    Soft Palate Modification Using a Collagen Crosslinking Reagent for Equine Dorsal Displacement of the Soft Palate and Other Upper Airway Breathing Disorders

    The mechanical properties of the soft palate can be associated with breathing abnormalities. Dorsal displacement of the soft palate (DDSP) is a naturally occurring equine soft palate disorder caused by displacement of the caudal edge of the soft palate. Snoring and a more serious, sometimes life-threatening, condition called obstructive sleep apnea (OSA) are forms of sleep-related breathing disorders in humans which may involve the soft palate. The goal of this study was to investigate the effect of injecting the protein crosslinker genipin into the soft palate to modify its mechanical properties for the treatment of equine DDSP with potential implications for the treatment of snoring and OSA in humans. Ex vivo experiments consisted of mechanical testing and a wind tunnel study to examine the effect of genipin on the mechanical properties, displacement, and vibration of equine soft palates. A pilot in vivo study was completed using DDSP and control horses to test the safety and effectiveness of injecting a genipin reagent into the soft palate. The wind tunnel testing demonstrated a greater than 50% decrease in transient deformation and a greater than 33% decrease in steady-state vibrations for all doses of genipin tested. Ultimate tensile stress, yield stress, and Young’s modulus were higher in the genipin-treated distal soft palate specimens by 52%, 53%, and 63%, respectively. The pilot in vivo study showed a reduction of snoring loudness in all DDSP horses and elimination of DDSP in at least one of three horses. The difficulty of using a 1-meter-long endoscopic injection needle contributed to a consistent overinjection of the equine soft palates, causing excessive stretching (pillowing) and related degradation of the tissue. These ex vivo and in vivo results demonstrated reduced vibration amplitude and flaccidity and increased strength of genipin-treated soft palates, suggesting that genipin crosslinking could become an effective and safe treatment for soft palate related breathing abnormalities

    Low-cost automated vectors and modular environmental sensors for plant phenotyping

High-throughput plant phenotyping in controlled environments (growth chambers and glasshouses) is often delivered via large, expensive installations, leading to limited access and the increased relevance of “affordable phenotyping” solutions. We present two robot vectors for automated plant phenotyping under controlled conditions. Using 3D-printed components and readily-available hardware and electronic components, these designs are inexpensive, flexible and easily modified to multiple tasks. We present a design for a thermal imaging robot for high-precision time-lapse imaging of canopies and a Plate Imager for high-throughput phenotyping of roots and shoots of plants grown on media plates. Phenotyping in controlled conditions requires multi-position spatial and temporal monitoring of environmental conditions. We also present a low-cost sensor platform for environmental monitoring based on inexpensive sensors, microcontrollers and internet-of-things (IoT) protocols

    Pasture Production and Grazing System in ASSIST Project: Outcome and Replication Viability in Nigeria for Sustainable Ruminant Production

    As important as pasture is to productive management of cattle, sheep and goats, intensive development of grassland for ruminant animals is rare in Nigeria largely due to no conscious effort at pasture cultivation in the country. Consequently, nomadic and free grazing systems remain a practice in the country, with resultant poor animal productivity and agro-related conflicts between ruminant keepers and crop farmers and/or agro-processors. The need to revert this situation underscored the exploration of the potential value of the innovative pasture production and grazing management system tested under the Achieving Sustainable Agricultural Systems (ASSIST) programmme in Rothamsted Research, United Kingdom, for application in Nigeria. The project, which cultivated swards of different species such as permanent pasture, grass mix (“soft fibre” cultivars) and multispecies swards, was managed as continuous stocking orcell-rotational grazing systems under drained and undrained soil conditions. Conscientious management and weekly monitoring of the pasture, using calibrated plate-meter, not only made it possible to ensure adequate herbage availability to the reared ruminants but also enhanced productive performance of the farm animals. Although, the common types of swards in Nigeria differ from that of ASSIST project, a modified application of the paddock development model could enhance quality paddock development in the country for sustainable grazing and productive ruminant management

