    Health and Safety Issues and Perceptions of Forest Harvesting Contractors in Ireland

    The aim of this study was to examine the extent and severity of accidents and long-term health problems, and to evaluate perceptions of health and safety issues among forest harvesting contractors and sub-contractors in Ireland. A postal questionnaire was distributed to 450 forest harvesting (sub)contractors with the co-operation of the Irish Forestry Contractors Association (IFCA) and Coillte Teoranta (the State Forestry Company, referred to as Coillte). The questionnaire was completed and returned by 61 respondents. The majority of these were chainsaw, harvester and forwarder operators. The occurrence of an accident (requiring medical attention) in the last three years was reported by two respondents, while long-term health problems were reported by 23 (40%) of the respondents. Results showed that long-term health problems were not significantly correlated to age or training level. The two work elements in forest harvesting operations that were found to be most hazardous were operating a chainsaw and machine maintenance. When asked about the perceived dangers in the industry and the obstacles to improvements, 59% of the respondents identified harvesting work as dangerous, while 62% described financial pressure as the main barrier to improvements in safety, followed by the pressure of work (39%) and a lack of adequate training (36%). Suggestions on ways to improve the overall health and safety situation within forest harvesting included improved training and safety education (45%) and reduced work pressure (36%)

    Differential Regulation of Primitive Myelopoiesis in the Zebrafish by Spi-1/Pu.1 and C/ebp1

    The zebrafish has become a powerful tool for analysis of vertebrate hematopoiesis. Zebrafish, unlike mammals, have a robust primitive myeloid pathway that generates both granulocytes and macrophages. It is not clear how this unique primitive myeloid pathway relates to mammalian definitive hematopoiesis. In this study, we show that the two myeloid subsets can be distinguished using RNA in situ hybridization. Using a morpholino-antisense gene knockdown approach, we have characterized the hematopoietic defects resulting from knockdown of the myeloid transcription factor gene pu.1 and the unique zebrafish gene c/ebp1. Severe reduction of pu.1 resulted in complete loss of primitive macrophage development, with effects on granulocyte development only with maximal knockdown. Reduction of c/ebp1 did not ablate initial macrophage or granulocyte development, but resulted in loss of expression of the secondary granule gene lys C. These data reveal strong functional conservation of pu.1 between zebrafish primitive myelopoiesis and mammalian definitive myelopoiesis. Further, these results are consistent with a conserved role between c/ebp1 and mammalian C/EBPE, whose ortholog in zebrafish has not been identified.     Hair cortisol in the perinatal period mediates associations between maternal adversity and disrupted maternal interaction in early infancy

    Existing literature points to the possibility that cortisol could be one link between maternal adversity and poorer parenting quality, but most studies have examined salivary cortisol concentrations rather than hair cortisol concentrations. The current study examined hair cortisol concentration (HCC) during the third trimester of pregnancy as a mediator between maternal adversity indicators (childhood abuse, severe mental illness, symptomatic functioning) and maternal caregiving behavior at 4 months postpartum. Forty‐four women participated in the study: 30 with severe mental disorders, and 14 nonclinical controls. HCC was assessed during the third trimester of pregnancy (HCC‐P) and at 4 months postpartum (HCC‐4M). Sexual, physical, and emotional abuse were assessed by the Adverse Childhood Experiences Study Questionnaire. Maternal disrupted interaction was reliably coded from mother–infant video interactions during a Still‐Face Procedure. Mediation models indicated that maternal HCC‐P and HCC‐4M mediated associations between maternal psychopathology (severe mental illness, symptomatic functioning) and maternal disrupted interaction at 4 months. Maternal HCC at 4 months also mediated associations between experienced childhood abuse and overall disrupted interaction. Our findings indicate that HCC may be a potential early biomarker for future caregiving challenges among mothers with severe mental illness and histories of childhood abuse

    N6-Methyladenosine Inhibits Local Ribonucleolytic Cleavage to Stabilize mRNAs in Arabidopsis

    N6-methyladenosine (m6A) is a dynamic, reversible, covalently modified ribonucleotide that occurs predominantly toward 30 ends of eukaryotic mRNAs and is essential for their proper function and regulation. In Arabidopsis thaliana, many RNAs contain at least one m6A site, yet the transcriptome-wide function of m6A remains mostly unknown. Here, we show that manym6A-modified mRNAs in Arabidopsis have reduced abundance in the absence of this mark. The decrease in abundance is due to transcript destabilization caused by cleavage occurring 4 or 5 nt directly upstream of unmodified m6A sites. Importantly, we also find that, upon agriculturally relevant salt treatment, m6A is dynamically deposited on and stabilizes transcripts encoding proteins required for salt and osmotic stress response. Overall, our findings reveal that m6A generally acts as a stabilizing mark through inhibition of site-specific cleavage in plant transcriptomes, and this mechanism is required for proper regulation of the salt-stress-responsive transcriptome

Understanding the functioning and organisation of social work within various country contexts is critical in order to formulate knowledge around its overall impact, successes and challenges, allowing social workers to learn from one another and build professional consolidation. The profession is mapped out in 10 countries, reflecting on its structure, identity and development. Although the profession is developing globally, it is also experiencing significant challenges. Key insights, conclusions and recommendations for future research are presented

    Atypical B cells and impaired SARS-CoV-2 neutralization following heterologous vaccination in the elderly

    Suboptimal responses to a primary vaccination course have been reported in the elderly, but there is little information regarding the impact of age on responses to booster third doses. Here, we show that individuals 70 years or older (median age 73, range 70-75) who received a primary two-dose schedule with AZD1222 and booster third dose with mRNA vaccine achieve significantly lower neutralizing antibody responses against SARS-CoV-2 spike pseudotyped virus compared with those younger than 70 (median age 66, range 54-69) at 1 month post booster. Impaired neutralization potency and breadth post third dose in the elderly is associated with circulating "atypical" spike-specific B cells expressing CD11c and FCRL5. However, when considering individuals who received three doses of mRNA vaccine, we did not observe differences in neutralization or enrichment in atypical B cells.     Exploring urban metabolism—Towards an interdisciplinary perspective

However, in the anthropocene human forces such as industrialization and urbanization are mobilizing people, goods and information at an increasing pace and as such have a large impact on urban energy and material flows. In this white paper, we develop a combined natural and social science perspective on urban metabolism. More specifically, innovative conceptual and methodological interdisciplinary approaches are identified and discussed to enhance the understanding of the forces that shape urban metabolism, and how these forces affect urban living and the environment. A challenging research agenda on urban metabolism is also presented
