Genomic DNA isolation from Artemisia species grown in cold desert high altitude of India
Artemisia which produces a large number of secondary metabolites is naturally found in cold desert high altitude environment of India. Secondary metabolites such as alkaloids, flavonoids, phenols, polysaccharides and terpenes represent a significant barrier to the extraction of pure genomic DNA. Thus, in this study, the DNA extraction protocol to extract pure genomic DNA from different Artemisia species was tailored. The protocol was based on the CTAB method with slight modifications. In the study, 1.6 M NaCl, 2% cetyltrimethyl ammonium bromide (CTAB), 3% polyvinylpyrrolidone (PVP) and 0.5% β-mercaptoethanol was used in the extraction buffer. The incubation period was kept for 1 h at 65°C with one-tenth of the volume of warm (55°C) 10% CTAB solution during the extraction process. This study described a reliable protocol for extracting good quality and optimum amount of DNA from Artemisia species suitable for PCR analysis.Key words: Artemisia, genomic DNA isolation, PCR amplification, secondary metabolites
Microbial Fingerprinting of Potential Biodegrading Organisms

The biological responses to various pollutants show variations as the living system behaves differently in their sensitivities to the same types of pollutants. The relative response and activity depend upon the duration of exposure to the specific pollutant. It is impossible to stop various activities leading to environmental pollution; however, pollutants can be eliminated from the environment using the microorganisms. Application of biological processes can be executed in order to get rid of toxic pollutants through their biodegradation. The pollutants like hydrocarbons, heavy metals, chlorinated hydrocarbons, nitro-aromatic compounds, non-chlorinated herbicides and pesticides, organophosphates, radionuclides can lead to serious health and environmental problems. The main objective of this paper is to evaluate the effects of pollutants on the living beings and environment, microbial responses to pollution, and distribution of various biodegrading microorganisms in the environment. Profiling of biodegrading microorganisms, microbial biosensor to detect environmental pollution, and strain improvement through genetic manipulation to enhance the biodegradation process have been discussed in detail
Social factors and obesity: an investigation of the role of health behaviours
OBJECTIVES: This study evaluated a behavioural model of the relation between social factors and obesity, in which differences in body mass index (BMI) across sociodemographic groups were hypothesized to be attributable to social group differences in health behaviours affecting energy expenditure (physical activity, diet and alcohol consumption and weight control). METHODS: A total of 8667 adults who participated in the 1995 Australian National Health and Nutrition Surveys provided data on a range of health factors including objectively measured height and weight, health behaviours, and social factors including family status, employment status, housing situation and migration status. RESULTS: Social factors remained significant predictors of BMI after controlling for all health behaviours. Neither social factors alone, nor health behaviours alone, adequately explained the variance in BMI. Gender-specific interactions were found between social factors and individual health behaviours. CONCLUSIONS: These results suggest that social factors moderate the relation between BMI and weight-related behaviours, and that the mechanisms underlying sociodemographic group differences in obesity may vary among men and women. Additional factors are likely to act in conjunction with current health behaviours to explain variation in obesity prevalence across sociodemographic groups.<br /
Identification of Mannose Interacting Residues Using Local Composition
BACKGROUND: Mannose binding proteins (MBPs) play a vital role in several biological functions such as defense mechanisms. These proteins bind to mannose on the surface of a wide range of pathogens and help in eliminating these pathogens from our body. Thus, it is important to identify mannose interacting residues (MIRs) in order to understand mechanism of recognition of pathogens by MBPs. RESULTS: This paper describes modules developed for predicting MIRs in a protein. Support vector machine (SVM) based models have been developed on 120 mannose binding protein chains, where no two chains have more than 25% sequence similarity. SVM models were developed on two types of datasets: 1) main dataset consists of 1029 mannose interacting and 1029 non-interacting residues, 2) realistic dataset consists of 1029 mannose interacting and 10320 non-interacting residues. In this study, firstly, we developed standard modules using binary and PSSM profile of patterns and got maximum MCC around 0.32. Secondly, we developed SVM modules using composition profile of patterns and achieved maximum MCC around 0.74 with accuracy 86.64% on main dataset. Thirdly, we developed a model on a realistic dataset and achieved maximum MCC of 0.62 with accuracy 93.08%. Based on this study, a standalone program and web server have been developed for predicting mannose interacting residues in proteins (http://www.imtech.res.in/raghava/premier/). CONCLUSIONS: Compositional analysis of mannose interacting and non-interacting residues shows that certain types of residues are preferred in mannose interaction. It was also observed that residues around mannose interacting residues have a preference for certain types of residues. Composition of patterns/peptide/segment has been used for predicting MIRs and achieved reasonable high accuracy. It is possible that this novel strategy may be effective to predict other types of interacting residues. This study will be useful in annotating the function of protein as well as in understanding the role of mannose in the immune system