    Acute Effects of Aerosolized S-Nitrosoglutathione in Cystic Fibrosis

    This is the publisher's version, also available electronically from http://www.atsjournals.org/doi/abs/10.1164/ajrccm.165.7.2105032#.VJCPDXs4d41.S-Nitrosoglutathione (GSNO), a naturally occurring constituent of airway lining fluid, enhances ciliary motility, relaxes airway smooth muscle, inhibits airway epithelial amiloride-sensitive sodium transport, and prevents pathogen replication. Remarkably, airway levels of GSNO are low in patients with cystic fibrosis (CF). We hypothesized that replacement of airway GSNO would improve gas exchange in CF. In a double-blind, placebo controlled study, we administered 0.05 ml/kg of 10 mM GSNO or phosphate buffered saline by aerosol to patients with CF and followed oxygen saturation, spirometry, respiratory rate, blood pressure, heart rate, and expired nitric oxide (NO). Nine patients received GSNO and 11 placebo. GSNO inhalation was associated with a modest but sustained increase in oxygen saturation at all time points. Expired NO increased in the low ppb range with GSNO treatment, peaking at 5 minutes but remaining above baseline at 30 minutes. There were no adverse effects. We conclude that GSNO is well tolerated in patients with CF and improves oxygenation through a mechanism that may be independent of free NO. Further, GSNO breakdown increases expired NO. We suggest that therapy aimed at restoring endogenous GSNO levels in the CF airway may merit study

    An enhanced functional ability questionnaire (faVIQ) to measure the impact of rehabilitation services on the visually impaired

    AIM To develop a short, enhanced functional ability Quality of Vision (faVIQ) instrument based on previous questionnaires employing comprehensive modern statistical techniques to ensure the use of an appropriate response scale, items and scoring of the visual related difficulties experienced by patients with visual impairment. METHODS Items in current quality-of-life questionnaires for the visually impaired were refined by a multi-professional group and visually impaired focus groups. The resulting 76 items were completed by 293 visually impaired patients with stable vision on two occasions separated by a month. The faVIQ scores of 75 patients with no ocular pathology were compared to 75 age and gender matched patients with visual im pairm ent. RESULTS Rasch analysis reduced the faVIQ items to 27. Correlation to standard visual metrics was moderate (r=0.32-0.46) and to the NEI-VFQ was 0.48. The faVIQ was able to clearly discriminate between age and gender matched populations with no ocular pathology and visual impairment with an index of 0.983 and 95% sensitivity and 95% specificity using a cut off of 29. CONCLUSION The faVIQ allows sensitive assessm ent of quality-of-life in the visually im paired and should support studies which evaluate the effectiveness of low vision rehabilitation services. © Copyright International Journal of Ophthalmology Press

    Late-season snowfall is associated with decreased offspring survival in two migratory arctic-breeding songbird species

    While the effect of weather on reproduction has been studied for many years in avian taxa, the rapid pace of climate change in arctic regions has added urgency to this question by changing the weather conditions species experience during breeding. Given this, it is important to understand how factors such as temperature, rain, snowfall, and wind affect reproduction both directly and indirectly (e.g. through their effects on food availability). In this study, we ask how weather factors and food availability influence daily survival rates of clutches in two arctic-breeding migratory songbirds: the Lapland longspur Calcarius lapponicus , a circumpolar breeder, and Gambel’s white-crowned sparrow Zonotrichia leucophrys gambelii , which breeds in shrubby habitats across tundra, boreal and continental climates. To do this, we monitored clutch survival in these two species from egg-lay through fledge at field sites located near Toolik Field Station (North Slope, Alaska) across 5 yr (2012–2016). Our results indicate that snowfall and cold temperatures decreased offspring survival rates in both species; although Lapland longspurs were more susceptible to snowfall. Food availability, quantified by pitfall sampling and sweep-net sampling methods, had minimal effects on offspring survival. Some climate models predict increased precipitation for the Arctic with global warming, and in the Toolik region, total snow accumulation may be increasing. Placed in this context, our results suggest that changes in snow storms with climate change could have substantial consequences for reproduction in migratory songbirds breeding in the North American Arctic

    Late Holocene isotope hydrology of Lake Qinghai, NE Tibetan Plateau: effective moisture variability and atmospheric circulation changes

