    Nano-scale corrosion mechanism of T91 steel in static lead-bismuth eutectic: a combined APT, EBSD, and STEM investigation

    T91 steel is a candidate material for structural components in lead-bismuth-eutectic (LBE) cooled systems, for example fast reactors and solar power plants [1]. However, the corrosion mechanisms of T91 in LBE remain poorly understood. In this study, we have analysed the static corrosion of T91 in liquid LBE using a range of characterisation techniques at increasingly smaller scales. A unique pattern of liquid metal intrusion was observed that does not appear to correlate with the grain boundary network. Upon closer inspection, electron backscatter diffraction (EBSD) reveals a change in the morphology of grains at the LBE-exposed surface, suggesting a local phase transition. Energy dispersive X-ray (EDX) maps show that Cr is depleted in the T91 material near the LBE interface. Furthermore, we observed the dissolution of all Cr-enriched precipitates in this region. Although the corrosion is conducted in an oxygen deficient environment, both scanning transmission electron microscopy (STEM) and atom probe tomography (APT) reveal a thin surface oxide layer (presumably wüstite) at the LBE-steel interface. Using electron energy loss spectroscopy (EELS) in the STEM, as well as APT, the atomic scale elemental redistribution and 3D morphology of the corrosion interface is investigated. By combining results from these different techniques, several types of oxide phases and structures can be identified. Based on this detailed nano-scale information, we propose potential mechanisms of T91 corrosion in LBE

    Examination of Acid-Fast Bacilli in Sputum Using Modified Light Microscope with Homemade Light Emitting Diode Additional Attachment

    Typical clinical symptoms and chest X-ray is a marker of Tuberculosis (TB) sufferers. However, the diagnosis of TB in adults should be supported by microscopic examination. Currently, Bacilli microscopic examination of acid-fast bacilli (AFB) in sputum by Ziehl-Neelsen (ZN) coloring is the most widely used. However, for reasons of convenience, especially for laboratories with a considerable amount of smear samples, and due to higher sensitivity compared with ZN staining, the World Health Organization (WHO) has recommended the use of auramine-O-staining (fluorochrome  staining), which is visualized by light emitting diode (LED) fluorescence microscopy. The aim of this study was to evaluate the performance of modified light microscope with homemade LED additional attachment for examination of AFB in sputum using auramine-O-staining method. We compared the sensitivity and specificity of 2 kinds of AFB in sputum methods: ZN and fluorochrome, using culture on Lowenstein-Jensen media as the gold standard. The results showed auramine-O-staining gives more proportion of positive findings (81%) compared to the ZN method (70%). These results demonstrated that the sensitivity of auramine-O-staining was higher than ZN, however it gives more potential false positive results than ZN. The sensitivity of auramine-O-staining in detecting AFB in sputum was 100% while the specificity was 88%

    Subdiffusion and weak ergodicity breaking in the presence of a reactive boundary

    We derive the boundary condition for a subdiffusive particle interacting with a reactive boundary with finite reaction rate. Molecular crowding conditions, that are found to cause subdiffusion of larger molecules in biological cells, are shown to effect long-tailed distributions with identical exponent for both the unbinding times from the boundary to the bulk and the rebinding times from the bulk. This causes a weak ergodicity breaking: typically, an individual particle either stays bound or remains in the bulk for very long times.     US GLOBEC: Program Goals, Approaches, and Advances

    This special issue summarizes the major achievements of the US Global Ocean Ecosystem Dynamics (GLOBEC) program and celebrates its accomplishments. The articles grew out of a final symposium held in October 2009 under the auspices of the National Academy of Sciences Ocean Studies Board (http://usglobec.org/Symposium). This special issue updates the US GLOBEC "mid-life" Oceanography issue (Vol. 15, No. 2, 2002, http://tos.org/oceanography/archive/15-2.html), which put forward many of the goals and activities of the program, but was published while field work was still being conducted and results had yet to be synthesized across regional programs. The present special issue highlights the advances in understanding achieved through the synthesis of regional studies and pan-regional comparisons

    Circulating resistin levels and risk of multiple myeloma in three prospective cohorts

    BACKGROUND: Resistin is a polypeptide hormone secreted by adipose tissue. A prior hospital-based case-control study reported serum resistin levels to be inversely associated with risk of multiple myeloma (MM). To date, this association has not been investigated prospectively. METHODS: We measured resistin concentrations for pre-diagnosis peripheral blood samples from 178 MM cases and 358 individually matched controls from three cohorts participating in the MM cohort consortium. RESULTS: In overall analyses, higher resistin levels were weakly associated with reduced MM risk. For men, we observed a statistically significant inverse association between resistin levels and MM (odds ratio, 0.44; 95% confidence interval (CI) 0.24-0.83 and 0.54; 95% CI 0.29-0.99, for the third and fourth quartiles, respectively, vs the lowest quartile; Ptrend=0.03). No association was observed for women. CONCLUSIONS: This study provides the first prospective evidence that low circulating resistin levels may be associated with an increased risk of MM, particularly for men

    Physiological concentrations of bile acids down-regulate agonist induced secretion in colonic epithelial cells

