Combining a pollen and macrofossil synthesis with climate simulations for spatial reconstructions of European climate using Bayesian filtering
Probabilistic spatial reconstructions of past climate states are valuable to quantitatively study the climate system under different forcing conditions because they combine the information contained in a proxy synthesis into a comprehensible product. Unfortunately, they are subject to a complex uncertainty structure due to complicated proxyâclimate relations and sparse data, which makes interpolation between samples difficult. Bayesian hierarchical models feature promising properties to handle these issues, like the possibility to include multiple sources of information and to quantify uncertainties in a statistically rigorous way.
We present a Bayesian framework that combines a network of pollen and macrofossil samples with a spatial prior distribution estimated from a multi-model ensemble of climate simulations. The use of climate simulation output aims at a physically reasonable spatial interpolation of proxy data on a regional scale. To transfer the pollen data into (local) climate information, we invert a forward version of the probabilistic indicator taxa model. The Bayesian inference is performed using Markov chain Monte Carlo methods following a Metropolis-within-Gibbs strategy.
Different ways to incorporate the climate simulations into the Bayesian framework are compared using identical twin and cross-validation experiments. Then, we reconstruct the mean temperature of the warmest and mean temperature of the coldest month during the mid-Holocene in Europe using a published pollen and macrofossil synthesis in combination with the Paleoclimate Modelling Intercomparison Project Phase III mid-Holocene ensemble. The output of our Bayesian model is a spatially distributed probability distribution that facilitates quantitative analyses that account for uncertainties
Zur Theorie des Kaufverhaltens bei Second-Hand-GĂŒtern
Der nachfolgende Beitrag nÀhert sich dem PhÀnomen einer asymmetrischen Informationsverteilung
aus der verhaltenswissenschaftlichen Perspektive. Die zutage geförderten
Erkenntnisse beruhen auf theoretischen Ăberlegungen und einer schriftlichen Befragung von
Verbrauchern. Das theoretische Fundament der Arbeit konstituieren die mikroökonomische
Theorie des Konsumentenverhaltens, die Theorie des sozialen Vergleichs, das Konzept der
sozialen Bezugsgruppe, die Theorie der kognitiven Dissonanz sowie die Theorie des
wahrgenommenen Kaufrisikos. Das auf dieser Basis rekonstruierte Kausalmodell wird mittels
des Programmpakets LISREL mit empirischem Datenmaterial konfrontiert.
Die Frage, ob und auf welche Art und Weise eine asymmetrische Informationsverteilung
zwischen Marktpartnern deren Handeln beeinfluĂt, zog von seiten der verhaltenswissenschaftlich
orientierten Konsumentenforschung bislang wenig Aufmerksamkeit auf
sich. Diese LĂŒcke nehmen die Autoren zum AnlaĂ, sich dem angesprochenen PhĂ€nomen aus
der verhaltenswissenschaftlichen Perspektive zu nÀhern. Hierzu generieren sie zunÀchst auf der
Basis theoretischer Ăberlegungen ein Kausalmodell, das sie in einem nĂ€chsten Schritt anhand
von empirischem Datenmaterial ĂŒberprĂŒfen. Hierbei zeigt sich, daĂ die auf einem Markt
bestehende Informationsasymmetrie das Agieren von Individuen nachhaltig beeinflussen kann.
Beleben lieĂe sich die Nachfrage auf einem solchen Markt insbesondere durch die Reduktion
des wahrgenommenen Kaufrisikos, beispielsweise mittels einer Funktionsgarantie, eines
GĂŒtesiegels oder dem Aufbau von Reputation seitens des Anbieters, sowie durch die
Verbesserung der Haltung, die die soziale Bezugsgruppe zum Erwerb eines entsprechenden
GerÀtes einnimmt
How lipid flippases can modulate membrane structure
AbstractPhospholipid flippases, are proteins able to translocate phospholipids from one side of a membrane to the other even against a gradient of concentration and thereby able to establish, or annihilate, a transmembrane asymmetrical lipid distribution. This lipid shuttling forms new membrane structures, in particular vesicles, which are associated with diverse physiological functions in eukaryotic cells such as lipid and protein traffic via vesicles between organelles or towards the plasma membrane, and the stimulation of fluid phase endocytosis. The transfer of lipids is also responsible for the triggering of membrane associated events such as blood coagulation, the recognition and elimination of apoptotic or aged cells, and the regulation of phosphatidylserine dependent enzymes. Exposure of new lipid-head groups on a membrane leaflet by rapid flip-flop can serve as a specific signal and, upon recognition, can be the cause of physiological modifications. Membrane bending is one of the mechanisms by which such activities can be triggered. We show that the lateral membrane tension is an important physical factor for the regulation of the size of the membrane invaginations. Finally, we suggest in this review that this diversity of functions benefits from the diversity of the lipids existing in a cell and the ability of proteins to recognize specific messenger molecules
A virtual centre at the interface of basic and applied weather and climate research
The Hans-Ertel Centre for Weather Research is a network of German
universities, research institutes and the German Weather Service (Deutscher
Wetterdienst, DWD). It has been established to trigger and intensify basic
research and education on weather forecasting and climate monitoring. The
performed research ranges from nowcasting and short-term weather forecasting
to convective-scale data assimilation, the development of parameterizations
