    Remediation of chromium(VI) at polypyrrole-coated titanium

    The application of conducting polypyrrole (PPy) and polyaniline (PAni) coated substrates in remediation of chromium, Cr(VI), is an area of considerable interest. Here, we discuss the implementation of PPy-coated titanium as a new material for the reduction of Cr(VI) to the less toxic trivalent state, Cr(III). An alkaline-peroxide based etching process was used to ensure the adhesion of the PPy coatings to the underlying titanium. The PPy films showed excellent resistance to acidic Cr(VI) solutions and remained adherent after continuous exposure to the solutions. In order to optimise the remediation process a number of experimental parameters were investigated, including the thickness of the PPy coating, the reduction potential used in pre-treatment of the PPy and the degree of solution agitation. The durability of the materials on exposure to the Cr(VI) test solutions made them suitable for repeated remediation experiments. Following several test-runs, the cleanup efficiency of the material was found to decrease slightly, however, increasing the exposure/experiment time resulted in significantly improved cleanup ability

    Efficient depletion of host DNA contamination in malaria clinical sequencing.

    The cost of whole-genome sequencing (WGS) is decreasing rapidly as next-generation sequencing technology continues to advance, and the prospect of making WGS available for public health applications is becoming a reality. So far, a number of studies have demonstrated the use of WGS as an epidemiological tool for typing and controlling outbreaks of microbial pathogens. Success of these applications is hugely dependent on efficient generation of clean genetic material that is free from host DNA contamination for rapid preparation of sequencing libraries. The presence of large amounts of host DNA severely affects the efficiency of characterizing pathogens using WGS and is therefore a serious impediment to clinical and epidemiological sequencing for health care and public health applications. We have developed a simple enzymatic treatment method that takes advantage of the methylation of human DNA to selectively deplete host contamination from clinical samples prior to sequencing. Using malaria clinical samples with over 80% human host DNA contamination, we show that the enzymatic treatment enriches Plasmodium falciparum DNA up to ∼9-fold and generates high-quality, nonbiased sequence reads covering >98% of 86,158 catalogued typeable single-nucleotide polymorphism loci

    1 m long multilayer-coated deformable piezoelectric bimorph mirror for adjustable focusing of high-energy X-rays

    The Diamond Light Source (DLS) beamline I15-1 measures atomic pair distribution functions (PDF) using scattering of 40-80 keV X-rays. A unique focusing element was needed to condense these X-rays from an initial large cross section (11.0 mm H × 4.2 mm V) into a required spot size of FWHM ≈680 μm (H) × 20 μm (V) at a variable position between the sample and the detector. The large numerical aperture is achieved by coating a silicon substrate over 1 m long with three multilayer stripes of Bragg angle 4.2 mrad. One stripe selects X-rays of each energy 40.0, 65.4, and 76.6 keV. Sixteen piezoelectric bimorph actuators attached to the sides of the mirror substrate adjusted the reflecting surface’s shape. Focal spots of vertical width < 15 μm were obtained at three positions over a 0.92 m range, with fast, easy switching from one focal position to another. Minimized root mean square slope errors were close to 0.5 μrad after subtraction of a uniform curvature. Reflectivity curves taken along each stripe showed consistent high peaks with generally small angular variation of peak positions. This is the first application of a 1 m long multilayercoated bimorph mirror at a synchrotron beamline. Data collected with its help on a slice of a lithium ion battery’s cathode are presented

    Natural Wormholes as Gravitational Lenses

    Visser has suggested traversable 3-dimensional wormholes that could plausibly form naturally during Big Bang inflation. A wormhole mouth embedded in high mass density might accrete mass, giving the other mouth a net *negative* mass of unusual gravitational properties. The lensing of such a gravitationally negative anomalous compact halo object (GNACHO) will enhance background stars with a time profile that is observable and qualitatively different from that recently observed for massive compact halo objects (MACHOs) of positive mass.     Distant field BHB stars and the mass of the Galaxy II: Photometry and spectroscopy of UKST candidates 16<B<19.5, 11<R<52 kpc

