    Import 05/08/2014Tato diplomová práce popisuje kapalinový paprsek nejen jako nástroj pro obrábění či řezání, ale také jako efektivní nástroj při těžbě uhlí. V první části je seznámení s kapalinovým paprskem jakožto řezným nástrojem, jeho popis, funkce a vlastnosti. Další část se věnuje použití kapalinového paprsku jako nástroje pro dobývání uhlí, který je nazýván hydromonitor. V dalších kapitolách se věnujeme dobývacím kombajnům a těžené surovině uhlí. Hlavní náplní této práce je porovnání klasického dobývacího kombajnu a hydromonitoru, analýza celkové hodnoty vytěženého uhlí v jednotce tuna/hodina a porovnání, který z uvedených nástrojů je při těžbě černého uhlí efektivnější a ekonomicky výhodnější.This thesis describes a liquid jet not only as a tool for machining or cutting, but also as an effective tool in coal mining. The first part is the introduction of the fluid jet as a cutting tool, a description, function and properties. The next section is devoted to the use of liquid jet as a tool for mining coal, which is called Hydromonitors. In the following chapters we deal with mining combines a raw material coal mined. The main concern of this work is the comparison of classical mining shearer and Hydromonitors, analysis of the total value of the coal in the unit ton / hour, compared to which of these tools is the extraction of coal efficiently and economically.Prezenční545 - Institut ekonomiky a systémů řízenívelmi dobř

    Schizophyllum commune: An unexploited source for lignocellulose degrading enzymes

    Lignocellulose represents the most abundant source of carbon in the Earth. Thus, fraction technology of the biomass turns up as an emerging technology for the development of biorefineries. Saccharification and fermentation processes require the formulation of enzymatic cocktails or the development of microorganisms (naturally or genetically modified) with the appropriate toolbox to produce a cost-effective fermentation technology. Therefore, the search for microorganisms capable of developing effective cellulose hydrolysis represents one of the main challenges in this era. Schizophyllum commune is an edible agarical with a great capability to secrete a myriad of hydrolytic enzymes such as xylanases and endoglucanases that are expressed in a high range of substrates. In addition, a large number of protein-coding genes for glycoside hydrolases, oxidoreductases like laccases (Lacs; EC 1.10.3.2), as well as some sequences encoding for lytic polysaccharide monooxygenases (LPMOs) and expansins-like proteins demonstrate the potential of this fungus to be applied in different biotechnological process. In this review, we focus on the enzymatic toolbox of S. commune at the genetic, transcriptomic, and proteomic level, as well as the requirements to be employed for fermentable sugars production in biorefineries.     A Genome Wide Association Study of arabinoxylan content in 2-row spring barley grain

    In barley endosperm arabinoxylan (AX) is the second most abundant cell wall polysaccharide and in wheat it is the most abundant polysaccharide in the starchy endosperm walls of the grain. AX is one of the main contributors to grain dietary fibre content providing several health benefits including cholesterol and glucose lowering effects, and antioxidant activities. Due to its complex structural features, AX might also affect the downstream applications of barley grain in malting and brewing. Using a high pressure liquid chromatography (HPLC) method we quantified AX amounts in mature grain in 128 spring 2-row barley accessions. Amounts ranged from ~ 5.2 μg/g to ~ 9 μg/g. We used this data for a Genome Wide Association Study (GWAS) that revealed three significant quantitative trait loci (QTL) associated with grain AX levels which passed a false discovery threshold (FDR) and are located on two of the seven barley chromosomes. Regions underlying the QTLs were scanned for genes likely to be involved in AX biosynthesis or turnover, and strong candidates, including glycosyltransferases from the GT43 and GT61 families and glycoside hydrolases from the GH10 family, were identified. Phylogenetic trees of selected gene families were built based on protein translations and were used to examine the relationship of the barley candidate genes to those in other species. Our data reaffirms the roles of existing genes thought to contribute to AX content, and identifies novel QTL (and candidate genes associated with them) potentially influencing the AX content of barley grain. One potential outcome of this work is the deployment of highly associated single nucleotide polymorphisms markers in breeding programs to guide the modification of AX abundance in barley grain

The following chapters are devoted to onlookers educational programs, theatre-goers club, and are supplied with examples from history and present of Prague theatres as well. The pivotal part of the work brings universal view at audience development. It describes possible motives and even barriers of theatre visit rate, segmentation of onlookers, and concrete instruments how to evolve the relation between theatre ? onlooker. The semifinal chapter describes Prague theatre-goer scope. At the end there is shown the problem of Divadlo Na Zábradlí theatre-goers. The whole work is completed with statistic dates and results of research ?Theatre onTV?, whereof was used eight questions of the authoress of this work

    Import 11/07/2012Tato bakalářská práce je zaměřena na problematiku týkající se kapalinového paprsku. V první části práce se věnuji čistě kapalinovému paprsku, jeho specifickým vlastnostem, rozdělení, zda je paprsek s abrazivem či bez abraziva a jeho hlavní činností. Druhá část práce pojednává o ekonomické stránce problému, především o ceně a spotřebě elektrické energie a o dalších nezbytných poplatcích souvisejících s provozem. Dále se pak zabývám vlastním výpočtem účinnosti kapalinového paprsku a jeho ekonomickému parametru. Závěr práce je zaměřen na srovnání dané analýzy s jinými publikacemi podobného či stejného charakteru.This thesis focuses on issues concerning the liquid jet. The first part of my work is devoted to purely liquid beam, its specific characteristics, splits, if the beam is with or without abrasive blasting and its main activities. The second part of my work is devoted to economic section of the beam liquid, which deals with mainly price and consumption of electricity and other necessary fees associated with the operation. I am also involved his own calculation of efficiency liquid jet and its economic parameters.
