    Effect of surface roughness and sterilization on bacterial adherence to ultra-high molecular weight polyethylene

    AbstractSterilization with ethylene oxide (EO) and gas plasma (GP) are well-known methods applied to ultra-high molecular weight polyethylene (UHMWPE) surfaces in the belief that they prevent major material changes caused by gamma irradiation. However, the influence of these surface sterilization methods on bacterial adherence to UHMWPE is unknown. UHMWPE samples with various degrees of roughness (0.3, 0.8 and 2.0 µm) were sterilized with either GP or EO. The variations in hydrophobicity, surface free energy and surface functional groups were investigated before and after sterilization. Sterilized samples were incubated with either Staphylococcus aureus or Staphylococcus epidermidis in order to study bacterial adherence to these materials. Fewer bacteria adhered to UHMWPE after sterilization with EO than after sterilization with GP, especially to the smoothest surfaces. No changes in chemical composition of the UHMWPE surface due to sterilization were observed using X-ray photoemission spectroscopy analysis. The decreased bacterial adherence to UHMWPE found at the smoothest surfaces after sterilization with EO was not directly related to changes in chemical composition. Increased bacterial adherence to rougher surfaces was associated with increased polar surface energy of EO-sterilized surfaces

    A Test of Pre-Main-Sequence Lithium Depletion Models

    Despite the extensive study of lithium depletion during pre-main-sequence contraction, studies of individual stars show discrepancies between ages determined from the HR diagram and ages determined from lithium depletion (Song et al. 2002, White & Hillenbrand 2005) indicating open questions in the pre-main-sequence evolutionary models. To further test these models, we present high resolution spectra for members of the Beta Pictoris Moving Group (BPMG), which is young and nearby. We measure equivalent widths of the 6707.8 Angstrom Li I line in these stars and use them to determine lithium abundances. We combine the lithium abundance with the predictions of pre-main-sequence evolutionary models in order to calculate a lithium depletion age for each star. We compare this age to the age predicted by the HR diagram of the same model. We find that the evolutionary models under-predict the amount of lithium depletion for the BPMG given its nominal HR diagram age of ~12 Myr (Zuckerman et al. 2001), particularly for the mid-M stars, which have no observable Li I line. This results in systematically older ages calculated from lithium depletion isochrones than from the HR diagram. We suggest that this discrepancy may be related to the discrepancy between measured M-dwarf radii and the smaller radii predicted by evolutionary models.Comment: Accepted by ApJ; 21 pages, 5 figure

    Discovery of novel DNA methylation biomarkers for non-invasive sporadic breast cancer detection in the Latino population

We described a panel of 38 differentially methylated CpG positions that was able to cluster breast cancer patients (BCP) and controls, and that also recapitulated methylation differences in 12 primary breast tumors and their matched normal breast tissue. Moving forward, we simplified the detection method to improve its applicability in a clinical setting and used an independent well-characterized cohort of 80 leukocyte DNA samples from BCP and 80 healthy controls to validate methylation results at specific cancer-related genes. Our investigations identified methylation at CYFIP1 as a novel epigenetic biomarker candidate for sporadic breast cancer in the Uruguayan population. These results provide a proof-of-concept for the design of larger studies aimed at validating biomarker panels for the Latin American population

    Argonaute proteins: Why are they so important for the legume–rhizobia symbiosis?

