    Supervision is an act of direct observations and periodically by the supervisor of the work performed by subordinates and then if there are any problems, immediately given guidance or assistance that is directly in order to cope. Satisfaction is a person's emotional response to the work he does. Job satisfaction is also inseparable from the role of the various parties, and one of them is the role of supervision. Aim for the head room supervision relationship is known to nurse satisfaction in hospitals implementing Liunkendage Tahuna. The study design is observational analytic cross-sectional approach. The population of the entire nurse practitioner working in eight wards in hospitals Liunkendage Tahuna. Sampling using purposive sampling. Data processed through the analysis of univariate and bivariate Chi Square. The results obtained by the analysis of the probability (p) = 0.001 <α (0.05). Conclusion, the relationship supervision chief nurse executive room with satisfaction. Advice, For the hospital in order to make improvements to the quality of head room to be included in the training and special education for the head of the room

    Sensitivity and specificity of detection methods for erythropoietin doping in cyclists

    Recombinant human erythropoietin (rHuEPO) is used as doping a substance. Anti-doping efforts include urine and blood testing and monitoring the athlete biological passport (ABP). As data on the performance of these methods are incomplete, this study aimed to evaluate the performance of two common urine assays and the ABP. In a randomized, double-blinded, placebo-controlled trial, 48 trained cyclists received a mean dose of 6000 IU rHuEPO (epoetin beta) or placebo by weekly injection for eight weeks. Seven timed urine and blood samples were collected per subject. Urine samples were analyzed by sarcosyl-PAGE and isoelectric focusing methods in the accredited DoCoLab in Ghent. A selection of samples, including any with false presumptive findings, underwent a second sarcosyl-PAGE confirmation analysis. Hematological parameters were used to construct a module similar to the ABP and analyzed by two evaluators from an Athlete Passport Management Unit. Sensitivity of the sarcosyl-PAGE and isoelectric focusing assays for the detection of erythropoietin abuse were 63.8% and 58.6%, respectively, with a false presumptive finding rate of 4.3% and 6%. None of the false presumptive findings tested positive in the confirmation analysis. Sensitivity was highest between 2 and 6 days after dosing, and dropped rapidly outside this window. Sensitivity of the ABP was 91.3%. Specificity of the urine assays was high; however, the detection window of rHuEPO was narrow, leading to questionable sensitivity. The ABP, integrating longitudinal data, is more sensitive, but there are still subjects that evade detection. Combining these methods might improve performance, but will not resolve all observed shortcomings

    The Saccharomyces cerevisiae CWH8 gene is required for full levels of dolichol-linked oligosaccharides in the endoplasmic reticulum and for efficient N-glycosylation

    The Saccharomyces cerevisiae mutant cwh8 was previously found to have an anomalous cell wall. Here we show that the cwh8 mutant has an N-glycosylation defect. We found that cwh8 cells were resistant to vanadate and sensitive to hygromycin B, and produced glycoforms of invertase and carboxypeptidase Y with a reduced number of N-chains. We have cloned the CWH8 gene. We found that it was nonessential and encoded a putative transmembrane protein of 239 amino acids. Comparison of the in vitro oligosaccharyl transferase activities of membrane preparations from wild type or cwh8Δ cells revealed no differences in enzyme kinetic properties indicating that the oligosaccharyl transferase complex of mutant cells was not affected. cwh8Δ cells also produced normal dolichols and dolichol-linked oligosaccharide intermediates including the full-length form Glc3Man9GlcNAc2. The level of dolichol-linked oligosaccharides in cwh8Δ cells was, however, reduced to about 20% of the wild type. We propose that inefficient N-glycosylation of secretory proteins in cwh8Δ cells is caused by an insufficient supply of dolichol-linked oligosaccharide substrat

