    The use of the anesthetic metomidate hydrochloride as an aid in open water capture of young-of-the-year rockfish

    Many species of young-of-the-year (YOY) rockfish, Sebastes sp. move inshore into shallow reef areas in central California each year. We monitored the dates of arrival, relative abundance, species composition, lengths, and growth rates of these arrivals to help assess recruitment variability and stock success. We tested the usefulness of the anesthetic metomidate hydrochloride as a non-lethal method to collect YOY rockfish to assess growth, abundance, and species composition. Earlier experiments by other researchers illustrated the usefulness of metomidate to collect benthic-oriented adult rockfish. Our research tested the possibility of using metomidate hydrochloride to collect open-water and benthic-oriented YOY rockfish. The anesthetic was applied and the fish captured by divers. Several concentrations of metomidate were tested, premixed both in fresh and salt water. The anesthetic was not very useful in collecting open water YOY rockfish as the fish rapidly left the area of application and avoided being anesthetized. Increasing the concentration of metomidate did not increase success of capture of open-water YOY rockfish. However, the anesthetic metomidate worked very well in capturing benthic-oriented YOY rockfishes. (22pp.

    HD 101088, An Accreting 14 AU Binary in Lower Centaurus Crux With Very Little Circumstellar Dust

    We present high resolution (R=55,000) optical spectra obtained with MIKE on the 6.5 m Magellan Clay Telescope as well as Spitzer MIPS photometry and IRS low resolution (R~60) spectroscopy of the close (14 AU separation) binary, HD 101088, a member of the ~12 Myr old southern region of the Lower Centaurus Crux (LCC) subgroup of the Scorpius-Centaurus OB association. We find that the primary and/or secondary is accreting from a tenuous circumprimary and/or circumsecondary disk despite the apparent lack of a massive circumbinary disk. We estimate a lower limit to the accretion rate of > 1x10^-9 solar masses per year, which our multiple observation epochs show varies over a timescale of months. The upper limit on the 70 micron flux allows us to place an upper limit on the mass of dust grains smaller than several microns present in a circumbinary disk of 0.16 moon masses. We conclude that the classification of disks into either protoplanetary or debris disks based on fractional infrared luminosity alone may be misleading.Comment: 8 pages, 2 figures, ApJ accepte

    Determination of an optimal response cut-off able to predict progression-free survival in patients with well-differentiated advanced pancreatic neuroendocrine tumours treated with sunitinib: an alternative to the current RECIST-defined response.

    BACKGROUND: Sunitinib prolongs progression-free survival (PFS) in patients with advanced pancreatic neuroendocrine tumours (pNET). Response Evaluation Criteria in Solid Tumors (RECIST)-defined partial responses (PR; classically defined as ⩾30% size decrease from baseline) are infrequent. METHODS: Individual data of pNET patients from the phase II [NCT00056693] and pivotal phase III [NCT00428597] trials of sunitinib were analysed in this investigator-initiated, post hoc study. The primary objective was to determine the optimal RECIST (v.1.0) response cut-off value to identify patients who were progression-free at 11 months (median PFS in phase III trial); and the most informative time-point (highest area under the curve (AUC) by receiver operating characteristic (ROC) analysis and logistic regression) for prediction of benefit (PFS) from sunitinib. RESULTS: Data for 237 patients (85 placebo; 152 sunitinib (n=66.50 mg \u274-weeks on/2-weeks off\u27 schedule; n=86 \u2737.5 mg continuous daily dosing (CDD)\u27)) and 788 scans were analysed. The median PFS for sunitinib and placebo were 9.3 months (95% CI 7.6-12.2) and 5.4 months (95% CI 3.5-6.01), respectively (hazard ratio (HR) 0.43 (95% CI 0.29-0.62); P CONCLUSIONS: A 10% reduction within marker lesions identifies pNET patients benefiting from sunitinib treatment with implications for maintenance of dose intensity and future trial design

    Discovery and analysis of p-mode and g-mode oscillations in the A-type primary of the eccentric binary HD 209295

    We have discovered both intermediate-order gravity mode and low-order pressure mode pulsation in the same star, HD 209295. It is therefore both a Gamma Doradus and a Delta Scuti star, which makes it the first pulsating star to be a member of two classes. The star is a single-lined spectroscopic binary with an orbital period of 3.10575 d and an eccentricity of 0.352. Weak pulsational signals are found in both the radial velocity and line-profile variations, allowing us to show that the two highest-amplitude Gamma Doradus pulsation modes are consistent with l=1 and |m|=1. In our 280 h of BVI multi-site photometry we detected ten frequencies in the light variations, one in the Delta Scuti regime and nine in the Gamma Doradus domain. Five of the Gamma Doradus frequencies are exact integer multiples of the orbital frequency. This observation leads us to suspect they are tidally excited. Results of theoretical modeling (stability analysis, tidal excitation) were consistent with the observations. We could not detect the secondary component of the system in infrared photometry, suggesting that it may not be a main-sequence star. Archival data of HD 209295 show a strong ultraviolet excess, the origin of which is not known. The orbit of the primary is consistent with a secondary mass of M > 1.04 Msun indicative of a neutron star or a white dwarf companion.Comment: 18 pages, 18 figures, accepted for publication in MNRAS, shortened abstrac

