    Probabilistic risk assessment of solar particle events considering the cost of countermeasures to reduce the aviation radiation dose

    太陽フレアによる被ばくの脅威から航空機搭乗者を「合理的」に護る --経済的損失リスクの定量化により最適な航空機運用指針の策定が可能に--. 京都大学プレスリリース. 2021-09-03.Cosmic-ray exposure to flight crews and passengers, which is called aviation radiation exposure, is an important topic in radiological protection, particularly for solar energetic particles (SEP). We therefore assessed the risks associated with the countermeasure costs to reduce SEP doses and dose rates for eight flight routes during five ground level enhancements (GLE). A four-dimensional dose-rate database developed by the Warning System for Aviation Exposure to Solar Energetic Particles, WASAVIES, was employed in the SEP dose evaluation. As for the cost estimation, we considered two countermeasures; one is the cancellation of the flight, and the other is the reduction of flight altitudes. Then, we estimated the annual occurrence frequency of significant GLE events that would bring the maximum flight route dose and dose rate over 1.0 mSv and 80 μSv/h, respectively, based on past records of GLE as well as historically large events observed by the cosmogenic nuclide concentrations in tree rings and ice cores. Our calculations suggest that GLE events of a magnitude sufficient to exceed the above dose and dose rate thresholds, requiring a change in flight conditions, occur once every 47 and 17 years, respectively, and their conservatively-estimated annual risks associated with the countermeasure costs are up to around 1.5 thousand USD in the cases of daily-operated long-distance flights

However, iPSC-derived differentiated cells are often heterogenous and need purification using a flow cytometer, which has high cost and time consumption for large-scale purification. MicroRNAs (miRNAs) can be used as cell selection markers, because their activity differs between cell types. Here, we show miRNA-responsive ON and OFF switch mRNAs for robust cell purification. The ON switch contains a miRNA-target sequence after the polyadenylate tail, triggering translational activation by sensing the target miRNA. By designing RNA-only circuits with miRNA-ON and -OFF switch mRNAs that encode a lethal ribonuclease, Barnase, and its inhibitor, Barstar, we efficiently purified specific cell types, including human iPSCs and differentiated cardiomyocytes, without flow cytometry. Synthetic mRNA circuits composed of ON and OFF switches provide a safe, versatile, and time-saving method to purify various cell types for biological and clinical applications

    Calcium sparks enhance the tissue fluidity within epithelial layers and promote apical extrusion of transformed cells

    In vertebrates, newly emerging transformed cells are often apically extruded from epithelial layers through cell competition with surrounding normal epithelial cells. However, the underlying molecular mechanism remains elusive. Here, using phospho-SILAC screening, we show that phosphorylation of AHNAK2 is elevated in normal cells neighboring RasV12 cells soon after the induction of RasV12 expression, which is mediated by calcium-dependent protein kinase C. In addition, transient upsurges of intracellular calcium, which we call calcium sparks, frequently occur in normal cells neighboring RasV12 cells, which are mediated by mechanosensitive calcium channel TRPC1 upon membrane stretching. Calcium sparks then enhance cell movements of both normal and RasV12 cells through phosphorylation of AHNAK2 and promote apical extrusion. Moreover, comparable calcium sparks positively regulate apical extrusion of RasV12-transformed cells in zebrafish larvae as well. Hence, calcium sparks play a crucial role in the elimination of transformed cells at the early phase of cell competition

    Synchrotron Emission from Elliptical Galaxies Consequent to AGN outbursts

    Both radiative and mechanical feedback from Active Galactic Nuclei have been found to be important for the evolution of elliptical galaxies. We compute how a shock may be driven from a central black hole into the gaseous envelope of an elliptical galaxy by mechanical as well as radiative feedback (in the form of nuclear winds) using high resolution 1-D hydrodynamical simulations. We calculate the synchrotron emission from the electron cosmic rays accelerated by the shocks (not the jets) in the central part of elliptical galaxies, and we also study the synchrotron spectrum's evolution using the standard diffusive shock acceleration mechanism, which is routinely applied to the scaled volume case of supernova remnants. We find good agreement quantitatively between the synchrotron radio emission produced via this mechanism with extant observations of elliptical galaxies which are undergoing outbursts. Additionally, we also find that synchrotron optical and X-ray emission can co-exist inside elliptical galaxies during a certain phase of evolution subsequent to central outbursts. In fact, our calculations predict a synchrotron luminosity of 1.3×106L\sim 1.3\times 10^6 L_{\odot} at the frequency 5 GHz (radio band), of 1.1×106L\sim 1.1\times 10^6 L_{\odot} at 4.3×10144.3\times10^{14} Hz (R band, corresponding to the absolute magnitude -10.4), and of 1.5×107L\sim 1.5\times 10^{7} L_{\odot} at 2.4×10172.4\times10^{17} Hz (soft X-ray, 0.5 -- 2.0 keV band).Comment: 15 pages, 11 figures, resubmitted to ApJ, add one more figure, typos correcte
