    Is foster caring associated with an earlier transition to adulthood for caregivers’ own children? ONS Longitudinal Study

    This study investigates whether the existing children in a fostering household differ from young people in non-caregiving households in the timing of their transitions to key adult roles, known to affect later health and life chances. Using data from the ONS Longitudinal Study, we pooled records from census years 1971-2001 and linked them to follow-up records from 1981-2011. We identified 2656 children living with a foster child and compared their profiles on the “big five” transitions to roles of adulthood — finishing school; leaving home; finding work and becoming financially independent; getting married; and having children — with those of other children without a foster child in the household (N=209,453). We fitted logistic and multinomial models that controlled for childhood socioeconomic and demographic confounders to estimate the proportion achieving the five roles measured in early adulthood. We found a modest but reliably higher proportion of caregivers’ children having achieved the transition to adulthood. There was some evidence that caregivers’ children might cope better with the transition to adulthood if they were older than the foster child or were female. The findings suggest that supporting foster parents with delaying their children’s transition to adulthood could become part of the role of supervising social workers

    The lifelong health and wellbeing trajectories of people who have been in care: Findings from the Looked-after Children Grown up Project

    The overarching aim of the Looked-after Children Grown Up Project (LACGro) was to use the unique data in the Office for National Statistics Longitudinal Study to build a comprehensive picture up to mid-life of the health and social functioning of care experienced members of the study. To achieve that, we set ourselves a number of objectives: • To determine whether children in residential care, kinship care and in foster care have different health and social experiences 10 to 40 years later compared to children in parental homes. • To explore if children cared for in residential settings do better or worse than children in kinship or foster care, and if children in kinship care fare better than children in foster care. • To understand if any differences in health and social difficulties vary by gender or ethnicity/migration status for those who have been looked after in childhood. • To investigate if care experiences and their outcomes differ by the census in which children are observed. • To find out what evidence there is for later resilience. Are there continuities or discontinuities in outcomes? • To explore if there is evidence that a long-term experience of care predicts better or worse outcomes than a short-term experience. • To consider if caregivers’ children are affected in the long-term by living with a child in care. Through the research, our goal was to describe the scale of inequalities in outcomes for children cared for in different places, of different ages and d identities, and to begin to understand how these factors interact to produce inequalities

    Strenuous Exercise Increases the Risk of Oxidative Stress in Ironman Triathlon Participants

    Regular physical activity has been linked to greater overall health. Literature review and studies have also defined regular physical activity as a reducer of life-threatening illnesses such as cardiovascular disease, diabetes and obesity. However, long increments of strenuous exercise can produce oxidative stress and muscle fatigue in the human body. The increase in oxygen consumption during strenuous exercise leads to elevated reactive oxygen species (ROS). Cells continuously produce free radicals and reactive oxygen species as part of metabolic processes in the body. These free radicals are neutralized by an antioxidant defense system in the body consisting of enzymes, such as catalase, and non-enzymatic antioxidants. An Ironman Triathlon consists of a 2.4-mile (3.86 km) swim, a 112-mile (180.25 km) bicycle ride and a marathon 26.2-mile (42.2 km) run, raced in that order and without a break. It is widely considered by athletes to be one of the most demanding sporting events in the world. It is hypothesized that a physically challenging event such as the Ironman Triathlon can be linked to elevated cortisol levels, increased occurrence of DNA damage, elevated concentrations of ROS, and consequently increased oxidative stress in humans. In order to derive conclusive results regarding the hypothesis, groups containing athletes who completed the full Ironman race, the half Ironman race, and a control group of moderately active individuals were established and individuals were required to report Garmin Smartwatch health and wellness data. The half Ironman consists of a 1.2-mile (1.93 km) swim, a 56-mile (90.12 km) bicycle ride and a marathon 13.1-mile (21.1 km) run, raced in that order and without a break. Several protocols were then applied to derive data necessary to complete the research. After the participants were selected, their saliva was collected in a non-invasive fashion and was used in the Elisa Saliva Kit to determine cortisol concentration. The saliva samples were also utilized to perform DNA and RNA extraction; and the resulting products were analyzed for quantity and quality of the DNA and RNA. Real time PCR allows scientists to monitor PCR while it is occuring. In this technique, luminescence is produced by reporter molecules as the PCR products increase with every cycle. To determine ROS concentration, the ROS-Glo assay, which provides a light signal that is proportional to the ROS in a given sample, was utilized. An additional marker of oxidative stress is 8-oxo-2-deoxyguanosine(8-oxo-dG). The OxiSelect™ Oxidative DNA Damage ELISA uses antibody and antigen interactions to report the concentration of 8-oxo-dG in a sample. Furthermore, the results indicate an increase in enzymatic indicators of elevated ROS, elevated cortisol levels, and disruption of sleep in the participating athletes after the race. In conclusion, the athletes who completed the full Ironman triathlon experienced increased amounts of oxidative stress than their less active counterparts in the control group, as was denoted by the elevated cortisol levels, increased 8-oxo-dG concentrations, and increased ROS concentrations.Such a rigorous event negatively impacted participants and caused oxidative stress.Faculty Sponsor: Noelle Cutte

    Federal Research, Additional Actions Needed to Improve Public Access to Research Results: Report to Congressional Requesters [with Responses from Federal Agencies]