    Unlike most other land plants, legumes can fulfill their nitrogen needs through the establishment of symbioses with nitrogen-fixing soil bacteria (rhizobia). Through this symbiosis, fixed nitrogen is incorporated into the food chain. Because of this ecological relevance, the genetic mechanisms underlying the establishment of the legume–rhizobia symbiosis (LRS) have been extensively studied over the past decades. During this time, different types of regulators of this symbiosis have been discovered and characterized. A growing number of studies have demonstrated the participation of different types of small RNAs, including microRNAs, in the different stages of this symbiosis. The involvement of small RNAs also indicates that Argonaute (AGO) proteins participate in the regulation of the LRS. However, despite this obvious role, the relevance of AGO proteins in the LRS has been overlooked and understudied. Here, we discuss and hypothesize the likely participation of AGO proteins in the regulation of the different steps that enable the establishment of the LRS. We also briefly review and discuss whether rhizobial symbiosis induces DNA damages in the legume host. Understanding the different levels of LRS regulation could lead to the development of improved nitrogen fixation efficiency to enhance sustainable agriculture, thereby reducing dependence on inorganic fertilizers.Fil: Valdés-López, Oswaldo.     Methods for Analysis of Matrix Metalloproteinase Regulation of Neutrophil-Endothelial Cell Adhesion

    Recent evidence indicates novel role for matrix metalloproteinases (MMPs), in particular gelatinase A (MMP-2), in the regulation of vascular biology that are unrelated to their well-known proteolytic breakdown of matrix proteins. We have previously reported that MMP-2 can modulate vascular reactivity by cleavage of the Gly32-Leu33 bound in big endothelin-1 (ET-1) yielding a novel vasoactive peptide ET-1[1-32]. These studies were conducted to investigate whether gelatinolytic MMPs could affect neutrophil-endothelial cell attachment. ET-1[1-32] produced by MMP-2 up-regulated CD11b/CD18 expression on human neutrophils, thereby promoted their adhesion to cultured endothelial cells. ET-1[1-32] evoked release of gelatinase B (MMP-9), which in turn cleaved big ET-1 to yield ET-1[1-32], thus revealing a self-amplifying loop for ET-1[1-32] generation. ET-1[1-32] was rather resistant to cleavage by neutrophil proteases and further metabolism of ET-1[1-32] was not a prerequisite for its biological actions on neutrophils. The neutrophil responses to ET-1[1-32] were mediated via activation of ET(A)receptors through activation of the Ras/Raf-1/MEK/ERK signaling pathway. These results suggest a novel role for gelatinase A and B in the regulation of neutrophil functions and their interactions with endothelial cells.     An online air-sea exchange model framework for trace gases powered by machine- learning

    The ocean emits a wide range of trace gases, such as volatile organic compounds, or sulfur-,nitrogen-, and halogen-containing compounds. Many of these gases play critical roles in the atmosphere, including aerosol and cloud formation, tropospheric and stratospheric ozone budget, as well as the self-cleaning capacity of the atmosphere. Most chemistry-climate models use prescribed oceanic emissions (often derived from observations). These prescribed (offline) emissions generally do not respond to changes in local conditions. A process-level representation of the bi-directional oceanic emissions of trace gases remains challenging, mainly because the ocean biogeochemicalprocesses controlling the natural synthesis of these compounds in the seawater remain poorly understood. We present a new online air-sea exchange framework for the NCAR CESM2, with an observationally trained machine-learning emulator to couple the ocean biogeochemistry with the air-sea exchange. This machine-learning based approach so far has been tested for a number of important trace gases, including dimethyl sulfide (DMS), acetone, bromoform (CHBr 3 ), and dibromomethane (CH 2 Br 2 ), and the preliminary results are evaluated with observations around the globe. This new model framework is more skillful than the widely used top-down approaches for representing the seasonal/spatial variations and the annual means of atmospheric concentrations. The new approach improves the model predictability for the coupled earth system model, and can be used as a basis for investigating the future ocean emissions and feedbacks under climate change.Fil: Wang, Siyuan.     Association between heavy metals and metalloids in topsoil and mental health in the adult population of Spain

