    The effects of mining machinery noise of different frequencies on the behaviour, faecal corticosterone and tissue morphology of wild mice (Mus musculus)

    Mining noise has a wide variety of frequency spectra and is a potential source of stress for wildlife. We evaluated the effects of mining machinery noise on behaviour and associated physiological parameters at two isoenergetic frequency ranges: high (>2 kHz) and low (≤2 kHz), the latter being less audible to mice, our model species. Effects of these two frequency spectra on behaviour, organ morphology and faecal corticosterone of wild mice were compared with a control treatment with no extra auditory stimuli. The mice exposed to high frequency noise spent less time in their nest than those exposed to low frequency noise or those in the control treatment, and they spent more time circling, especially anticlockwise, which in conjunction with elevated faecal corticosterone levels may reflect a greater right brain hemisphere stress-related response, particularly in females. Low frequency mining noise reduced grooming and circling, suggesting decreased physiological arousal due to mild stress. Low frequencies were also associated with increased faecal corticosterone in males compared to controls, which may be related to gender-based differences of the ear canal that affect frequency sensitivity. In conclusion, high frequency and low frequency mining machinery noise produced stress-related responses that may be important for the animals’ welfare and survival

    Using the universal P2/P8 primers, we were able to obtain the gene segments of chromo-helicase-DNA binding protein (CHD)-Z and CHD-W from ten species of ardeid birds including Chinese egret (Egretta eulophotes), little egret (E. garzetta), eastern reef egret (E. sacra), great egret (Ardea alba), grey heron (A. cinerea), Chinese pond-heron (Ardeola bacchus), cattle egret (Bubulcus ibis), black-crowned night-heron (Nycticorax nycticorax), cinnamon bittern (Ixobrychus cinnamomeus) and yellow bittern (I. sinensis). Based on conserved regions inside the P2/P8-derived sequences, we designed new PCR primers for sex identification in these ardeid species. Using agarose gel electrophoresis, the PCR products showed two bands for females (140 bp derived from CHD-W and the other 250 bp from CHD-ZW), whereas the males showed only the 250 bp band.     Fundulus as the premier teleost model in environmental biology : opportunities for new insights using genomics

In this paper we summarize an extensive body of work examining the adaptive responses of Fundulus species to environmental conditions, and describe how this research has contributed importantly to our understanding of physiology, gene regulation, toxicology, and ecological and evolutionary genetics of teleosts and other vertebrates. These explorations have reached a critical juncture at which advancement is hindered by the lack of genomic resources for these species.     Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung’s disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung’s disease) from 264 hospitals (89 in high-income countries, 166 in middleincome countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in lowincome countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. Interpretation Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between lowincome, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030

    Silvopastoral systems: A feasible step towards environmentally sustainable livestock systems in Mexico

    In Mexico, the cattle industry is related to employment and economic growth, and provides food security for people living in poverty; nonetheless, livestock production is also related to the decrease of biodiversity and environmental services. A feasible option to maintain food security while utilizing natural resources in a sustainable manner is the conversion to silvopastoral systems (SPS; associations of grasslands with trees and shrubs). Mexico has the potential to successfully convert livestock systems in the tropics to SPS, as they occur unintendedly in the Mexican Tropics. Recently, Mexican researchers have focused on exploring the variables involving the implementation of SPS, including animal welfare indicators. Recent findings have shown positive results in body condition, flight distance (a measure of fear to humans), foraging time in relation to temperature and affiliative interactions. Such benefits are also related to humane livestock practices, which contemplate ethical livestock production in environmentally friendly systems, especially when this activity has the potential to improve food security in an overpopulated world

    Mining noise has been identified as a source of stress for wildlife but the effects on the behaviour and physiology of individuals are largely unknown. We evaluated the effects of exposing wild mice to mining machinery noise for 3 weeks at two amplitude ranges: high (HA; 70–75 dB (A)) and low (LA; 60–65 dB (A)) on their behaviour, organ properties and faecal corticosterone levels, when compared with a control treatment (no extra auditory stimuli, below 55 dB (A)). Both noise treatments increased circling to the left, compared to the control (P = 0.001) which corresponds to right hemispheric processing of the stress response; however only the high amplitude noise increased circling to the right (left hemisphere activation; P < 0.0001), which may inhibit stress arousal by right hemisphere. Total circling behaviour (to the left and right) was just increased in mice exposed to high amplitude noise, compared with those in the control and low amplitude noise (P < 0.0001), but in the low amplitude noise faecal corticosterone was increased (P = 0.003). We hypothesize that dopamine-related stereotypies during high amplitude mining noise were a coping mechanism that prevented a physiological response. Noise-exposed males tended to have smaller spleens (P = 0.003) and females in the HA treatment tended to have a smaller adrenal cortex/medulla ratio (P = 0.007).     Assessing Sustainability in Cattle Silvopastoral Systems in the Mexican Tropics Using the SAFA Framework

