    Hair and salivary cortisol and their relationship with lifestyle, mood and cognitive outcomes in premanifest Huntington’s disease

    Salivary cortisol dysrhythmias have been reported in some, but not all studies assessing hypothalamic–pituitary–adrenal (HPA) axis function in Huntington’s disease (HD). These differences are presumed to be due to environmental influences on temporal salivary cortisol measurement. Further exploration of HPA-axis function using a more stable and longer-term measure, such as hair cortisol, is needed to confirm earlier findings. This study aimed to evaluate hair and salivary cortisol concentrations and their associations with clinical and lifestyle outcomes in individuals with premanifest HD (n = 26) compared to healthy controls (n = 14). Participants provided saliva and hair samples and data were collected on clinical disease outcomes, mood, cognition, physical activity, cognitive reserve, sleep quality and social network size to investigate relationships between clinical and lifestyle outcomes and cortisol concentrations. Hair and salivary cortisol concentrations did not significantly differ between the premanifest HD and control groups. No significant associations were observed between hair or salivary cortisol concentrations and cognitive, mood or lifestyle outcomes. However, hair cortisol concentrations were significantly associated with disease outcomes in individuals with premanifest HD. Significant associations between hair cortisol concentrations and measures of disease burden and onset may suggest a potential disease marker and should be explored longitudinally in a larger sample of individuals with HD

    Evaluation of a fecal shedding test to detect badger social groups infected with Mycobacterium bovis

    Bovine Tuberculosis (bTB) is an economically important disease affecting the cattle industry in England and Wales. bTB, caused by Mycobacterium bovis, also causes disease in the Eurasian badger (Meles meles), a secondary maintenance host. Disease transmission between these two species is bidirectional. Infected badgers shed M. bovis in their faeces. The UK Animal and Plant Health Agency (APHA) organised a comparative trial to determine the performance of tests in detecting M. bovis in badger faeces for the Department for Environment, Food, and Rural Affairs (DEFRA). Here we present the performance of the existing Warwick Fast24-qPCR test, and its modified version based on a high-throughput DNA extraction method (Fast96-qPCR). We found Fast24-qPCR to have a sensitivity of 96.7% (95%CI 94.5-99%, n=244) and a specificity of 99% (95%CI 97.8-100%, n=292). Fast96-qPCR requires further optimisation. Determining the disease status of badger social groups requires multiple tests per group. Therefore to increase specificity further, we independently repeated the Fast24-qPCR test on positive samples, increasing stringency by requiring a 2nd positive result. Fast24-qPCR with repeat testing had a sensitivity of 87.3% (95%CI 83.1-91.5%, n=244), and a specificity of 100% (95%CI 100-100, n=201) on an individual sample level. At the social group level, this repeat testing gives Fast24-qPCR high herd specificity, while testing multiple samples per group provides high herd sensitivity. With Fast24-qPCR we provide a social group level test with sufficient specificity and sensitivity to monitor shedding in badgers via latrine sampling, delivering a potentially valuable tool to measure the impacts of bTB control measures

    Schaffnerella Rediscovered! (Gramineae, Chloridoideae)

    From 1876 to 1880 in San Luis PotosĂ­, Mexico, J. G. Schaffner made the first collections of a small grass that later was named Schaffnerella gracilis (Chloridoideae). The monotypic genus apparently was not encountered again by botanists until 2001, when, during a targeted search, we discovered it in the Sierra de San Miguelito growing along the RĂ­o Potosino, ca. 6 air km southwest of the city of San Luis PotosĂ­. Most of the 100-150 plants encountered along a 3-km stretch of the RĂ­o Potosino above the village of Escalerillas and reservoir EI Potosino were growing in a moist alluvium of rock and sand. Historically known to occur some 10 km or more downstream near Morales, a village at the western edge of the city of San Luis PotosĂ­, S. gracilis has been impacted adversely by the creation of dams. Much additional field work is required to determine the geographic range and frequency of the species. Also needed are life history and population-level studies

