    Rhipicephalus bursa Sialotranscriptomic Response to Blood Feeding and Babesia ovis Infection: Identification of Candidate Protective Antigens

    Ticks are among the most prevalent blood-feeding arthropods, and they act as vectors and reservoirs for numerous pathogens. Sialotranscriptomic characterizations of tick responses to blood feeding and pathogen infections can offer new insights into the molecular interplay occurring at the tick-host-pathogen interface. In the present study, we aimed to identify and characterize Rhipicephalus bursa salivary gland (SG) genes that were differentially expressed in response to blood feeding and Babesia ovis infection. Our experimental approach consisted of RNA sequencing of SG from three different tick samples, fed-infected, fed-uninfected, and unfed-uninfected, for characterization and inter-comparison. Overall, 7,272 expressed sequence tags (ESTs) were constructed from unfed-uninfected, 13,819 ESTs from fed-uninfected, and 15,292 ESTs from fed-infected ticks. Two catalogs of transcripts that were differentially expressed in response to blood feeding and B. ovis infection were produced. Four genes coding for a putative vitellogenin-3, lachesin, a glycine rich protein, and a secreted cement protein were selected for RNA interference functional studies. A reduction of 92, 65, and 51% was observed in vitellogenin-3, secreted cement, and lachesin mRNA levels in SG, respectively. The vitellogenin-3 knockdown led to increased tick mortality, with 77% of ticks dying post-infestation. The reduction of the secreted cement protein-mRNA levels resulted in 46% of ticks being incapable of correctly attaching to the host and significantly lower female weights post-feeding in comparison to the control group. The lachesin knockdown resulted in a 70% reduction of the levels associated with B. ovis infection in R. bursa SG and 70% mortality. These results improved our understanding of the role of tick SG genes in Babesia infection/proliferation and tick feeding. Moreover, lachesin, vitellogenin-3, and secreted cement proteins were validated as candidate protective antigens for the development of novel tick and tick-borne disease control measures.FCT for funds to GHTM UID/Multi/04413/2013.info:eu-repo/semantics/publishedVersio

    Controlling quantum entanglement through photocounts

    We present a protocol to generate and control quantum entanglement between the states of two subsystems (the system S{\cal S}) by making measurements on a third subsystem (the monitor M{\cal M}), interacting with S{\cal S}. For the sake of comparison we consider first an ideal, or instantaneous projective measurement, as postulated by von Neumann. Then we compare it with the more realistic or generalized measurement procedure based on photocounting on M{\cal M}. Further we consider that the interaction term (between S{\cal S} and M{\cal M}) contains a quantum nondemolition variable of S{\cal S} and discuss the possibility and limitations for reconstructing the initial state of S{\cal S} from information acquired by photocounting on M{\cal M}.Comment: 12 pages, 3 figures, accepted for publication in Phys.     Recent advances in neutrinoless double beta decay search

    Even after the discovery of neutrino flavour oscillations, based on data from atmospheric, solar, reactor, and accelerator experiments, many characteristics of the neutrino remain unknown. Only the neutrino square-mass differences and the mixing angle values have been estimated, while the value of each mass eigenstate still hasn't. Its nature (massive Majorana or Dirac particle) is still escaping. Neutrinoless double beta decay (0ν0\nu-DBD) experimental discovery could be the ultimate answer to some delicate questions of elementary particle and nuclear physics. The Majorana description of neutrinos allows the 0ν0\nu-DBD process, and consequently either a mass value could be measured or the existence of physics beyond the standard should be confirmed without any doubt. As expected, the 0ν0\nu-DBD measurement is a very difficult field of application for experimentalists.     Limits on different Majoron decay modes of 100^{100}Mo and 82^{82}Se for neutrinoless double beta decays in the NEMO-3 experiment

    The NEMO-3 tracking detector is located in the Fr\'ejus Underground Laboratory. It was designed to study double beta decay in a number of different isotopes. Presented here are the experimental half-life limits on the double beta decay process for the isotopes 100^{100}Mo and 82^{82}Se for different Majoron emission modes and limits on the effective neutrino-Majoron coupling constants. In particular, new limits on "ordinary" Majoron (spectral index 1) decay of 100^{100}Mo (T1/2>2.71022T_{1/2} > 2.7\cdot10^{22} y) and 82^{82}Se (T1/2>1.51022T_{1/2} > 1.5\cdot10^{22} y) have been obtained. Corresponding bounds on the Majoron-neutrino coupling constant are <(0.41.9)104 < (0.4-1.9) \cdot 10^{-4} and <(0.661.7)104< (0.66-1.7) \cdot 10^{-4}.Comment: 23 pages includind 4 figures, to be published in Nuclear Physics

    Effects of supplementation of tropical fruit processing by-products on lipid profile, retinol levels and intestinal function in Wistar rats

    Abstract Fruits agro-industrial by-products may have a great variety of bioactive compounds that promote health. Thus, the effects of supplementation with acerola, cashew and guava processing by-products for 28 days on retinol level, lipid profile and on some aspects related to intestinal function in rats were investigated. The animals supplemented with different fruit by-products presented similar weight gain, faecal pH values and intestinal epithelial structures; however, they showed higher moisture and Lactobacillus spp. and Bifidobacterium spp. counts in faeces compared to the control group. Supplementation with the cashew by-product decreased the blood glucose, acerola and guava by-products reduced serum lipid levels and all fruit by-products tested increased serum and hepatic retinol. The results indicated that acerola and guava by-products possess a potential hypolipidemic effect. The three fruit by-products increase the hepatic retinol deposition and the faecal populations of beneficial bacterial groups and modulated aspects of intestinal function. The findings of this study can contribute to sustainable fruticulture and support future clinical studies with the supplementation of by-products