A Novel Triterpenoid Isolated from the Root Bark of Ailanthus excelsa Roxb (Tree of Heaven), AECHL-1 as a Potential Anti-Cancer Agent
We report here the isolation and characterization of a new compound Ailanthus excelsa chloroform extract-1 (AECHL-1) (C(29)H(36)O(10); molecular weight 543.8) from the root bark of Ailanthus excelsa Roxb. The compound possesses anti-cancer activity against a variety of cancer cell lines of different origin.AECHL-1 treatment for 12 to 48 hr inhibited cell proliferation and induced death in B16F10, MDA-MB-231, MCF-7, and PC3 cells with minimum growth inhibition in normal HEK 293. The antitumor effect of AECHL-1 was comparable with that of the conventional antitumor drugs paclitaxel and cisplatin. AECHL-1-induced growth inhibition was associated with S/G(2)-M arrests in MDA-MB-231, MCF-7, and PC3 cells and a G(1) arrest in B16F10 cells. We observed microtubule disruption in MCF-7 cells treated with AECHL-1 in vitro. Compared with control, subcutaneous injection of AECHL-1 to the sites of tumor of mouse melanoma B16F10 implanted in C57BL/6 mice and human breast cancer MCF-7 cells in athymic nude mice resulted in significant decrease in tumor volume. In B16F10 tumors, AECHL-1 at 50 microg/mouse/day dose for 15 days resulted in increased expression of tumor suppressor proteins P53/p21, reduction in the expression of the oncogene c-Myc, and downregulation of cyclin D1 and cdk4. Additionally, AECHL-1 treatment resulted in the phosphorylation of p53 at serine 15 in B16F10 tumors, which seems to exhibit p53-dependent growth inhibitory responses.The present data demonstrate the activity of a triterpenoid AECHL-1 which possess a broad spectrum of activity against cancer cells. We propose here that AECHL-1 is a futuristic anti-cancer drug whose therapeutic potential needs to be widely explored for chemotherapy against cancer
Prevalence, genetic diversity and antiretroviral drugs resistance-associated mutations among untreated HIV-1-infected pregnant women in Gabon, central Africa
BACKGROUND: In Africa, the wide genetic diversity of HIV has resulted in
emergence of new strains, rapid spread of this virus in sub-Saharan populations
and therefore spread of the HIV epidemic throughout the continent.
METHODS: To determine the prevalence of antibodies to HIV among a high-risk
population in Gabon, 1098 and 2916 samples were collected from pregnant women in
2005 and 2008, respectively. HIV genotypes were evaluated in 107 HIV-1-positive
samples to determine the circulating subtypes of strains and their resistance to
antiretroviral drugs (ARVs).
RESULTS: The seroprevalences were 6.3% in 2005 and 6.0% in 2008. The main subtype
was recombinant CRF02_AG (46.7%), followed by the subtypes A (19.6%), G (10.3%),
F (4.7%), H (1.9%) and D (0.9%) and the complex recombinants CRF06_cpx (1.9%) and
CRF11_cpx (1.9%); 12.1% of subtypes could not be characterized. Analysis of ARVs
resistance to the protease and reverse transcriptase coding regions showed
mutations associated with extensive subtype polymorphism. In the present study,
the HIV strains showed reduced susceptibility to ARVs (2.8%), particularly to
protease inhibitors (1.9%) and nucleoside reverse transcriptase inhibitors
(0.9%).
CONCLUSIONS: The evolving genetic diversity of HIV calls for continuous
monitoring of its molecular epidemiology in Gabon and in other central African
countries
Multiple order-up-to policy for mitigating bullwhip effect in supply chain network
This paper proposes a multiple order-up-to policy based inventory replenishment scheme to mitigate the bullwhip effect in a multi-stage supply chain scenario, where various transportation modes are available between the supply chain (SC) participants. The proposed policy is similar to the fixed order-up-to policy approach where replenishment decision “how much to order” is made periodically on the basis of the predecided order-up-to inventory level. In the proposed policy, optimal multiple order-up-to levels are assigned to each SC participants, which provides decision making reference point for deciding the transportation related order quantity. Subsequently, a mathematical model is established to define optimal multiple order-up-to levels for each SC participants that aims to maximize overall profit from the SC network. In parallel, the model ensures the control over supply chain pipeline inventory, high satisfaction of customer demand and enables timely utilization of available transportation modes. Findings from the various numerical datasets including stochastic customer demand and lead times validate that—the proposed optimal multiple order-up-to policy based inventory replenishment scheme can be a viable alternative for mitigating the bullwhip effect and well-coordinated SC. Moreover, determining the multiple order-up-to levels is a NP hard combinatorial optimization problem. It is found that the implementation of new emerging optimization algorithm named bacterial foraging algorithm (BFA) has presented superior optimization performances. The robustness and applicability of the BFA algorithm are further validated statistically by employing the percentage heuristic gap and two-way ANOVA analysis

standard model, and most recent determinations from other experiments and of
comparable accuracy. We also report the first observation of the {\mathcal{B}}(B^0_s \to
