Mitochondria and endoplasmic reticulum interplay at the core of efavirenz-induced hepatic effects
El inhibidor de la transcriptasa inversa no análogo de nucleósido efavirenz (EFV) es uno de los fármacos más utilizados en la terapia antirretroviral combinada (TARc) empleada en el tratamiento de la infección por el virus de la inmunodeficiencia humana (VIH). Aunque en general se ha considerado seguro, existe una gran preocupación acerca de los efectos secundarios inducidos por las terapias que contienen EFV. Este fármaco se ha asociado con toxicidad hepática y trastornos metabólicos y, aunque los mecanismos implicados no están claros, evidencias recientes han señalado una acción mitocondrial específica de EFV acompañada por la inducción de estrés de retículo endoplasmático (RE)/respuesta a proteínas desplegadas en células hepáticas humanas cultivadas.
En el presente trabajo, con el objetivo de entender el papel de la mitocondria en los efectos inducidos por EFV, en primer lugar, hemos evaluado las acciones celulares de este fármaco en un modelo de células hepáticas que carecen de mitocondrias funcionales (células rho⁰ generadas en el fondo Hep3B). Además, hemos estudiado la dinámica mitocondrial, en células Hep3B wild-type, que depende entre otros procesos de la interacción entre las mitocondria y el RE. Esta interacción también se ha analizado mediante la evaluación de las membranas asociadas a la mitocondria (MAMs). Por último, con el fin de vincular aún más los dos efectos de EFV (mitocondria y RE), hemos analizado la expresión de LONP1, una proteasa mitocondrial altamente conservada cuya activación es un mecanismo adaptativo tanto en el estrés oxidativo como en el de RE. A lo largo del manuscrito, los efectos de EFV se han comparado con los inducidos por un inductor farmacológico clásico del estrés de RE, tapsigargina (TG), el agente mitotóxico típico rotenona (Rot) - un inhibidor estándar del complejo I - y el desacoplador de la OxPhos, CCCP.
Por un lado, las células rho⁰ tratadas con EFV mostraron una reducción importante en los parámetros indicativos de interferencia mitocondrial, tales como aumento de la producción de superóxido, alteraciones en la morfología y masa mitocondrial, y aumento de la expresión de LONP1. En consonancia con estos resultados, el efecto citotóxico (número de células, condensación de la cromatina, alteraciones del ciclo celular e inducción de la apoptosis) de EFV fue menos pronunciado en las células Hep3B deficientes de respiración que en las células wild-type. El efecto de EFV fue tanto similar como diferente de los efectos producidos por dos factores distintos de estrés mitocondrial, TG y Rot, dependiendo del parámetro estudiado.
Por otro lado, los marcadores de la dinámica mitocondrial se expresaron de forma diferente con los estímulos utilizados (EFV, TG, Rot y CCCP), lo que apunta a una especificidad del doble estrés mitocondrial/RE inducido por EFV. El tratamiento con EFV potenció la interacción entre la mitocondria y el RE, como se muestra por los experimentos de co-inmunoprecipitación de los complejos proteicos de las MAMs. Además, LONP1 fue regulado positivamente tanto a nivel de ARNm como de proteína en todas las condiciones. Sorprendentemente, tras el tratamiento con EFV, su presencia extramitocondrial (RE y MAMs) aumentó.
En conclusión, las células hepáticas que carecen de mitocondrias normales (rho⁰) son menos vulnerables a EFV. Este hallazgo puede explicar las reacciones hepáticas idiosincrásicas desencadenadas por los fármacos anti-VIH y también los diferentes grados de susceptibilidad al daño hepático observados en pacientes sometidos a terapia antirretroviral. El doble efecto mitochondria-RE específico inducido por EFV aumenta el contenido de MAMs y esto está asociado con el aumento de la expresión extramitochondrial de LONP1. Este es el primer informe de este fenómeno en células de mamífero y sugiere una función novedosa de LONP1 relacionada con las MAMs.The non-nucleoside analogue reverse transcriptase inhibitor efavirenz (EFV) is among the most widely used drugs in the combined antiretroviral therapy (cART) employed in the treatment of human immunodeficiency virus (HIV) infection. Although generally considered safe, there is a concern about the side effects induced by EFV-containing therapies. It has been associated with hepatic toxicity and metabolic disturbances and, although the mechanisms involved are not clear, recent evidence has pinpointed a specific mitochondrial action of EFV accompanied by the induction of an endoplasmic reticulum (ER) stress/unfolded protein response in cultured human hepatic cells.