for numerical weather prediction models, climate monitoring and the
communication and use of forecast information. Scientific findings from the
network contribute to better understanding of the life-cycle of shallow and
deep convection, representation of uncertainty in ensemble systems, effects of
unresolved variability, regional climate variability, perception of forecasts
and vulnerability of society. Concrete developments within the research
network include dual observation-microphysics composites, satellite forward
operators, tools to estimate observation impact, cloud and precipitation
system tracking algorithms, large-eddy-simulations, a regional reanalysis and
a probabilistic forecast test product. Within three years, the network has
triggered a number of activities that include the training and education of
young scientists besides the centre's core objective of complementing DWD's
internal research with relevant basic research at universities and research
institutes. The long term goal is to develop a self-sustaining research
network that continues the close collaboration with DWD and the national and
international research community
Endocytosis controlled by monolayer area asymmetry
Endozytose erfordert hohe MembrankrĂŒmmung und fĂŒhrt zu FlĂ€chenĂ€nderungen der MembranhĂ€lften. Dies kann durch eine OberflĂ€chendifferenz zwischen den Schichten initiiert werden, die durch geĂ€nderte Lipidzusammensetzungen hervorgerufen werden kann. Daher wurde die Hypothese aufgestellt, dass Lipid-Transporter zu Beginn der Endozytose fĂŒr verĂ€nderte FlĂ€chenverhĂ€ltnisse verantwortlich sind. Um den Einfluss verĂ€nderter FlĂ€chen auf Endozytose zu untersuchen, wurden die OberflĂ€chenverhĂ€ltnisse der Membran durch Zugabe von Phospholipiden verĂ€ndert und anschlieĂend Endozytose gemessen. AbhĂ€ngig von der Sorte wurden die Lipide nur in die Ă€uĂere Schicht eingebaut oder auch auf die innere Seite transportiert, wodurch die entsprechende Seite vergröĂert wurde. Die Zugabe verschiedener Aminophospholipide, die auf die innere Membranseite transportiert werden, fĂŒhrte zu gesteigerter âbulk flowâ Endocytose in K562-Zellen. DarĂŒber hinaus deuten die Ergebnisse darauf hin, dass Clathrin-vermittelte Endozytose von Hep2-Zellen ebenfalls stimuliert wurde. Umgekehrt hatte die Zugabe von Lipiden, die auf der Ă€uĂeren HĂ€lfte bleiben, reduzierte âbulk flowâ- oder Clathrin-vermittelte Endozytose in verschiedenen Zelllinien zur Folge. Bemerkenswert ist, dass auch Clathrin-vermittelte Endozytose durch die Lipidzugabe beeinflusst wurde, obwohl gerade in diesem Weg viele Proteine involviert sind, die KrĂŒmmung induzieren können. Dies passt zu einem neuen Modell wie Lipidtransporter in Endozytose involviert sind. Durch den Transport von Lipiden und die zusĂ€tzliche Interaktion mit Endozytoseproteinen, könnten diese Transporter zwei Mechanismen zur Erzeugung von KrĂŒmmung miteinander verbinden: MembrankrĂŒmmung induziert durch eine FlĂ€chenasymmetrie zwischen den MembranhĂ€lften und durch Wechselwirkung mit Proteinen. Die Ergebnisse dieser Arbeit deuten darauf hin, dass die fĂŒr Endozytose notwendige KrĂŒmmung durch die durch Lipidtransport induzierte FlĂ€chenasymmetrie der Membranschichten unterstĂŒtzt wird.Endocytic engulfment requires high local membrane curvature and causes significant area changes of the membrane leaflets. This can be initiated by differences between the surface areas of the two monolayers related to leaflet specific modulation of lipid composition. Thus, it was proposed that lipid translocators, pumping phospholipids from the outer to the inner leaflet, account for monolayer area asymmetry as an early step in endocytosis. To elucidate the influence of this asymmetry on endocytosis, surface area relation was altered by adding exogenous phospholipids to living cells and changes in endocytic activity were quantified. Depending on the lipid species, exogenous lipids were only incorporated into the outer layer or subsequently translocated across the plasma membrane thereby increasing either the outer or inner surface area. Addition of different analogues of aminophospholipids, which are translocated to the inner leaflet, led to an enhancement of bulk flow endocytosis in K562 cells. Moreover, our data indicate that clathrin-mediated endocytosis of Hep2 cells was stimulated as well. Inversely, addition of phospholipids, which remain on the outer layer, reduced bulk flow or clathrin-mediated endocytosis in various cell lines. Notably, also clathrin-mediated endocytosis was influenced by the addition of lipids, although many proteins noted for their ability to induce membrane curvature are known to be implicated in this pathway. This corroborates a recent model how aminophospholipid translocases are implicated in endocytosis. Upon translocating lipids and additionally interacting with endocytic accessory proteins, lipid translocators could integrate two processes to generate curvature: membrane bending based on monolayer area asymmetry and protein-related mechanisms. Collectively, findings in the present study suggest that curvature generation in endocytosis is supported by the induction of monolayer area asymmetry mediated by the translocation of lipids

Syftet med uppsatsen Àr att undersöka hur litteraturundervisning bedrivs i teori och praktik i
svenska och svenska som andrasprÄk pÄ gymnasiet. TvÄ underliggande frÄgestÀllningar har
stÀllts för att kunna besvara huvudproblematiken. Dessa frÄgestÀllningar Àr:
1. PÄ vilket sÀtt skiljer sig kursplanerna pÄ gymnasiet för svenska jÀmfört med svenska
som andrasprÄk nÀr det gÀller litteraturundervisning?
