    Proteolysis in milk during experimental Escherichia coli mastitis

    This work consisted of the intramammary infections (IMI) of 8 heifers by high doses of Escherichia coli to study both the proteolytic activity in milk and the resulting peptides. Therefore, a milking kinetic has been followed, and several parameters have been studied, such as proteose peptones ( PP) fraction ( quantitative and qualitative changes), plasmin activity (PA), milk somatic cell count (SCC), and bacterial count. A qualitative study of milk proteins and PP was performed by sodium dodecyl sulfate-PAGE, and the peptides recovered in PP during the acute phase of inflammation were amino-terminal micro-sequenced. A BSA increase in milk over time supported the hypothesis of an increase in the permeability of the epithelial barrier. A significant increase in PP content, considered to be an indicator of proteolysis, was observed from postinfusion hours (PIH) 12 to 48. Both the E. coli bacterial count and the SCC increased from PIH 3 to 216. Plasmin activity was increased noticeably from PIH 15 to 24. The respective increases in SCC, bacterial count, and PA suggest their involvement in a global mechanism responsible for the increase in proteolysis in milk after E. coli challenge. Somatic cell count and E. coli may be involved from PIH 3 to 216, and PA involvement might be highlighted during the maximum proteolysis, from PIH 15 to 24. A qualitative study of PP fraction by electrophoresis revealed the apparition of 5 peptide bands: P1 and P2 previously recovered during the lipopolysaccharide challenge, and E1 (27.0 kDa), E2 (15.5 kDa), and E3 (9.0 kDa) were specific to E. coli challenge; E1, E2, and E3 contained casein fragments.     Application d’une structure métamatériau sous forme triangulaire pour la réalisation de Filtres Coupe Bande

    Dans ce papier, nous présentons deux filtres coupe bande obtenus grâce à l’application des split triangle resonators (STRs). C’est une nouvelle particule que nous avons proposée et qui présente un ε et μ simultanément négatifs dans la même bande de fréquence. Ainsi, nous appliquons le résonateur sur deux composants microonde afin d’évaluer son impact sur leurs performances. Le premier filtre est basé sur la technologie microruban avec des STRs imprimés sur la masse. Le second filtre est dans la même catégorie et à l’inverse du premier, les STRs sont imprimés sur la ligne conductrice. Le substrat utilisé est le FR4. Nous avons procédé à une étude par simulation et avons aussi réalisé et fait une étude expérimentale des deux filtres. Les résultats expérimentaux et par simulation donnent des résultats avec une bonne similitude

    Some theoretical aspects in computational analysis of adhesive lap joints

    This paper is devoted to the numerical analysis of bidimensional bonded lap joints. For this purpose, the stress singularities occurring at the intersections of the adherend-adhesive interfaces with the free edges are first investigated and a method for computing both the order and the intensity factor of these singularities is described briefly. After that, a simplified model, in which the adhesive domain is reduced to a line, is derived by using an asymptotic expansion method. Then, assuming that the assembly debonding is produced by a macro-crack propagation in the adhesive, the associated energy release rate is computed. Finally, a homogenization technique is used in order to take into account a preliminary adhesive damage consisting of periodic micro-cracks. Some numerical results are presented

    The effect of stabilizing and destabilizing salts on the catalytic behavior of ribonuclease A (RNase A) was investigated at pH 7.5 and 25 degrees C, using spectrophotometric, viscometric and molecular dynamic methods. The changes in the distance between N(epsilon2) of His(12) and N(delta1) of His(119) at the catalytic center of RNase A upon the addition of sodium sulfate, sodium hydrogen sulfate and sodium thiocyanate were evaluated by molecular dynamic methods. The compactness and expansion in terms of Stokes radius of RNase A upon the addition of sulfate ions as kosmotropic salts, and thiocyanate ion as a chaotropic salt, were estimated by viscometric measurements. Enzyme activity was measured using cytidine 2', 3'-cyclic monophosphate as a substrate. The results from the measurements of distances between N(epsilon2) of His(12) and N(delta1) of His(119) and Stokes radius suggest (i) that the presence of sulfate ions decreases the distance between the catalytic His residues and increases the globular compactness, and (ii) that there is an expansion of the enzyme surface as well as elongation of the catalytic center in the presence of thiocyanate ion. These findings are in agreement with activity measurements

