Proteomic study of the membrane components of signalling cascades of Botrytis cinerea controlled by phosphorylation

Protein phosphorylation and membrane proteins play an important role in the infection of plants by phytopathogenic fungi, given their involvement in signal transduction cascades. Botrytis cinerea is a well-studied necrotrophic fungus taken as a model organism in fungal plant pathology, given its broad host range and adverse economic impact. To elucidate relevant events during infection, several proteomics analyses have been performed in B. cinerea, but they cover only 10% of the total proteins predicted in the genome database of this fungus. To increase coverage, we analysed by LC-MS/MS the first-reported overlapped proteome in phytopathogenic fungi, the “phosphomembranome” of B. cinerea, combining the two most important signal transduction subproteomes. Of the 1112 membrane-associated phosphoproteins identified, 64 and 243 were classified as exclusively identified or overexpressed under glucose and deproteinized tomato cell wall conditions, respectively. Seven proteins were found under both conditions, but these presented a specific phosphorylation pattern, so they were considered as exclusively identified or overexpressed proteins. From bioinformatics analysis, those differences in the membrane-associated phosphoproteins composition were associated with various processes, including pyruvate metabolism, unfolded protein response, oxidative stress response, autophagy and cell death. Social Laboratory: Innovation and Experimentation for a Formal and Inclusive Economy

El libro tuvo como objeto definir el diseño y la estructura del modelo de negocio de un Laboratorio Social (esto es, un laboratorio centrado en la creación de prototipos y gestión de soluciones prometedoras, formado por personas de diversos sectores y con distintas experiencias para atacar la raíz y el núcleo de los problemas sociales) para una economía formal e inclusiva. A partir de tres fases (recopilación de datos, análisis de la información y cocreación del modelo de negocio), se obtuvo la estructura del modelo de negocio del Laboratorio Social, que cuenta con características que lo diferencian de instituciones similares: es sostenible y está en capacidad de brindar servicios a las entidades del ecosistema, opera en red para ser flexible y eficiente, aborda el fenómeno de la informalidad desde una perspectiva multienfoque o interdisciplinar y tiene un mayor enfoque en los territorios, profundidad de análisis y capacidad de monitoreo sobre el conocimiento que genera. Además, resultado de una investigación de casi dos años, este libro también plantea las etapas para el desarrollo del Laboratorio Social (planeación, consolidación y sostenibilidad), de tal forma que pueda ajustarse a las necesidades de investigación y de la población objetivo, escalar y consolidarse.This book aimed to define the design and structure of the business model of a Social Laboratory (i.e., a laboratory focused on prototyping and managing promising solutions, made up of people from different sectors and with different experiences to attack the root and core of social problems) for a formal and inclusive economy. Through three phases (data collection, information analysis, and co-creation of the business model) the structure of the Social Laboratory’s business model was developed, which has characteristics that differentiate it from similar institutions: it is sustainable and able to provide services to ecosystem entities, operates in a network to be flexible and efficient, addresses the phenomenon of informality from a multi-focus or interdisciplinary perspective, and has a greater focus on territories, depth of analysis, and monitoring capacity about the knowledge it generates. In addition, as the result of almost two years of research, this book also proposes stages for the development of the Social Laboratory (planning, consolidation, and sustainability) so that it can be adjusted to the research and the needs of the target population, scaled up, and consolidated

Characterizing Informality: Analysis of Migration, Gender, Youth, Education, and Technology in Informality

