Assessing Child Development: A Critical Review and the Sustainable Child Development Index (SCDI)
Children are an important stakeholder group for sustainable development, as they represent the interface between current and future generations. A comprehensive assessment of child development (CD) in the context of sustainable development is still missing. In this paper, as a first step, a literature review is conducted to identify relevant aspects and gaps related to the assessment of CD. The main issues of CD are categorized into seven themes: health, education, safety, economic status, relationships, participation, and newly proposed environmental aspects. The corresponding subthemes and criteria are classified accordingly (e.g., nutrition, child mortality, immunization, etc., are assigned to the theme health). However, gaps in current studies, such as the heterogeneous classification of relevant aspects, regional and societal bias in addressing certain aspects, the limited number of subthemes, and criteria and the missing inclusion of environmental aspects impede the assessment of sustainable child development. To address the existing gaps, a comprehensive framework, the Sustainable Child Development Index (SCDI), is proposed. The SCDI is based on sustainable development as the core value, considers relevant aspects of CD with regard to newly-proposed environmental aspects and includes 26 aspects on an outcome and 37 indicators on a context level to tackle the heterogeneous classifications and interdependencies of relevant aspects. Identification and purification of a bovine liver mitochondrial NAD+-glycohydrolase
AbstractNonenzymatic ADP-ribosylation of mitochondrial proteins is thought to play a role in the regulation of Ca2+ efflux from mitochondria. It has been shown that intramitochondrial ADP-ribose is generated by a specific NAD+ glycohydrolase, which catalizes hydrolysis of NAD+ to ADP-ribose and nicotinamide. We purified this enzyme from bovine liver mitochondrial membranes. The final preparation had a 1660-fold purified enzyme activity and contained a main protein band with an apparent molar mass of 32,000 in a SDS-polyacrylamide gel. The identity of this protein band with NAD+-glycohydrolase was verified by renaturation of its enzymatic activity. Partial amino acid sequence information was obtained from two enzyme fragments after proteolytic cleavage of the protein band in the SDS-polyacrylamide gel. Searches in protein databases revealed that an arginine ADP-ribosyl hydrolase harbours two stretches of amino acids that are highly similar to the partial NAD+-glycohydrolase sequences
Microlensing Detections of Planets in Binary Stellar Systems
We demonstrate that microlensing can be used for detecting planets in binary
stellar systems. This is possible because in the geometry of planetary binary
systems where the planet orbits one of the binary component and the other
binary star is located at a large distance, both planet and secondary companion
produce perturbations at a common region around the planet-hosting binary star
and thus the signatures of both planet and binary companion can be detected in
the light curves of high-magnification lensing events. We find that identifying
planets in binary systems is optimized when the secondary is located in a
certain range which depends on the type of the planet. The proposed method can
detect planets with masses down to one tenth of the Jupiter mass in binaries
with separations <~ 100 AU. These ranges of planet mass and binary separation
are not covered by other methods and thus microlensing would be able to make

Short Communication: Lack of Support for Socially Connected HIV-1 Transmission among Young Adult Black Men Who Have Sex with Men

We explore the phylogenetic relationships among HIV sequences sampled from young adult black men who have sex with men (YAB-MSM), who are connected through peer referral/social ties and who attend common venues. Using 196 viral sequences sampled from the peripheral blood mononuclear cells of 10 individuals, our preliminary phylogenetic results indicate that these socially connected YAB-MSM are infected with distantly related viruses and provide no evidence for viral transmission between network members. Our results suggest that HIV-prevention strategies that target young adult MSM should extend beyond their network members and local community
Towards 3D printed multifunctional immobilization for proton therapy: initial materials characterization
Purpose: 3D printing technology is investigated for the purpose of patient immobilization during proton therapy. It potentially enables a merge of patient immobilization, bolus range shifting, and other functions into one single patient-speci c structure. In this rst step, a set of 3D printed materials is characterized in detail, in terms of structural and radiological properties, elemental composition, directional dependence, and structural changes induced by radiation damage. These data will serve as inputs for the design of 3D printed immobilization structure prototypes. Methods: Using four di erent 3D printing techniques, in total eight materials were subjected to testing. Samples with a nominal dimension of 20Ă20Ă80 mm3 were 3D printed. The geometrical printing accuracy of each test sample was measured with a dial gage. To assess the mechanical response of the samples, standardized compression tests were performed to determine the Youngâs modulus. To investigate the e ect of radiation on the mechanical response, the mechanical tests were performed both prior and after the administration of clinically relevant dose levels (70 Gy), multiplied with a safety factor of 1.4. Dual energy computed tomography (DECT) methods were used to calculate the relative electron density to water Ïe, the e ective atomic number Ze , and the proton stopping power ratio (SPR) to water SPR. In order to validate the DECT based calculation of radiological properties, beam measurements were performed on the 3D printed samples as well. Photon irradiations were performed to measure the photon linear attenuation coe cients, while proton irradiations were performed to measure the proton range shift of the samples. The direc- tional dependence of these properties was investigated by performing the irradiations for di erent orientations of the samples. Results: The printed test objects showed reduced geometric printing accuracy for 2 materials (deviation > 0.25 mm). Compression tests yielded Youngâs moduli ranging from 0.6 to 2940 MPa. No deterioration in the mechanical response was observed after exposure of the samples to 100 Gy in a therapeutic MV photon beam. The DECT-based characterization yielded Ze ranging from 5.91 to 10.43. The SPR and Ïe both ranged from 0.6 to 1.22. The measured photon attenuation coe cients at clinical energies scaled linearly with Ïe. Good agreement was seen between the DECT estimated SPR and the measured range shift, except for the higher Ze . As opposed to the photon attenuation, the proton range shifting appeared to be printing orientation dependent for certain materials. Conclusions: In this study, the rst step toward 3D printed, multifunctional immobilization was performed, by going through a candidate clinical work ow for the rst time: from the material printing to DECT characterization with a veri cation through beam measurements. Besides a proof of concept for beam modi cation, the mechanical response of printed materials was also investigated to assess their capabilities for positioning functionality. For the studied set of printing techniques and materials, a wide variety of mechanical and radiological properties can be selected from for the intended purpose. Moreover the elaborated hybrid DECT methods aid in performing in-house quality assurance of 3D printed components, as these methods enable the estimation of the radiological properties relevant for use in radiation therapy
Lagrangian transport through an ocean front in the North-Western Mediterranean Sea
We analyze with the tools of lobe dynamics the velocity field from a
numerical simulation of the surface circulation in the Northwestern
Mediterranean Sea. We identify relevant hyperbolic trajectories and their
manifolds, and show that the transport mechanism known as the `turnstile',
previously identified in abstract dynamical systems and simplified model flows,
is also at work in this complex and rather realistic ocean flow. In addition
nonlinear dynamics techniques are shown to be powerful enough to identify the
key geometric structures in this part of the Mediterranean. In particular the
North Balearic Front, the westernmost part of the transition zone between
saltier and fresher waters in the Western Mediterranean is interpreted in terms
of the presence of a semipermanent ``Lagrangian barrier'' across which little
transport occurs. Our construction also reveals the routes along which this
transport happens. Topological changes in that picture, associated with the
crossing by eddies and that may be interpreted as the breakdown of the front,

