    Régime alimentaire de Rupicola rupicola (Cotingidae) en Guyane française : relations avec la frugivorie et la zoochorie

    The diet of the Guianan Cock-of-the-Rock (Rupicola rupicola) was studied in a pristine lowland rainforest, near an inselberg in French Guiana. Droppings were collected under a nest and observations were conducted on a radio-tagged female. Nestlings were mostly fed fruit, but not exclusively so, as insects, small snakes, and lizards were also delivered to the chicks. The characteristics of the fruits consumed, and of the fruiting plants, were given special attention. The fruit diet is highly diversified : 65 species belonging to 31 families were recorded as ingested in a little more than one month. Remains of fifty-two species belonging to 26 families were identified under the nest itself. A quantification by feeding units (representing what the bird actually takes on the plant) shows that three quarters of the diet were contributed by only a quarter of the fruit species. Two thirds of the feeding units were contributed by only four plant families. Half the number of fruit species and two thirds of the feeding units were drupes, generally 10 to 20 mm in breadth, with seeds 5-1 5 mm wide. 83 % of the fruits were uniformly coloured. Taken together, black and red fruits were three times more abundant than all the others. The pulp of more than two thirds of the fruits amounted to at least three fifths of the fruit itself. A large proportion of the fruits consumed were those of canopy trees and lianas. More than three quarters of the fruits were from plants with large simultaneous crop production. These fruits were plucked out on the wing by the adult female which patroled a home range of 9.7 ha when feeding her brood, going from one fruiting tree to another. These results are compared with the scanty littérature data on the diet of the two Rupicola species. Cocks-of-the-rocks cannot be considered as specialized frugivores, sensu Snow (1981).     Increased Asymmetric and Multi-Daughter Cell Division in Mechanically Confined Microenvironments

    As the microenvironment of a cell changes, associated mechanical cues may lead to changes in biochemical signaling and inherently mechanical processes such as mitosis. Here we explore the effects of confined mechanical environments on cellular responses during mitosis. Previously, effects of mechanical confinement have been difficult to optically observe in three-dimensional and in vivo systems. To address this challenge, we present a novel microfluidic perfusion culture system that allows controllable variation in the level of confinement in a single axis allowing observation of cell growth and division at the single-cell level. The device is capable of creating precise confinement conditions in the vertical direction varying from high (3 µm) to low (7 µm) confinement while also varying the substrate stiffness (E = 130 kPa and 1 MPa). The Human cervical carcinoma (HeLa) model with a known 3N+ karyotype was used for this study. For this cell line, we observe that mechanically confined cell cycles resulted in stressed cell divisions: (i) delayed mitosis, (ii) multi- daughter mitosis events (from 3 up to 5 daughter cells), (iii) unevenly sized daughter cells, and (iv) induction of cell death. In the highest confined conditions, the frequency of divisions producing more than two progeny was increased an astounding 50-fold from unconfined environments, representing about one half of all successful mitotic events. Notably, the majority of daughter cells resulting from multipolar divisions were viable after cytokinesis and, perhaps suggesting another regulatory checkpoint in the cell cycle, were in some cases observed to re-fuse with neighboring cells post-cytokinesis. The higher instances of abnormal mitosis that we report in confined mechanically stiff spaces, may lead to increased rates of abnormal, viable, cells in the population. This work provides support to a hypothesis that environmental mechanical cues influences structural mechanisms of mitosis such as geometric orientation of the mitotic plane or planes

    Neanderthal firewood management: Evidence from Stratigraphic Unit IV of Abric del Pastor (Eastern Iberia)

    This paper presents anthracological data from Abric del Pastor (Alcoi, Spain), a Middle Paleolithic rock shelter site. Analysis of 1077 wood charcoal remains from Stratigraphic Unit IV (S.U. IV), collected within archaeological combustion structures and from loose sediment outside of structures, allowed us to characterise the local landscape, as well as to approach the interaction between Neanderthal groups and their local environment. Taxonomic identification suggests that firewood was gathered from nearby sources, with predominance of juniper (Juniperus sp.) followed by thermophilous shrubby taxa. Additional analysis focussing on post-depositional processes affecting charcoal have shown features indicative of biodegradation and mechanical action. The results of this study contribute significant anthracological data towards our understanding of Late Pleistocene Mediterranean landscapes and Neanderthal forest management in this region

