    Dynamically Programmable Magnetic Fields for Controlled Movement of Cells Loaded with Iron Oxide Nanoparticles

    Cell-based therapies are becoming increasingly prominent in numerous medical contexts, particularly in regenerative medicine and the treatment of cancer. However, since the efficacy of the therapy is largely dependent on the concentration of therapeutic cells at the treatment area, a major challenge associated with cell-based therapies is the ability to move and localize therapeutic cells within the body. In this article, a technique based on dynamically programmable magnetic fields is successfully demonstrated to noninvasively aggregate therapeutic cells at a desired location. Various types of therapeutically relevant cells (neural stem cells, monocytes/macrophages, and chimeric antigen receptor T cells) are loaded with iron oxide nanoparticles and then focused at a particular site using externally controlled electromagnets. These experimental results serve as a readily scalable prototype for designing an apparatus that patients can wear to focus therapeutic cells at the anatomical sites needed for treatment

    Strangers in the Room: Unpacking Perceptions of 'Smartness' and Related Ethical Concerns in the Home

    The increasingly widespread use of 'smart' devices has raised multifarious ethical concerns regarding their use in domestic spaces. Previous work examining such ethical dimensions has typically either involved empirical studies of concerns raised by specific devices and use contexts, or alternatively expounded on abstract concepts like autonomy, privacy or trust in relation to 'smart homes' in general. This paper attempts to bridge these approaches by asking what features of smart devices users consider as rendering them 'smart' and how these relate to ethical concerns. Through a multimethod investigation including surveys with smart device users (n=120) and semi-structured interviews (n=15), we identify and describe eight types of smartness and explore how they engender a variety of ethical concerns including privacy, autonomy, and disruption of the social order. We argue that this middle ground, between concerns arising from particular devices and more abstract ethical concepts, can better anticipate potential ethical concerns regarding smart devices.Comment: 10 pages, 1 figure.     Cognitive and mood functioning in borderline and schizotypal personality disorders

    Research suggests many shared clinical features across individuals with Schizotypal Personality Disorder (SPD) and Borderline Personality Disorder (BPD), including problems with attention/ executive functioning and mood. Therefore, aspects of these areas of functioning were compared in SPD and BPD to better characterize their respective difficulties. BPD, SPD, and healthy control (HC) participants were administered measures of cognitive and mood functioning. Compared with healthy controls, SPD patients performed significantly worse on aspects of the Delayed-Matching- to-Sample task, a measure of short-term visual memory abilities; however, the individuals with BPD did not differ from healthy controls. Neither of the patient groups differed from HC’s on measures of processing speed or planning. With regard to mood functioning, the BPD group exhibited significantly higher levels of affective disturbance (e.g., sadness, fear, anger) compared with the SPD patients and HCs. Overall, findings suggest different patterns of fronto-subcortical weakness in each patient group. While SPD patients exhibited relative weakness with short-term memory, BPD patient performance on such measures did not reveal relative weakness compared with HCs but did implicate problems with mood

    An original phylogenetic approach identified mitochondrial haplogroup T1a1 as inversely associated with breast cancer risk in BRCA2 mutation carriers

    Introduction: Individuals carrying pathogenic mutations in the BRCA1 and BRCA2 genes have a high lifetime risk of breast cancer. BRCA1 and BRCA2 are involved in DNA double-strand break repair, DNA alterations that can be caused by exposure to reactive oxygen species, a main source of which are mitochondria. Mitochondrial genome variations affect electron transport chain efficiency and reactive oxygen species production. Individuals with different mitochondrial haplogroups differ in their metabolism and sensitivity to oxidative stress. Variability in mitochondrial genetic background can alter reactive oxygen species production, leading to cancer risk. In the present study, we tested the hypothesis that mitochondrial haplogroups modify breast cancer risk in BRCA1/2 mutation carriers. Methods: We genotyped 22,214 (11,421 affected, 10,793 unaffected) mutation carriers belonging to the Consortium of Investigators of Modifiers of BRCA1/2 for 129 mitochondrial polymorphisms using the iCOGS array. Haplogroup inference and association detection were performed using a phylogenetic approach. ALTree was applied to explore the reference mitochondrial evolutionary tree and detect subclades enriched in affected or unaffected individuals. Results: We discovered that subclade T1a1 was depleted in affected BRCA2 mutation carriers compared with the rest of clade T (hazard ratio (HR) = 0.55; 95% confidence interval (CI), 0.34 to 0.88; P = 0.01). Compared with the most frequent haplogroup in the general population (that is, H and T clades), the T1a1 haplogroup has a HR of 0.62 (95% CI, 0.40 to 0.95; P = 0.03). We also identified three potential susceptibility loci, including G13708A/rs28359178, which has demonstrated an inverse association with familial breast cancer risk. Conclusions: This study illustrates how original approaches such as the phylogeny-based method we used can empower classical molecular epidemiological studies aimed at identifying association or risk modification effects.Peer reviewe

