    Evaluation of reproductive strategies in captive California yellowtail (Seriola dorsalis) using genetic parentage analyses

    A good understanding of spawning dynamics of species in aquaculture is vital in order to maximize egg production and quality as well as efficient allocation of food and space resources. The breeding program of California yellowtail (CYT; Seriola dorsalis previously Seriola lalandi) at Hubbs-SeaWorld Research Institute (HSWRI) is set up such that up to 30 wild caught brood fish can broadcast spawn in a group setting, just as CYT reproduce in the wild. The population of brood fish were originally caught offshore from Southern California, and are maintained under ambient sea water and natural lighting conditions. The spawning season at HSWRI lasted from March to September during the 2013 and 2014 study period. Reproductive output from this population of cultured CYT was evaluated through microsatellite-based parentage analyses whereby the percent contribution of offspring was determined across all spawning events over two years. Methods were first tested to determine the minimum sample size required to accurately describe the parental contributions in this specific group spawning environment. To do this, five spawns were selected from the 2013 season based on spawn volume, which was presumed to represent a high number of female parents thus representing the most complex spawning dynamic. For these five spawning events, there were 19 brood fish present, representing all possible parents. Actual assignment of samples sizes between 47 and over 300 was assessed for each spawn. Except in one instance, the parental contribution from sample size of 47 CYT offspring analyzed per spawn, was not statistically significantly different than a sample size of over 300 offspring per spawn (average P-value = 0.65). Simulated subsampling via computational bootstrapping, and subsequent statistical analysis, indicated that a sample size of 30 offspring per spawn was adequate to accurately describe the parental contributions. Based on this information, a samples size of 47 was used so that samples from two spawns could be run on a single 96 well plate, including one negative control sample per spawn. This constitutes one of the first studies of sample size quality control for genetic parentage contribution for an aquaculture species. Offspring were then analyzed from every spawning event (N = 130) over two spawning seasons to characterize spawning events of CYT. Analyzing all spawns during multiple spawning seasons allowed for determination of individual contribution levels, spawn pairings, and analysis of female fecundity. The breeding population consisted of 19 brood fish in 2013, and 37 in 2014; both years were roughly 50: 50 male – female. Brood fish added to the population in 2014 were smaller in terms of mass and total lengths. Spawning events tended to have one female contribute (primary female), with relatively equal paternal contribution during both the spawning events and season, for a possible lottery polygyny (Nunney, 1993) spawning system. One female in particular contributed 40% of all offspring during the two years, spawning nearly every 5-6 days during the spawning seasons. The larger females (~21 kg) had an average batch fecundity of ~490,000 eggs per spawn, while smaller females (~8.4 kg) only spawned 35,000 eggs per spawn. Annual and batch spawning totals were correlated to female mass (P \u3c 0.0005). All smaller brood fish spawned fewer times in the season than larger brood fish. This work constitutes the first-ever study of reproductive strategy (lottery polygyny) and parental contribution for a carangid species at the individual-level over several spawning events

    Plasmodium falciparum Rab1A localizes to rhoptries in schizonts

    Over-expression of a GFP-PfRab1A fusion protein in Plasmodium falciparum schizonts produces a punctate pattern of fluorescence typical of rhoptries, secretory organelles involved in host cell invasion. The GFP-positive bodies were purified by a combination of differential and density gradient centrifugation and their protein content determined by MS/MS sequencing. Consistent with the GFP rhoptry-like pattern of transgenic parasites, four of the 19 proteins identified have been previously described to be rhoptry-associated and another four are ER or ER-associated proteins. Confirmation that GFP-PfRab1A decorates rhoptries was obtained by its co-localization with Rap1 and Ron4 in late phase schizonts. We conclude that PfRab1A potentially regulates vesicular traffic from the endoplasmic reticulum to the rhoptries in Apicomplexa parasites