    The sub-humid native rainforest in Yucatan is one of the most endangered in Mexico. Cattle production is one of the main causes of land use change and silvopastoral systems are a feasible alternative. This work compares the sustainable performance of silvopastoral (native and intensive) and monoculture cattle farms in the state of Yucatan using the Sustainability Assessment for Food and Agriculture (SAFA) framework. Questionnaires and semi-structured interviews were applied in 9 farms. Responses were fed to the SAFA Tool to obtain sustainability polygons. Percentages of SAFA themes positively and negatively valuated were calculated. Native farms had positive ratings for Participation, Land, Biodiversity and Cultural Diversity, whereas intensive excelled on Holistic Management. Native farms had limited ratings for Decent Livelihood. Native farms (and one intensive silvopastoral farm) had the highest percentages of themes positively valuated compared to monocultures (and one intensive silvopastoral farm), which scored the lowest. Positive evaluations identified native systems as an option for sustainable production; however, areas of opportunity in all farms were discovered. This is the first comparative study using SAFA to evaluate differences in farming systems in the Mexican tropics, providing valuable information to generate policies and incentives on sustainable livestock production, as well as for improving evaluation tools for local application

    Effects of Enrichment Type, Presentation and Social Status on Enrichment Use and Behavior of Sows&mdash;Part 2: Free Access Stall Feeding

    Continuing with previous research by our group in an ESF system, four types of enrichment treatments were assessed in gestating sows housed in Free Access Stalls: (1) Constant: constant provision of wood on chain; (2) Rotate: rotation of rope, straw and wood; (3) Stimulus: rotation of enrichments with an acoustic cue; and (4) Control: no enrichment. Treatments had a 12 day-duration. Four groups (28 &plusmn; 2 sows) were studied from weeks 6 to 14 of gestation. Groups received all treatments in random order. Three dominant and 3 subordinates per pen were selected using a feed competition test. Digital photos were collected at 10 min intervals for 8 h on days 1, 8, 10 and 12 to record interactions with enrichment. Skin lesions were assessed on days 1 and 12, and salivary cortisol was assessed in weeks 6, 10 and 14 of gestation. More enrichment use was observed in Rotate and Stimulus treatments compared to Constant, and more sows contacted enrichment when straw was provided in the Rotate and Stimulus treatments. There was no difference in the amount of enrichment use by dominants and subordinates, no cortisol concentration elevation in subordinate sows nor any difference in lesion scores. In conclusion, social status had little impact and feeding system is important to reduce stress and aggression

    Oncogenic KRAS Induces NIX-Mediated Mitophagy to Promote Pancreatic Cancer.

    Activating KRAS mutations are found in nearly all cases of pancreatic ductal adenocarcinoma (PDAC), yet effective clinical targeting of oncogenic KRAS remains elusive. Understanding of KRAS-dependent PDAC-promoting pathways could lead to the identification of vulnerabilities and the development of new treatments. We show that oncogenic KRAS induces BNIP3L/NIX expression and a selective mitophagy program that restricts glucose flux to the mitochondria and enhances redox capacity. Loss of Nix restores functional mitochondria to cells, increasing demands for NADPH reducing power and decreasing proliferation in glucose-limited conditions. Nix deletion markedly delays progression of pancreatic cancer and improves survival in a murine (KPC) model of PDAC. Although conditional Nix ablation in vivo initially results in the accumulation of mitochondria, mitochondrial content eventually normalizes via increased mitochondrial clearance programs, and pancreatic intraepithelial neoplasia (PanIN) lesions progress to PDAC. We identify the KRAS-NIX mitophagy program as a novel driver of glycolysis, redox robustness, and disease progression in PDAC. SIGNIFICANCE: NIX-mediated mitophagy is a new oncogenic KRAS effector pathway that suppresses functional mitochondrial content to stimulate cell proliferation and augment redox homeostasis. This pathway promotes the progression of PanIN to PDAC and represents a new dependency in pancreatic cancer.This article is highlighted in the In This Issue feature, p. 1143.Includes: M.P.
