    Anti-depressive effectiveness of olanzapine, quetiapine, risperidone and ziprasidone: a pragmatic, randomized trial

    <p>Abstract</p> <p>Background</p> <p>Efficacy studies indicate anti-depressive effects of at least some second generation antipsychotics (SGAs). The Bergen Psychosis Project (BPP) is a 24-month, pragmatic, industry-independent, randomized, head-to-head comparison of olanzapine, quetiapine, risperidone and ziprasidone in patients acutely admitted with psychosis. The aim of the study is to investigate whether differential anti-depressive effectiveness exists among SGAs in a clinically relevant sample of patients acutely admitted with psychosis.</p> <p>Methods</p> <p>Adult patients acutely admitted to an emergency ward for psychosis were randomized to olanzapine, quetiapine, risperidone or ziprasidone and followed for up to 2 years. Participants were assessed repeatedly using the Positive and Negative Syndrome Scale - Depression factor (PANSS-D) and the Calgary Depression Scale for Schizophrenia (CDSS).</p> <p>Results</p> <p>A total of 226 patients were included. A significant time-effect showing a steady decline in depressive symptoms in all medication groups was demonstrated. There were no substantial differences among the SGAs in reducing the PANSS-D score or the CDSS sum score. Separate analyses of groups with CDSS sum scores > 6 or ≤6, respectively, reflecting degree of depressive morbidity, revealed essentially identical results to the primary analyses. There was a high correlation between the PANSS-D and the CDSS sum score (r = 0.77; p < 0.01).</p> <p>Conclusions</p> <p>There was no substantial difference in anti-depressive effectiveness among olanzapine, quetiapine, risperidone or ziprasidone in this clinically relevant sample of patients acutely admitted to hospital for symptoms of psychosis.     Highly mixed impacts of near-future climate change on stock productivity proxies in the North East Atlantic

    Impacts of climate change on ocean productivity sustaining world fisheries are predominantly negative but vary greatly among regions. We assessed how 39 fisheries resources—ranging from data-poor to data-rich stocks—in the North East Atlantic are most likely affected under the intermediate climate emission scenario RCP4.5 towards 2050. This region is one of the most productive waters in the world but subjected to pronounced climate change, especially in the northernmost part. In this climate impact assessment, we applied a hybrid solution combining expert opinions (scorings)—supported by an extensive literature review—with mechanistic approaches, considering stocks in three different large marine ecosystems, the North, Norwegian and Barents Seas. This approach enabled calculation of the directional effect as a function of climate exposure and sensitivity attributes (life-history schedules), focusing on local stocks (conspecifics) across latitudes rather than the species in general. The resulting synopsis (50–82°N) contributes substantially to global assessments of major fisheries (FAO, The State of World Fisheries and Aquaculture, 2020), complementing related studies off northeast United States (35–45°N) (Hare et al., PLoS One, 2016, 11, e0146756) and Portugal (37–42°N) (Bueno-Pardo et al., Scientific Reports, 2021, 11, 2958). Contrary to prevailing fisheries forecasts elsewhere, we found that most assessed stocks respond positively. However, the underlying, extensive environmental clines implied that North East Atlantic stocks will develop entirely different depending upon the encountered stressors: cold-temperate stocks at the southern and Arctic stocks at the northern fringes appeared severely negatively impacted, whereas warm-temperate stocks expanding from south were found to do well along with cold-temperate stocks currently inhabiting below-optimal temperatures in the northern subregion.publishedVersio

    Integration of micro-gravity and geodetic data to constrain shallow system mass changes at Krafla Volcano, N Iceland

    New and previously published micro-gravity data are combined with InSAR data, precise levelling and GPS measurements to produce a model for the processes operating at Krafla volcano, 20 years after its most recent eruption. The data have been divided into two periods: from 1990 to 1995 and from 1996 to 2003 and show that the rate of deflation at Krafla is decaying exponentially. The net micro-gravity change at the centre of the caldera is shown, using the measured Free Air Gradient, to be -85 ÎźGal for the first and -100 ÎźGal for the second period. After consideration of the effects of water extraction by the geothermal power station within the caldera, the net gravity decreases are -73 Âą 17 ÎźGal for the first and -65 Âą 17 ÎźGal for the second period. These decreases are interpreted in terms of magma drainage. Following a Mogi point source model we calculate the mass decrease to be ~2 x 1010 kg/yr reflecting a drainage rate of ~0.23 m3/s, similar to the ~0.13 m3/s drainage rate previously found at Askja volcano, N-Iceland. Based on the evidence for deeper magma reservoirs and the similarity between the two volcanic systems, we suggest a pressure-link between Askja and Krafla at deeper levels (at the lower crust or the crust-mantle boundary). After the Krafla fires, co-rifting pressure decrease of a deep source at Krafla stimulated the subsequent inflow of magma, eventually affecting conditions along the plate boundary in N-Iceland, as far away as Askja. We anticipate that the pressure of the deeper reservoir at Krafla will reach a critical value and eventually magma will rise from there to the shallow magma chamber, possibly initiating a new rifting episode. We have demonstrated that by examining micro-gravity and geodetic data, our knowledge of active volcanic systems can be significantly improved