    Citrus limonoids (CLs) are a group of highly oxygenated terpenoid secondary metabolites found mostly in the seeds, fruits and peel tissues of citrus fruits such as lemons, limes, oranges, pumellos, grapefruits, bergamots, and mandarins. Represented by limonin, the aglycones and glycosides of CLs have shown to display numerous pharmacological activities including anticancer, antimicrobial, antioxidant, antidiabetic and insecticidal among others. In this review, the chemistry and pharmacology of CLs are systematically scrutinised through the use of medicinal chemistry tools and structure-activity relationship approach. Synthetic derivatives and other structurally-related limonoids from other sources are include in the analysis.     Poverty is one of the most pressing global problems of the world today. One promising option to provide employment opportunities for the inhabitants of underdeveloped rural areas is the development of ‘pro-poor tourism’. Poverty alleviation tourism can generate environmental benefits in addition to economic, social and cultural ones. This study aims to discuss the possibilities of poverty alleviation of rural communities through the development of community-based ecotourism. The introductory part of the study briefly defines the theoretical frameworks of the concept of poverty and ecotourism in the context of sustainable development. Using the specific example of the Mid-Himalayan Rural village of Pastanga (India), show the potential areas for developing community-based ecotourism with respect to the local development opportunities. In the same way that is draw on our own research as well as available statistical data from the East Sikkim and Questionnaire Survey-2021. The case study presents the current extent of adventure trekking tourism and highlights the trends, characteristics and significance of trekking in Pastanga-Khedi eco-trail. The paper also propose strategies for exploring the trekking potential through the emphasis on ecotourism in the study area. In conclusion, we propose solutions for the regulated development of the area in a participatory way by the concerned stakeholders by implementing activities related to the Community-Based Ecotourism

    Rational Design of Mechanism-Based Inhibitors and Activity-Based Probes for the Identification of Retaining α-l-Arabinofuranosidases

    Identifying and characterizing the enzymes responsible for an observed activity within a complex eukaryotic catabolic system remains one of the most significant challenges in the study of biomass-degrading systems. The debranching of both complex hemicellulosic and pectinaceous polysaccharides requires the production of α-l-arabinofuranosidases among a wide variety of coexpressed carbohydrate-active enzymes. To selectively detect and identify α-l-arabinofuranosidases produced by fungi grown on complex biomass, potential covalent inhibitors and probes which mimic α-l-arabinofuranosides were sought. The conformational free energy landscapes of free α-l-arabinofuranose and several rationally designed covalent α-l-arabinofuranosidase inhibitors were analyzed. A synthetic route to these inhibitors was subsequently developed based on a key Wittig-Still rearrangement. Through a combination of kinetic measurements, intact mass spectrometry, and structural experiments, the designed inhibitors were shown to efficiently label the catalytic nucleophiles of retaining GH51 and GH54 α-l-arabinofuranosidases. Activity-based probes elaborated from an inhibitor with an aziridine warhead were applied to the identification and characterization of α-l-arabinofuranosidases within the secretome of A. niger grown on arabinan. This method was extended to the detection and identification of α-l-arabinofuranosidases produced by eight biomass-degrading basidiomycete fungi grown on complex biomass. The broad applicability of the cyclophellitol-derived activity-based probes and inhibitors presented here make them a valuable new tool in the characterization of complex eukaryotic carbohydrate-degrading systems and in the high-throughput discovery of α-l-arabinofuranosidases

    Maternity management in SMEs: a transdisciplinary review and research agenda

    This paper provides a transdisciplinary critical review of the literature on maternity management in small- and medium-sized enterprises (SMEs), embedded within the wider literatures on maternity in the workplace. The key objectives are to describe what is known about the relations that shape maternity management in smaller workplaces and to identify research directions to enhance this knowledge. The review is guided by theory of organizational gendering and small business management, conceptualising adaptions to maternity as a process of mutual adjustment and dynamic capability within smaller firms’ informally negotiated order, resource endowments and wider labour and product/service markets. A context sensitive lens is also applied. The review highlights the complex range of processes involved in SME maternity management and identifies major research gaps in relation to pregnancy, maternity leave and the return to work (family-friendly working and breastfeeding) in these contexts. This blind spot is surprising as SMEs employ the majority of women worldwide. A detailed agenda for future research is outlined, building on the gaps identified by the review and founded on renewed theoretical direction