    Landscape approaches for ecosystem management in Mediterranean Islands

    This book presents a series of essays, drawing on the twm concepts of ecosystem management and landscape approaches, to elucidate and reflect on the present situation and future evolution of Mediterranean islands. This publication brings together contributions from Mediterranean individuals, non-Mediterranean individuals, islanders and non-islanders there is, after all, no geographical limit on who and what we can learn from. The essays presented here each contribute a specific perspective on the future evolution of Mediterranean islands. This book presents a series of essays, drawing on the twin concepts of ecosystem management and landscape approaches, to elucidate and reflect on the present situation and future evolution of Mediterranean islands. This publication brings together contributions from Mediterranean individuals, non-Mediterranean individuals, islanders and non-islanders; there is, after all, no geographical limit on who and what we can learn from. The essays presented here each contribute a specific perspective on the future evolution of Mediterranean islands. Following this introductory chapter, the first section of the book focuses on the contributions that can be made by the discipline of landscape ecology. loannis Vogiatzakis and Geoffrey Griffiths first explain the concepts and relevance of landscape ecology, also presenting and discussing a range of applied tools that can facilitate landscape planning in Mediterranean islands. Louis F. Cassar then reviews the 'offshoot' discipline of restoration ecology, making a strong case for offsetting the environmental damage inflicted on natural ecosystems over millennia of human occupation, with constructive efforts to effectively restore and/or rehabilitate ecosystems. The two following chapters bring the socio-economic dimension into the discussion. Godfrey Baldacchino first presents two contrasting paradigms for the development of island territories, reviewing the dual influences of ecological and economic factors, and exploring ways in which the two can be brought together in successful development strategies. Gordon Cordina and Nadia Farrugia then address the demographic dimension of development, presenting a model to explain the economic costs of high population densities on islands. The third block of chapters expands on the relevance of social and cultural dynamics to the management of Mediterranean Islands. Isil Cakcï, Nur Belkayali and Ilkden Tazebay explain the evolution of the concept of a 'cultural landscape', focusing on the challenges of managing change in landscapes with strong heritage values. The chapter concludes with a case study on the Turkish island of Gökçeada (lmbros), which is experiencing major challenges in balancing the conservation of a cultural landscape on the one hand, and the management of inevitable change, on the other. Elisabeth Conrad then discusses the role of social capital in managing the landscape resources of Mediterranean islands, reviewing the potential for this intangible social fabric to facilitate or impede the sustainable evolution of island territories. The fourth section includes four chapters, each of which addresses a different aspect relevant to policy development and implementation in Mediterranean islands. Salvino Busuttil presents an essay outlining the political influences on the management of coastal landscapes, the latter so relevant to Mediterranean island territories. The essay derives from the author's professional experience in various policy-related institutions for environmental management within the Mediterranean region. Maggie Roe then reflects on issues of landscape sustainability, focusing on the neglected aspect of intelligence. She discusses ways in which landscape