    La mucosa del tracto reproductor femenino (TRF) debe conciliar las funciones reproductoras con la inmunidad para permitir la fecundación y mantener el embarazo. De esta forma, las hormonas sexuales femeninas no solo preparan el ambiente necesario para la reproducción, sino que también lo coordinan con el sistema inmune. Por ejemplo, a lo largo del ciclo ovárico existen variaciones en el número de neutrófilos en la vagina. Así, durante la fase folicular, los neutrófilos desaparecen dellumen vaginal, a la vez que aumenta la concentración de estradiol (E2). Tras la ovulación, los neutrófilos vuelven a infiltrarse en la vagina a lo largo de la fase lútea, coincidiendo con el pico de progesterona (P4)..

    Dependence of Induced Biological Damage on the Energy Distribution and Intensity of Clinical Intra-Operative Radiotherapy Electron Beams

    The survival fraction of epithelial HaCaT cells was analysed to assess the biological damage caused by intraoperative radiotherapy electron beams with varying energy spectra and intensities. These conditions were achieved by irradiating the cells at different depths in water using nominal 6 MeV electron beams while consistently delivering a dose of 5 Gy to the cell layer. Furthermore, a Monte Carlo simulation of the entire irradiation procedure was performed to evaluate the molecular damage in terms of molecular dissociations induced by the radiation. A significant agreement was found between the molecular damage predicted by the simulation and the damage derived from the analysis of the survival fraction. In both cases, a linear relationship was evident, indicating a clear tendency for increased damage as the averaged incident electron energy and intensity decreased for a constant absorbed dose, lowering the dose rate. This trend suggests that the radiation may have a more pronounced impact on surrounding healthy tissues than initially anticipated. However, it is crucial to conduct additional experiments with different target geometries to confirm this tendency and quantify the extent of this effect

    Estradiol-based therapies predispose women to vaginal infections. Moreover, it has long been known that neutrophils are absent from the vaginal lumen during the ovulatory phase (high estradiol). However, the mechanisms that regulate neutrophil influx to the vagina remain unknown. We investigated the neutrophil transepithelial migration (TEM) into the vaginal lumen. We revealed that estradiol reduces the CD44 and CD47 epithelial expression in the vaginal ectocervix and fornix, which retain neutrophils at the apical epithelium through the estradiol receptor-alpha. In contrast, luteal progesterone increases epithelial expression of CD44 and CD47 to promote neutrophil migration into the vaginal lumen and Candida albicans destruction. Distinctive to vaginal mucosa, neutrophil infiltration is contingent to sex hormones to prevent sperm from neutrophil attack; although it may compromise immunity during ovulation. Thus, sex hormones orchestrate tolerance and immunity in the vaginal lumen by regulating neutrophil TEM.The authors thank the units of flow cytometry, cell culture and statistical analysis.     Stress in the pre-slaughter stage is a critical period in the production chain because it causes losses in animal live weight, affects carcass yield and quality, and leads to a loss of animal welfare due to an alteration in the homeostasis of the organism as a result of exposure to stressful situations. This review identifies and describes the stressors that have been reported in the last 5 years as the main causes of meat quality deterioration, including animal handling activities by farm or plant personnel (transport, loading and unloading, in the slaughter plant), environmental conditions (temperature, vibrations), waiting time prior to slaughter and mixing of lots, water and food fasting period, and fatigue states, as well as the biomarkers routinely used for the study of conditions associated with stress. Although research on the subject is abundant, the biochemical mechanisms involved in obtaining certain organoleptic characteristics of meat with respect to stress levels and the individual variations observable in different groups of animals are still not completely clear, and more research is needed on stress biomarkers that allow a more specific, effective and non-invasive identification and measurement.El estrés en la etapa de prefaenado es un periodo crítico dentro de la cadena productiva al provocar pérdidas en el peso vivo del animal, afectaciones en el rendimiento y calidad de la canal y pérdida del bienestar animal debido a una alteración de homeóstasis del organismo por la exposición a situaciones estresantes. En la presente revisión se identifican y describen los factores estresores que se han reportado en los últimos 5 años como principales causas de deterioro de la calidad de la carne entre los cuales se incluyen actividades de manejo de los animales por parte del personal de granja o planta (transporte, carga y descarga, en planta de sacrificio), condiciones ambientales (temperatura, vibraciones), tiempo de espera previo al sacrificio y mezcla de lotes, periodo de ayuno de agua y comida, y estados de fatiga, así como los biomarcadores usados rutinariamente para el estudio de condiciones asociadas al estrés. Aunque las investigaciones sobre el tema son abundantes, aún no es totalmente claro los mecanismos bioquímicos involucrados en la obtención de ciertas características organolépticas de la carne con respecto a niveles de estrés y las variaciones individuales observables en diferentes grupos de animales, así mismo, se requiere mayor investigación en los biomarcadores de estrés que permitan una identificación y medición más específica, efectiva y no invasiva

    Biomarkers Associated with Regorafenib First-Line Treatment Benefits in Metastatic Colorectal Cancer Patients: REFRAME Molecular Study

