    This thesis focuses on race relations during the 1937 in Louisville. The dominant narrative of the 1937 flood in Louisville is that the city united while facing mutual adversity and rebuilding the city. In this story, the waters of the flood washed away any social or racial distinctions, rendering everyone equal during the crisis. Despite this popular narrative, the reality of race relations during the flood was much more complicated. Louisville’s race relations from the nineteenth century until well into the twentieth century have been described by historian George C. Wright as “polite racism.” This complex and unequal relationship between whites and African Americans persisted throughout the flood.     Reducing Lead Times in a two-process cell using lean and simulation.

    An existing power system production process includes two operations and produces twelve different part types. The first operation fills WIP carts used by the second operation. A combined lean and discrete event simulation study supported by the analysis of order history information stored in a corporate information system is presented. The goal was to identify operations alternatives that could be used to reduce customer lead time from the current 3 to 3 days to 1 to 3 days. The application of lean methods included the examination of the order history data that showed that 80% of parts ship to a single primary customer and 20% to many secondary customers. The lean part of the study further concluded that the number of WIP carts should equal the number of different products, that each WIP cart should be associated with one and only one product and that each WIP cart should be should refilled daily after orders are processed. Using a discrete event simulation model, four order processing sequencing alternatives for improving on-time delivery were evaluated. The percent of orders delivered in 1 day was maximized, with the lowest variance, by sorting all orders for the primary customer first from smallest in size to largest. The orders for the same product from the remaining customers are processed immediately after the order for the same product from the primary customer. The value of the synergistic effect of combining lean tools with simulation supported by order data extracted from the corporate information system is demonstrated

    A Case Study of Laser Wind Sensor Performance Validation by Comparison to an Existing Gage

    A case study concerning validation of wind speed measurements made by a laser wind sensor mounted on a 190 square foot floating platform in Muskegon Lake through comparison with measurements made by pre-existing cup anemometers mounted on a met tower on the shore line is presented. The comparison strategy is to examine the difference in measurements over time using the paired-t statistical method to identify intervals when the measurements were equivalent and to provide explanatory information for the intervals when the measurements were not equivalent. The data was partitioned into three sets: not windy (average wind speed measured by the cup anemometers ≤ 6.7m/s) windy but no enhanced turbulence (average wind speed measured by the cup anemometers \u3e 6.7m/s), and windy with enhanced turbulence associated with storm periods. For the not windy data set, the difference in the average wind speeds was equal in absolute value to the precision of the gages and not statistically significant. Similar results were obtained for the windy with no enhanced turbulence data set and the average difference was not statistically significant (α=0.01). The windy with enhanced turbulence data set showed significant differences between the buoy mounted laser wind sensor and the on-shore mast mounted cup anemometers. The sign of the average difference depended on the direction of the winds. Overall, validation evidence is obtained in the absence of enhanced turbulence.     Surface-enhanced Raman scattering measurement from a lipid bilayer encapsulating a single decahedral nanoparticle mediated by an optical trap

    We present a new technique for the study of model membranes on the length-scale of a single nanosized liposome. Silver decahedral nanoparticles have been encapsulated by a model unilamellar lipid bilayer creating nano-sized lipid vesicles. The metal core has two roles (i) increasing the polarizability of vesicles, enabling a single vesicle to be isolated and confined in an optical trap, and (ii) enhancing Raman scattering from the bilayer, via the high surface-plasmon field at the sharp vertices of the decahedral particles. Combined this has allowed us to measure a Raman fingerprint from a single vesicle of 50 nmdiameter, containing just ∼104 lipid molecules in a bilayer membrane over a surface area of <0.01 µm2, equivalent to a volume of approximately 1 zepto-litre. Raman scattering is a weak and inefficient process and previous studies have required either a substantially larger bilayer area in order to obtain a detectable signal, or the tagging of lipid molecules with a chromophore to provide an indirect probe of the bilayer. Our approach is fully label-free and bio-compatible and, in the future, it will enable much more localized studies of the heterogeneous structure of lipid bilayers and of membrane-bound components than is currently possible

    Severity of cardiovascular disease outcomes among patients with HIV is related to markers of inflammation and coagulation