    Despite the biological plausibility of the association between heavy metal exposure and mental health disorders, epidemiological evidence remains scarce. The objective was to estimate the association between heavy metals and metalloids in soil and the prevalence of mental disorders in the adult population of Spain. Methods Individual data came from the Spanish National Health Survey 2011–2012, 18,073 individuals residing in 1772 census sections. Mental health was measured with the 12-item General Health Questionnaire. The concentration estimates of heavy metal and metalloid levels in topsoil (upper soil horizon) came from the Geochemical Atlas of Spain based on 13,317 soil samples. Levels of lead (Pb), arsenic (As), cadmium (Cd) and manganese (Mn) were estimated in each census section by “ordinary Kriging”. Odds ratios (OR) were calculated by multilevel logistic regression models. Results Compared with the lowest Pb concentration levels quartile, the OR for the second quartile was 1.29 (95%CI: 1.11–1.50), increasing progressively to 1.37 (95%CI: 1.17–1.60) and 1.51 (95%CI: 1.27–1.79) in the third and fourth quartiles, respectively. For As, the association was observed in the third and fourth quartiles: 1.21 (95%CI: 1.04–1.41) and 1.42 (95% CI: 1.21–1.65), respectively. Cd was associated also following a gradient from the second quartile: 1.34 (95%CI: 1.15–1.57) through the fourth: 1.84 (95%CI: 1.56–2.15). In contrast, Mn only showed a positive association at the second quartile. Additionally, individuals consuming vegetables > once a day the OR for the fourth quartile of Pb concentration, vs. the first, increased to 2.93 (95%CI: 1.97–4.36); similarly for As: 3.00 (95%CI: 2.08–4.31), and for Cd: 3.49 (95%CI: 2.33–5.22). Conclusions Living in areas with a higher concentration of heavy metals and metalloids in soil was associated with an increased probability of having a mental disorder.     Autonomic nervous system assessment in critically ill patients undergoing a cognitive rehabilitation therapy

    Recent clinical and electrophysiological studies reveal a high incidence of autonomic nervous system (ANS) dysfunction in patients treated in Intensive Care Units (ICUs). Cognitive rehabilitation (CR) is a behavioral therapy that has proven to be effective improving cognitive deficits in clinical populations with abnormalities in brain activation patterns. A total of 17 critically ill patients received CR aimed to improve the ANS status, which was quantified in terms of HRV. The CR included cognitive exercises aimed to improve prefrontal activation. HRV was obtained during pre-CR, CR and post-CR. Power in the low (PLF) and high (PHF) frequency bands related to sympathetic and parasympathetic systems was computed. PHF was obtained within a band centered at respiratory rate. Comparing with baseline values, 7 patients showed an increased PHF in post-CR, suggesting an increase of parasympathetic activity

    Modeling the Accretion Disk around the High-mass Protostar GGD 27-MM1

    Recent high angular resolution (≃40 mas) ALMA observations at 1.14 mm resolve a compact (R ≃ 200 au), flattened dust structure perpendicular to the HH 80─81 jet emanating from the GGD 27-MM1 high-mass protostar, making it a robust candidate for a true accretion disk. The jet─disk system (HH 80─81/GGD 27-MM1) resembles those found in association with low- and intermediate-mass protostars. We present radiative transfer models that fit the 1.14 mm ALMA dust image of this disk, which allow us to obtain its physical parameters and predict its density and temperature structure. Our results indicate that this accretion disk is compact (R disk ≃ 170 au) and massive (≃5 M ☉), at about 20% of the stellar mass of ≃20 M ☉. We estimate the total dynamical mass of the star─disk system from the molecular line emission, finding a range between 21 and 30 M ☉, which is consistent with our model. We fit the density and temperature structures found by our model with power-law functions. These results suggest that accretion disks around massive stars are more massive and hotter than their low-mass siblings, but they still are quite stable. We also compare the temperature distribution in the GGD 27─MM1 disk with that found in low- and intermediate-mass stars and discuss possible implications for the water snow line. We have also carried out a study of the distance based on Gaia DR2 data and the population of young stellar objects in this region and from the extinction maps. We conclude that the source distance is within 1.2 and 1.4 kpc, closer than what was derived in previous studies (1.7 kpc).Fil: Añez López, N..