    © 2021 by the authors.First-line treatment with regorafenib in frail metastatic colorectal cancer (mCRC) patients has shown some benefit. To accurately identify such patients before treatment, we studied blood biomarkers and primary tumor molecules. We unveiled serum microRNAs (miRNAs), single-nucleotide polymorphisms (SNPs) in angiogenic-related genes, and Notch 1 expression as biomarkers associated with response or toxicity. MicroRNA array profiling and genotyping of selected SNPs were performed in the blood of fragile mCRC patients treated with regorafenib. Notch 1 and CRC-associated miRNA expression was also analyzed in tumors. High levels of miR-185-5p in serum, rs7993418 in the vascular endothelial growth factor receptor 1 (VEGFR1) gene, and Notch 1 expression in biopsies were associated with a favorable response to treatment. Serum levels of miR-126-3p and miR-152-3p and tumor expression of miR-92a-1-5p were associated with treatment toxicity, particularly interesting in patients exhibiting comorbidities, and high levels of miR-362-3p were associated with asthenia. Additionally, several miRNAs were associated with the presence of metastasis, local recurrence, and peritoneal metastasis. Besides, miRNAs determined in primary tumors were associated with tumor-node-metastasis (TNM) staging. The rs2305948 and rs699947 SNPs in VEGFR2 and VEGFA, respectively, were markers of poor prognosis correlating with locoregional relapse, a higher N stage, and metastatic shedding.     Real-world effectiveness of caplacizumab vs the standard of care in immune thrombotic thrombocytopenic purpura

    Immune thrombotic thrombocytopenic purpura (iTTP) is a thrombotic microangiopathy caused by anti-ADAMTS13 antibodies. Caplacizumab is approved for adults with an acute episode of iTTP in conjunction with plasma exchange (PEX) and immunosuppression. The objective of this study was to analyze and compare the safety and efficacy of caplacizumab vs the standard of care and assess the effect of the concomitant use of rituximab. A retrospective study from the Spanish TTP Registry of patients treated with caplacizumab vs those who did not receive it was conducted. A total of 155 patients with iTTP (77 caplacizumab, 78 no caplacizumab) were included. Patients initially treated with caplacizumab had fewer exacerbations (4.5% vs 20.5%; P <.05) and less refractoriness (4.5% vs 14.1%; P <.05) than those who were not treated. Time to clinical response was shorter when caplacizumab was used as initial treatment vs caplacizumab used after refractoriness or exacerbation. The multivariate analysis showed that its use in the first 3 days after PEX was associated with a lower number of PEX (odds ratio, 7.5; CI, 2.3-12.7; P <.05) and days of hospitalization (odds ratio, 11.2; CI, 5.6-16.9; P <.001) compared with standard therapy. There was no difference in time to clinical remission in patients treated with caplacizumab compared with the use of rituximab. No severe adverse event was described in the caplacizumab group. In summary, caplacizumab reduced exacerbations and refractoriness compared with standard of care regimens. When administered within the first 3 days after PEX, it also provided a faster clinical response, reducing hospitalization time and the need for PEX

    Towards precision medicine: defining and characterizing adipose tissue dysfunction to identify early immunometabolic risk in symptom-free adults from the GEMM family study

    Interactions between macrophages and adipocytes are early molecular factors influencing adipose tissue (AT) dysfunction, resulting in high leptin, low adiponectin circulating levels and low-grade metaflammation, leading to insulin resistance (IR) with increased cardiovascular risk. We report the characterization of AT dysfunction through measurements of the adiponectin/leptin ratio (ALR), the adipo-insulin resistance index (Adipo-IRi), fasting/postprandial (F/P) immunometabolic phenotyping and direct F/P differential gene expression in AT biopsies obtained from symptom-free adults from the GEMM family study. AT dysfunction was evaluated through associations of the ALR with F/P insulin-glucose axis, lipid-lipoprotein metabolism, and inflammatory markers. A relevant pattern of negative associations between decreased ALR and markers of systemic low-grade metaflammation, HOMA, and postprandial cardiovascular risk hyperinsulinemic, triglyceride and GLP-1 curves was found. We also analysed their plasma non-coding microRNAs and shotgun lipidomics profiles finding trends that may reflect a pattern of adipose tissue dysfunction in the fed and fasted state. Direct gene differential expression data showed initial patterns of AT molecular signatures of key immunometabolic genes involved in AT expansion, angiogenic remodelling and immune cell migration. These data reinforce the central, early role of AT dysfunction at the molecular and systemic level in the pathogenesis of IR and immunometabolic disorders

    En 2018, Treinta y nueve estudiantes femeninas del X ciclo del periodo 2018-1 de la escuela de Psicología de una universidad particular del cono norte de Lima, ellas han descrito el romance de su personas cercanas a ellas. Se analizaron los 266 casos de parejas de enamorados estaban el rango de 17 a 25 años. El instrumento que se usó fue una CUESTIONARIO CUPIDO que es una escala nominal de conductas donde las estudiantes de psicología clasificaban los comportamientos en categorías preestablecidas, claramente definidas y mutuamente excluyentes. La conclusión más importante del estudio observacional fue que las parejas analizadas en un 64.4 % la mujer ero atrajo al varón y después el varón emo conectó con la mujer. También han observado otro estilo de romance cuando el varón ero atrajo a la mujer por su cuerpo atracción y después la mujer fue la que emo conectó con el varón y observaron este hecho en un 35.6 % de los casos