    Why GAO Did This Study Research and development helps catalyze breakthroughs that improve the overall health and wellbeing of our society. Federal research and development expenditures averaged about 135billionannuallyforfiscalyears2015to2017.AccordingtoOSTP,providingfreepublicaccesstofederallyfundedresearchresultscanimproveboththeimpactandaccountabilityofthisimportantfederalinvestment.InFebruary2013,OSTPdirectedfederalagencieswithmorethan135 billion annually for fiscal years 2015 to 2017. According to OSTP, providing free public access to federally funded research results can improve both the impact and accountability of this important federal investment. In February 2013, OSTP directed federal agencies with more than 100 million in annual research and development expenditures to develop a plan to support increased public access to the results of federally funded research. GAO was asked to examine public access to federally funded research results. This report examines the extent of agencies’ (1) progress implementing plans to increase public access to federally funded research results and (2) coordination on public access plan implementation. GAO administered a questionnaire to 19 federal agencies selected based on annual research and development expenditure amounts, among other criteria; reviewed agency documents; and interviewed officials from 11 agencies, OSTP, and 21 stakeholder organizations. What GAO Found The 19 agencies that GAO reviewed have made progress implementing their plans to increase public access to federally funded research results (publications and data), as called for in a 2013 Office of Science and Technology Policy (OSTP) memorandum. However, some agencies have not fully implemented some aspects of their plans, in particular those related to data access and mechanisms to ensure researchers comply with public access requirements. Agencies are coordinating with each other and with nonfederal stakeholders to implement public access plans, including through an interagency group led by OSTP and five other agencies. However, the group has not fully implemented selected leading practices identified by GAO that can enhance and sustain interagency collaboration, such as defining and articulating common outcomes. For example, according to OSTP staff, key outcomes have not yet been decided upon. Agency officials and stakeholders identified several challenges to implementing public access plans that interagency coordination might help them address, such as • Absence of common standards in several areas; • Measuring effectiveness of public access plan implementation; and • Balancing providing public access with safeguarding sensitive information. By taking steps to fully implement relevant leading collaboration practices, the interagency group could help agencies better marshal their collective efforts to address common challenges to public access plan implementation. What GAO Recommends GAO is making 37 recommendations to 16 agencies to promote full and effective implementation of agency public access plans. For example, GAO recommends that OSTP and 5 agencies leading a public access interagency group take steps to fully implement selected leading collaboration practices. Of the 16 agencies, 15 agreed with GAO’s recommendations while 1 (OSTP) disagreed. GAO continues to believe the recommendation to OSTP is warranted

    Hydrogen activation by [NiFe]-hydrogenases

    Hydrogenase-1 (Hyd-1) from Escherichia coli is a membrane-bound enzyme that catalyses the reversible oxidation of molecular H2 The active site contains one Fe and one Ni atom and several conserved amino acids including an arginine (Arg(509)), which interacts with two conserved aspartate residues (Asp(118) and Asp(574)) forming an outer shell canopy over the metals. There is also a highly conserved glutamate (Glu(28)) positioned on the opposite side of the active site to the canopy. The mechanism of hydrogen activation has been dissected by site-directed mutagenesis to identify the catalytic base responsible for splitting molecular hydrogen and possible proton transfer pathways to/from the active site. Previous reported attempts to mutate residues in the canopy were unsuccessful, leading to an assumption of a purely structural role. Recent discoveries, however, suggest a catalytic requirement, for example replacing the arginine with lysine (R509K) leaves the structure virtually unchanged, but catalytic activity falls by more than 100-fold. Variants containing amino acid substitutions at either or both, aspartates retain significant activity. We now propose a new mechanism: heterolytic H2 cleavage is via a mechanism akin to that of a frustrated Lewis pair (FLP), where H2 is polarized by simultaneous binding to the metal(s) (the acid) and a nitrogen from Arg(509) (the base)

    Tele-branding in TVIII: the network as brand and the programme as brand

    In the era of TVIII, characterized by deregulation, multimedia conglomeration, expansion and increased competition, branding has emerged as a central industrial practice. Focusing on the case of HBO, a particularly successful brand in TVIII, this article argues that branding can be understood not simply as a feature of television networks, but also as a characteristic of television programmes. It begins by examining how the network as brand is constructed and conveyed to the consumer through the use of logos, slogans and programmes. The role of programmes in the construction of brand identity is then complicated by examining the sale of programmes abroad, where programmes can be seen to contribute to the brand identity of more than one network. The article then goes on to examine programme merchandising, an increasingly central strategy in TVIII. Through an analysis of different merchandising strategies the article argues that programmes have come to act as brands in their own right, and demonstrates that the academic study of branding not only reveals the development of new industrial practices, but also offers a way of understanding the television programme and its consumption by viewers in a period when the texts of television are increasingly extended across a range of media platforms

    B-cell Zone Reticular Cell Microenvironments Shape CXCL13 Gradient Formation

    Through the formation of concentration gradients, morphogens drive graded responses to extracellular signals, thereby fine-tuning cell behaviors in complex tissues. Here we show that the chemokine CXCL13 forms both soluble and immobilized gradients. Specifically, CXCL13+ follicular reticular cells form a small-world network of guidance structures, with computer simulations and optimization analysis predicting that immobilized gradients created by this network promote B-cell trafficking. Consistent with this prediction, imaging analysis show that CXCL13 binds to extracellular matrix components in situ, constraining its diffusion. CXCL13 solubilization requires the protease cathepsin B that cleaves CXCL13 into a stable product. Mice lacking cathepsin B display aberrant follicular architecture, a phenotype associated with effective B cell homing to but not within lymph nodes. Our data thus suggest that reticular cells of the B cell zone generate microenvironments that shape both immobilized and soluble CXCL13 gradient
