    A new squat lobster of the genus Munidopsis (Crustacea, Decapoda, Munidopsidae) from the Mediterranean Sea

The species (M. ariadne) is morphologically closely related to M. maunga Schnabel and Bruce, 2006, from New Zealand. The species is also close to M. polymorpha Koelbel, 1892, from Lanzarote, Canary Islands, and M. talismani A. Milne Edwards and Bouvier, 1894, from the NW Africa.     Screening of Human Tumor Antigens for CD4+ T Cell Epitopes by Combination of HLA-Transgenic Mice, Recombinant Adenovirus and Antigen Peptide Libraries

    BACKGROUND: As tumor antigen-specific CD4+ T cells can mediate strong therapeutic anti-tumor responses in melanoma patients we set out to establish a comprehensive screening strategy for the identification of tumor-specific CD4+ T cell epitopes suitable for detection, isolation and expansion of tumor-reactive T cells from patients. METHODS AND FINDINGS: To scan the human melanoma differentiation antigens TRP-1 and TRP-2 for HLA-DRB1*0301-restricted CD4+ T cell epitopes we applied the following methodology: Splenocytes of HLA-DRB1*0301-transgenic mice immunized with recombinant adenovirus encoding TRP-1 (Ad5.TRP-1) or TRP-2 (Ad5.TRP-2) were tested for their T cell reactivity against combinatorial TRP-1- and TRP-2-specific peptide libraries. CD4+ T cell epitopes thus identified were validated in the human system by stimulation of peripheral blood mononuclear cells (PBMC) from healthy donors and melanoma patients. Using this strategy we observed that recombinant Ad5 induced strong CD4+ T cell responses against the heterologous tumor antigens. In Ad5.TRP-2-immunized mice CD4+ T cell reactivity was detected against the known HLA-DRB1*0301-restricted TRP-2(60-74) epitope and against the new epitope TRP-2(149-163). Importantly, human T cells specifically recognizing target cells loaded with the TRP-2(149-163)-containing library peptide or infected with Ad5.TRP-2 were obtained from healthy individuals, and short term in vitro stimulation of PBMC revealed the presence of epitope-reactive CD4+ T cells in melanoma patients. Similarly, immunization of mice with Ad5.TRP-1 induced CD4+ T cell responses against TRP-1-derived peptides that turned out to be recognized also by human T cells, resulting in the identification of TRP-1(284-298) as a new HLA-DRB1*0301-restricted CD4+ T cell epitope. CONCLUSIONS: Our screening approach identified new HLA-DRB1*0301-restricted CD4+ T cell epitopes derived from melanoma antigens. This strategy is generally applicable to target antigens of other tumor entities and to different HLA class II molecules even without prior characterization of their peptide binding motives

    A Candidate H1N1 Pandemic Influenza Vaccine Elicits Protective Immunity in Mice

    BackgroundIn 2009 a new pandemic disease appeared and spread globally. The recent emergence of the pandemic influenza virus H1N1 first isolated in Mexico and USA raised concerns about vaccine availability. We here report our development of an adenovirus-based influenza H1N1 vaccine tested for immunogenicity and efficacy to confer protection in animal model.MethodsWe generated two adenovirus(Ad5)-based influenza vaccine candidates encoding the wildtype or a codon-optimized hemagglutinin antigen (HA) from the recently emerged swine influenza isolate A/California/04/2009 (H1N1)pdm. After verification of antigen expression, immunogenicity of the vaccine candidates were tested in a mouse model using dose escalations for subcutaneous immunization. Sera of immunized animals were tested in microneutalization and hemagglutination inhibition assays for the presence of HA-specific antibodies. HA-specific T-cells were measured in IFN? Elispot assays. The efficiency of the influenza vaccine candidates were evaluated in a challenge model by measuring viral titer in lung and nasal turbinate 3 days after inoculation of a homologous H1N1 virus.Conclusions/SignificanceA single immunization resulted in robust cellular and humoral immune response. Remarkably, the intensity of the immune response was substantially enhanced with codon-optimized antigen, indicating the benefit of manipulating the genetic code of HA antigens in the context of recombinant influenza vaccine design. These results highlight the value of advanced technologies in vaccine development and deployment in response to infections with pandemic potential. Bugün geleneksel olarak hizmet işletmesi olarak tanımlanabilecek işletmelerin yanında somut ögelerin ağırlıklı olduğu ürünler üreten işletmeler de kendilerini hizmet sunan işletmeler olarak tanımlamaktadırlar. Bu durum hizmetlerin ekonomik hayattaki önemini göstermektedir. En temel fonksiyonu finansal sistem içinde tasarruf edilen fonları toplayıp, fon ihtiyacı duyan kesime aktarmak olan bankalar da birer hizmet işletmesidir. Finans sektörünün en önemli aktörlerinden olan bankalar işlevleri ve sundukları hizmetlerle ekonominin de temel yapı taşlarındandır. Hizmetler genellikle üretildikleri yerde tüketilmek zorunda olduklarından hizmet işletmeleri için hizmetlerin dağıtımında doğrudan dağıtım mümkün olan tek dağıtım kanalıdır. Son zamanlara kadar bankalar da ürettikleri hizmetleri şubeler aracılığıyla doğrudan hizmetin üretildiği yerde sunmaktaydılar. Fakat teknolojik gelişmelere bağlı olarak oluşan elektronik tabanlı alternatif dağıtım kanalları sayesinde banka hizmetleri şubelerden bağımsız olarak sunulabilmiştir. Bu çalışmada bankacılık hizmetlerinin pazarlanması konusu incelenmiş, banka hizmetlerinin sunumunda hizmet sunumu ile hizmet üreticisinin birbirinden ayrılmasını sağlayan elektronik tabanlı dağıtım kanallarından olan kredi kartlarının Halkbank'taki uygulaması genelde banka düzeyinde, özelde Samsun Bölge Koordinatörlüğü düzeyinde incelenerek alternatif dağıtım kanallarının banka hizmetlerinin pazarlanmasındaki etkisi araştırılmıştır. In most of the developed economies service sector is the largest part of the national economy. In addition to the business that are traditionally defined as service business, businesses that are producing concrete element dominated products defines themselves as service businesses. This proves the importance of services sector for economic activity. Banks whose basic function is to collect saved funds and channel those funds to those who need them are an example of service businesses. As being the most important agent of the financial sector, banks are one of the most important fundamental elements of the economy by the services they provide and functions they perform. As services should be consumed at the place where they are produced direct distribution is the only available distribution channel for services businesses. Until recently, banks have been delivering their services through their branch networkæ however as a result of technological improvements in alternative electronic data based distribution, they are able to provide their services without depending on the branch network.     CLPFFD-PEG functionalized NIR-absorbing hollow gold nanospheres and gold nanorods inhibit β-amyloid aggregation

    Gold nanoparticles with specific optical properties in combination with the CLPFFD peptide that exhibits selectivity for beta-amyloid (A beta) aggregates are promising photothermal absorbers for application in Alzheimer's disease therapy. We report on hollow gold nanospheres (HAuNS) and gold nanorods (AuNR), which exhibit strong plasmonic near infrared (NIR) absorbance in the optical window of biological tissue and which are functionalized with CLPFFD in two different ways. Therefore the peptide was either directly bound to the particle surface or indirectly to a particle-protecting polyethylene glycol (PEG) ligand shell, thereby reducing the CLPFFD density on the surfaces of both types of particles. Fully PEGylated particles were used for comparison. The effects on cell viability and the fundamental suitability of the HAuNS and AuNR conjugates as photothermal absorbers to inhibit A beta-fibrillation are analysed in vitro.
