    What Do Closed Source Data Tell Us About Lone Actor Terrorist Behavior? A Research Note

    This article contributes to the growing body of knowledge on lone-actor terrorism with the incorporation of closed-source data. The analyses presented investigate the antecedent behaviors of U.K.-based lone-actor terrorists leading up to their planning or conducting a terrorist event. The results suggest that prior to their attack or arrest the vast majority of lone-actor terrorists each demonstrated elements concerning (a) their grievance, (b) an escalation in their intent to act, (c) gaining capability—both psychologically and technically and (d) attack planning. The results also disaggregate our understanding of lone-actor terrorists in two ways. First, we compare the behaviors of the jihadist actors to those of the extreme-right. Second, we visualize Borum’s (2012) continuums of loneness, direction, and motivation. Collectively the results provide insight into the threat assessment and management of potential lone actors

    Abstract: One of the most exciting and frustrating times in the treatment process for a client in recovery is the period when that individual moves into aftercare. There are many challenges and obstacles to maintaining his or her clean/ sober status, and support systems are key to a client's success in aftercare. Unfortunately, a group of professionals who can have a very strong impact on that success-those in Vocational Rehabilitation (VR)-are often left out of the system. Treatment and aftercare counselors may have a good understanding of many of the social services needed by clients who are transitioning into aftercare, but most are not aware of, or are under-informed about, the scope of services offered by VR that can meet the client's rehabilitation needs. Clients in recovery from substance abuse are eligible to apply for VR services. Bringing VR counselors into the process and encouraging them to be active participants in the aftercare of the client can therefore help a client prevent relapse and become a contributing, successful member of society. However, if treatment and aftercare counselors are unfamiliar with VR programs, they will not include VR counselors in aftercare planning and service provision. In an effort to assist such partnerships to be established and maintained, this article will discuss VR and its history, briefly outline its case management format, and discuss the limitations of the format. It will touch upon ways to incorporate VR staff into the aftercare process. This article will also discuss partnership efforts in the state of Oregon between American Indian-based treatment center and the Oregon Department of Human Services; Office of Vocational Rehabilitation Services (OVRS), and outline suggestions to allow the reader to create and maintain ties for improving collaboration in their communities

    Performance of concrete in fire: a review of the state of the art, with a case study of the windsor tower fire

    This paper provides a “State of the Art” review on current research into the effects of fire exposures upon concrete. The principal influences of high temperature in concrete are loss of compressive strength and spalling, the forcible ejection of material from a member. Though a lot of information has been gathered on both phenomena, there remains a need for a broader understanding of the response of concrete structures to different heating regimes and the performance of complete concrete structures subjected to realistic fire exposures. There is a lack of information derived from large-scale tests on concrete buildings in natural fires. Besides undertaking further fire tests, lessons can also be learnt from real fires and the University of Edinburgh has embarked upon detailed studies of the serious fire in the Windsor Tower, Madrid. In order to properly characterise the fire and the performance of the structure a data-gathering exercise has been undertaken and computer modelling tools are being applied in order to obtain better insights into the structural response. There remains some uncertainty about the precise mechanism of fire spread, but an external route is likely, facilitated to some degree by the glazed curtain walling construction; lack of fire protection on the steelwork was the major reason for the subsequent partial collapse of the upper floors and the localised failure of a concrete portal frame can be attributed to the same reason

    The tip of brucella O-Polysaccharide is a potent epitope in response to brucellosis infection and enables short synthetic antigens to be superior diagnostic reagents

    Brucellosis is a global disease and the world’s most prevalent zoonosis. All cases in livestock and most cases in humans are caused by members of the genus Brucella that possess a surface O-polysaccharide (OPS) comprised of a rare monosaccharide 4-deoxy-4-formamido-D-mannopyranose assembled with α1,2 and α1,3 linkages. The OPS of the bacterium is the basis for serodiagnostic tests for brucellosis. Bacteria that also contain the same rare monosaccharide can induce antibodies that cross-react in serological tests. In previous work we established that synthetic oligosaccharides, representing elements of the Brucella A and M polysaccharide structures, were excellent antigens to explore the antibody response in the context of infection, immunisation and cross reaction. These studies suggested the existence of antibodies that are specific to the tip of the Brucella OPS. Sera from naturally and experimentally Brucella abortus-infected cattle as well as from cattle experimentally infected with the cross-reactive bacterium Yersinia enterocolitica O:9 and field sera that cross react in conventional serological assays were studied here with an expanded panel of synthetic antigens. The addition of chemical features to synthetic antigens that block antibody binding to the tip of the OPS dramatically reduced their polyclonal antibody binding capability providing conclusive evidence that the OPS tip (non-reducing end) is a potent epitope. Selected short oligosaccharides, including those that were exclusively α1,2 linked, also demonstrated superior specificity when evaluated with cross reactive sera compared to native smooth lipopolysaccharide (sLPS) antigen and capped native OPS. This surprising discovery suggests that the OPS tip epitope, even though common to both Brucella and Y. enterocolitica O:9, has more specific diagnostic properties than the linear portion of the native antigens. This finding opens the way to the development of improved serological tests for brucellosis