    MagAO Imaging of Long-period Objects (MILO). II. A Puzzling White Dwarf around the Sun-like Star HD 11112

The American Astronomical Society. All rights reserved.HD 11112 is an old, Sun-like star that has a long-term radial velocity (RV) trend indicative of a massive companion on a wide orbit. Here we present direct images of the source responsible for the trend using the Magellan Adaptive Optics system. We detect the object (HD 11112B) at a separation of 2\fasec 2 (100 AU) at multiple wavelengths spanning 0.6-4 \microns ~and show that it is most likely a gravitationally-bound cool white dwarf. Modeling its spectral energy distribution (SED) suggests that its mass is 0.9-1.1 \msun, which corresponds to very high-eccentricity, near edge-on orbits from Markov chain Monte Carlo analysis of the RV and imaging data together. The total age of the white dwarf is >2σ>2\sigma discrepant with that of the primary star under most assumptions. The problem can be resolved if the white dwarf progenitor was initially a double white dwarf binary that then merged into the observed high-mass white dwarf. HD 11112B is a unique and intriguing benchmark object that can be used to calibrate atmospheric and evolutionary models of cool white dwarfs and should thus continue to be monitored by RV and direct imaging over the coming years.Peer reviewedFinal Published versio

    Modeling the Afferent Dynamics of the Baroreflex Control System

    In this study we develop a modeling framework for predicting baroreceptor firing rate as a function of blood pressure. We test models within this framework both quantitatively and qualitatively using data from rats. The models describe three components: arterial wall deformation, stimulation of mechanoreceptors located in the BR nerve-endings, and modulation of the action potential frequency. The three sub-systems are modeled individually following well-established biological principles. The first submodel, predicting arterial wall deformation, uses blood pressure as an input and outputs circumferential strain. The mechanoreceptor stimulation model, uses circumferential strain as an input, predicting receptor deformation as an output. Finally, the neural model takes receptor deformation as an input predicting the BR firing rate as an output. Our results show that nonlinear dependence of firing rate on pressure can be accounted for by taking into account the nonlinear elastic properties of the artery wall. This was observed when testing the models using multiple experiments with a single set of parameters. We find that to model the response to a square pressure stimulus, giving rise to post-excitatory depression, it is necessary to include an integrate-and-fire model, which allows the firing rate to cease when the stimulus falls below a given threshold. We show that our modeling framework in combination with sensitivity analysis and parameter estimation can be used to test and compare models. Finally, we demonstrate that our preferred model can exhibit all known dynamics and that it is advantageous to combine qualitative and quantitative analysis methods

    Enabling microbial syringol conversion through structure-guided protein engineering

    Microbial conversion of aromatic compounds is an emerging and promising strategy for valorization of the plant biopolymer lignin. A critical and often rate-limiting reaction in aromatic catabolism is O-aryl-demethylation of the abundant aromatic methoxy groups in lignin to form diols, which enables subsequent oxidative aromatic ring-opening. Recently, a cytochrome P450 system, GcoAB, was discovered to demethylate guaiacol (2-methoxyphenol), which can be produced from coniferyl alcohol-derived lignin, to form catechol. However, native GcoAB has minimal ability to demethylate syringol (2,6-dimethoxyphenol), the analogous compound that can be produced from sinapyl alcohol-derived lignin. Despite the abundance of sinapyl alcohol-based lignin in plants, no pathway for syringol catabolism has been reported to date. Here we used structure-guided protein engineering to enable microbial syringol utilization with GcoAB. Specifically, a phenylalanine residue (GcoA-F169) interferes with the binding of syringol in the active site, and on mutation to smaller amino acids, efficient syringol O-demethylation is achieved. Crystallography indicates that syringol adopts a productive binding pose in the variant, which molecular dynamics simulations trace to the elimination of steric clash between the highly flexible side chain of GcoA-F169 and the additional methoxy group of syringol. Finally, we demonstrate in vivo syringol turnover in Pseudomonas putida KT2440 with the GcoA-F169A variant. Taken together, our findings highlight the significant potential and plasticity of cytochrome P450 aromatic O-demethylases in the biological conversion of lignin-derived aromatic compounds
