    Can variability in the effect of opioids on refractory breathlessness be explained by genetic factors?

Objectives: Opioids modulate the perception of breathlessness with a considerable variation in response, with poor correlation between the required opioid dose and symptom severity. The objective of this hypothesis-generating, secondary analysis was to identify candidate single nucleotide polymorphisms (SNP) from those associated with opioid receptors, signalling or pain modulation to identify any related to intensity of breathlessness while on opioids. This can help to inform prospective studies and potentially lead to better tailoring of opioid therapy for refractory breathlessness. Setting: 17 hospice/palliative care services (tertiary services) in 11 European countries. Participants: 2294 people over 18 years of age on regular opioids for pain related to cancer or its treatment. Primary outcome measures: The relationship between morphine dose, breathlessness intensity (European Organisation for Research and Treatment of Cancer Core Quality of Life Questionnaire; EORTCQLQC30 question 8) and 112 candidate SNPs from 25 genes (n=588). Secondary outcome measures: The same measures for people on oxycodone (n=402) or fentanyl (n=429). Results: SNPs not in Hardy-Weinberg equilibrium or with allele frequencies ( < 5%) were removed. Univariate associations between each SNP and breathlessness intensity were determined with Benjamini-Hochberg false discovery rate set at 20%. Multivariable ordinal logistic regression, clustering over country and adjusting for available confounders, was conducted with remaining SNPs. For univariate morphine associations, 1 variant on the 5-hydroxytryptamine type 3B (HTR3B) gene, and 4 on the ÎČ-2-arrestin gene (ARRB2) were associated with more intense breathlessness. 1 SNP remained significant in the multivariable model: people with rs7103572 SNP (HTR3B gene; present in 8.4% of the population) were three times more likely to have more intense breathlessness (OR 2.86; 95% CIs 1.46 to 5.62; p=0.002). No associations were seen with fentanyl nor with oxycodone. Conclusions: This large, exploratory study identified 1 biologically plausible SNP that warrants further study in the response of breathlessness to morphine therapy

    This study is an investigation of tertiary students' learning approaches and outcomes, and the relationship between these aspects of learning. The research context was a topic in a first-year biology unit, taught in both on-campus (internal) and distance (external) modes at a regional Australian university. The study followed three research themes. The first was to identify the approaches to learning adopted by students. The second was to identify qualitative variation in learning outcomes, using and testing recent developments in the SOLO model. The third theme was to investigate the relationship between learning approaches and outcomes. These three themes included comparisons of internal and external students

    Incentive-based approaches to sustainable fisheries

    The failures of traditional target-species management have led many to propose an ecosystem approach to fisheries to promote sustainability. The ecosystem approach is necessary, especially to account for fishery-ecosystem interactions, but by itself is not sufficient to address two important factors contributing to unsustainable fisheries — inappropriate incentives bearing on fishers, and the ineffective governance that frequently exists in commercial, developed fisheries managed primarily by total harvest limits and input-controls. We contend that much greater emphasis must be placed on fisher motivation when managing fisheries. Using evidence from more than a dozen ‘natural experiments’ in commercial fisheries, we argue that incentive-based approaches that better specify community, individual harvest, or territorial rights and also price ecosystem services — coupled with public research, monitoring and effective oversight — promote sustainable fisheries.incentives, sustainability, rights, fisheries management

    Using examples from more than a dozen fisheries, we highlight the failures of ‘command control’ management and show that approaches that empower fishers with the incentives and the mandate to be co-custodians of the marine environment can promote sustainability. Evidence is provided that where harvesters share well-defined management responsibilities over fish, and experience both the pain of overexploitation and the gains from conservation, they are much more likely to protect fish stocks and habitat. The key insight is that to maintain marine ecosystems for present and future generations, fishing incentives must be compatible with long-term goals of sustainability.incentives, sustainability, rights, fisheries management

    Regional Structural Orientation of the Mount Sharp Group Revealed by In Situ Dip Measurements and Stratigraphic Correlations on the Vera Rubin Ridge

    Ground‐based bedding orientation measurements are critical to determine the geologic history and processes of sedimentation in Gale crater, Mars. We constrain the dip of lacustrine strata of the Blunts Point, Pettegrove Point, and Jura members of the Murray formation using a combination of regional stratigraphic correlations and bed attitude measurements from stereo Mastcam images taken by the Mars Science Laboratory Curiosity rover. In situ bed attitude measurements using a principal component analysis‐based regression method reveal a wide range of dips and dip azimuths owing to a combination of high stereo errors, postdepositional deformation of strata (e.g., fracturing, rotation, and impact cratering), and different primary depositional dips. These constrain regional dips to be within several degrees of horizontal on average. Stratigraphic correlations between targets observed in the Glen Torridon trough and at the Pettegrove Point‐Jura member contact of Vera Rubin ridge (VRR) constrain dips to be between 3°SE and 2°NW, consistent with nearly flat strata deposited horizontally on an equipotential surface. The Jura member is determined to be stratigraphically equivalent to the northern portion of the Glen Torridon trough. Rover‐based dip magnitudes are generally significantly shallower than the orientation of VRR member contacts measured from High Resolution Imaging Science Experiment‐based traces, suggesting the sedimentary strata and VRR member contacts may be discordant