research, knowledge and understanding can feed directly into frameworks for 'sustainable' landscape planning. In the subsequent chapter, Adrian Phillips takes from his substantial experience with international landscape policy, reviewing the gradual emergence of international and national landscape 'tools', to draw out lessons for application in Mediterranean islands. In the final chapter of this section, Riccardo Priore and Damiano Galla present a comprehensive discussion of the European Landscape Convention, the first international instrument to focus exclusively on landscape. The authors explain the innovative character of this convention, and discuss its potential implementation in Mediterranean islands. The publication concludes with a series of case studies, highlighting specific constraints, experiences and opportunities in different Mediterranean islands. Theano Terkenli explores the landscapes of tourism in Mediterranean islands - perhaps no other industry has played such a fundamental role in shaping the evolution of Mediterranean landscapes in recent years. The author reviews the theoretical relationship between landscape and tourism across Mediterranean islands, before focusing on the specific case of the Greek Cycladic islands. In the following chapter, Alex Camilleri, Isabella Colombini and Lorenzo Chelazzi present an in-depth review of the context and challenges being faced on a number of minor Mediterranean islands, namely those of the Tuscan archipelago (Elba, Giglio, Capraia, Montecristo, Pianosa, Gorgona and Giannutri), and Comino, the latter forming part of the Maltese archipelago. The comparison between these various islands enables an appreciation of both commonalities across these islands, as well as considerations that are specific to the context of each in dividual island. JeremyBoissevain then adopts an anthropological lens to review the cautionary tale of landscape change in Malta, exploring underlying causes of landscape destruction and limited civil engagement. In the subsequent chapter, Jala Makhzoumi outlines the richness of Mediterranean islands' rural landscapes, focusing on olive landscapes in Cyprus. Her research demonstrates the economic and ecological robustness of various olive cultivation practices, and whilst warning of several threats to such sustainable regimes, she outlines strategies for reconfiguring our approach to rural heritage, in order to integrate such assets into sustainable development strategies. Finally, Stephen Morse concludes the section with an evaluation of sustainable development indicators, and the contribution that these can make towards enhancing the management of Mediterranean island territories. He illustrates his arguments with reference to the two island states of Malta and Cyprus. To conclude, in the final chapter of this publication, we review key insights emerging from the various chapters, and summarize considerations for ecosystem management and sustainable development in Mediterranean Islands.     Modern academic librarianship demands reconfigured skillsets and expertise to meet the rapidly evolving, data-driven needs of today’s students, faculty, and researchers as they pursue digital scholarship and produce immense amounts of research data. These rapid developments in user needs and research services speak to an urgent need to provide training for academic librarians to support digital scholarship. We seek to address this need in “Digging Deeper, Reaching Further: Libraries Empowering Users to Mine the HathiTrust Digital Library Resources,” a three-year project funded by a Laura Bush 21st Century Librarian grant award from the Institute of Museum and Library Services.     The Biosurveillance Analytics Resource Directory (BARD): Facilitating the Use of Epidemiological Models for Infectious Disease Surveillance