    Minimal information for studies of extracellular vesicles 2018 (MISEV2018): a position statement of the International Society for Extracellular Vesicles and update of the MISEV2014 guidelines

    El artículo incluye más de 400 autoresThe last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities.     UV wavelengths experienced during development affect larval newt visual sensitivity and predation efficiency

    We experimentally investigated the influence of developmental plasticity of ultraviolet (UV) visual sensitivity on predation efficiency of the larval smooth newt, Lissotriton vulgaris. We quantified expression of SWS1 opsin gene (UV-sensitive protein of photoreceptor cells) in the retinas of individuals who had developed in the presence (UV+) or absence (UV-) of UV light (developmental treatments), and tested their predation efficiency under UV+ and UV- light (testing treatments). We found that both SWS1 opsin expression and predation efficiency were significantly reduced in the UV- developmental group. Larvae in the UV- testing environment displayed consistently lower predation efficiency regardless of their developmental treatment. These results prove for the first time, we believe, functional UV vision and developmental plasticity of UV sensitivity in an amphibian at the larval stage. They also demonstrate that UV wavelengths enhance predation efficiency and suggest that the magnitude of the behavioural response depends on retinal properties induced by the developmental lighting environment

    A Machine Checked Model of Idempotent MGU Axioms For Lists of Equational Constraints

    We present formalized proofs verifying that the first-order unification algorithm defined over lists of satisfiable constraints generates a most general unifier (MGU), which also happens to be idempotent. All of our proofs have been formalized in the Coq theorem prover. Our proofs show that finite maps produced by the unification algorithm provide a model of the axioms characterizing idempotent MGUs of lists of constraints. The axioms that serve as the basis for our verification are derived from a standard set by extending them to lists of constraints. For us, constraints are equalities between terms in the language of simple types. Substitutions are formally modeled as finite maps using the Coq library Coq.FSets.FMapInterface.     Thermoresponsive Micropatterned Substrates for Single Cell Studies

    We describe the design of micropatterned surfaces for single cell studies, based on thermoresponsive polymer brushes. We show that brushes made of poly(N-isopropylacrylamide) grafted at high surface density display excellent protein and cell anti-adhesive properties. Such brushes are readily patterned at the micron scale via deep UV photolithography. A proper choice of the adhesive pattern shapes, combined with the temperature-dependent swelling properties of PNIPAM, allow us to use the polymer brush as a microactuator which induces cell detachment when the temperature is reduced below C

    MFGE8 does not influence chorio-retinal homeostasis or choroidal neovascularization in vivo

    Purpose: Milk fat globule-epidermal growth factor-factor VIII (MFGE8) is necessary for diurnal outer segment phagocytosis and promotes VEGF-dependent neovascularization. The prevalence of two single nucleotide polymorphisms (SNP) in MFGE8 was studied in two exsudative or “wet” Age-related Macular Degeneration (AMD) groups and two corresponding control groups. We studied the effect of MFGE8 deficiency on retinal homeostasis with age and on choroidal neovascularization (CNV) in mice. Methods: The distribution of the SNP (rs4945 and rs1878326) of MFGE8 was analyzed in two groups of patients with “wet” AMD and their age-matched controls from Germany and France. MFGE8-expressing cells were identified in Mfge8+/− mice expressing ß-galactosidase. Aged Mfge8+/− and Mfge8−/− mice were studied by funduscopy, histology, electron microscopy, scanning electron microscopy of vascular corrosion casts of the choroid, and after laser-induced CNV. Results: rs1878326 was associated with AMD in the French and German group. The Mfge8 promoter is highly active in photoreceptors but not in retinal pigment epithelium cells. Mfge8−/− mice did not differ from controls in terms of fundus appearance, photoreceptor cell layers, choroidal architecture or laser-induced CNV. In contrast, the Bruch's membrane (BM) was slightly but significantly thicker in Mfge8−/− mice as compared to controls. Conclusions: Despite a reproducible minor increase of rs1878326 in AMD patients and a very modest increase in BM in Mfge8−/− mice, our data suggests that MFGE8 dysfunction does not play a critical role in the pathogenesis of AMD
