Orthogonal Decomposition Methods for Turbulent Heat Transfer Analysis with Application to Gas Turbines
Gas turbine engines are the main propulsion source for world wide aviation and are also used for power generation. Even though they rely mainly on fossil fuel and emit climate active gasses, their importance is not likely to decrease in the future. But more efficient ways of using finite resources and hence reducing emissions have to be found. Thus, the interest to improve engine efficiency is growing. Considering the efficiency of the underlying thermodynamic cycle, an increase can be achieved by raising the turbine inlet temperature or compression ratio. Due to the complex nature of the underlying flow physics, however, the aero-thermal processes are still not fully understood. For this reason, one needs to perform research at high spatial and
temporal resolution, in turn creating the need for effective means of postprocessing the large amounts of data.
This dissertation addresses both sides of the problem - using high-scale, high resolution simulations as well as effective post processing techniques. As an example for the latter, a temporal highly resolved data set from wall pressure measurements of a transonic compressor stage is analyzed using proper orthogonal decomposition. The underlying experiments were performed by collaborators at Technical University Darmstadt. To decompose signals into optimal orthogonal basis functions based on temporal correlations including temperature, a formal mathematical framework is developed. A method to rank the reduced order representations with respect to heat transfer effectiveness is presented. To test both methods, a Reynolds-averaged Navier-Stokes (RANS) simulation and large eddy simulation (LES) are performed on turbulent heat transfer in a square duct with one single row of pin fins. While the LES results show closer agreement to experiments, both simulations unveil flow parts that do not contribute to heat transfer augmentation and can be considered wasteful. From the most effective mode, a wall contour for the same domain is derived and applied. In the wall contoured domain, energy in wasteful modes decreased, heat transfer increased and the temperature fluctuations at the wall decreased.
Another stagnating boundary layer flow is examined in a direct numerical simulation of a first stage stator vane. Elevated levels of free stream turbulence and integral length scale are generated to simulate the features of combustor exit flow. The horseshoe vortex dynamics cause an increase in endwall heat transfer upstream of the vane. The link between energy optimal orthogonal basis functions and flow structures is examined using this data and the reduced order heat transfer analysis shows high energy modes with comparatively low impact on turbulent heat transfer. The analysis further shows that there are multiple horseshoe vortices that oscillate upstream of the blade, vanish, regenerate and can also merge. There is a punctual correlation of intense vortex dynamics and peaks in the orthogonal temperature basis function.
For all data considered, the link between the energy optimal orthogonal basis functions and flow structures is neither guaranteed to exist nor straightforward to
establish. The orthogonal expansion locks onto flow parts with high fluctuating kinetic energy - which might or might not be the ones that are looked for. The heat
transfer ranking eliminates this problem and is valid independently of how certain basis functions are interpreted
Matrix-comparative genomic hybridization from multicenter formalin-fixed paraffin-embedded colorectal cancer tissue blocks
<p>Abstract</p> <p>Background</p> <p>The identification of genomic signatures of colorectal cancer for risk stratification requires the study of large series of cancer patients with an extensive clinical follow-up. Multicentric clinical studies represent an ideal source of well documented archived material for this type of analyses.</p> <p>Methods</p> <p>To verify if this material is technically suitable to perform matrix-CGH, we performed a pilot study using macrodissected 29 formalin-fixed, paraffin-embedded tissue samples collected within the framework of the EORTC-GI/PETACC-2 trial for colorectal cancer. The scientific aim was to identify prognostic genomic signatures differentiating locally restricted (UICC stages II-III) from systemically advanced (UICC stage IV) colorectal tumours.</p> <p>Results</p> <p>The majority of archived tissue samples collected in the different centers was suitable to perform matrix-CGH. 5/7 advanced tumours displayed 13q-gain and 18q-loss. In locally restricted tumours, only 6/12 tumours showed a gain on 13q and 7/12 tumours showed a loss on 18q. Interphase-FISH and high-resolution array-mapping of the gain on 13q confirmed the validity of the array-data and narrowed the chromosomal interval containing potential oncogenes.</p> <p>Conclusion</p> <p>Archival, paraffin-embedded tissue samples collected in multicentric clinical trials are suitable for matrix-CGH analyses and allow the identification of prognostic signatures and aberrations harbouring potential new oncogenes.</p