In the present work, in order to understand the role of mitochondria in the effects of EFV, firstly, we have assessed the cellular actions of this drug in a model of hepatic cells that lack functional mitochondria (rho⁰ cells generated in Hep3B background). In addition, we have studied mitochondrial dynamics, in wild-type Hep3B cells, which depends among other processes on the interaction between mitochondria and ER. This interaction has also been analysed by evaluation of mitochondrial-associated membranes (MAMs). Finally, in order to further link the two effects of EFV (mitochondria and ER), we have analysed the expression of LONP1, a highly conserved mitochondrial protease whose activation is an adaptive mechanism in both oxidative and ER stress. Throughout the manuscript, the effects of EFV have been compared to those induced by the classic pharmacological inducer of ER stress thapsigargin (TG), the typical mitotoxic agent rotenone (Rot) - a standard complex I inhibitor - and the uncoupler of OxPhos CCCP.
On the one hand, EFV-treated rho⁰ cells exhibited a substantial reduction in parameters indicative of mitochondrial interference, such as increased superoxide production, mitochondrial mass/morphology alterations and enhanced expression of LONP1. In line with these results, the cytotoxic effect (cell number, chromatin condensation, cell cycle alterations and induction of apoptosis) of EFV was less pronounced in Hep3B respiration-depleted cells than in wild-type cells. The effect of EFV was both similar and different from those of two distinct mitochondrial stressors, TG and Rot, depending on the parameter studied.
On the other hand, markers of mitochondrial dynamics were expressed differentially with the stimuli used (EFV, TG, Rot and CCCP), which points to a specificity of the dual ER/mitochondrial stress induced by EFV. EFV treatment enhanced mitochondria/ER interorganelle interaction, as shown by co-immunoprecipitation experiments of MAMs protein partners. In addition, LONP1 was upregulated at mRNA and protein levels under all conditions. Surprisingly, upon treatment with EFV, its extramitochondrial presence (ER and MAMs) increased.
In conclusion, hepatic cells lacking normal mitochondria (rho⁰) are less vulnerable to EFV. This finding may account for the idiosyncratic hepatic reactions triggered by anti-HIV drugs and may also explain the different degrees of susceptibility to liver damage seen in patients undergoing antiretroviral therapy. The specific dual mitochondria-ER effect induced by EFV enhances MAMs content and this is associated with increased extramitochondrial LONP1 expression. This is the first report of this phenomenon in mammalian cells and suggests a novel MAMs-linked function of LONP1

Material

and methods.
Retrospective observational study of historical cohorts,
includes 151 children
admitted with the diagnosis of CAP in the
Hospital
General
University of Castellón between 2016 and 2017.
Results
.
Patients admitted with NAC with antibiotherapy and good clinical course
present a median reduction of CRP with respect to admission of 81.41
% (IQR 73.66
-
87.43%). In 93% of patients, it decreased more than 50%. The presence of pleural
effusion is associate
d with a lower reduction of CRP and it
is associated with longer
duration of fever prior to the antibiotic.
Conclusion.