    This is the second in a series of papers presenting a new calculation of the mass of the Galaxy based on radial velocities and distances for a sample of faint 16 < B < 21.3 field blue horizontal-branch (BHB) stars. We present accurate BV CCD photometry and spectra for 142 candidate A-type stars selected from ub_jr photometry of UK Schmidt telescope plates in six high-Galactic-latitude fields. Classification of these candidates produces a sample of 60 BHB stars at distances of 11-52 kpc from the Sun (mean 28 kpc), with heliocentric line-of-sight velocities accurate to 15 km/s, and distance errors < 10%. We provide a summary table listing coordinates and velocities of these stars. The measured dispersion of the radial component of the Galactocentric velocity for this sample is 108+-10 km/s, in agreement with a recent study of the distant halo by Sirko and coworkers. Measurements of the Ca II K line indicate that nearly all the stars are metal-poor with a mean [Fe/H] = -1.8 with dispersion 0.5. Subsequent papers will describe a second survey of BHBs to heliocentric distances 70 < R < 125 kpc and present a new estimate of the mass of the Galaxy.Comment: 16 pages, 15 figures.     The Science of Sungrazers, Sunskirters, and Other Near-Sun Comets

    This review addresses our current understanding of comets that venture close to the Sun, and are hence exposed to much more extreme conditions than comets that are typically studied from Earth. The extreme solar heating and plasma environments that these objects encounter change many aspects of their behaviour, thus yielding valuable information on both the comets themselves that complements other data we have on primitive solar system bodies, as well as on the near-solar environment which they traverse. We propose clear definitions for these comets: We use the term near-Sun comets to encompass all objects that pass sunward of the perihelion distance of planet Mercury (0.307 AU). Sunskirters are defined as objects that pass within 33 solar radii of the Sun’s centre, equal to half of Mercury’s perihelion distance, and the commonly-used phrase sungrazers to be objects that reach perihelion within 3.45 solar radii, i.e. the fluid Roche limit. Finally, comets with orbits that intersect the solar photosphere are termed sundivers. We summarize past studies of these objects, as well as the instruments and facilities used to study them, including space-based platforms that have led to a recent revolution in the quantity and quality of relevant observations. Relevant comet populations are described, including the Kreutz, Marsden, Kracht, and Meyer groups, near-Sun asteroids, and a brief discussion of their origins. The importance of light curves and the clues they provide on cometary composition are emphasized, together with what information has been gleaned about nucleus parameters, including the sizes and masses of objects and their families, and their tensile strengths. The physical processes occurring at these objects are considered in some detail, including the disruption of nuclei, sublimation, and ionisation, and we consider the mass, momentum, and energy loss of comets in the corona and those that venture to lower altitudes. The different components of comae and tails are described, including dust, neutral and ionised gases, their chemical reactions, and their contributions to the near-Sun environment. Comet-solar wind interactions are discussed, including the use of comets as probes of solar wind and coronal conditions in their vicinities. We address the relevance of work on comets near the Sun to similar objects orbiting other stars, and conclude with a discussion of future directions for the field and the planned ground- and space-based facilities that will allow us to address those science topics

    β2-adrenoceptor-induced modulation of transglutaminase 2 transamidase activity in cardiomyoblasts

    Tissue transglutaminase 2 (TG2) is modulated by protein kinase A (PKA) mediated phosphorylation: however, the precise mechanism(s) of its modulation by G-protein coupled receptors coupled to PKA activation are not fully understood. In the current study we investigated the potential regulation of TG2 activity by the β2-adrenoceptor in rat H9c2 cardiomyoblasts. Transglutaminase transamidation activity was assessed using amine-incorporating and protein cross-linking assays. TG2 phosphorylation was determined via immunoprecipitation and Western blotting. The long acting β2-adrenoceptor agonist formoterol induced time- and concentration-dependent increases in TG2 transamidation. Increases in TG2 activity were reduced by the TG2 inhibitors Z-DON (Benzyloxycarbonyl-(6-Diazo-5-oxonorleucinyl)-L-valinyl-L-prolinyl-L-leucinmethylester) and R283 (1,3,dimethyl-2[2-oxo-propyl]thio)imidazole chloride). Responses to formoterol were blocked by pharmacological inhibition of PKA, extracellular signal-regulated kinase 1 and 2 (ERK1/2), or phosphatidylinositol 3-kinase (PI-3K) signalling. Furthermore, the removal of extracellular Ca2+ also attenuated formoterol-induced TG2 activation. Fluorescence microscopy demonstrated TG2-induced biotin-X-cadaverine incorporation into proteins. Formoterol increased the levels of TG2-associated phosphoserine and phosphothreonine, which were blocked by inhibition of PKA, ERK1/2 or PI-3K signalling. Subsequent proteomic analysis identified known (e.g. lactate dehydrogenase A chain) and novel (e.g. Protein S100-A6) protein substrates for TG2. Taken together, the data obtained suggest that β2-adrenoceptor-induced modulation of TG2 represents a novel paradigm in β2-adrenoceptor cell signalling, expanding the repertoire of cellular functions responsive to catecholamine stimulation