    The 10th Biennial Hatter Cardiovascular Institute workshop: cellular protection—evaluating new directions in the setting of myocardial infarction, ischaemic stroke, and cardio-oncology

    Due to its poor capacity for regeneration, the heart is particularly sensitive to the loss of contractile cardiomyocytes. The onslaught of damage caused by ischaemia and reperfusion, occurring during an acute myocardial infarction and the subsequent reperfusion therapy, can wipe out upwards of a billion cardiomyocytes. A similar program of cell death can cause the irreversible loss of neurons in ischaemic stroke. Similar pathways of lethal cell injury can contribute to other pathologies such as left ventricular dysfunction and heart failure caused by cancer therapy. Consequently, strategies designed to protect the heart from lethal cell injury have the potential to be applicable across all three pathologies. The investigators meeting at the 10th Hatter Cardiovascular Institute workshop examined the parallels between ST-segment elevation myocardial infarction (STEMI), ischaemic stroke, and other pathologies that cause the loss of cardiomyocytes including cancer therapeutic cardiotoxicity. They examined the prospects for protection by remote ischaemic conditioning (RIC) in each scenario, and evaluated impasses and novel opportunities for cellular protection, with the future landscape for RIC in the clinical setting to be determined by the outcome of the large ERIC-PPCI/CONDI2 study. It was agreed that the way forward must include measures to improve experimental methodologies, such that they better reflect the clinical scenario and to judiciously select combinations of therapies targeting specific pathways of cellular death and injury

    Chemical Magnetoreception: Bird Cryptochrome 1a Is Excited by Blue Light and Forms Long-Lived Radical-Pairs

    Cryptochromes (Cry) have been suggested to form the basis of light-dependent magnetic compass orientation in birds. However, to function as magnetic compass sensors, the cryptochromes of migratory birds must possess a number of key biophysical characteristics. Most importantly, absorption of blue light must produce radical pairs with lifetimes longer than about a microsecond. Cryptochrome 1a (gwCry1a) and the photolyase-homology-region of Cry1 (gwCry1-PHR) from the migratory garden warbler were recombinantly expressed and purified from a baculovirus/Sf9 cell expression system. Transient absorption measurements show that these flavoproteins are indeed excited by light in the blue spectral range leading to the formation of radicals with millisecond lifetimes. These biophysical characteristics suggest that gwCry1a is ideally suited as a primary light-mediated, radical-pair-based magnetic compass receptor

    New genetic loci implicated in fasting glucose homeostasis and their impact on type 2 diabetes risk.

    Levels of circulating glucose are tightly regulated. To identify new loci influencing glycemic traits, we performed meta-analyses of 21 genome-wide association studies informative for fasting glucose, fasting insulin and indices of beta-cell function (HOMA-B) and insulin resistance (HOMA-IR) in up to 46,186 nondiabetic participants. Follow-up of 25 loci in up to 76,558 additional subjects identified 16 loci associated with fasting glucose and HOMA-B and two loci associated with fasting insulin and HOMA-IR. These include nine loci newly associated with fasting glucose (in or near ADCY5, MADD, ADRA2A, CRY2, FADS1, GLIS3, SLC2A2, PROX1 and C2CD4B) and one influencing fasting insulin and HOMA-IR (near IGF1). We also demonstrated association of ADCY5, PROX1, GCK, GCKR and DGKB-TMEM195 with type 2 diabetes. Within these loci, likely biological candidate genes influence signal transduction, cell proliferation, development, glucose-sensing and circadian regulation. Our results demonstrate that genetic studies of glycemic traits can identify type 2 diabetes risk loci, as well as loci containing gene variants that are associated with a modest elevation in glucose levels but are not associated with overt diabetes

    Coexisting Cyclic Parthenogens Comprise a Holocene Species Flock in Eubosmina

    Background: Mixed breeding systems with extended clonal phases and weak sexual recruitment are widespread in nature but often thought to impede the formation of discrete evolutionary clusters. Thus, cyclic parthenogens, such as cladocerans and rotifers, could be predisposed to ‘‘species problems’ ’ and a lack of discrete species. However, species flocks have been proposed for one cladoceran group, Eubosmina, where putative species are sympatric, and there is a detailed paleolimnological record indicating a Holocene age. These factors make the Eubosmina system suitable for testing the hypotheses that extended clonal phases and weak sexual recruitment inhibit speciation. Although common garden experiments have revealed a genetic component to the morphotypic variation, the evolutionary significance of the morphotypes remains controversial. Methodology/Principal Findings: In the present study, we tested the hypothesis of a single polymorphic species (i.e., mixing occurs but selection maintains genes for morphology) in four northern European lakes where the morphotypes coexist. Our evidence is based on nuclear DNA sequence, mitochondrial DNA sequence, and morphometric analysis of coexisting morphotypes. We found significant genetic differentiation, genealogical exclusivity, and morphometric differentiation for coexisting morphotypes.