    Epidemiological modeling for infectious disease is important for disease management and its routine implementation needs to be facilitated through better description of models in an operational context. A standardized model characterization process that allows selection or making manual comparisons of available models and their results is currently lacking. A key need is a universal framework to facilitate model description and understanding of its features. Los Alamos National Laboratory (LANL) has developed a comprehensive framework that can be used to characterize an infectious disease model in an operational context. The framework was developed through a consensus among a panel of subject matter experts. In this paper, we describe the framework, its application to model characterization, and the development of the Biosurveillance Analytics Resource Directory (BARD; http://brd.bsvgateway.org/brd/), to facilitate the rapid selection of operational models for specific infectious/communicable diseases. We offer this framework and associated database to stakeholders of the infectious disease modeling field as a tool for standardizing model description and facilitating the use of epidemiological models

    The reductive activation of CO2 across a Ti═Ti double bond: synthetic, structural, and mechanistic studies

    [Image: see text] The reactivity of the bis(pentalene)dititanium double-sandwich compound Ti(2)Pn(†)(2) (1) (Pn(†) = 1,4-{Si(i)Pr(3)}(2)C(8)H(4)) with CO(2) is investigated in detail using spectroscopic, X-ray crystallographic, and computational studies. When the CO(2) reaction is performed at −78 °C, the 1:1 adduct 4 is formed, and low-temperature spectroscopic measurements are consistent with a CO(2) molecule bound symmetrically to the two Ti centers in a μ:η(2),η(2) binding mode, a structure also indicated by theory. Upon warming to room temperature the coordinated CO(2) is quantitatively reduced over a period of minutes to give the bis(oxo)-bridged dimer 2 and the dicarbonyl complex 3. In situ NMR studies indicated that this decomposition proceeds in a stepwise process via monooxo (5) and monocarbonyl (7) double-sandwich complexes, which have been independently synthesized and structurally characterized. 5 is thermally unstable with respect to a μ-O dimer in which the Ti–Ti bond has been cleaved and one pentalene ligand binds in an η(8) fashion to each of the formally Ti(III) centers. The molecular structure of 7 shows a “side-on” bound carbonyl ligand. Bonding of the double-sandwich species Ti(2)Pn(2) (Pn = C(8)H(6)) to other fragments has been investigated by density functional theory calculations and fragment analysis, providing insight into the CO(2) reaction pathway consistent with the experimentally observed intermediates. A key step in the proposed mechanism is disproportionation of a mono(oxo) di-Ti(III) species to yield di-Ti(II) and di-Ti(IV) products. 1 forms a structurally characterized, thermally stable CS(2) adduct 8 that shows symmetrical binding to the Ti(2) unit and supports the formulation of 4. The reaction of 1 with COS forms a thermally unstable complex 9 that undergoes scission to give mono(μ-S) mono(CO) species 10. Ph(3)PS is an effective sulfur transfer agent for 1, enabling the synthesis of mono(μ-S) complex 11 with a double-sandwich structure and bis(μ-S) dimer 12 in which the Ti–Ti bond has been cleaved

    Tracer transport timescales and the observed Atlantic-Pacific lag in the timing of the Last Termination

The definitive version was published in Paleoceanography 27 (2012): PA3225, doi:10.1029/2011PA002273.The midpoint of the Last Termination occurred 4,000 years earlier in the deep Atlantic than the deep Pacific according to a pair of benthic foraminiferal δ18O records, seemingly implying an internal circulation shift because the lag is much longer than the deep radiocarbon age. Here a scenario where the lag is instead caused by regional surface boundary condition changes, delays due to oceanic transit timescales, and the interplay between temperature and seawater δ18O (δ18Ow) is quantified with a tracer transport model of the modern-day ocean circulation. Using an inverse method with individual Green functions for 2,806 surface sources, a time history of surface temperature and δ18Ow is reconstructed for the last 30,000 years that is consistent with the foraminiferal oxygen-isotope data, Mg/Ca-derived deep temperature, and glacial pore water records. Thus, in the case that the ocean circulation was relatively unchanged between glacial and modern times, the interbasin lag could be explained by the relatively late local glacial maximum around Antarctica where surface δ18Ow continues to rise even after the North Atlantic δ18Ow falls. The arrival of the signal of the Termination is delayed at the Pacific core site due to the destructive interference of the still-rising Antarctic signal and the falling North Atlantic signal. This scenario is only possible because the ocean is not a single conveyor belt where all waters at the Pacific core site previously passed the Atlantic core site, but instead the Pacific core site is bathed more prominently by waters with a direct Antarctic source.G.G. is supported by NSF grant OIA-1124880 and the WHOI Arctic Research Initiative.2013-03-0

    Vaccine delivery by penetratin: mechanism of antigen presentation by dendritic cells

    Cell-penetrating peptides (CPP) or membrane-translocating peptides such as penetratin from Antennapedia homeodomain or TAT from human immunodeficiency virus are useful vectors for the delivery of protein antigens or their cytotoxic (Tc) or helper (Th) T cell epitopes to antigen-presenting cells. Mice immunized with CPP containing immunogens elicit antigen-specific Tc and/or Th responses and could be protected from tumor challenges. In the present paper, we investigate the mechanism of class I and class II antigen presentation of ovalbumin covalently linked to penetratin (AntpOVA) by bone marrow-derived dendritic cells with the use of biochemical inhibitors of various pathways of antigen processing and presentation. Results from our study suggested that uptake of AntpOVA is via a combination of energy-independent (membrane fusion) and energy-dependent pathways (endocytosis). Once internalized by either mechanism, multiple tap-dependent or independent antigen presentation pathways are accessed while not completely dependent on proteasomal processing but involving proteolytic trimming in the ER and Golgi compartments. Our study provides an understanding on the mechanism of antigen presentation mediated by CPP and leads to greater insights into future development of vaccine formulations
