em relação à marcação de trabalhos para casa, aos alunos. Para identificar os pontos de discórdia, foram abordados os principais elementos que os definem: que são; os seus protagonistas, quando e como se realizam e para que é que servem. Com base nesses elementos, foram entrevistadas duas pessoas com opiniões opostas sobre os trabalhos para casa. Em concreto, a Diretora Pedagógica e o Professor Titular de uma turma da mesma instituição. O estudo revela que a divergência sobre os trabalhos para casa não reside no valor das atividades escolares além das aulas, como estratégia de aprendizagem em alunos do ensino básico. As divergências situam-se mais em elementos não essenciais do trabalho para casa: o local (ser em casa ou noutro lugar) e o tempo (para evitar inconvenientes em relação a períodos de descanso de pais e crianças)

Jornalismo digital : as práticas de produção dos conteúdos noticiosos no jornal Observador on time

O presente Relatório de Estágio propõe um debate acerca da temática do jornalismo digital, tendo como estudo de caso o jornal Observador On Time. As plataformas digitais, nomeadamente o jornal em questão, começam cada vez mais a adquirir um lugar de relevo na forma como se consome e distribui informação. Deste modo, a partir do cruzamento de fatores relacionados ao tema, tais como as próprias características de produção de conteúdos para o online, tentámos verificar como é que, hoje em dia, o jornalismo funciona no seu novo habitat. Perspetivando neste ângulo, o objetivo deste estudo prende-se, particularmente, com a curiosidade sobre o fenómeno a ser estudado: perceber como é que é realizada a produção jornalística num jornal que nasceu em ambiente digital, nomeadamente quanto à exequibilidade das potencialidades da Web na sua produção noticiosa. Para tal, decidimos elaborar inquéritos por questionário aos jornalistas do Observador, de forma a tentar entender como se relacionam com o meio, complementando essa informação com uma análise de conteúdos informativos da secção Mundo do próprio jornal. Ainda que o Observador seja um nativo do ambiente digital, a verdade é que os dados recolhidos se mostraram bastante aquém do esperado. A implementação de características como a hipertextualidade, multimedialidade, interatividade e instantaneidade traduziram-se num valor não apenas reduzido como também mecanizado.The present internship Report proposes a debate on the subject of digital journalism, having as a case study the location of the internship itself, the journal Observador On Time. Digital platforms, notably the newspaper in question, are increasingly beginning to gain prominence in the way that information is nowadays consumed and distributed. Therefore, by using the cross-referencing of factors related to the theme, as well as the characteristics of online content production, we will try to verify how today’s journalism works in its new habitat. From this point of view, the goal of this study is, in particular, based on the curiosity about the phenomenon to be studied: to perceive how journalistic production is carried out in a newspaper that was born in a digital environment, namely as to the feasibility of the Web's potentialities in its news production. Considering this, we decided to carry out questionnaire surveys on the journalists in order to understand how they relate to the medium, complementing this information with an analysis of the journal's own information content. Although the Observador is a native of the digital environment, the truth is that the data collected proved to be much less than expected, as the values of implementation of characteristics such as hypertextuality, multimedia, interactivity and instantaneity were very reduced and mechanized

A identidade através do brinquedo traduzida em criação artística

Relatório da Prática de Ensino Supervisionada, Mestrado em Ensino de Artes Visuais no 3º Ciclo do Ensino Básico e no Ensino Secundário, Universidade de Lisboa, Instituto de Educação, 2021O projeto A identidade através do brinquedo traduzida em criação artística desenvolvido no âmbito da prática de ensino supervisionada foi realizado na Escola Profissional Gustave Eiffel – polo Lumiar, com um grupo de 20 adolescentes do curso Técnico de Apoio à Infância, no âmbito da disciplina de expressão plástica. A falta de habitabilidade da palavra criatividade, familiaridade com o processo artístico e com a linguagem da obra de arte contemporânea eram lacunas no vocabulário e identidade do público-alvo. O presente projeto pretendeu consciencializar para a obra de arte contemporânea, criatividade plástica e da importância da arte na construção e expressão da própria identidade, indo ao encontro das lacunas encontradas na tentativa de conseguir colmatá-las. Trabalhar estas questões de forma a não desencadear medos, receios e bloqueios, foi uma preocupação e um cuidado inicial imprescindíveis para a fluidez do processo. A adolescência, fase de crescimento em que o grupo alvo se encontrava, foi igualmente tida em consideração, um período de transição da infância para a idade adulta, indissociável da palavra metamorfose, bem como da palavra identidade. Neste contexto, surgiu a ideia de trabalhar a partir do objeto brinquedo, familiar e nutrido de memórias, vivências, sentimentos e emoções de outrora, contendo em si os ingredientes propícios para o ato criativo e, ponto de partida, para diferentes perspetivas com novas formas de pensar, olhar e interpretar. A transformação que a identidade sofre desde a infância até à adolescência, foi simbolicamente a mesma que o objeto brinquedo padeceu ao ser recriado / interpretado através do ato criativo com uma linguagem artística contemporânea. Posteriormente integrado numa instalação grupal, a marca criativa individual versus criação coletiva, evidenciou a força do contexto grupal, fazendo sobressair cada criação individual. Neste projeto, educação e arte abraçam a emergência de contribuir para o processo de construção interno dos indivíduos, sendo a arte um campo privilegiado de atuação, desenvolvendo uma nova perspetiva da arte e, principalmente, de si mesmo.The art project “Identity through toy translated into artistic creation”, developed in the scope of supervised teaching practice, was carried out at the Escola Profissional Gustave Eiffel – Lumiar school, with a group of 20 students, from the Child Care Technician course, within the subject of plastic expression. The lack of habitability of the word creativity, familiarity with the artistic process and with the language of contemporary art were flaws in the vocabulary and identity of the target audience. This project aimed to raise awareness for contemporary art work, plastic creativity and the importance of art in the construction and expression of one's own identity, meeting the gaps found in the attempt to fill them. Working on these issues so as not to trigger fears and blockages was an essential initial care and concern for the fluidity of the process. Adolescence, the growth phase in which the target group was, and also taken into account, a period of transition from childhood to adulthood, inseparable from the word metamorphosis, as well as from the word identity. In this context, the idea of working from the object-toy arose, familiar and nourished by memories, experiences, feelings and emotions of the past, contained in itself favourable elements for the creative act and, acting as a starting point, for other perspectives, demanding new forms of thinking, watching and interpreting. The transformation that one’s identity undergoes from childhood to adolescence was symbolically the same that affected the object-toy when recreated / interpreted through the creation act with a contemporary artistic language. Later integrated in a group installation, the individual creative brand versus collective creation, highlighted the strength of the group context, making each individual creation stand out. In this project, education and art embrace the emergence of contributing to the internal construction process of the individuals, with art being a privileged field of action, developing a new perspective on art and, above all, on itself
