    Dextran sulfate from Leuconostoc mesenteroides B512F exerts potent antiviral activity against SARS-CoV-2 in vitro and in vivo

    The emergent human coronavirus SARS-CoV-2 and its resistance to current drugs makes the need for new potent treatments for COVID-19 patients strongly necessary. Dextran sulfate (DS) polysaccharides have long demonstrated antiviral activity against different enveloped viruses in vitro. However, their poor bioavailability has led to their abandonment as antiviral candidates. Here, we report for the first time the broad-spectrum antiviral activity of a DS-based extrapolymeric substance produced by the lactic acid bacterium Leuconostoc mesenteroides B512F. Time of addition assays with SARS-CoV-2 pseudoviruses in in vitro models confirm the inhibitory activity of DSs in the early stages of viral infection (viral entry). In addition, this exopolysaccharide substance also reports broad-spectrum antiviral activity against several enveloped viruses such as SARS-CoV-2, HCoV229E, HSV-1, in in vitro models and in human lung tissue. The toxicity and antiviral capacity of DS from L. mesenteroides was tested in vivo in mouse models which are susceptible to SARS-CoV-2 infection. The described DS, administered by inhalation, a new route of administration for these types of polymers, shows strong inhibition of SARS-CoV-2 infection in vivo, significantly reducing animal mortality and morbidity at non-toxic doses.     The importance of conserving Mexico's tomato agrodiversity to research plant biochemistry under different climates

    Tomatoes are important to agriculture, human nutrition and cuisines globally. However, many commercial tomato varieties, including the saladette that dominates the North American market, are highly sensitive to environmental changes that impact yields and critical biochemical pathways including carotenoids and isoprenoids that influence nutritional content and flavour. We highlight the potential of tomato agrodiversity, notably its genetic diversity, as an undervalued research tool for understanding environmental regulation of plant biochemistry under different climates. Yet, tomato genetic diversity in Mexico, the major centre of tomato domestication, is not formally described or protected. We propose that transdisciplinary efforts are essential to identify, conserve and research these globally significant genetic resources

    Environmental Volunteering Programme for Seagrass Monitoring

    Las praderas de fanerógamas marinas sufren actualmente un declive global como consecuencia de la presión humana ejercida sobre las áreas costeras y estuáricas. Frente a esta situación, la comunidad científica ha respondido creando redes de seguimiento de las praderas basadas en la participación ciudadana a través del voluntariado. Mediante la cooperación entre la Oficina Verde y el área de Ecología de la Universidad de Cádiz (UCA) se ha creado el Voluntariado Ambiental de Fanerógamas Marinas (FAMAR) en el Parque Natural Bahía de Cádiz. Este programa pretende implicar activamente a la sociedad, transmitir la necesidad de conservación de estos valiosos ecosistemas e inculcar un sentimiento de responsabilidad sobre el entorno natural en general. Tras un año de funcionamiento y con 80 voluntarios registrados, FAMAR ha cubierto con creces las expectativas de participación y han podido plantearse futuras líneas de trabajo tanto a corto como a medio y largo plazo.Seagrass meadows are currently declining worldwide, mainly as a consequence of human activities on coastal and estuarine zones. Scientists have acted against this situation by setting up seagrass monitoring networks. These networks are based on a public participation through volunteer work. Aware of this global threat on seagrasses, the Ecology research group of the University of Cadiz (UCA) together with the Green Office of this University have developed the "Environmental Volunteering Programme for Seagrass Monitoring at Cadiz Bay (FAMAR)”. The main objectives of this programme are to involve the society in seagrass conservation and to transmit a feeling of environmental responsibility. After one year of FAMAR launching, the participation level of volunteers has surpassed by far our initial expectations. FAMAR has recorded 80 volunteers, mainly UCA undergraduate students.     Urbanisation affects ecosystem functioning more than structure in tropical streams

    Urbanisation poses a clear threat to tropical freshwater streams, yet fundamental knowledge gaps hinder our ability to effectively conserve stream biodiversity and preserve ecosystem functioning. Here, we studied the impact of urbanisation on structural and functional ecosystem responses in low-order streams in Singapore, a tropical city with a mosaic landscape of protected natural forests, managed buffer zones (between forest and open-country habitats), and built-up urban areas. We quantified an urbanisation gradient based on landscape, in-stream, and riparian conditions, and found an association between urbanisation and pollution-tolerant macroinvertebrates (e.g. freshwater snail and worm species) in litter bags. We also found greater macroinvertebrate abundance (mean individuals bag−1; forest: 30.3, buffer: 70.1, urban: 109.0) and richness (mean taxa bag−1; forest: 4.53, buffer: 4.75, urban: 7.50) in urban streams, but similar diversity across habitats. Higher levels of primary productivity (measured from algal accrual on ceramic tiles) and microbial decomposition (measured from litter-mass loss in mesh bags) at urban sites indicate rapid microbial activity at higher light, temperature, and nutrient levels. We found that urbanisation affected function 32% more than structure in the studied tropical streams, likely driven by greater algal growth in urban streams. These changes in ecological processes (i.e. ecosystem functioning) possibly lead to a loss of ecosystem services, which would negatively affect ecology, society, and economy. Our results point to possible management strategies (e.g. increasing vegetation density through buffer park creation) to reduce the impacts of urbanisation, restore vital ecosystem functions in tropical streams, and create habitat niches for native species