Una porción importante del mercado laboral en los países del sur global es la población informal. Tan solo en Colombia, la informalidad representó el 58,2 % de la fuerza laboral entre septiembre y noviembre de 2022. Más aún, teniendo en cuenta el papel que desempeña el capital social en los mercados y en las interacciones sociales de diversos grupos poblacionales, y el acceso al financiamiento en la inclusión de las dinámicas de mercado y la promoción de ecosistemas emprendedores, es necesario analizar la heterogeneidad presente en los niveles de bienestar, capital social, acceso financiero y avance tecnológico. Lo anterior, con el objetivo de establecer políticas dirigidas a reducir las diferencias socioeconómicas en términos de empleo, nacionalidad, género y grupos socioeconómicos. Por esto, el libro busca analizar diferentes características, valoraciones y comportamientos, en términos de bienestar social, capital social, inclusión financiera y uso tecnológico en diferentes subgrupos informales: 1) ocupaciones (vendedores ambulantes, recicladores y domiciliarios), 2) género (hombres, mujeres u otros), 3) nacionalidad (colombianos y migrantes) y 4) jóvenes (menores de 30 años) y mayores de 30 años.A significant portion of the labor market in the countries of the Global South consists of the informal population. In Colombia alone, informality represented 58.2% of the labor force between September and November 2022. Moreover, considering the role of social capital in markets and the social interactions of various population groups, as well as access to financing in the inclusion of market dynamics and promotion of entrepreneurial ecosystems, it is necessary to analyze the heterogeneity present in the levels of well-being, social capital, financial access, and technological progress to establish policies aimed at reducing socioeconomic differences in terms of employment, nationality, gender, and socioeconomic groups. Thus, the book seeks to analyze different characteristics, assessments, and behaviors regarding social welfare, social capital, financial inclusion, and technological use in different informal subgroups based on (i) occupation (street vendors, recyclers, and home-based vendors), (ii) gender (men, women, or others), (iii) nationality (Colombians and migrants), and (iv) age (young people under 30 years old and people over 30 years)

Microcredits: a promise that does not meet expectations

Nacieron para favorecer a los microempresarios, pero resulta que cuando el Banco de la República baja la tasa de interés, es el único producto en el que esta corrección no se refleja. El efecto es completamente diferente al esperado. Para llegar a esta conclusión los investigadores exploraron un histórico de los últimos 10 años de la Superintendencia Financiera de Colombia, la entidad que controla y maneja la información oficial, tanto de las tasas entregadas como de los desembolsos hechos por las entidades financieras, e igualmente revisaron las tasas del Banco de la República en ese mismo periodo de tiempo. Con esta información hicieron la correlación y análisis de los datos y así evidenciaron ese comportamiento de las tasas para los microcréditos.They were born to favor micro-entrepreneurs, but it turns out that when the Bank of the Republic lowers the interest rate, it is the only product in which this correction is not reflected. The effect is completely different than expected. To reach this conclusion, the researchers explored a history of the last 10 years of the Financial Superintendence of Colombia, the entity that controls and manages official information, both on the rates delivered and on the disbursements made by financial entities, and also reviewed the rates of the Banco de la República in that same period of time. With this information they made the correlation and analysis of the data and thus evidenced that behavior of the rates for microcredits

Ocupando la calle: tecnología y conocimiento en la informalidad

El estudio de la informalidad laboral ha estado dominado por una visión económica que limita la comprensión del fenómeno al omitir las reflexiones de los trabajadores informales sobre sus propias experiencias de vida dentro de la ocupación. En la investigación que presentamos en este libro, utilizamos la metodología de photo-elicitation para profundizar en la comprensión del impacto de la tecnología y el conocimiento en tres actividades informales: el reciclaje, la venta ambulante y los domicilios a través de plataformas móviles. Para ello, profundizamos, por un lado, en las tecnologías físicas y digitales que estos trabajadores utilizan y transforman y, por otro, en los procesos que les permiten agregar conocimiento para llevar a cabo su labor diaria. Epistasis-driven identification of SLC25A51 as a regulator of human mitochondrial NAD import

About a thousand genes in the human genome encode for membrane transporters. Among these, several solute carrier proteins (SLCs), representing the largest group of transporters, are still orphan and lack functional characterization. We reasoned that assessing genetic interactions among SLCs may be an efficient way to obtain functional information allowing their deorphanization. Here we describe a network of strong genetic interactions indicating a contribution to mitochondrial respiration and redox metabolism for SLC25A51/MCART1, an uncharacterized member of the SLC25 family of transporters. Through a combination of metabolomics, genomics and genetics approaches, we demonstrate a role for SLC25A51 as enabler of mitochondrial import of NAD, showcasing the potential of genetic interaction-driven functional gene deorphanization