CRP is an analytica
l parameter predictive of clinical improvement,
although it
’
s use is not justified in patients with CAP who progress favorably. However,
it would be useful if they present a pleural effusion to determine the duration of
intravenous antibiotic therapy.Introducción

A Virtual Lab Environment For Improving Students’ Understanding Of Infiltration And Runoff Processes
Infiltration of water in the soil is a key process in the hydrological cycle and its understanding is paramount for students of any course related to Hydrology. Different equations exist, which, in function of boundary conditions and the level of detail required in any particular case can become complex to solve in an analytical way. Water movement in the soil can be described by the Richards equation, but its solution requires numerical methods. Therefore, amongst the most used ways of calculating water infiltration are the Green and Ampt method, Horton’s equation, Philip’s equation and the Curve Number method. In all these equations, different soil parameters intervene that can be measured in the field or estimated from soil properties. The understanding of the physical meaning of these parameters, their relative importance and of the differences between the abovementioned methods is important for students. Infiltration measurements in the field or lab are time-consuming. We therefore developed a tool that helps the students to calculate infiltration in a step-by-step way, and which helps to easily change the input parameter values so as to understand the importance of the different variables. This virtual lab environment was coded in Matlab and used for the first time during the academic year 2013-2014. A Comparison of Myoelectric Control Modes for an Assistive Robotic Virtual Platform
In this paper, we propose a daily living situation where objects in a kitchen can be grasped and stored in specific containers using a virtual robot arm operated by different myoelectric control modes. The main goal of this study is to prove the feasibility of providing virtual environments controlled through surface electromyography that can be used for the future training of people using prosthetics or with upper limb motor impairments. We propose that simple control algorithms can be a more natural and robust way to interact with prostheses and assistive robotics in general than complex multipurpose machine learning approaches. Additionally, we discuss the advantages and disadvantages of adding intelligence to the setup to automatically assist grasping activities. The results show very good performance across all participants who share similar opinions regarding the execution of each of the proposed control modes
Influencia del porcentaje de piedra pómez sobre la resistencia a la compresión y peso unitario en un concreto estructural para pórticos, Trujillo – 2019
La presente investigación se realizó en la Universidad Privada del Norte - Trujillo, la cual
consistió en determinar la influencia del porcentaje de piedra pómez en el aumento de la
resistencia a la compresión y peso unitario del concreto. Para desarrollar el estudio, se utilizó
un diseño experimental, en su categoría de experimental puro, utilizando un muestreo
probabilístico, la técnica fue la observación y para el análisis de datos, la inferencia estadística
con una confiabilidad al 95%.
Se aplicó diferentes porcentajes de piedra pómez al concreto convencional reemplazando al
agregado grueso en 5%, 10%, 15%, 20%, 25% y 30%; para disminuir su peso unitario y
optimizar la resistencia, obteniendo que el porcentaje óptimo es de 10%, alcanzando un f’c. de
277 kg/cm2 a los 28 días y una disminución de peso unitario de 2360 kg/m3, en comparación
con un diseño patrón con un f’c. de 210 kg/cm2 y 2420 kg/m3 en peso unitario.
Además, se realizó 3 modelaciones estructurales sismo estático de un edificio de 6 pisos para
evaluar el comportamiento sísmico de la estructura, Edificio A: según el ACI, con un peso
específico de 2400 Kg/m3 y un f’c=210 kg/cm2; Edificio B: conformado por un concreto patrón
de laboratorio, con un peso específico de 2520 kg/m3 y un f’c= 238 kg/cm2 y Edificio C:
conformado por un concreto de 10% de sustitución de piedra pómez, con un peso específico
de 2460 kg/m3 y un f’c=277 kg/cm2, obteniendo de éste último resultados de distorsiones entre
los valores de 0.003 mm y 0.007 mm, los cuales se encuentran en los parámetros establecidos
por la NTP E.030, y mencionando que el Edificio A y B no cumplen con dicha normativa.
Finalmente, se realizó un análisis de costos para 1m3 de concreto patrón y con 10 % de piedra
pómez obteniendo que éste último aumenta su costo en S/.7.24, siendo su costo de producción
S/.269.80 y S/.277.04 respectivamente
Adaptation to noise in amplitude modulation detection without the medial olivocochlear reflex
[EN]The detection of amplitude modulation (AM) in quiet or in noise improves when the AM carrier is preceded by noise, an effect that has been attributed to the medial olivocochlear reflex (MOCR). We investigate whether this improvement can occur without the MOCR by measuring AM sensitivity for cochlear implant (CI) users, whose MOCR effects are circumvented as a result of the electrical stimulation provided by the CI. AM detection thresholds were measured monaurally for short (50 ms) AM probes presented at the onset (early condition) or delayed by 300 ms (late condition) from the onset of a broadband noise. The noise was presented ipsilaterally, contralaterally and bilaterally to the test ear. Stimuli were processed through an experimental, time-invariant sound processing strategy. On average, thresholds were 4 dB better in the late than in the early condition and the size of the improvement was similar for the three noise lateralities
