The Short-Term Opening of Cyclosporin A-Independent Palmitate/Sr2+-Induced Pore Can Underlie Ion Efflux in the Oscillatory Mode of Functioning of Rat Liver Mitochondria
Mitochondria are capable of synchronized oscillations in many variables, but the underlying mechanisms are still unclear. In this study, we demonstrated that rat liver mitochondria, when exposed to a pulse of Sr2+ ions in the presence of valinomycin (a potassium ionophore) and cyclosporin A (a specific inhibitor of the permeability transition pore complex) under hypotonia, showed prolonged oscillations in K+ and Sr2+ fluxes, membrane potential, pH, matrix volume, rates of oxygen consumption and H2O2 formation. The dynamic changes in the rate of H2O2 production were in a reciprocal relationship with the respiration rate and in a direct relationship with the mitochondrial membrane potential and other indicators studied. The pre-incubation of mitochondria with Ca2+(Sr2+)-dependent phospholipase A2 inhibitors considerably suppressed the accumulation of free fatty acids, including palmitic and stearic acids, and all spontaneous Sr2+-induced cyclic changes. These data suggest that the mechanism of ion efflux from mitochondria is related to the opening of short-living pores, which can be caused by the formation of complexes between Sr2+(Ca2+) and endogenous long-chain saturated fatty acids (mainly, palmitic acid) that accumulate due to the activation of phospholipase A2 by the ions. A possible role for transient palmitate/Ca2+(Sr2+)-induced pores in the maintenance of ion homeostasis and the prevention of calcium overload in mitochondria under pathophysiological conditions is discussed
NOVOSIBIRSK REGION PROGRAM OF THE LIVER TRANSPLANTATION IN CHILDREN
The aim of the study was to analyze the early and late outcomes of pediatric liver transplantation (LTx) program in the Novosibirsk region. Material and Methods. The retrospective study included 24 patients aged 72 ± 74.8 months (from 4 to 212 months, median – 20 months) with a body weight of 21.7 ± 18.1 kg (from 4.5 to 55 kg, median – 12.5 kg). Two (8.3 %) cadaveric whole liver grafts, 19 (79.2 %) living donor liver grafts, and 3 (12.5 %) cadaveric liver fragments (reduced-size or split-liver) were transplanted. Results. The features of vascular and biliary reconstruction in different types of LTx are discussed. The incidence of vascular and biliary complications was 8.3 and 20.8%, respectively. The patients stayed in the intensive care unit for 9 ± 5.1 days (from 4 to 22 days, median 8 days) and total length of hospital stay was for 40 ± 25.4 days (from 19 to 136 days, median 32 days). Two (8.3 %) recipients had early graft dysfunction. Perioperative mortality (up to 90 days) was absent. The overall 5-year patient and graft survival rates were 95 and 88 %, respectively. Conclusion. The Novosibirsk region has a pediatric LTx program with outcomes comparable to the data of the leading world and Russian centers
Present Status and Future Programs of the n_TOF Experiment
This is an Open Access article distributed under the terms of the Creative Commons Attribution-Noncommercial License 3.0, which permits unrestricted use, distribution, and reproduction in any noncommercial medium, provided the original work is properly citedThe neutron time-of-flight facility n_TOF at CERN, Switzerland, operational since 2001, delivers neutrons using the Proton Synchrotron (PS) 20 GeV/c proton beam impinging on a lead spallation target. The facility combines a very high instantaneous neutron flux, an excellent time of flight resolution due to the distance between the experimental area and the production target (185 meters), a low intrinsic background and a wide range of neutron energies, from thermal to GeV neutrons. These characteristics provide a unique possibility to perform neutron-induced capture and fission cross-section measurements for applications in nuclear astrophysics and in nuclear reactor technology. The most relevant measurements performed up to now and foreseen for the future will be presented in this contribution. The overall efficiency of the experimental program and the range of possible measurements achievable with the construction of a second experimental area (EAR-2), vertically located 20 m on top of the n_TOF spallation target, might offer a substantial improvement in measurement sensitivities. A feasibility study of the possible realisation of the installation extension will be also presented
Measurement of 73 Ge(n,γ) cross sections and implications for stellar nucleosynthesis