    A holistic neuropsychological approach to cognitive remediation for a community-based mixed psychiatric sample

    Background: Improved real world functioning is the ultimate goal of cognitive rehabilitation (which was developed for an acquired brain injury population), however, cognitive remediation for psychiatric populations focuses primarily on cognitive interventions (e.g., computerized cognitive training) and utilizes cognitive test results as outcomes. A broader range of neuropsychological interventions and outcome measures, incorporating real-world measures of functioning, is recommended for cognitive remediation program evaluation. Objective: To determine the feasibility and explore the effectiveness of a holistic cognitive remediation program administered by clinical neuropsychologists for a community-based mixed psychiatric treatment-seeking sample. Method: Twenty-five adults of mixed psychiatric aetiology were referred for a 10-week intervention (including four hours of weekly individual and group-based sessions). A broad array of outcomes was assessed post-intervention. Functional status, self-reported cognitive symptoms and quality of life was assessed at 11.3 months follow-up. Results: Eighteen of the referred participants (72%) completed the intervention. Completers showed: a high rate of functional cognitive goal attainment; increased employment rates; improved symptoms of psychological distress and quality of life; reduced self-report of cognitive difficulties; and improved auditory attention span and verbal memory. Self-report of reduced cognitive difficulties and improved quality of life was maintained approximately one year later. The majority of participants reported very high levels of satisfaction with the program. Conclusions: This intervention was acceptable to participants and associated with high satisfaction rates and gains in cognitive, psychological and functional outcomes. Findings suggest there are multiple benefits to adopting an intervention program that is holistic, individualized to the goals of the patient and facilitated by trained neuropsychologists

    ACMiner: Extraction and Analysis of Authorization Checks in Android's Middleware

    Billions of users rely on the security of the Android platform to protect phones, tablets, and many different types of consumer electronics. While Android's permission model is well studied, the enforcement of the protection policy has received relatively little attention. Much of this enforcement is spread across system services, taking the form of hard-coded checks within their implementations. In this paper, we propose Authorization Check Miner (ACMiner), a framework for evaluating the correctness of Android's access control enforcement through consistency analysis of authorization checks. ACMiner combines program and text analysis techniques to generate a rich set of authorization checks, mines the corresponding protection policy for each service entry point, and uses association rule mining at a service granularity to identify inconsistencies that may correspond to vulnerabilities. We used ACMiner to study the AOSP version of Android 7.1.1 to identify 28 vulnerabilities relating to missing authorization checks. In doing so, we demonstrate ACMiner's ability to help domain experts process thousands of authorization checks scattered across millions of lines of code

    Pathogen quantitation in complex matrices: a multi-operator comparison of DNA extraction methods with a novel assessment of PCR inhibition

Available from the publisher via the DOI in this record.BACKGROUND: Mycobacterium bovis is the aetiological agent of bovine tuberculosis (bTB), an important recrudescent zoonosis, significantly increasing in British herds in recent years. Wildlife reservoirs have been identified for this disease but the mode of transmission to cattle remains unclear. There is evidence that viable M. bovis cells can survive in soil and faeces for over a year. METHODOLOGY/PRINCIPAL FINDINGS: We report a multi-operator blinded trial for a rigorous comparison of five DNA extraction methods from a variety of soil and faecal samples to assess recovery of M. bovis via real-time PCR detection. The methods included four commercial kits: the QIAamp Stool Mini kit with a pre-treatment step, the FastDNA® Spin kit, the UltraClean™ and PowerSoil™ soil kits and a published manual method based on phenol:chloroform purification, termed Griffiths. M. bovis BCG Pasteur spiked samples were extracted by four operators and evaluated using a specific real-time PCR assay. A novel inhibition control assay was used alongside spectrophotometric ratios to monitor the level of inhibitory compounds affecting PCR, DNA yield, and purity. There were statistically significant differences in M. bovis detection between methods of extraction and types of environmental samples; no significant differences were observed between operators. Processing times and costs were also evaluated. To improve M. bovis detection further, the two best performing methods, FastDNA® Spin kit and Griffiths, were optimised and the ABI TaqMan environmental PCR Master mix was adopted, leading to improved sensitivities. CONCLUSIONS: M. bovis was successfully detected in all environmental samples; DNA extraction using FastDNA® Spin kit was the most sensitive method with highest recoveries from all soil types tested. For troublesome faecal samples, we have used and recommend an improved assay based on a reduced volume, resulting in detection limits of 4.25×10(5) cells g(-1) using Griffiths and 4.25×10(6) cells g(-1) using FastDNA® Spin kit.This work was supported by Biotechnology and Biological Sciences Research Council BBSRCBB/E020925/1 grant to OC and EMHW, and Department for Environment, Food and Rural Affairs, DEFRA SE3231 grant to EMHW and OC.     The variability and seasonality of the environmental reservoir of Mycobacterium bovis shed by wild European badgers