    High throughput method for analysis of repeat number for 28 phase variable loci of C. jejuni strain NCTC11168

    Mutations in simple sequence repeat tracts are a major mechanism of phase variation in several bacterial species including Campylobacter jejuni. Changes in repeat number of tracts located within the reading frame can produce a high frequency of reversible switches in gene expression between ON and OFF states. The genome of C. jejuni strain NCTC11168 contains 29 loci with polyG/polyC tracts of seven or more repeats. This protocol outlines a method for rapidly determining ON/OFF states of these 28 phase-variable loci in a large number of individual colonies. The method combines a series of multiplex PCR assays with a GeneScan assay and automated extraction of tract length, repeat number and expression state. This high throughput, multiplex assay has utility for detecting shifts in phase variation states within and between populations over time and for exploring the effects of phase variation on adaptation to differing selective pressures. An important output of this assay is combinatorial expression states that cannot be determined by other methods. This method can be adapted to analysis of phase variation in other C. jejuni strains and in a diverse range of bacterial species

    Clinical supervision for clinical psychology students in Uganda: an initial qualitative exploration

    Background Burn out in clinical psychologists working in low income countries has been reported. Clinical supervisory structures do not yet exist in Uganda. A way to decrease levels of burn out and increase quality of care for people with mental illness is through clinical supervision. The aim of this study was to explore the initial experiences of supervision for clinical psychology students in Uganda to ascertain whether or not clinical supervision is culturally appropriate, and what aspects of supervision had been helpful and unhelpful. Methods A qualitative design with thematic analysis was utilized. A focus group was held with 12 second year clinical psychology students to ask their experiences of receiving supervision. Results Data analysis created five themes. Firstly, the negative emotions that resulted from the training processed were discussed, and how supervision helped and did not help the students to manage these. Secondly, the students voiced that supervision helped them to learn through observational experiences, co-therapist roles and parallel processes within the supervisory relationship. Thirdly, supervision had taught the clinical psychology students their role as a clinical psychology student, how to act within the Ugandan mental health system and skills to conduct therapy. Fourthly, suggestions for the future of supervision were given, with the students requesting for it to start earlier in the training, for supervisors who can meet with the students on a regular basis to be selected and for the training the students receive at university to match the skills required on their placements, with a request for more practical techniques rather than theory. The final theme related to left over miscellaneous data, such as the students agreeing with each other. Conclusions The students stated that supervision was helpful overall, implying that clinical supervision is culturally appropriate for clinical psychology students in Uganda. Suggestions for future supervision were given. In order to decrease high levels of staff burn out in the mental health systems in Uganda, supervisory structures with an emphasis on self care need to be established

    Antimicrobial Stewardship from Policy to Practice: Experiences from UK Antimicrobial Pharmacists

    Antimicrobial stewardship in the UK has evolved dramatically in the last 15 years. Factors driving this include initial central funding for specialist pharmacists and mandatory reductions in healthcare-associated infections (particularly Clostridium difficile infection). More recently, the introduction of national stewardship guidelines, and an increased focus on stewardship as part of the UK five-year antimicrobial resistance strategy, have accelerated and embedded developments. Antimicrobial pharmacists have been instrumental in effecting changes at an organizational and national level. This article describes the evolution of the antimicrobial pharmacist role, its impact, the progress toward the actions listed in the five-year resistance strategy, and novel emerging areas in stewardship in the UK

    Targeted glycoproteomic identification of cancer cell glycosylation

    GalMBP is a fragment of serum mannose-binding protein that has been modified to create a probe for galactose-containing ligands. Glycan array screening demonstrated that the carbohydrate-recognition domain of GalMBP selectively binds common groups of tumor-associated glycans, including Lewis-type structures and T antigen, suggesting that engineered glycan-binding proteins such as GalMBP represent novel tools for the characterization of glycoproteins bearing tumor-associated glycans. Blotting of cell extracts and membranes from MCF7 breast cancer cells with radiolabeled GalMBP was used to demonstrate that it binds to a selected set of high molecular weight glycoproteins that could be purified from MCF7 cells on an affinity column constructed with GalMBP. Proteomic and glycomic analysis of these glycoproteins by mass spectrometry showed that they are forms of CD98hc that bear glycans displaying heavily fucosylated termini, including Lewisx and Lewisy structures. The pool of ligands was found to include the target ligands for anti-CD15 antibodies, which are commonly used to detect Lewisx antigen on tumors, and for the endothelial scavenger receptor C-type lectin, which may be involved in tumor metastasis through interactions with this antigen. A survey of additional breast cancer cell lines reveals that there is wide variation in the types of glycosylation that lead to binding of GalMBP. Higher levels of binding are associated either with the presence of outer-arm fucosylated structures carried on a variety of different cell surface glycoproteins or with the presence of high levels of the mucin MUC1 bearing T antigen
