Partindo de uma exposição da metodologia criada pelo teatrólogo russo, em seus trabalhos publicados, é exposta uma experiência com um grupo de não atores, através de um projeto de pesquisação, cujo resultado é demonstrado em relatos pessoais dos próprios “alunos”, onde são refletidas as trajetórias individuais como uma grande influência no trabalho criador

    Geografias e cinemas : a (des)construção espacial através da imagem cinematográfica

O Cinema é parte desta abordagem à Geografia, como uma expressão artística e cultural, e também pelo seu alcance comercial e industrial, o que faz do cinema um agente de transformação do espaço. Este O objetivo dessa dissertação é analisar o potencial de transformação do espaço na produção cinematográfica tendo em vista, principalmente, sua veiculação e distribuição. Sendo assim é apresentado um estudo da relação entre geografia e cinema tendo como referenciais empíricos os filmes "‘Cidade de Deus’", “Tropa de Elite”, “Tropa de Elite 2 – O Inimigo agora é outro”, e para ampliar a abordagem conceitual há também breves análises dos filmes: ‘Babel’, “Blade Runner – O Caçador de Andróides” e “A Origem” assim como referências a outros filmes brasileiros e internacionais. Esta análise constitui uma pesquisa qualitativa utilizando-se da decupagem fílmica e análise relacional que leva ao cinema as diversas categorias da geografia numa busca pelas características que o definem como agente de transformação espacial através das imagens, sons, da sua contribuição ao imaginário e sua atuação nas políticas de cultura e informação do país, que resulta em um mecanismo cíclico de formação de conceitos e imaginário sobre as esferas de debate elucidadas pela composição fílmica. ______________________________________________________________________________ ABSTRACTGeography and the Arts have common features in their development because in their areas of possibilities are present: society, nature, imagination, and therefore the space. Cinema is part of this approach to Geography as a cultural and artistic expression, and also for its industrial and commercial reach, which makes the film an agent of transformation space. The objective of this dissertation is to analyze the potential transformation of space in film production viewing, especially, its broadcasting and distribution. Thus it is presented a study of the relationship between Geography and cinema with empirical framework based on films: “City of God”, "Elite Squad", "Elite Squad 2 - The Enemy within" and to enlarge the approach there is the conceptual references to movies: “Babel”, “Blade Runner” and “Inception” as well as references to other Brazilian and international films. This analysis is a qualitative research using filmic decoupage and relational analysis which leads to the Cinema the various categories of Geography in a search for the characteristics that will define it as an agent of space transformation through imagery, sounds, its contribution to the imaginary and its performance in political culture and country information, which results in a cyclical wheel mechanism of concepts and imagery on the sphere of subjects elucidated by filmic composition

    Antimicrobial synergism against different lineages of methicillin-resistant Staphylococcus aureus carrying SCCmec IV

    Aim To evaluate the synergistic activity of antimicrobial drugs against lineages of methicillin-resistant Staphylococcus aureus (MRSA) carrying SCCmec IV. The biofilm production and related genes were also detected. Methods and Results Forty two MRSA isolates were tested for biofilm production and related genes. Biofilm/biomass susceptibility to gentamicin (G), linezolid (L), rifampicin (R) and vancomycin (V) was determined for six isolates from three lineages prevalent in Rio de Janeiro hospitals in concentrations ranging from 0·25 to 64 μg ml−1. Biomass was evaluated by microtitre plate test and number of viable cells (CFU cm−2) and inspected by epifluorescence microscopy. All isolates presented the icaA and sasG genes, but only 38% were biofilm producers. There were 50 and 45% biomass reductions when concentrations ≥4 μg ml−1 of R or L and ≥16 μg ml−1 of G or V, respectively, were used. Synergism tests produced a 55% biomass reduction with R2lg ml1 + G16lg ml1 , R2lg ml1 + L2lg ml1 , R2lg ml1 + V4lg ml1 , and L2lg ml1 + V4lg ml1 . Number of viable cells was reduced from 2 to 3 logs with R2lg ml1 + L2lg ml1 and R2lg ml1 + V4lg ml1 . Conclusions Synergisms involving R plus L and R plus V caused important reductions in biofilm/biomass and the number of viable cells. Drug combinations should be considered in the chemotherapies of MRSA-SCCmec IV infections. Significance and Impact of the Study Biofilms in MRSA infections restrict the clinical choice of antimicrobials.     Antimicrobial Action of 1,10-Phenanthroline-Based Compoundson Carbapenemase-Producing Acinetobacter Baumannii Clinical Strains: Efficacy Against Planktonic- and Biofilm-Growing Cells

    Therapeutic options are limited for patients infected with Acinetobacter baumannii due to its multidrug-resistance profile. So, the search for new antimicrobials against this gram-negative bacterial pathogen has become a worldwide priority. The present study aimed to evaluate the effects of 1,10-phenanthroline (phen), 1,10-phenanthroline-5,6-dione (phendione), [Ag(phendione)2]ClO4 (Ag-phendione) and [Cu(phendione)3](ClO4)2.4H2O (Cu-phendione) on 26 carbapenemase-producing A. baumannii strains. The susceptibility to carbapenems was performed by detecting the metallo-beta-lactamase (MBL) genes by PCR and by determining the MIC. Also, disk diffusion method was applied to evaluate the susceptibility to other antimicrobial classes. The test compounds were evaluated on both planktonic- and biofilm-growing bacterial cells. The results revealed that all A. baumannii strains had the intrinsic blaOXA-51 gene, and at least one of the blaOXA-23 or blaOXA-24 genes. The geometric mean MIC and minimum bactericidal concentration (MBC) values, respectively, were as follows: Cu-phendione (1.56 and 2.30 μM), Ag-phendione (2.48 and 3.63 μM), phendione (9.44 and 9.70 μM) and phen (70.46 and 184.28 μM). The test compounds (at 0.5×MIC) affected the biofilm formation and disrupted the mature biofilm, in a typically dose-dependent manner, reducing biomass and viability parameters. Collectively, silver- and copper-phendione derivatives presented potent antimicrobial action against planktonic- and biofilm-forming cells of carbapenemase-producing A. baumannii