Available from the publisher via the DOI in this record.The incidence of Mycobacterium bovis, the causative agent of bovine tuberculosis, has been increasing in UK cattle herds resulting in substantial economic losses. The European badger (Meles meles) is implicated as a wildlife reservoir of infection. One likely route of transmission to cattle is through exposure to infected badger urine and faeces. The relative importance of the environment in transmission remains unknown, in part due to the lack of information on the distribution and magnitude of environmental reservoirs. Here we identify potential infection hotspots in the badger population and quantify the heterogeneity in bacterial load; with infected badgers shedding between 1 × 10(3)- 4 × 10(5) M. bovis cells g(-1) of faeces, creating a substantial and seasonally variable environmental reservoir.     How journal rankings can suppress interdisciplinary research. A comparison between Innovation Studies and Business & Management

    This study provides quantitative evidence on how the use of journal rankings can disadvantage interdisciplinary research in research evaluations. Using publication and citation data, it compares the degree of interdisciplinarity and the research performance of a number of Innovation Studies units with that of leading Business & Management schools in the UK. On the basis of various mappings and metrics, this study shows that: (i) Innovation Studies units are consistently more interdisciplinary in their research than Business & Management schools; (ii) the top journals in the Association of Business Schools' rankings span a less diverse set of disciplines than lower-ranked journals; (iii) this results in a more favourable assessment of the performance of Business & Management schools, which are more disciplinary-focused. This citation-based analysis challenges the journal ranking-based assessment. In short, the investigation illustrates how ostensibly 'excellence-based' journal rankings exhibit a systematic bias in favour of mono-disciplinary research. The paper concludes with a discussion of implications of these phenomena, in particular how the bias is likely to affect negatively the evaluation and associated financial resourcing of interdisciplinary research organisations, and may result in researchers becoming more compliant with disciplinary authority over time.Comment: 41 pages, 10 figure

    Performance of a non-invasive test for detecting mycobacterium bovis shedding in European badger (meles meles) populations

    The incidence of Mycobacterium bovis, the causative agent of bovine tuberculosis, in cattle herds in the United Kingdom is increasing, resulting in substantial economic losses. The European badger (Meles meles) is implicated as a wildlife reservoir and is the subject of control measures aimed at reducing incidence in cattle populations. Understanding the epidemiology of M. bovis in badger populations is essential to direct control interventions and understand disease spread; however, accurate diagnosis in live animals is challenging and currently uses invasive methods. Here we present a non-invasive diagnostic procedure and sampling regime using field sampling of latrines and detection of M. bovis with qPCR, the results of which strongly correlate with the results of immunoassay testing in the field at the social group level. This method allows M. bovis infection in badger populations to be monitored without trapping and provides additional information on the quantity of bacterial DNA shed. Our approach may therefore provide valuable insights into the epidemiology of bovine tuberculosis in badger populations and inform disease control interventions