    Kynurenine 3-Monooxygenase Inhibition in Blood Ameliorates Neurodegeneration

    SummaryMetabolites in the kynurenine pathway, generated by tryptophan degradation, are thought to play an important role in neurodegenerative disorders, including Alzheimer's and Huntington's diseases. In these disorders, glutamate receptor-mediated excitotoxicity and free radical formation have been correlated with decreased levels of the neuroprotective metabolite kynurenic acid. Here, we describe the synthesis and characterization of JM6, a small-molecule prodrug inhibitor of kynurenine 3-monooxygenase (KMO). Chronic oral administration of JM6 inhibits KMO in the blood, increasing kynurenic acid levels and reducing extracellular glutamate in the brain. In a transgenic mouse model of Alzheimer's disease, JM6 prevents spatial memory deficits, anxiety-related behavior, and synaptic loss. JM6 also extends life span, prevents synaptic loss, and decreases microglial activation in a mouse model of Huntington's disease. These findings support a critical link between tryptophan metabolism in the blood and neurodegeneration, and they provide a foundation for treatment of neurodegenerative diseases

    Tachykinins Stimulate a Subset of Mouse Taste Cells

    The tachykinins substance P (SP) and neurokinin A (NKA) are present in nociceptive sensory fibers expressing transient receptor potential cation channel, subfamily V, member 1 (TRPV1). These fibers are found extensively in and around the taste buds of several species. Tachykinins are released from nociceptive fibers by irritants such as capsaicin, the active compound found in chili peppers commonly associated with the sensation of spiciness. Using real-time Ca2+-imaging on isolated taste cells, it was observed that SP induces Ca2+ -responses in a subset of taste cells at concentrations in the low nanomolar range. These responses were reversibly inhibited by blocking the SP receptor NK-1R. NKA also induced Ca2+-responses in a subset of taste cells, but only at concentrations in the high nanomolar range. These responses were only partially inhibited by blocking the NKA receptor NK-2R, and were also inhibited by blocking NK-1R indicating that NKA is only active in taste cells at concentrations that activate both receptors. In addition, it was determined that tachykinin signaling in taste cells requires Ca2+-release from endoplasmic reticulum stores. RT-PCR analysis further confirmed that mouse taste buds express NK-1R and NK-2R. Using Ca2+-imaging and single cell RT-PCR, it was determined that the majority of tachykinin-responsive taste cells were Type I (Glial-like) and umami-responsive Type II (Receptor) cells. Importantly, stimulating NK-1R had an additive effect on Ca2+ responses evoked by umami stimuli in Type II (Receptor) cells. This data indicates that tachykinin release from nociceptive sensory fibers in and around taste buds may enhance umami and other taste modalities, providing a possible mechanism for the increased palatability of spicy foods

    Enhancing surface production of negative ions using nitrogen doped diamond in a deuterium plasma

    The production of negative ions is of significant interest for applications including mass spectrometry, particle acceleration, material surface processing, and neutral beam injection for magnetic confinement fusion. Methods to improve the efficiency of the surface production of negative ions, without the use of low work function metals, are of interest for mitigating the complex engineering challenges these materials introduce. In this study we investigate the production of negative ions by doping diamond with nitrogen. Negatively biased (−20-20 V or −130-130 V), nitrogen doped micro-crystalline diamond films are introduced to a low pressure deuterium plasma (helicon source operated in capacitive mode, 2 Pa, 26 W) and negative ion energy distribution functions (NIEDFs) are measured via mass spectrometry with respect to the surface temperature (30∘^{\circ}C to 750∘^{\circ}C) and dopant concentration. The results suggest that nitrogen doping has little influence on the yield when the sample is biased at −130-130 V, but when a relatively small bias voltage of −20-20 V is applied the yield is increased by a factor of 2 above that of un-doped diamond when its temperature reaches 550∘^{\circ}C. The doping of diamond with nitrogen is a new method for controlling the surface production of negative ions, which continues to be of significant interest for a wide variety of practical applications