    Livestock Origin for a Human Pandemic Clone of Community-Associated Methicillin-Resistant Staphylococcus aureus

    ABSTRACT The importance of livestock as a source of bacterial pathogens with the potential for epidemic spread in human populations is unclear. In recent years, there has been a global increase in community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) infections of healthy humans, but an understanding of the different evolutionary origins of CA-MRSA clones and the basis for their recent expansion is lacking. Here, using a high-resolution phylogenetic approach, we report the discovery of two emergent clones of human epidemic CA-MRSA which resulted from independent livestock-to-human host jumps by the major bovine S. aureus complex, CC97. Of note, one of the new clones was isolated from human infections on four continents, demonstrating its global dissemination since the host jump occurred over 40 years ago. The emergence of both human S. aureus clones coincided with the independent acquisition of mobile genetic elements encoding antimicrobial resistance and human-specific mediators of immune evasion, consistent with an important role for these genetic events in the capacity to survive and transmit among human populations. In conclusion, we provide evidence that livestock represent a reservoir for the emergence of new human-pathogenic S. aureus clones with the capacity for pandemic spread. These findings have major public health implications highlighting the importance of surveillance for early identification of emergent clones and improved transmission control measures at the human-livestock interface. IMPORTANCE Animals are the major source of new pathogens affecting humans. However, the potential for pathogenic bacteria that originally were found in animals to switch hosts and become widely established in human populations is not clear. Here, we report the discovery of emergent clones of methicillin-resistant Staphylococcus aureus (MRSA) that originated in livestock and switched to humans, followed by host-adaptive evolution and epidemic spread in global human populations. Our findings demonstrate that livestock can act as a reservoir for the emergence of new human bacterial clones with potential for pandemic spread, highlighting the potential role of surveillance and biosecurity measures in the agricultural setting for preventing the emergence of new human pathogens

    Epidemiological aspects of american cutaneous leishmaniasis and phlebotomine sandfly population, in the municipality of monte negro, state of Rondônia, Brazil

    Introduction: This work was carried out on the purpose of identifying the species of phlebotomine sandflies in the municipality of Monte Negro, state of Rondonia, Brazil, that may have been transmitting the American cutaneous leishmaniasis (ACL), and concisely describe epidemiological aspects of disease. Methods: The epidemiologic and socioeconomical indicators were obtained from government institutions and the local Municipal Secretary of Health. Phlebotomine sandflies were captured using CDC light traps between July 2006 to July 2008. The total of 1,240 of female sandflies were examined by PCR method directed to k-DNA. Results: There has been a significant decrease in the incidence of ACL of about 50% over the last ten years in the municipality. A total of 1,935 specimens of 53 sandfly species were captured, three of the genus Brumptomyia genus and 50 of the genus Lutzomyia. The predominant species was Lutzomyia acanthopharynx, Lutzomyia whitmani, Lutzomyia geniculata and Lutzomyia davisi. None were positive for Leishmania sp. Conclusions: Four sandflies species were found in the State of Rondonia for the first time: Brumptomyia brumpti, Lutzomyia tarapacaensis, Lutzomyia melloi and Lutzomyia lenti. The presence of Lutzomyia longipalpis, was also captured. Socioeconomical improvement of Brazilian economy and the increase of environmental surveillance in the last 15 years collaborated in the decrease of people exposed to vectors, reducing the incidence of ACL

    Accuracy of phenotypic methicillin susceptibility methods in the detection of Staphylococcus aureus isolates carrying different SCCmec types

    A total of 138 isolates, 118 methicillin-resistant Staphylococcus aureus (MRSA) isolates (staphylococcal cassette chromosome type II, 20 isolates, type III, 39 isolates and type IV, 59 isolates) and 20 methicillin-sensitive S. aureus isolates were evaluated by phenotypic methods: cefoxitin and oxacillin disk diffusion (DD), agar dilution (AD), latex agglutination (LA), oxacillin agar screening (OAS) and chromogenic agar detection. All methods showed 100% specificity, but only the DD tests presented 100% sensitivity. The sensitivity of the other tests ranged from 82.2% (OAS)-98.3% (AD). The LA test showed the second lowest sensitivity (86.4%). The DD test showed high accuracy in the detection of MRSA isolates, but there was low precision in the detection of type IV isolates by the other tests, indicating that the genotypic characteristics of the isolates should be considered

    First report of the blaOXA-58 gene in a clinical isolate of Acinetobacter baumannii in Rio de Janeiro, Brazil

    Carbapenemase production is an important mechanism of carbapenem resistance among nonfermentative Gram-negative isolates. This study aimed to report the detection of blaOXA-58 gene in multiresistant clinical isolates of Acinetobacter baumannii recovered from inpatients in a public hospital. Polymerase chain reaction tests were performed to detect the blaOXA-23-like, blaOXA-24-like, blaOXA-58-like and blaOXA-51-like genes. The blaOXA-58 and blaOXA-23 genes were detected in one and three isolates, respectively. Sequencing of the blaOXA-58-like amplicon revealed 100% identity with the A. baumannii blaOXA-58 gene listed in the GenBank database. This is the first report of an OXA-58-producing A. baumannii isolate in Rio de Janeiro, Brazil
