    Salinity dependence of the distribution of multicellular magnetotactic prokaryotes in a hypersaline lagoon

    Candidatus Magnetoglobus multicellularis is an unusual magnetotactic multicellular microorganism composed of a highly organized assemblage of gram-negative bacterial cells. In this work, the salinity dependence of Ca. M. multicellularis and its abundance in the hypersaline Araruama Lagoon, Brazil were studied. Viability experiments showed that Ca. M. multicellularis died in salinities >55‰ and < 40‰. Low salinities were also observed to modify the cellular assemblage. In microcosms prepared with different salinities, the microorganism grew better at intermediate salinities whereas in high or low salinities, the size of the population did not increase over time. The concentrations of Ca. M. multicellularis in the lagoon were related to salinity; sites with lower and higher salinities than the lagoon average contained less Ca. M. multicellularis. These results demonstrate the influence of salinity on the survival and distribution of Ca. M. multicellularis in the environment. In sediments, the abundance of Ca. M. multicellularis ranged from 0 to 103 microorganisms/ml, which represented 0.001% of the counts of total bacteria. The ability of Ca. M. multicellularis to accumulate iron and sulfur in high numbers of magnetosomes (up to 905 per microorganism) suggests that its impact on the sequestration of these elements (0.1% for biogenic bacterial iron) is not proportional to its abundance in the lagoon.     Effects of carbonated water injection on the pore system of a carbonate rock (coquina)

    CO2 injection is a well-known Enhanced Oil Recovery (EOR) technique that has been used for years to improve oil extraction from carbonate rock and other oil reservoirs. Optimal functioning of CO2 injection requires a thorough understanding of how this method affects the petrophysical properties of the rocks. We evaluated pore-scale changes in these properties, notably porosity and absolute permeability, following injection of CO2-saturated water in two coquina outcrop samples from the Morro do Chaves Formation in Brazil. The coquinas are close analogues of Pre-salt oil reservoirs off the coast of southern Brazil. The effects of carbonated water injection were evaluated using a series of experimental and numerical steps before and after coreflooding: cleaning, basic petrophysics, microtomography (microCT) imaging, nuclear magnetic resonance (NMR) analyses, and pore network modeling (PNM). Our study was motivated by an earlier experiment which did not show the development of a wormhole in the center of the sample, with a concomitant increase in permeability of the coquina as often noted in the literature. We instead observed a substantial decrease in the absolute permeability (between 71 and 77%), but with little effect on the porosity and no wormhole formation. While all tests were carried out on both samples, here we present a comprehensive analysis for one of the samples to illustrate changes at the pore network level. Different techniques were used for the pore-scale analyses, including pore network modeling using PoreStudio, and software developed by the authors to enable a statistical analysis of the pore network.     Post-Weaning Protein Malnutrition Increases Blood Pressure and Induces Endothelial Dysfunctions in Rats

    Malnutrition during critical periods in early life may increase the subsequent risk of hypertension and metabolic diseases in adulthood, but the underlying mechanisms are still unclear. We aimed to evaluate the effects of post-weaning protein malnutrition on blood pressure and vascular reactivity in aortic rings (conductance artery) and isolated-perfused tail arteries (resistance artery) from control (fed with Labina®) and post-weaning protein malnutrition rats (offspring that received a diet with low protein content for three months). Systolic and diastolic blood pressure and heart rate increased in the post-weaning protein malnutrition rats. In the aortic rings, reactivity to phenylephrine (10−10–3.10−4 M) was similar in both groups. Endothelium removal or L-NAME (10−4 M) incubation increased the response to phenylephrine, but the L-NAME effect was greater in the aortic rings from the post-weaning protein malnutrition rats. The protein expression of the endothelial nitric oxide isoform increased in the aortic rings from the post-weaning protein malnutrition rats. Incubation with apocynin (0.3 mM) reduced the response to phenylephrine in both groups, but this effect was higher in the post-weaning protein malnutrition rats, suggesting an increase of superoxide anion release. In the tail artery of the post-weaning protein malnutrition rats, the vascular reactivity to phenylephrine (0.001–300 µg) and the relaxation to acetylcholine (10−10–10−3 M) were increased. Post-weaning protein malnutrition increases blood pressure and induces vascular dysfunction. Although the vascular reactivity in the aortic rings did not change, an increase in superoxide anion and nitric oxide was observed in the post-weaning protein malnutrition rats. However, in the resistance arteries, the increased vascular reactivity may be a potential mechanism underlying the increased blood pressure observed in this model

    Indução da atividade fagocitária e produção de óxido nítrico numa população natural de Trypanosoma cruzi I e II do Estado do Paraná, Brasil

    Twelve strains of Trypanosoma cruzi isolated from wild reservoirs, triatomines, and chronic chagasic patients in the state of Paraná, southern Brazil, and classified as T. cruzi I and II, were used to test the correlation between genetic and biological diversity. The Phagocytic Index (PI) and nitric-oxide (NO) production in vitro were used as biological parameters. The PI of the T. cruzi I and II strains did not differ significantly, nor did the PI of the T. cruzi strains isolated from humans, triatomines, or wild reservoirs. There was a statistical difference in the inhibition of NO production between T. cruzi I and II and between parasites isolated from humans and the strains isolated from triatomines and wild reservoirs, but there was no correlation between genetics and biology when the strains were analyzed independently of the lineages or hosts from which the strains were isolated. There were significant correlations for Randomly Amplified Polymorphic Deoxyribonucleic acid (RAPD) and biological parameters for T. cruzi I and II, and for humans or wild reservoirs when the lineages or hosts were considered individually.Doze cepas de Trypanosoma cruzi isoladas de reservatórios silvestres, triatomíneos e de pacientes chagásicos crônicos do Estado do Paraná, Brasil, classificadas como Tc I e II foram usadas para avaliar a correlação entre genética e diversidade biológica. Há diferença estatística na inibição da produção de ON entre T. cruzi I e II e entre parasitos isolados de humanos e de cepas isoladas de triatomíneos e reservatórios silvestres, mas não foi observada correlação entre genética e biologia quando as cepas foram analisadas independentemente da linhagem ou hospedeiros das quais elas foram isoladas.     The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & Nemésio 2007; Donegan 2008, 2009; Nemésio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016

    Resumo: Os quelônios são importantes como fonte alimentar e econômica para as comunidades da região amazônica. Sendo assim, a caça, a pesca e a procura por ovos destes animais tem ocorrido frequentemente. Podocnemis expansa (Tartaruga da Amazônia) é o maior quelônio de água doce da América do Sul. É uma espécie largamente distribuída, porém, nota-se que aspectos anatômicos da espécie são insuficientemente descritos. O objetivo desse estudo foi descrever a cavidade orofaríngea desses quelônios. Foram utilizadas 12 cabeças de P. expansa adultas, de ambos os sexos, com idade entre 3 a 8 anos, com peso corpóreo de 5 a 22kg (média de 7,5kg). P. expansa conta com a presença de rafontecas afiadas e bem desenvolvidas que, associadas à musculatura potente da mandíbula e à língua volumosa e bem distribuída no assoalho da orofaringe, atuam na apreensão e deglutição do alimento, garantindo uma maior adaptação em diversos ambientes