    The Hellenistic and early Roman common-ware pottery from Tel Anafa.

    In this dissertation the corpus of common-ware pottery from the site of Tel Anafa, in Israel's Upper Galilee, is presented. This material, representing the basic kitchen equipment of the site's inhabitants, comes from three main periods of occupation: mid 3rd c. BC (early Hellenistic), late 2nd-early 1st c. BC (late Hellenistic) and early 1st c. AD (early Roman). The pottery has been analyzed and organized by both fabric and shape, leading to an initial broad distinction between vessels intended for table, kitchen or storage use. Within these groups the corpus is presented by shape, and includes an overview of the origin and development of the form, its specific regional and chronological range and its history at Anafa. A close analysis of the assemblage in its entirety has shown that common-ware pottery can provide information on historical, economic and social aspects of ancient societies. For Tel Anafa, the ceramic corpus has shown that in the early Hellenistic period the inhabitants were linked economically to the southern Phoenician coast. That area's prosperity was not shared by the inhabitants of Anafa, who instead lived the most marginal of lifestyles. In contrast, the late Hellenistic period was a time of great wealth, with the inhabitants building and decorating a large peristyle court house, similar to the villas on Delos. The pottery shows clearly that these cosmopolitan residents were Phoenicians, probably from Tyre or Sidon, and that their culinary patterns were essentially Greek. A gap in occupation throughout much of the 1st c. BC ended with a re-occupation in the early 1st c. AD. The introduction of several new common-ware shapes reveals these new occupants to be of Italic origin, while the dramatically increased reliance on vessels imported from the lower Galilee shows that the site's economic affiliation shifted southward, perhaps in response to Roman tolls between Syria and Judaea.Ph.D.ArchaeologyUniversity of Michiganhttp://deepblue.lib.umich.edu/bitstream/2027.42/161771/1/8812857.pd

    Relating health policy to women's health outcomes

    Individuals' social and economic circumstances, including socioeconomic status and medical care availability, are central to health outcomes, particularly for women. These factors are often mediated by governmental policies. This exploratory study found associations between women's health outcomes and state-level policies related to women's health. Outcomes were mortality rates for four leading causes of death for women in the US (heart disease, stroke, lung cancer, and breast cancer), infant mortality, and a mental health outcome variable. State policies on key women's health issues were evaluated on the degree to which they adequately protected women's health. Our regression models accounted for significant variance in mortality rates and substantial variance in the mental health outcome. Policies affecting access to care (Medicaid eligibility and efforts to expand Medicaid) and community (environmental health tracking and violence against women) were significantly associated with mortality outcomes.     Interpersonal Pathoplasticity in Individuals With Generalized Anxiety Disorder

    Recent theories of generalized anxiety disorder (GAD) have emphasized interpersonal and personality functioning as important aspects of the disorder. We examined heterogeneity in interpersonal problems in 2 studies of individuals with GAD (n = 47 and n = 83). Interpersonal subtypes were assessed with the Inventory of Interpersonal Problems-Circumplex (Alden, Wiggins, & Pincus, 1990). Across both studies, individuals with GAD exhibited heterogeneous interpersonal problems, and cluster analyses of these patients\u27 interpersonal characteristics yielded 4 replicable clusters, identified as intrusive, exploitable, cold, and nonassertive subtypes. Consistent with our pathoplasticity hypotheses, clusters did not differ with GAD severity, anxiety severity, or depression severity. Clusters in Study 2 differed on rates of personality disorders, including avoidant personality disorder, further providing support for the validity of interpersonal subtypes. The presence of interpersonal subtypes in GAD may have important implications for treatment planning and efficacy. © 2011 American Psychological Association
