Caracterización genética de ovinos criollos colombianos
ABSTRACT: indigenous breeds are important for poor farmers because of their natural selection against harsh environments and adaptation to regional conditions. However, inbreeding of indigenous sheep populations has increased in Colombia due to indiscriminate cross-breeding with foreign animals and lack of reproductive controls, with subsequent loss in productivity, which poses a great risk for the conservation of valuable genes. Objective: to determine the genetic diversity in Colombian indigenous sheep by using a panel of 10 microsatellite molecular markers. Methods: blood samples from 362 individuals from 43 farms in 11 Colombian provinces were genotyped and analyzed for a panel of 10 microsatellite markers. Results: a total of 134 alleles were found (13.4 alleles/locus on average) with a range of observed and expected heterozygosity of 0.428 to 0.831 and 0.615 to 0.855, respectively, and 0.742 polymorphic information content (PIC). The average Wright F-statistics (FIS ) of the breeds was 0.107, suggesting moderate levels of inbreeding. Colombian sheep showed a low level of genetic differentiation among breeds (FST = 0.054) and STRUCTURE analysis showed complex patterns of admixture in the breeds. Conclusion: overall, Colombian sheep have high genetic variability, which is very important for future conservation programs and genetic improvement.RESUMEN: las razas animales autóctonas son importantes para los agricultores de escasos recursos a causa de su selección natural contra el duro ambiente y su adaptación a condiciones regionales. Sin embargo en Colombia, debido al cruce indiscriminado con razas foráneas y a la falta de control de la reproducción, ha aumentado la consanguinidad en las poblaciones de ovinos criollos y por lo tanto la pérdida en la productividad, lo que supone un gran riesgo para la conservación de genes valiosos. Objetivo: determinar la diversidad genética en razas criollas de ovinos colombianos utilizando análisis por microsatélites. Métodos: se visitaron 43 granjas localizadas en 11 departamentos del país, en las cuales se tomaron muestras de sangre a 362 individuos. Las muestras fueron genotipadas y analizadas para un panel de 10 marcadores microsatélites. Resultados: un total de 134 alelos fueron encontrados (13,4 alelos/locus en promedio), con un rango de heterocigocidad observada y esperada de 0,428 a 0,831 y 0,615 a 0,855, respectivamente, y un contenido de información polimórfica (PIC) promedio de 0,742. El Wright F-statistics (FIS) promedio de las razas evaluadas fue 0,107, lo cual sugiere que las razas tienen niveles moderados de consanguinidad. Las ovejas colombianas presentaron un bajo grado de diferenciación genética entre las distintas razas (FST = 0,054) y el análisis de STRUCTURE mostro complejos patrones de mezcla en las razas estudiadas. Conclusión: en términos generales, las ovejas colombianas presentan una alta variabilidad genética lo cual es muy importante para futuros programas de conservacion y mejoramiento genetico
Sucrose esters from Physalis peruviana calyces with anti-inflammatory activity
Physalis peruviana is a native plant from the South American Andes and is widely used in tra- ditional Colombian medicine of as an anti-inflam- matory medicinal plant, specifically the leaves, calyces, and small stems in poultice form. Pre- vious studies performed by our group on P. pe- ruviana calyces showed potent anti-inflamma- tory activity in an enriched fraction obtained from an ether total extract. The objective of the present study was to obtain and elucidate the ac- tive compounds from this fraction and evaluate their anti-inflammatory activity in vivo and in vi- tro. The enriched fraction of P. peruviana was pu- rified by several chromatographic methods to ob- tain an inseparable mixture of two new sucrose esters named peruviose A (1) and peruviose B (2). Structures of the new compounds were eluci- dated using spectroscopic methods and chemical transformations. The anti-inflammatory activity of the peruvioses mixture was evaluated using λ-carrageenan-induced paw edema in rats and lipopolysaccharide-activated peritoneal macro- phages. Results showed that the peruvioses did not produce side effects on the liver and kidneys and significantly attenuated the inflammation in- duced by λ-carrageenan in a dosage-dependent manner, probably due to an inhibition of nitric oxide and prostaglandin E2, which was demon-
strated in vitro. To our knowledge, this is the first report of the presence of sucrose esters in P. pe- ruviana that showed a potent anti-inflammatory effect. These results suggest the potential of su- crose esters from the Physalis genus as a novel natural alternative to treat inflammatory diseases
Tuberculosis due to Mycobacterium bovis and Mycobacterium caprae in sheep
Tuberculosis was diagnosed in three flocks of sheep in Galicia, Spain, in 2009 and 2010. Two flocks were infected with Mycobacterium bovis and one flock was infected with Mycobacterium caprae. Infection was confirmed by the comparative intradermal tuberculin test, bacteriology, molecular analysis and histopathology. Sheep have the potential to act as a reservoir for tuberculosis
Cold ischemia >4 hours increases heart transplantation mortality. An analysis of the Spanish heart transplantation registry
[Abstract]
Background.