    Perceptual Advantage of Animal Facial Attractiveness: Evidence From b-CFS and Binocular Rivalry

Here we examined whether this preference for attractiveness is exclusive to human faces by measuring participants’ sensitivity to the attractiveness of cat and tiger faces. Experiment 1 measured the time taken to break continuous flash suppression (b-CFS), whereas Experiment 2 measured the dominant time in binocular rivalry (BR). The results showed that attractive cat faces were detected more quickly (Experiment 1) and dominated for longer time in visual awareness (Experiment 2). However, no effect of attractiveness was found for tiger faces in Experiment 1, while attractive tiger faces also dominated for longer time in visual awareness in Experiment 2. The results provide first evidence that the preference for attractive animal faces can be shown involuntarily or without apparent conscious control. The findings suggest that human preference for facial attractiveness may contain an aesthetic element rather than being a purely adaptive means for mate choice

    Cardio-metabolic risk factors and cortical thickness in a neurologically healthy male population: results from the psychological, social and biological determinants of ill health (pSoBid) study

    &lt;p&gt;Introduction: Cardio-metabolic risk factors have been associated with poor physical and mental health. Epidemiological studies have shown peripheral risk markers to be associated with poor cognitive functioning in normal healthy population and in disease. The aim of the study was to explore the relationship between cardio-metabolic risk factors and cortical thickness in a neurologically healthy middle aged population-based sample.&lt;/p&gt; &lt;p&gt;Methods: T1-weighted MRI was used to create models of the cortex for calculation of regional cortical thickness in 40 adult males (average age = 50.96 years), selected from the PSOBID study. The relationship between cardio-vascular risk markers and cortical thickness across the whole brain, were examined using the general linear models. The relationship with various covariates of interest was explored.&lt;/p&gt; &lt;p&gt;Results: Lipid fractions with greater triglyceride content (TAG, VLDL and LDL) were associated with greater cortical thickness pertaining to a number of regions in the brain. Greater C reactive protein (CRP) and Intercellular adhesion molecule (ICAM-1) levels were associated with cortical thinning pertaining to perisylvian regions in the left hemisphere. Smoking status and education status were significant covariates in the model.&lt;/p&gt; &lt;p&gt;Conclusions: This exploratory study adds to a small body of existing literature increasingly showing a relationship between cardio-metabolic risk markers and regional cortical thickness involving a number of regions in the brain in a neurologically normal middle aged sample. A focused investigation of factors determining the inter-individual variations in regional cortical thickness in the adult brain could provide further clarity in our understanding of the relationship between cardio-metabolic factors and cortical structures.&lt;/p&gt

    Size resolved characterization of the polysaccharidic and proteinaceous components of sea spray aerosol

    Dissolved organic polymers released by phytoplankton and bacteria abiologically self-assemble in surface ocean waters into nano-to micro-sized gels containing polysaccharides, proteins, lipids and other components. These gels concentrate in the sea surface microlayer (SML), where they can potentially contribute to sea spray aerosol (SSA). Sea spray is a major source of atmospheric aerosol mass over much of the earth’s surface, and knowledge of its properties (including the amount and nature of the organic content), size distributions and fluxes are fundamental for determining its role in atmospheric chemistry and climate. Using a cascade impactor, we collected size-fractionated aerosol particles from ambient air and from freshly generated Sea Sweep SSA in the western North Atlantic Ocean together with biological and chemical characterization of subsurface and SML waters. Spectrophotometric methods were applied to quantify the polysaccharide-containing transparent exopolymer (TEP) and protein-containing Coomassie stainable material (CSM) in these particles and waters. This study demonstrates that both TEP and CSM in surface ocean waters are aerosolized with sea spray with the greatest total TEP associated with particles &lt;180 nm in diameter and &gt;5 000 nm. The higher concentrations of TEP and CSM in particles &gt;5 000 nm most likely reflects collection of microorganism cells and/or fragments. The greater concentration of CSM in larger size particles may also reflect greater stability of proteinaceous gels compared to polysaccharide-rich gels in surface waters and the SML. Both TEP and CSM were measured in the ambient marine air sample with concentrations of 2.1 ± 0.16 ÎŒg Xanthan Gum equivalents (XG eq.) m−3 and 14 ± 1.0 ÎŒg bovine serum albumin equivalents (BSA eq.) m−3. TEP in Sea Sweep SSA averaged 4.7 ± 3.1 ÎŒg XG eq. m−3 and CSM 8.6 ± 7.3 ÎŒg BSA eq. m−3. This work shows the transport of marine biogenic material across the air-sea interface through primary particle emission and the first demonstration of particle size discriminated TEP and CSM characterization of SSA and ambient aerosol under field conditions