    Symphysiotomy in Zimbabwe; Postoperative Outcome, Width of the Symphysis Joint, and Knowledge, Attitudes and Practice among Doctors and Midwives

    BACKGROUND: Obstructed labour remains one of the leading causes of maternal and foetal death and morbidity in poorly resourced areas of the world, where the 24 hours availability of a caesarean section (CS) cannot be guaranteed, and the CS related mortality rate is still high. In these settings, reinstatement of symphysiotomy has been advocated. The objectives were, in1994; to study perioperative and long-term complications of symphysiotomy and compare them to those related to CS for similar indications, in 1996; to measure the symphyseal width after symphysiotomy and compare it to that after normal vaginal delivery, and, in 1998; to assess knowledge, attitudes and practice related to symphysiotomy among doctors and midwives in Zimbabwe. METHODS AND FINDINGS: Thirty-four women who had undergone symphysiotomy and 29 women who had undergone a CS for obstructed labour were interviewed. The symphyseal widths of 19 women with a previous symphysiotomy were compared to that of 92 women with previous normal vaginal deliveries, using ultrasound technique. Forty-one doctors and 39 midwives, in three central hospitals and seven district hospitals in Zimbabwe, were interviewed about symphysiotomy. None of the 34 women reported serious soft tissue injuries or infections post symphysiotomy. Long-term complications after symphysiotomy do not differ notably from those after CS for similar indications. The intra-articular width of the symphysis pubis is increased after a symphysiotomy. Seventy-nine of the 80 interviewed health care workers knew about symphysiotomy. One obstetrician had performed symphysiotomies. Two-thirds of the participants considered symphysiotomy an obsolete and second-class operation, but lifesaving and appropriate in remote areas of Zimbabwe. Ten of 13 midwives in remote areas wanted to carry out symphysiotomies themselves. CONCLUSIONS: No severe complications due to symphysiotomy were revealed in this study. The results suggest that a modest permanent enlargement of the pelvis post symphysiotomy (together with the absence of a scarred uterus) may facilitate subsequent vaginal delivery. Doctors and midwives working in district hospitals have a more positive attitude to symphysiotomies than the colleagues in central hospitals. Obstetricians (who would have to do the teaching), working in the large urban hospitals almost exclude symphysiotomy as an alternative management in Zimbabwe

    Microbiome assembly of avian eggshells and their potential as transgenerational carriers of maternal microbiota

    The microbiome is essential for development, health and homeostasis throughout an animal's life. Yet, the origins and transmission processes governing animal microbiomes remain elusive for non-human vertebrates, oviparous vertebrates in particular. Eggs may function as transgenerational carriers of the maternal microbiome, warranting characterisation of egg microbiome assembly. Here, we investigated maternal and environmental contributions to avian eggshell microbiota in wild passerine birds: woodlark Lullula arborea and skylark Alauda arvensis. Using 16S rRNA gene sequencing, we demonstrated in both lark species, at the population and within-nest levels, that bacterial communities of freshly laid eggs were distinct from the female cloacal microbiome. Instead, soil-borne bacteria appeared to thrive on freshly laid eggs, and eggshell microbiota composition strongly resembled maternal skin, body feather and nest material communities, sources in direct contact with laid eggs. Finally, phylogenetic structure analysis and microbial source tracking underscored species sorting from directly contacting sources rather than in vivo-transferred symbionts. The female-egg-nest system allowed an integrative assessment of avian egg microbiome assembly, revealing mixed modes of symbiont acquisition not previously documented for vertebrate eggs. Our findings illuminated egg microbiome origins, which suggested a limited potential of eggshells for transgenerational transmission, encouraging further investigation of eggshell microbiome functions in vertebrates

    Young [alpha/Fe]-enhanced stars discovered by CoRoT and APOGEE: What is their origin?

    We report the discovery of a group of apparently young CoRoT red-giant stars exhibiting enhanced [_/Fe] abundance ratios (as determined from APOGEE spectra) with respect to solar values. Their existence is not explained by standard chemical evolution models of the Milky Way, and shows that the chemical-enrichment history of the Galactic disc is more complex. We find similar stars in previously published samples for which isochrone-ages could be reliably obtained, although in smaller relative numbers. This might explain why these stars have not previously received attention. The young [_/Fe]-rich stars are much more numerous in the CoRoT APOGEE (CoRoGEE) inner-field sample than in any other high-resolution sample available at present because only CoRoGEE can explore the inner-disc regions and provide ages for its field stars. The kinematic properties of the young [_/Fe]-rich stars are not clearly thick-disc like, despite their rather large distances from the Galactic mid-plane. Our tentative interpretation of these and previous intriguing observations in the Milky Way is that these stars were formed close to the end of the Galactic bar, near corotation – a region where gas can be kept inert for longer times than in other regions that are more frequently shocked by the passage of spiral arms. Moreover, this is where the mass return from older inner-disc stellar generations is expected to be highest (according to an inside-out disc-formation scenario), which additionally dilutes the in-situ gas. Other possibilities to explain these observations (e.g., a recent gas-accretion event) are also discussed