Cold ischemia time (CIT) has been associated to heart transplantation (HT) prognosis. However, there is still uncertainty regarding the CIT cutoff value that might have relevant clinical implications.
Methods.
We analyzed all adults that received a first HT during the period 2008–2018. CIT was defined as the time between the cross-clamp of the donor aorta and the reperfusion of the heart. Primary outcome was 1-month mortality.
Results.
We included 2629 patients, mean age was 53.3 ± 12.1 years and 655 (24.9%) were female. Mean CIT was 202 ± 67 min (minimum 20 min, maximum 600 min). One-month mortality per CIT quartile was 9, 12, 13, and 19%. One-year mortality per CIT quartile was 16, 19, 21, and 28%. CIT was an independent predictor of 1-month mortality, but only in the last quartile of CIT >246 min (odds ratio 2.1, 95% confidence interval 1.49–3.08, p < .001). We found no relevant differences in CIT during the study period. However, the impact of CIT in 1-month and 1-year mortality decreased with time (p value for the distribution of ischemic time by year 0.01), particularly during the last 5 years.
Conclusions.
Although the impact of CIT in HT prognosis seems to be decreasing in the last years, CIT in the last quartile (>246 min) is associated with 1-month and 1-year mortality. Our findings suggest the need to limit HT with CIT > 246 min or to use different myocardial preservation systems if the expected CIT is >4 h
Venoarterial Extracorporeal Membrane Oxygenation with or without Simultaneous Intra-Aortic Balloon Pump Support as a Direct Bridge to Heart Transplantation: Results from a Nationwide Spanish Registry
[Abstract]
OBJECTIVES
To investigate the potential clinical benefit of an intra-aortic balloon pump (IABP) in patients supported with venoarterial extracorporeal membrane oxygenation (VA-ECMO) as a bridge to heart transplantation (HT).
METHODS
We studied 169 patients who were listed for urgent HT under VA-ECMO support at 16 Spanish institutions from 2010 to 2015. The clinical outcomes of patients under simultaneous IABP support (n = 73) were compared to a control group of patients without IABP support (n = 96).
RESULTS
There were no statistically significant differences between the IABP and control groups with regard to the cumulative rates of transplantation (71.2% vs 81.2%, P = 0.17), death during VA-ECMO support (20.6% vs 14.6%, P = 0.31), transition to a different mechanical circulatory support device (5.5% vs 5.2%, P = 0.94) or weaning from VA-ECMO support due to recovery (2.7% vs 0%, P = 0.10). There was a higher incidence of bleeding events in the IABP group (45.2% vs 25%, P = 0.006; adjusted odds ratio 2.18, 95% confidence interval 1.02–4.67). In-hospital postoperative mortality after HT was 34.6% in the IABP group and 32.5% in the control group (P = 0.80). One-year survival after listing for urgent HT was 53.3% in the IABP group and 52.2% in the control group (log rank P = 0.75). Multivariate adjustment for potential confounders did not change this result (adjusted hazard ratio 0.94, 95% confidence interval 0.56–1.58).
CONCLUSIONS
In our study, simultaneous IABP therapy in transplant candidates under VA-ECMO support did not significantly reduce morbidity or mortality
Influence of Gender in Advanced Heart Failure Therapies and Outcome Following Transplantation
Biological differences between males and females change the course of different diseases and affect therapeutic measures' responses. Heart failure is not an exception to these differences. Women account for a minority of patients on the waiting list for heart transplantation or other advanced heart failure therapies. The reason for this under-representation is unknown. Men have a worse cardiovascular risk profile and suffer more often from ischemic heart disease. Conversely, transplanted women are younger and more frequently have non-ischemic cardiac disorders. Women's poorer survival on the waiting list for heart transplantation has been previously described, but this trend has been corrected in recent years. The use of ventricular assist devices in women is progressively increasing, with comparable results than in men. The indication rate for a heart transplant in women (number of women on the waiting list for millions of habitants) has remained unchanged over the past 25 years. Long-term results of heart transplants are equal for both men and women. We have analyzed the data of a national registry of heart transplant patients to look for possible future directions for a more in-depth study of sex differences in this area. We have analyzed 1-year outcomes of heart transplant recipients. We found similar results in men and women and no sex-related interactions with any of the factors related to survival or differences in death causes between men and women. We should keep trying to approach sex differences in prospective studies to confirm if they deserve a different approach, which is not supported by current evidence