A Chemical and Genetic Approach to the Mode of Action of Fumagillin
SummaryPrevious mode of action studies identified methionine aminopeptidase 2 (MetAP-2) as the target of the antiangiogenic natural product fumagillin and its drug candidate analog, TNP-470. We report here that TNP-470-mediated MetAP-2 inhibition blocks noncanonical Wnt signaling, which plays a critical role in development, cell differentiation, and tumorigenesis. Consistent with this finding, antisense MetAP-2 morpholino oligonucleotide injection in zebrafish embryos phenocopies gastrulation defects seen in noncanonical Wnt5 loss-of-function zebrafish mutants. MetAP-2 inhibition or depletion blocks signaling downstream of the Wnt receptor Frizzled, but upstream of Calmodulin-dependent Kinase II, RhoA, and c-Jun N-terminal Kinase. Moreover, we demonstrate that TNP-470 does not block the canonical Wnt/ÎČ-catenin pathway. Thus, TNP-470 selectively regulates noncanonical over canonical Wnt signaling and provides a unique means to explore and dissect the biological systems mediated by these pathways
Far-infrared Tamm polaritons in a microcavity with incorporated graphene sheet
Tamm polaritons (TPs) are formed at the interface between a semi-infinite periodic
dielectric structure (Bragg mirror) and another reflector. They couple to elementary excitations in
the materials that form the interface, such as metal plasmons or semiconductor excitons. Here we
discuss the formation of TPs in the far-infrared spectral range, in the optical-phonon reststrahlen
band of a polar semiconductor such as GaAs, attached to a Bragg reflector (BR). Their dispersion
relation and the frequency window for the TP existence are calculated for a GaAs-BR interface.
Microcavity structures containing a gap between the two reflectors are also considered, including
those containing an inserted graphene layer and the possibility of tuning of the TP states by

Major immunophenotypic abnormalities in patients with primary adrenal insufficiency of different etiology
INTRODUCTION
Patients with primary adrenal insufficiency (PAI) suffer from increased risk of infection, adrenal crises and have a higher mortality rate. Such dismal outcomes have been inferred to immune cell dysregulation because of unphysiological cortisol replacement. As the immune landscape of patients with different types of PAI has not been systematically explored, we set out to immunophenotype PAI patients with different causes of glucocorticoid (GC) deficiency.
METHODS
This cross-sectional single center study includes 28 patients with congenital adrenal hyperplasia (CAH), 27 after bilateral adrenalectomy due to Cushing's syndrome (BADx), 21 with Addison's disease (AD) and 52 healthy controls. All patients with PAI were on a stable GC replacement regimen with a median dose of 25 mg hydrocortisone per day. Peripheral blood mononuclear cells were isolated from heparinized blood samples. Immune cell subsets were analyzed using multicolor flow cytometry after four-hour stimulation with phorbol myristate acetate and ionomycin. Natural killer (NK-) cell cytotoxicity and clock gene expression were investigated.
RESULTS
The percentage of T helper cell subsets was downregulated in AD patients (Th1 p = 0.0024, Th2 p = 0.0157, Th17 p < 0.0001) compared to controls. Cytotoxic T cell subsets were reduced in AD (Tc1 p = 0.0075, Tc2 p = 0.0154) and CAH patients (Tc1 p = 0.0055, Tc2 p = 0.0012) compared to controls. NKCC was reduced in all subsets of PAI patients, with smallest changes in CAH. Degranulation marker CD107a expression was upregulated in BADx and AD, not in CAH patients compared to controls (BADx p < 0.0001; AD p = 0.0002). In contrast to NK cell activating receptors, NK cell inhibiting receptor CD94 was upregulated in BADx and AD, but not in CAH patients (p < 0.0001). Although modulation in clock gene expression could be confirmed in our patient subgroups, major interindividual-intergroup dissimilarities were not detected.
DISCUSSION
In patients with different etiologies of PAI, distinct differences in T and NK cell-phenotypes became apparent despite the use of same GC preparation and dose. Our results highlight unsuspected differences in immune cell composition and function in PAI patients of different causes and suggest disease-specific alterations that might necessitate disease-specific treatment