    In patients with bile acid malabsorption, high concentrations of bile acids enter the colon and stimulate Cl− and fluid secretion, thereby causing diarrhoea. However, deoxycholic acid (DCA), the predominant colonic bile acid, is normally present at lower concentrations where its role in regulating transport is unclear. Thus, the current study set out to investigate the effects of physiologically relevant DCA concentrations on colonic epithelial secretory function. Cl− secretion was measured as changes in short-circuit current across voltage-clamped T84 cell monolayers. At high concentrations (0.5–1 mM), DCA acutely stimulated Cl− secretion but this effect was associated with cell injury, as evidenced by decreased transepithelial resistance (TER) and increased lactate dehydrogenase (LDH) release. In contrast, chronic (24 hrs) exposure to lower DCA concentrations (10–200 μM) inhibited responses to Ca2+ and cAMP-dependent secretagogues without altering TER, LDH release, or secretagogue-induced increases in intracellular second messengers. Other bile acids – taurodeoxycholic acid, chenodeoxycholic acid and cholic acid – had similar antisecretory effects. DCA (50 μM) rapidly stimulated phosphorylation of the epidermal growth factor receptor (EGFr) and both ERK and p38 MAPKs (mitogen-activated protein kinases). The EGFr inhibitor, AG1478, and the protein synthesis inhibitor, cycloheximide, reversed the antisecretory effects of DCA, while the MAPK inhibitors, PD98059 and SB203580, did not. In summary, our studies suggest that, in contrast to its acute prosecretory effects at pathophysiological concentrations, lower, physiologically relevant, levels of DCA chronically down-regulate colonic epithelial secretory function. On the basis of these data, we propose a novel role for bile acids as physiological regulators of colonic secretory capacity

    Injectable citrate-modified Portland cement for use in vertebroplasty

    Abstract: The injectability of Portland cement (PC) with several citrate additives was investigated for use in clinical applications such as vertebroplasty (stabilization of a fractured vertebra with bone cement) using a syringe. A 2-wt % addition of sodium or potassium citrate with PC significantly improved cement inject-ability, decreased cement setting times from over 2 h to below 25 min, while increasing the compressive strength to a maxi-mum of 125 MPa. Zeta-potential measurements indicated that the citrate anion was binding to one or more of the positively charged species causing charged repulsion between cement particles which dispersed aggregates and caused the liquefying effect of the anion. Analysis of the hydrating phases of PC indi-cated that the early strength producing PC phase (ettringite) developed within the first 2 h of setting following addition of the citrate anion, while this did not occur in the control cement (PC only). Within 24 h ettringite developed in PC as well as calcium– silicate–hydrate (C–S–H), the major setting phase of PC, whereas cements containing citrate did not develop this phase. The evi-dence suggested that in the presence of citrate the cements lim-ited water supply appeared to be utilized for ettringite formation, producing the early strength of the citrate cements. The present study has demonstrated that it is possible to modify PC with citrate to both improve the injectability and crucially reduce the setting times of PC while improving the strength of the cement

    Non-contact, non-destructive mapping of thermal diffusivity and surface acoustic wave speed using transient grating spectroscopy

    We present new developments of the laser-induced transient grating spectroscopy (TGS) technique that enable the measurement of large area 2D maps of thermal diffusivity and surface acoustic wave speed. Additional capabilities include targeted measurements and the ability to accommodate samples with increased surface roughness. These new capabilities are demonstrated by recording large TGS maps of deuterium implanted tungsten, linear friction welded aerospace alloys and high entropy alloys with a range of grain sizes. The results illustrate the ability to view grain microstructure in elastically anisotropic samples, and to detect anomalies in samples, for example due to irradiation and previous measurements. They also point to the possibility of using TGS to quantify grain size at the surface of polycrystalline materials.Comment: The following article has been submitted to Review of Scientific Instruments.     Comparative venom-gland transcriptomics and venom proteomics of four Sidewinder Rattlesnake (\u3ci\u3eCrotalus cerastes\u3c/i\u3e) lineages reveal little differential expression despite individual variation

    Changes in gene expression can rapidly influence adaptive traits in the early stages of lineage diversification. Venom is an adaptive trait comprised of numerous toxins used for prey capture and defense. Snake venoms can vary widely between conspecific populations, but the influence of lineage diversification on such compositional differences are unknown. To explore venom differentiation in the early stages of lineage diversification, we used RNA-seq and mass spectrometry to characterize Sidewinder Rattlesnake (Crotalus cerastes) venom. We generated the first venom-gland transcriptomes and complementary venom proteomes for eight individuals collected across the United States and tested for expression differences across life history traits and between subspecific, mitochondrial, and phylotranscriptomic hypotheses. Sidewinder venom was comprised primarily of hemorrhagic toxins, with few cases of differential expression attributable to life history or lineage hypotheses. However, phylotranscriptomic lineage comparisons more than doubled instances of significant expression differences compared to all other factors. Nevertheless, only 6.4% of toxins were differentially expressed overall, suggesting that shallow divergence has not led to major changes in Sidewinder venom composition. Our results demonstrate the need for consensus venom-gland transcriptomes based on multiple individuals and highlight the potential for discrepancies in differential expression between different phylogenetic hypotheses