    Background-In the general population, raised levels of inflammatory markers are stronger predictors of fatal than nonfatal cardiovascular disease (CVD) events. People with HIV have elevated levels of interleukin-6 (IL-6), high-sensitivity C-reactive protein (hsCRP), and D-dimer; HIV-induced activation of inflammatory and coagulation pathways may be responsible for their greater risk of CVD. Whether the enhanced inflammation and coagulation associated with HIV is associated with more fatal CVD events has not been investigated. Methods and Results-Biomarkers were measured at baseline for 9764 patients with HIV and no history of CVD. Of these patients, we focus on the 288 that experienced either a fatal (n=74) or nonfatal (n=214) CVD event over a median of 5 years. Odds ratios (ORs) (fatal versus nonfatal CVD) (95% confidence intervals [CIs]) associated with a doubling of IL-6, D-dimer, hsCRP, and a 1-unit increase in an IL-6 and D-dimer score, measured a median of 2.6 years before the event, were 1.39 (1.07 to 1.79), 1.40 (1.10 to 1.78), 1.09 (0.93 to 1.28), and 1.51 (1.15 to 1.97), respectively. Of the 214 patients with nonfatal CVD, 23 died during follow-up. Hazard ratios (95% CI) for all-cause mortality were 1.72 (1.28 to 2.31), 1.73 (1.27 to 2.36), 1.44 (1.15 to 1.80), and 1.88 (1.39 to 2.55), respectively, for IL-6, D-dimer, hsCRP, and the IL-6 and D-dimer score. Conclusions-Higher IL-6 and D-dimer levels reflecting enhanced inflammation and coagulation associated with HIV are associated with a greater risk of fatal CVD and a greater risk of death after a nonfatal CVD even

    Prevalence and Correlates of At-Risk Drinking Among Older Adults: The Project SHARE Study

    At-risk drinking, excessive or potentially harmful alcohol use in combination with select comorbidities or medication use, affects about 10% of elderly adults and is associated with higher mortality. Yet, our knowledge is incomplete regarding the prevalence of different categories of at-risk drinking and their associations with patient demographics. To examine the prevalence and correlates of different categories of at-risk drinking among older adults. Cross-sectional analysis of survey data. Current drinkers ages 60 and older accessing primary care clinics around Santa Barbara, California (n = 3,308). At-risk drinkers were identified using the Comorbidity Alcohol Risk Evaluation Tool (CARET). At-risk alcohol use was categorized as alcohol use in the setting of 1) high-risk comorbidities or 2) high-risk medication use, and 3) excessive alcohol use alone. Adjusted associations of participant characteristics with at-risk drinking in each of the three at-risk categories and with at-risk drinking of any kind were estimated using logistic regression. Over one-third of our sample (34.7%) was at risk. Among at-risk individuals, 61.9% had alcohol use in the context of high-risk comorbidities, 61.0% had high-risk medication use, and 64.3% had high-risk alcohol behaviors. The adjusted odds of at-risk drinking of any kind were decreased and significant for women (odds ratio, OR = 0.41; 95% confidence interval: 0.35-0.48; p-value &lt; 0.001), adults over age 80 (OR = 0.55; CI: 0.43-0.72; p &lt; 0.001 vs. ages 60-64), Asians (OR = 0.40; CI: 0.20-0.80; p = 0.01 vs. Caucasians) and individuals with higher education levels. Similar associations were observed in all three categories of at-risk drinking. High-risk alcohol use was common among older adults in this large sample of primary care patients, and male Caucasians, those ages 60-64, and those with lower levels of education were most likely to have high-risk alcohol use of any type. Our findings could help physicians identify older patients at increased risk for problems from alcohol consumption

    Distinct endocrine effects of chronic haloperidol or risperidone administration in male rats

    Antipsychotic drugs have been used effectively for the treatment of schizophrenia symptoms, but they are often associated with metabolic side effects such as weight gain and endocrine disruptions. To investigate the possible mechanisms of antipsychotic-induced metabolic effects, we studied the impact of chronic administration of a typical antipsychotic drug (haloperidol) and an atypical antipsychotic (risperidone) to male rats on food intake, body weight, adiposity, and the circulating concentrations of hormones and metabolites that can influence energy homeostasis. Chronic (28 days) haloperidol administration had no effect on food intake, weight gain or adiposity in male rats, whereas risperidone treatment resulted in a transient reduction in food intake and significantly reduced body weight gain compared to vehicle-treated control rats. Whereas neither antipsychotic had any effect on serum lipid profiles, glucose tolerance or the circulating concentrations of hormones controlled by the hypothalamo-pituitary-thyroid (free T4), -adrenal (corticosterone), -somatotropic (IGF-1), or -gonadotropic axes (testosterone), haloperidol increased circulating insulin levels and risperidone increased serum glucagon levels. This finding suggests that haloperidol or risperidone induce distinct metabolic effects. Since metabolic disorders such as obesity and type 2 diabetes mellitus represent serious health issues, understanding antipsychotic-induced endocrine and metabolic effects may ultimately allow better control of these side effects
