    Thermal Decomposition of CxF2 x+1C(O)OONO2(x = 2, 3, 4)

    The atmospheric degradation of molecules containing the CxF2x+1C(O) moiety, such as perfluoroaldehydes CxF2x+1C(O)H (x = 2-4) formed in the degradation of telomeric alcohols, could lead to the formation of perfluoroacyl peroxynitrates CxF2x+1C(O)OONO2. The thermal decomposition of the CxF2x+1C(O)OONO2 family (x = 2, 3, 4) was investigated by infrared spectroscopy and computational models. Each peroxynitrate synthesis was performed through the photolysis of gas mixtures of the corresponding perfluoroaldehyde, chlorine, nitrogen dioxide, and oxygen. Kinetic analysis for the thermal decomposition of peroxynitrates were performed in the range from 297.0 to 313.7 K at a total pressure of 1000 mbar and the activation energy was experimentally determined. Experimental data were complemented with theoretical data using the Gaussian09 Program Suite. The structures of peroxynitrates were optimized using DFT methods. The activation energies were calculated and investigated taking into account the stereoelectronic effects and using theoretical calculations as well as NBO analysis. The influence of anomeric interaction over the O-N bond was evaluated for all the molecules. Analysis of the results shows that CxF2x+1C(O)OONO2 stability is independent of CxF2x+1 chain length, in contrast to the behavior for perfluoroalkyl peroxynitrates (CxF2x+1OONO2).Fil: Vila, Jesús Alberto.     Hyperleptinaemia, respiratory drive and hypercapnic response in obese patients

    Leptin is a powerful stimulant of ventilation in rodents. In humans, resistance to leptin has been consistently associated with obesity. Raised leptin levels have been reported in subjects with sleep apnoea or obesity-hypoventilation syndrome. The aim of the present study was to assess, by multivariate analysis, the possible association between respiratory centre impairment and levels of serum leptin. In total, 364 obese subjects (body mass index >or=30 kg.m(-2)) underwent the following tests: sleep studies, respiratory function tests, baseline and hypercapnic response (mouth occlusion pressure (P(0.1)), minute ventilation), fasting leptin levels, body composition and anthropometric measures. Subjects with airways obstruction on spirometry were excluded. Out of the 346 subjects undergoing testing, 245 were included in the current analysis. Lung volumes, age, log leptin levels, end-tidal carbon dioxide tension, percentage body fat and minimal nocturnal saturation were predictors for baseline P(0.1). The hypercapnic response test was performed by 186 subjects; log leptin levels were predictors for hypercapnic response in males, but not in females. Hyperleptinaemia is associated with a reduction in respiratory drive and hypercapnic response, irrespective of the amount of body fat. These data suggest the extension of leptin resistance to the respiratory centre

    Determinación de marcadores de origen botánico para mieles de citrus (citrus spp) de la región fronteriza: Concordia (República Argentina) – Salto (República Oriental del Uruguay)

    Argentina y Uruguay son productores de mieles de alta calidad que destinan en más de un 95% a exportación, especialmente a países de la CEE. Mercados exigentes que valorizan mieles diferenciadas por origen botánico y geográfico. La región fronteriza Concordia-Salto se caracteriza por ser una zona de producción citrícola, que cuenta con extensas plantaciones dedicadas a la producción de naranjas y mandarinas, lo que constituye una fuente de néctar para las colmenas instaladas en el área por apicultores de ambos países. La miel de cítricos es muy apreciada comercialmente con un valor agregado importante cuando es posible diferenciarla. Una de las dificultades que se presentan, es que debido a características propias la especie,  el número de gránulos de polen en la miel suele ser escaso, dificultando determinar origen botánico solo con este parámetro. El estudio complementario de las características físico-químicas y composición del aroma, resulta un aporte importante para diferenciar y valorizar estas mieles. El objetivo general de este proyecto es disponer en la región fronteriza Concordia-Salto de una miel tipificada de Citrus con el fin de aumentar su valor agregado y mejorar su posicionamiento en los mercados internacionales,  a partir de hallar componentes aromáticos marcadores de origen botánico

    Investigation of late Pleistocene and early Holocene palaeoenvironmental change at El Mirón cave (Cantabria, Spain): insights from carbon and nitrogen isotope analysis of red deer