2. Hur arbetar olika lÀrare i svenska respektive svenska som andrasprÄk med litteratur
och pÄ vilket sÀtt knyter de an till kursplanernas innehÄll?
För att fÄ svar pÄ dessa frÄgor genomfördes tvÄ olika undersökningar. För det första jÀmfördes
kursplanerna pÄ gymnasiet för svenska och svenska som andrasprÄk med fokus pÄ litteratur-
undervisning. För det andra intervjuades sex lÀrare frÄn olika gymnasieskolor, varav tvÄ
undervisar i svenska, tvÄ i svenska som andrasprÄk och tvÄ i bÄda Àmnen. De tre olika lÀrar-
kategorierna valdes för att fÄ ett sÄ brett perspektiv som möjligt pÄ hela problematiken.
Kursplanerna för svenska och svenska som andrasprÄk strÀvar efter att skapa likvÀrdiga mÄl
och syften bÄda Àmnen emellan, men det finns ÀndÄ vissa skillnader. Svenskeleven ska t.ex.
lÀsa nordiska verk och göra nedslag i olika epoker i litteraturhistorien, men i svenska som
andrasprÄk ligger fokus pÄ svensk 1900-talslitteratur. I det stora hela arbetar lÀrare med
litteratur pÄ liknande sÀtt. Det Àr endast en lÀrare som utmÀrker sig eftersom hon arbetar helt
individuellt till skillnad frÄn de andra som varierar undervisningen i större grad. Alla lÀrare Àr
ense om att lÀromedel inte Àr centrala i deras undervisning. LÀrarna Àr ocksÄ överens om att
Àmnena ska vara likvÀrdiga. De lÀrare som undervisar i svenska lÀgger större vikt vid
litteraturhistorisk orientering Àn andrasprÄkslÀrare. UtifrÄn ett jÀmförande perspektiv Àr det
framförallt lÀrarna som undervisar i svenska som andrasprÄk som vet mycket mer om det
andra Àmnet svenska, och de Àr dessutom mer engagerade Àn vad svensklÀrarna Àr i det andra
Àmnet svenska som andrasprÄk. De nationella kursplanerna tycks prÀgla litteraturunder-
visningen mer Àn vad lÀrarna Àr medvetna om. NÀr det gÀller de lokala kursplanerna sÄ Àr
andrasprÄkslÀrarna betydligt mer insatta Àn vad lÀrarna i svenska Àr
MĂ€rkte fĂŒr gebrauchte GĂŒter : Ein Ansatz auf transaktionskostentheoretischer, sozialpsychologischer und spieltheoretischer Basis
MĂ€rkte fĂŒr Erzeugnisse aus zweiter Hand existieren fĂŒr eine Vielzahl von Produktklassen und gewinnen an Einfluss auf die entsprechenden PrimĂ€rmĂ€rkte. Dennoch fanden Second hand-MĂ€rkte in der Forschung bislang nur geringe Beachtung. Martin Ohlwein analysiert in einem Gebrauchtwarenmarkt agierende FunktionstrĂ€ger und Determinanten, die die Entstehung eines solchen Marktes erklĂ€ren. Der Autor quantifiziert das Volumen der Transaktionen anhand ausgewĂ€hlter Produktkategorien. AnschlieĂend untersucht er die Einstellung Beteiligter sowohl auf der Nachfrager- als auch auf der Anbieterseite. A pollen-based reconstruction of summer temperature in central North America and implications for circulation patterns during medieval times
We present a reconstruction of mean summer temperature for the northern Midwest of the USA based on lacustrine pollen records from three different lakes in Wisconsin. The results suggest a relatively warm period during the earlier part of the record (~1200â1500 CE) followed by a cooler Little Ice Age (~1500â1900) and a subsequent warming to modern conditions. The reconstructed modern summer mean temperature is in good agreement with observations, and the decades of the 1930s to 1950s appear to be the warmest such period in the proxy record (through 1974).
Analyses of circulation features associated with the warmest summers in the recent climate record suggest a prevalence of continental ridging accompanied by generally dry conditions during these warm summers in the Midwest. Drought reconstruction using the Palmer Drought Severity Index (PDSI) and tree-ring records as predictors also yield relatively dry conditions in medieval times for the central US. As reported in a number of recent studies, possible forcingmechanisms include La Niña-like conditions in the equatorial Pacific andwarmer than average waters in the tropical Indo-western Pacific Ocean possibly coupled to a positive mode of the AMO/ NAO North Atlantic circulation pattern