Combined Patterns of IGHV Repertoire and Cytogenetic/Molecular Alterations in Monoclonal B Lymphocytosis versus Chronic Lymphocytic Leukemia
Background:Chronic lymphocytic leukemia (CLL)-like monoclonal B lymphocytosis (MBL) with (MBLhi) or without (MBLlo) absolute B-lymphocytosis precedes most CLL cases,the specific determinants for malignant progression remaining unknown.Methodology/Principal Findings:For this purpose, simultaneous iFISH and molecular analysis of well-established cytogenetic alterations of chromosomes 11, 12, 13, 14 and 17 together with the pattern of rearrangement of the IGHV genes were performed in CLL-like cells from MBL and CLL cases. Our results based on 78 CLL-like MBL and 117 CLL clones from 166 subjects living in the same geographical area, show the existence of three major groups of clones with distinct but partially overlapping patterns of IGHV gene usage, IGHV mutational status and cytogenetic alterations. These included a group enriched in MBLloclones expressing specific IGHV subgroups (e.g. VH3-23) with no or isolated good-prognosis cytogenetic alterations, a second group which mainly consisted of clinical MBLhiand advanced stage CLL with a skewed but different CLL-associated IGHV gene repertoire (e.g. VH1-69), frequently associated with complex karyotypes and poor-prognosis cytogenetic alterations, and a third group of clones with intermediate features, with prevalence of mutated IGHV genes, and higher numbers of del(13q)+clonal B-cells.Conclusions/Significance:These findings suggest that the specific IGHV repertoire and IGHV mutational status of CLL-like B-cell clones may modulate the type of cytogenetic alterations acquired, their rate of acquisition and/or potentially also their clinical consequences. Further long-term follow-up studies investigating the IGHV gene repertoire of MBLloclones in distinct geographic areas and microenvironments are required to confirm our findings and shed light on the potential role of some antigen-binding BCR specificities contributing to clonal evolution
Genomic imbalances in 5918 malignant epithelial tumors: an explorative meta-analysis of chromosomal CGH data
BACKGROUND: Chromosomal abnormalities have been associated with most human malignancies, with gains and losses on some genomic regions associated with particular entities. METHODS: Of the 15429 cases collected for the Progenetix molecular-cytogenetic database, 5918 malignant epithelial neoplasias analyzed by chromosomal Comparative Genomic Hybridization (CGH) were selected for further evaluation. For the 22 clinico-pathological entities with more than 50 cases, summary profiles for genomic imbalances were generated from case specific data and analyzed. RESULTS: With large variation in overall genomic instability, recurring genomic gains and losses were prominent. Most entities showed frequent gains involving 8q2, while gains on 20q, 1q, 3q, 5p, 7q and 17q were frequent in different entities. Loss "hot spots" included 3p, 4q, 13q, 17p and 18q among others. Related average imbalance patterns were found for clinically distinct entities, e.g. hepatocellular carcinomas (ca.) and ductal breast ca., as well as for histologically related entities (squamous cell ca. of different sites). CONCLUSION: Although considerable case-by-case variation of genomic profiles can be found by CGH in epithelial malignancies, a limited set of variously combined chromosomal imbalances may be typical for carcinogenesis. Focus on the respective regions should aid in target gene detection and pathway deduction
Refinement of 1p36 Alterations Not Involving PRDM16 in Myeloid and Lymphoid Malignancies
Fluorescence in situ hybridization was performed to characterize 81 cases of myeloid and lymphoid malignancies with cytogenetic 1p36 alterations not affecting the PRDM16 locus. In total, three subgroups were identified: balanced translocations (N = 27) and telomeric rearrangements (N = 15), both mainly observed in myeloid disorders; and unbalanced non-telomeric rearrangements (N = 39), mainly observed in lymphoid proliferations and frequently associated with a highly complex karyotype. The 1p36 rearrangement was isolated in 12 cases, mainly myeloid disorders. The breakpoints on 1p36 were more widely distributed than previously reported, but with identifiable rare breakpoint cluster regions, such as the TP73 locus. We also found novel partner loci on 1p36 for the known multi-partner genes HMGA2 and RUNX1. We precised the common terminal 1p36 deletion, which has been suggested to have an adverse prognosis, in B-cell lymphomas [follicular lymphomas and diffuse large B-cell lymphomas with t(14;18)(q32;q21) as well as follicular lymphomas without t(14;18)]. Intrachromosomal telomeric repetitive sequences were detected in at least half the cases of telomeric rearrangements. It is unclear how the latter rearrangements occurred and whether they represent oncogenic events or result from chromosomal instability during oncogenesis
