    A search for modifying genetic factors in CHEK2 : c.1100delC breast cancer patients

    The risk of breast cancer associated with CHEK2:c.1100delC is 2-threefold but higher in carriers with a family history of breast cancer than without, suggesting that other genetic loci in combination with CHEK2:c.1100delC confer an increased risk in a polygenic model. Part of the excess familial risk has been associated with common low-penetrance variants. This study aimed to identify genetic loci that modify CHEK2:c.1100delC-associated breast cancer risk by searching for candidate risk alleles that are overrepresented in CHEK2:c.1100delC carriers with breast cancer compared with controls. We performed whole-exome sequencing in 28 breast cancer cases with germline CHEK2:c.1100delC, 28 familial breast cancer cases and 70 controls. Candidate alleles were selected for validation in larger cohorts. One recessive synonymous variant, rs16897117, was suggested, but no overrepresentation of homozygous CHEK2:c.1100delC carriers was found in the following validation. Furthermore, 11 non-synonymous candidate alleles were suggested for further testing, but no significant difference in allele frequency could be detected in the validation in CHEK2:c.1100delC cases compared with familial breast cancer, sporadic breast cancer and controls. With this method, we found no support for a CHEK2:c.1100delC-specific genetic modifier. Further studies of CHEK2:c.1100delC genetic modifiers are warranted to improve risk assessment in clinical practice.Peer reviewe

    Balanskravet infördes i Sveriges kommuner 1 januari 2000. Detta innebar att intäkterna skall täcka kostnaderna. Sveriges kommuner hade en ekonomisk kris under 90-talet och för att balanskravet skulle kunna följas infördes blandmodellen. Detta innebar att pensioner som är intjänade fram till 1998 har lyfts ur balansräkningen och redovisas som en not, en ansvarsförbindelse. Eftersom inte samtliga pensionsskulder redovisas, ger blandmodellen en förskönad bild av verkligheten. Tanken om att varje generation skall bära sina egna kostnader brister då skattebetalarna inte bara bekostar dagens anställdas pension utan även får ta kostnaden för de som är pensionärer i dag. Detta kan tyckas vara fel men ändå helt enligt lagen. Man kan fråga sig om det är viktigare att följa balanskravet och blandmodellen än att redovisa en rättvisande bild. Att lyfta ut skulden medför att det egna kapitalet blir högre och på så sätt visas ett högre soliditetsmått. Soliditetsmåttet är viktigt eftersom det visar kommunernas betalningsförmåga på lång sikt. Att inte beräkna soliditetsmåttet inklusive skulden ger inte en korrekt bild av soliditeten, alltså blir även detta mått missvisande. På detta vis ger kommunernas redovisning en förvrängd bild av verkligheten, och man kan fråga sig om rättvisande bild bara är ett uttryck och saknar innebörd. Kommunerna agerar som om inte skulden existerar och kan på så vis använda pengar som inte finns. Vi har genom denna studie undersökt om blandmodellen ger en rättvisande bild av kommunernas redovisning samt hur soliditeten påverkas av detta. Studiens slutsats är att soliditetsmåtten som är beräknade enligt blandmodellen inte ger en rättvisande bild eftersom pensionsskulden inte är medräknad. Vi kan dessutom konstatera att utvecklingen av soliditetsmåttet har försämrats sedan blandmodellen infördes 2000.      

    CGGBP1 phosphorylation constitutes a telomere-protection signal

    The shelterin proteins are required for telomere integrity. Shelterin dysfunction can lead to initiation of unwarranted DNA damage and repair pathways at chromosomal termini. Interestingly, many shelterin accessory proteins are involved in DNA damage signaling and repair. We demonstrate here that in normal human fibroblasts, telomeric ends are protected by phosphorylation of CGG triplet repeat-binding protein 1 (CGGBP1) at serine 164 (S164). We show that serine 164 is a major phosphorylation site on CGGBP1 with important functions. We provide evidence that one of the kinases that can phosphorylate S164 CGGBP1 is ATR. Overexpression of S164A phospho-deficient CGGBP1 exerted a dominant-negative effect, causing telomeric dysfunction, accelerated telomere shortening, enhanced fusion of telomeres, and crisis. However, overexpression of wild-type or phospho-mimicking S164E CGGBP1 did not cause these effects. This telomere damage was associated with reduced binding of the shelterin protein POT1 to telomeric DNA. Our results suggest that CGGBP1 phosphorylation at S164 is a novel telomere protection signal, which can affect telomere-protective function of the shelterin complex

    Changes in proteins, physical stability and structure in directly heated UHT milk during storage at different temperatures

    Changes occurring in directly heated UHT milk were studied during storage at 5, 22, 30 and 40 °C. Industrially produced UHT milk samples were analysed for changes in enzymatic activity, protein modification, destabilisation of casein micelles and relocation of milk proteins in relation to sedimentation and gel formation. Sedimentation occurred at all temperatures, and the protein composition of the sediments reflected the composition of its liquid phase; however, there was no α-lactalbumin, β-lactoglobulin or κ-casein present in sediments. Tendrils composed of β-lactoglobulin and κ-casein were seen on casein micelles after UHT treatment and grew in length prior to gelation. High degrees of lactosylation of proteins and peptides were clearly correlated with the absence of gelation and long tendrils. Gelled samples showed complete hydrolysis of intact β-casein, and limited lactosylation of β-lactoglobulin and κ-casein

    Breast milk (BM) is the primary nutrition for infants and has a high content of lipids. Preterm infants receive expressed BM via tube feeding, and they are frequently treated with phototherapy. When parenteral nutrition (PN) is exposed to light and/or phototherapy, lipid peroxidation (LPO) increases. By light-protecting PN, morbidity and mortality are reduced in preterm infants through the reduction of oxidative stress. We aimed to investigate whether light-protecting breast milk could reduce LPO. Twelve mothers giving birth to a preterm infants of less than 32 weeks of gestational age were included. Transitional BM was collected and divided into three study groups; light-protected, ward light and phototherapy light. Baseline samples were collected after expression and the exposures started within one hour. Feeding syringe samples were exposed to light for 30 up to 360 min. Nasogastric tube samples were run through a tube under the same light conditions. Samples were stored in −80 °C until analyses of malondialdehyde (MDA), 4-hydroxynonenal (4-HNE) and total antioxidant capacity (TAC). There were no significant differences in MDA, 4-HNE or TAC levels observed between the different study groups.