© 2019 The Author(s). Published by Elsevier B.V.73 Ge(n,γ) cross sections were measured at the neutron time-of-flight facility n_TOF at CERN up to neutron energies of 300 keV, providing for the first time experimental data above 8 keV. Results indicate that the stellar cross section at kT=30 keV is 1.5 to 1.7 times higher than most theoretical predictions. The new cross sections result in a substantial decrease of 73 Ge produced in stars, which would explain the low isotopic abundance of 73 Ge in the solar system.Peer reviewe
New measurement of neutron capture resonances of 209Bi
The neutron capture cross section of Bi209 has been measured at the CERN n
TOF facility by employing the pulse-height-weighting technique. Improvements
over previous measurements are mainly because of an optimized detection system,
which led to a practically negligible neutron sensitivity. Additional
experimental sources of systematic error, such as the electronic threshold in
the detectors, summing of gamma-rays, internal electron conversion, and the
isomeric state in bismuth, have been taken into account. Gamma-ray absorption
effects inside the sample have been corrected by employing a nonpolynomial
weighting function. Because Bi209 is the last stable isotope in the reaction
path of the stellar s-process, the Maxwellian averaged capture cross section is
important for the recycling of the reaction flow by alpha-decays. In the
relevant stellar range of thermal energies between kT=5 and 8 keV our new
capture rate is about 16% higher than the presently accepted value used for
nucleosynthesis calculations. At this low temperature an important part of the
heavy Pb-Bi isotopes are supposed to be synthesized by the s-process in the He
shells of low mass, thermally pulsing asymptotic giant branch stars. With the
improved set of cross sections we obtain an s-process fraction of 19(3)% of the
solar bismuth abundance, resulting in an r-process residual of 81(3)%. The
present (n,gamma) cross-section measurement is also of relevance for the design
of accelerator driven systems based on a liquid metal Pb/Bi spallation target.Comment: 10 pages, 5figures, recently published in Phys. Rev.
Measurement of the neutron capture cross section of the s-only isotope 204Pb from 1 eV to 440 keV
The neutron capture cross section of 204Pb has been measured at the CERN
n_TOF installation with high resolution in the energy range from 1 eV to 440
keV. An R-matrix analysis of the resolved resonance region, between 1 eV and
100 keV, was carried out using the SAMMY code. In the interval between 100 keV
and 440 keV we report the average capture cross section. The background in the
entire neutron energy range could be reliably determined from the measurement
of a 208Pb sample. Other systematic effects in this measurement could be
investigated and precisely corrected by means of detailed Monte Carlo
simulations. We obtain a Maxwellian average capture cross section for 204Pb at
kT=30 keV of 79(3) mb, in agreement with previous experiments. However our
cross section at kT=5 keV is about 35% larger than the values reported so far.
The implications of the new cross section for the s-process abundance
contributions in the Pb/Bi region are discussed.Comment: 8 pages, 3 figures, article submitted to Phys. Rev.
Measurement of the Ge 70 (n,γ) cross section up to 300 keV at the CERN n-TOF facility
©2019 American Physical Society.Neutron capture data on intermediate mass nuclei are of key importance to nucleosynthesis in the weak component of the slow neutron capture processes, which occurs in massive stars. The (n,γ) cross section on Ge70, which is mainly produced in the s process, was measured at the neutron time-of-flight facility n-TOF at CERN. Resonance capture kernels were determined up to 40 keV neutron energy and average cross sections up to 300 keV. Stellar cross sections were calculated from kT=5 keV to kT=100 keV and are in very good agreement with a previous measurement by Walter and Beer (1985) and recent evaluations. Average cross sections are in agreement with Walter and Beer (1985) over most of the neutron energy range covered, while they are systematically smaller for neutron energies above 150 keV. We have calculated isotopic abundances produced in s-process environments in a 25 solar mass star for two initial metallicities (below solar and close to solar). While the low metallicity model reproduces best the solar system germanium isotopic abundances, the close to solar model shows a good global match to solar system abundances in the range of mass numbers A=60-80.Peer reviewedFinal Published versio
