    Tissue engineering for total meniscal substitution : Animal study in sheep model

    Objective: The aim of the study was to investigate the use of a novel hyaluronic acid/polycaprolactone material for meniscal tissue engineering and to evaluate the tissue regeneration after the augmentation of the implant with expanded autologous chondrocytes. Two different surgical implantation techniques in a sheep model were evaluated. Methods: Twenty-four skeletally mature sheep were treated with total medial meniscus replacements, while two meniscectomies served as empty controls. The animals were divided into two groups: cell-free scaffold and scaffold seeded with autologous chondrocytes. Two different surgical techniques were compared: in 12 animals, the implant was sutured to the capsule and to the meniscal ligament; in the other 12 animals, also a transtibial fixation of the horns was used. The animals were euthanized after 4 months. The specimens were assessed by gross inspection and histology. Results: All implants showed excellent capsular ingrowth at the periphery. Macroscopically, no difference was observed between cell-seeded and cell-free groups. Better implant appearance and integrity was observed in the group without transosseous horns fixation. Using the latter implantation technique, lower joint degeneration was observed in the cell-seeded group with respect to cell-free implants. The histological analysis indicated cellular infiltration and vascularization throughout the implanted constructs. Cartilaginous tissue formation was significantly more frequent in the cell-seeded constructs. Conclusion: The current study supports the potential of a novel HYAFF/polycaprolactone scaffold for total meniscal substitution. Seeding of the scaffolds with autologous chondrocytes provides some benefit in the extent of fibrocartilaginous tissue repair

    Globally Anisotropic High Porosity Silica Aerogels

    We discuss two methods by which high porosity silica aerogels can be engineered to exhibit global anisotropy. First, anisotropy can be introduced with axial strain. In addition, intrinsic anisotropy can result during growth and drying stages and, suitably controlled, it can be correlated with preferential radial shrinkage in cylindrical samples. We have performed small angle X-ray scattering (SAXS) to characterize these two types of anisotropy. We show that global anisotropy originating from either strain or shrinkage leads to optical birefringence and that optical cross-polarization studies are a useful characterization of the uniformity of the imposed global anisotropy.Comment: 18 pages, 14 figures, submitted to Journal of Non-Crystalline Solid

    Theory of ac electrokinetic behavior of spheroidal cell suspensions with an intrinsic dispersion

    The dielectric dispersion, dielectrophoretic (DEP) and electrorotational (ER) spectra of spheroidal biological cell suspensions with an intrinsic dispersion in the constituent dielectric constants are investigated. By means of the spectral representation method, we express analytically the characteristic frequencies and dispersion strengths both for the effective dielectric constant and the Clausius-Mossotti factor (CMF). We identify four and six characteristic frequencies for the effective dielectric spectra and CMF respectively, all of them being dependent on the depolarization factor (or the cell shape). The analytical results allow us to examine the effects of the cell shape, the dispersion strength and the intrinsic frequency on the dielectric dispersion, DEP and ER spectra. Furthermore, we include the local-field effects due to the mutual interactions between cells in a dense suspension, and study the dependence of co-field or anti-field dispersion peaks on the volume fractions.Comment: accepted by Phys.     Differentiating normal and problem gambling: a grounded theory approach.

    A previous study (Ricketts &amp; Macaskill, 2003) delineated a theory of problem gambling based on the experiences of treatment seeking male gamblers and allowed predictions to be made regarding the processes that differentiate between normal and problem gamblers. These predictions are the focus of the present study, which also utilised a grounded theory approach, but with a sample of male high frequency normal gamblers. The findings suggest that there are common aspects of gambling associated with arousal and a sense of achievement. The use of gambling to manage negative emotional states differentiated normal and problem gambling. Perceived self-efficacy , emotion management skills and perceived likelihood of winning money back were intervening variables differentiating problem and normal gamblers.</p

    A 750 mW, continuous-wave, solid-state laser source at 313 nm for cooling and manipulating trapped 9Be+ ions

    We present a solid-state laser system that generates 750 mW of continuous-wave single-frequency output at 313 nm. Sum-frequency generation with fiber lasers at 1550 nm and 1051 nm produces up to 2 W at 626 nm. This visible light is then converted to UV by cavity-enhanced second-harmonic generation. The laser output can be tuned over a 495 GHz range, which includes the 9Be+ laser cooling and repumping transitions. This is the first report of a narrow-linewidth laser system with sufficient power to perform fault-tolerant quantum-gate operations with trapped 9Be+ ions by use of stimulated Raman transitions.Comment: 9 pages, 4 figure

    The littoral sea cucumbers (Echinodermata: Holothuroidea) of Guam re-assessed - a diversity curve that still does not asymptote

    The Micronesian island of Guam has been an important site for the study of littoral tropical holothuriantaxonomy for almost 200 years. Despite substantial attention by both expeditions and resident taxonomists, new records arestill regularly added to the fauna, demonstrating the challenge of documenting even such large and well-known animals ina small hyper-diverse area. Guam is the type locality of species described by Quoy & Gaimard (1833) and Brandt (1835).A survey of the sea cucumber fauna by Rowe & Doty (1977) led to one of the most used guides for the identification oftropical Pacific sea cucumbers because of the color illustrations of living animals it presented. Focus on echinodermsincluding holothurians continued with numerous new records added in the following decades. Paulay (2003a) summarizedthe fauna last, recording 46-47 species. At this stage the fauna was thought to be well documented. A week-long workshopon holothurian systematics sponsored by the National Science Foundation PEET (Partnerships for Enhancing Expertise inTaxonomy) project in 2010 included a substantial field work component, sampling both during the day and night, withsnorkeling and SCUBA, across a variety of habitats. This survey yielded 40 species, including numerous new records andeven species. Further sampling by Kerr’s lab since the workshop has added additional records. The littoral holothuroidfauna of Guam now comprises 65 species in 17 genera and 7 families. Half of the 19 newly recorded species are the resultof unravelling cryptic species in complexes, the other half are based on new collections. Eleven species are known fromsingle specimens, suggesting that much still remains to be learned about the fauna

    Equilibrium and dynamical properties of two dimensional self-gravitating systems

    A system of N classical particles in a 2D periodic cell interacting via long-range attractive potential is studied. For low energy density UU a collapsed phase is identified, while in the high energy limit the particles are homogeneously distributed. A phase transition from the collapsed to the homogeneous state occurs at critical energy U_c. A theoretical analysis within the canonical ensemble identifies such a transition as first order. But microcanonical simulations reveal a negative specific heat regime near UcU_c. The dynamical behaviour of the system is affected by this transition : below U_c anomalous diffusion is observed, while for U > U_c the motion of the particles is almost ballistic. In the collapsed phase, finite NN-effects act like a noise source of variance O(1/N), that restores normal diffusion on a time scale diverging with N. As a consequence, the asymptotic diffusion coefficient will also diverge algebraically with N and superdiffusion will be observable at any time in the limit N \to \infty. A Lyapunov analysis reveals that for U > U_c the maximal exponent \lambda decreases proportionally to N^{-1/3} and vanishes in the mean-field limit.