    A sub-centennial-resolution record of lacustrine carbonate oxygen isotopes (ÎŽ<sup>18</sup>O<sub>C</sub>) from the closed-basin Lake Qinghai on the NE Tibetan Plateau shows pronounced variability over the past 1500 years. Changes in ÎŽ<sup>18</sup>O<sub>C</sub> in hydrologically closed lakes are often interpreted in terms of changing effective moisture. Under this interpretation our record would imply increasing effective moisture during the Little Ice Age (LIA) compared to the Medieval Warm Period (MWP). However, independent evidence from other archives strongly suggests the Asian summer monsoon was stronger during the MWP and weakened during the LIA. Controls other than effective moisture (the balance of water inputs over evaporative loss) must therefore have contributed to the ÎŽ<sup>18</sup>O<sub>C</sub> values. We propose the LIA signal in Lake Qinghai resulted from a reduction in evaporation caused by colder air temperatures, coupled with a decrease in oxygen isotope composition of input waters as a result of an increase in the relative importance of westerly-derived precipitation. Our results caution against simplistic interpretations of carbonate oxygen isotope records from hydrologically closed lakes and suggest all possible controlling factors must be taken into account in order to avoid misleading palaeoclimatic reconstructions

    Interactions between environmental contaminants and gastrointestinal parasites: novel insights from an integrative approach in a marine predator

    Environmental contaminants and parasites are ubiquitous stressors that can affect animal physiology and derive from similar dietary sources (co-exposure). To unravel their interactions in wildlife, it is thus essential to quantify their concurring drivers. Here, the relationship between blood contaminant residues (11 trace elements and 17 perfluoroalkyl substances) and nonlethally quantified gastrointestinal parasite loads was tested while accounting for intrinsic (sex, age, and mass) and extrinsic factors (trophic ecology inferred from stable isotope analyses and biologging) in European shags Phalacrocorax aristotelis. Shags had high mercury (range 0.65–3.21 ÎŒg g–1 wet weight, ww) and extremely high perfluorooctanoic acid (PFOA) and perfluorononanoic acid (PFNA) residues (3.46–53 and 4.48–44 ng g–1 ww, respectively). Males had higher concentrations of arsenic, mercury, PFOA, and PFNA than females, while the opposite was true for selenium, perfluorododecanoic acid (PFDoA), and perfluooctane sulfonic acid (PFOS). Individual parasite loads (Contracaecum rudolphii) were higher in males than in females. Females targeted pelagic-feeding prey, while males relied on both pelagic- and benthic-feeding organisms. Parasite loads were not related to trophic ecology in either sex, suggesting no substantial dietary co-exposure with contaminants. In females, parasite loads increased strongly with decreasing selenium:mercury molar ratios. Females may be more susceptible to the interactive effects of contaminants and parasites on physiology, with potential fitness consequences

    Spitzer View of Young Massive Stars in the LMC HII Complexes. II. N159

    The HII complex N159 in the Large Magellanic Cloud (LMC) is used to study massive star formation in different environments, as it contains three giant molecular clouds (GMCs) that have similar sizes and masses but exhibit different intensities of star formation. We identify candidate massive young stellar objects (YSOs) using infrared photometry, and model their SEDs to constrain mass and evolutionary state. Good fits are obtained for less evolved Type I, I/II, and II sources. Our analysis suggests that there are massive embedded YSOs in N159B, a maser source, and several ultracompact HII regions. Massive O-type YSOs are found in GMCs N159-E and N159-W, which are associated with ionized gas, i.e., where massive stars formed a few Myr ago. The third GMC, N159-S, has neither O-type YSOs nor evidence of previous massive star formation. This correlation between current and antecedent formation of massive stars suggests that energy feedback is relevant. We present evidence that N159-W is forming YSOs spontaneously, while collapse in N159-E may be triggered. Finally, we compare star formation rates determined from YSO counts with those from integrated H-alpha and 24 micron luminosities and expected from gas surface densities. Detailed dissection of extragalactic GMCs like the one presented here is key to revealing the physics underlying commonly used star formation scaling laws.Comment: 60 pages, 11 figures.