    Abstract: El Mirón Cavewas occupied by humans for over 40,000 years. Evidence of LateMousterian,Gravettian, Solutrean, Magdalenian, Azilian, Mesolithic, Neolithic, Chalcolithic, Bronze Age and Mediaeval occupations has been found in the cave. Understanding the local environmental conditions during the occupations is crucial for gaining an insight into the lifeways of El Mirón's inhabitants as they relied on the surrounding region and its natural resources for their subsistence. 170 bones of hunted red deer recovered from the cave were sampled for carbon and nitrogen stable isotope analyses with the aim of reconstructing the palaeoenvironment and palaeoclimate during the human occupation. The results show that the surrounding landscape underwent considerable environmental change during the Late Pleistocene and Early to Mid-Holocene. Shifts in d13C values between the Last Glacial Maximum, Heinrich stadial 1, Heinrich event 1, the Late-glacial interstadial and the onset of the Holocene are likely to reflect changes in water availability and temperature. Deer d15N generally increased over time indicating the regeneration of soil biological activity and nitrogen cycling, which was temporarily halted during the Younger Dryas. Comparison of the El Mirón results with those of 300 deer from other regions of Europe shows geographical variations in the timing and magnitude of the variation in d13C and d15N values. This variation tracks local climate (temperature andwater availability) and environmental (vegetation and forest development) change

    Genome-wide association analysis of dementia and its clinical endophenotypes reveal novel loci associated with Alzheimer's disease and three causality networks : The GR@ACE project

    Introduction: Large variability among Alzheimer's disease (AD) cases might impact genetic discoveries and complicate dissection of underlying biological pathways. Methods: Genome Research at Fundacio ACE (GR@ACE) is a genome-wide study of dementia and its clinical endophenotypes, defined based on AD's clinical certainty and vascular burden. We assessed the impact of known AD loci across endophenotypes to generate loci categories. We incorporated gene coexpression data and conducted pathway analysis per category. Finally, to evaluate the effect of heterogeneity in genetic studies, GR@ACE series were meta-analyzed with additional genome-wide association study data sets. Results: We classified known AD loci into three categories, which might reflect the disease clinical heterogeneity. Vascular processes were only detected as a causal mechanism in probable AD. The meta-analysis strategy revealed the ANKRD31-rs4704171 and NDUFAF6-rs10098778 and confirmed SCIMP-rs7225151 and CD33-rs3865444. Discussion: The regulation of vasculature is a prominent causal component of probable AD. GR@ACE meta-analysis revealed novel AD genetic signals, strongly driven by the presence of clinical heterogeneity in the AD series

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Multiancestry analysis of the HLA locus in Alzheimer’s and Parkinson’s diseases uncovers a shared adaptive immune response mediated by HLA-DRB1*04 subtypes

    Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson’s disease (PD) and Alzheimer’s disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. Protective HLA-DRB1*04 subtypes strongly bound the aggregation-prone tau PHF6 sequence, however only when acetylated at a lysine (K311), a common posttranslational modification central to tau aggregation. An HLA-DRB1*04-mediated adaptive immune response decreases PD and AD risks, potentially by acting against tau, offering the possibility of therapeutic avenues

The molecular geometry, Wiberg Index, NBO analysis and vibrational wavenumbers in the ground state have been calculated using a density functional method (B3LYP) with 6-31+G∗∗ and 6-311+G∗∗ basis set. Both compounds are stable as dimers in the solid phase, possessing C2 symmetry. The scaled theoretical wavenumbers showed very good agreement with the experimental values. This work contributes to the knowledge of important data which are rather scarce for quinazolinones. © The Royal Society of Chemistry 2015

    Priorización, limpieza viral, producción de semilla de calidad básica y devolución de cultivares nativos libres de virus

    Ocho instituciones colaboradoras (AGRUCO, CENDA, CPA, ACRA, PIABS, CESA, UTO, COPLA), y las comunidades a las que apoyan, han seleccionado en zonas con características extremas, cultivares de papa nativa cultivados la mayoría en alturas superiores a los 3,500 m. De ellos 24 fueron priorizados con PROINPA para su limpieza viral, en base a sus características sobresalientes de alta rusticidad frente a factores abióticos y por su importancia social en la elaboración de chuño (tubérculo deshidratado negro), tunta (tubérculo deshidratado blanco), waicu (tubérculo cocido con cáscara), y cocido en fresco. Con el uso de anticuerpos apropiados para la detección de variantes andinas de los virus comunes de la papa en DAS-ELISA, se ha limpiado de virus los 24 cultivares de papa nativa, de los cuales se ha producido semilla de calidad básica en la E.E. Toralapa y en condiciones in situ en camas protegidas construidas con materiales rústicos del lugar. Asimismo a la fecha se han producido tubérculossemillas de alta calidad en parcelas semillaras de los mismos agricultores. Los cultivares nativos de papa, limpios, han sido devueltos a sus centros de recolección a través de los sindicatos de agricultores e instituciones involucradas. Se ha realizado lacomparación preliminar de semilla libre de virus con la semilla proveniente de selección positiva y semilla del agricultor, con resultados muy satisfactorios. Se realizaron eventos de capacitación a 282 agricultores para el manejo de los cultivares libres de virus. La demanda al presente por los cultivares nativos limpios contribuirá al mantenimiento de la biodiversidad y apoyará a una agricultura más sostenible