Perfil del alumnado del grado de educación infantil: proyecto docente de la asignatura trastornos del desarrollo
A relevant aspect of the context in which a teaching project is framed is that related to the specific characteristics of the people who will be the protagonists of the learning process: the students of the degree program. Without a doubt, the students are the raw material of the universities, and the reason for their existence. Analyzing the characteristics of the students, their origin, objectives, interests, motivations, personal aspirations, etc., are essential elements in any project. In this work, a study is made of the university students of Pre-school Education in general, and of the students of the subject matter of study in particular, Development Disorders. Their origin, expectations and training and professional interests, their previous knowledge and learning styles, condition the teaching decisions. The conclusions drawn from this analysis should be reflected in the pedagogical and methodological decisions of the teaching plan.Un aspecto relevante del contexto en el que se encuadra un proyecto docente es el relativo a las características específicas de las personas que van a protagonizar el proceso de aprendizaje: los alumnos de la titulación. Sin duda alguna, los alumnos son la materia prima de las universidades, y la razón de su existencia. Analizar las características de los alumnos, su procedencia, objetivos, intereses, motivaciones, aspiraciones personales, etc., son elementos imprescindibles en todo proyecto. En este trabajo se realiza un estudio del perfil de los alumnos universitarios de Educación Infantil en general, y de los alumnos de una materia objeto de estudio en particular, Trastornos del desarrollo. Su procedencia, expectativas e intereses formativos y profesionales, sus conocimientos previos y estilos de aprendizaje, condicionan las decisiones docentes. Las conclusiones extraídas de este análisis deberán verse reflejadas en las decisiones pedagógicas y metodológicas del plan docente
The Neolithic of El Niño cave (Ayna, Albacete) in the Sierra del Segura context
Desde su excavación en el verano de 1973, la Cueva del Niño ha sido considerada un yacimiento de gran importancia para conocer el proceso de introducción de la economía de producción o de la tecnología cerámica en el sureste de la Península Ibérica, y especialmente para establecer cómo dicho proceso pudo haber afectado a las poblaciones ya asentadas en la región de la Sierra del Segura. Sin embargo, la información proporcionada por la Cueva del Niño resultaba muy fragmentaria, debido a la falta de un estudio de conjunto de las ocupaciones neolíticas del yacimiento. En este artículo, se presenta el análisis de las producciones cerámicas, la industria lítica y los restos de fauna, así como las dataciones existentes para los niveles holocenos del depósito. La revisión de las diversas evidencias proporcionadas por el yacimiento permite plantear que la Cueva del Niño habría actuado probablemente como una estación para el pastoreo y la caza, sirviendo como satélite de otros yacimientos de mayor entidad. No obstante, las limitaciones impuestas por el hecho de tratarse de una excavación efectuada hace cuarenta años impide aportar aspectos concluyentes sobre la dinámica del proceso de introducción del Neolítico en la Sierra del Segura.Since its excavation in the summer of 1973, El Niño cave has been considered a key site to understand the process of production economy and pottery technology introduction in South-eastern Iberian Peninsula, and especially to approach how such process could have affected people already settled in the Segura mountains. However, data from El Niño cave was very fragmentary, due to the lack of a broad study of Neolithic occupations of the site. In this paper, we present the analysis of pottery, lithic industry and faunal remains, as well as the existing dates from the site´s Holocene levels. The review of different evidence from the site allows suggesting that El Niño cave would have probably acted as a hunting and shepherding station, being a logistical site of larger places. However, limitations due to the fact that we are dealing with a 40- year-old excavation, prevent specifying how the process of Neolithic introduction in the Segura Mountains occurred
El Niño Cave (Aýna, Albacete, Spain): Late Middle Palaeolithic, Rock Art, and Neolithic Occupations from Inland Iberia
RESUMEN: La Cueva del Niño, situada en el reborde sudeste de la Meseta castellana, alberga una secuencia discontinua que abarca desde el Paleolítico Medio hasta el Neolítico, además de pinturas rupestres paleolíticas y de estilo Levantino. Se trata por lo tanto de un yacimiento clave para comprender la ocupación del interior de la Península Ibérica durante el Paleolítico y la Prehistoria Reciente. Este artículo recoge los principales resultados de un proyecto multidisciplinar destinado a definir las diferentes ocupaciones del yacimiento a lo largo de la Prehistoria.ABSTRACT: El Niño cave, located on the south-eastern border of the Spanish Meseta, hosts a discontinuous sequence including Middle Palaeolithic and Neolithic levels, along with Upper Palaeolithic and Levantine style paintings.
