Bei den seit 2003 laufenden Ausgrabungen wurden zahlreiche lithische und organische Siedlungsreste zu Tage gefördert, die dem mittleren und frühen Magdalénien, dem Badegoulien und dem Solutréen zugeordnet werden können. Die ältesten Schichten der Stratigraphie sind noch zu ergraben.The site of Le Petit Cloup Barrat opens on the edge of a small doline, 800 meters from the entrance of the famous decorated cave, Pech Merle. Though partially excavated at the beginning of the 20th century, this site has never been published. A test pit was opened in 2003, followed by excavations since 2004. The stratigraphic sequence, the base of which is still unknown, has yielded abundant lithic and organic remains attributed to the Middle and Lower Magdalenian, Badegoulian and Solutrean. In the upper ensemble attributed to the Magdalenian, numerous flint blade tools (end scrapers, burins, borers and retouched blades) coexist with backed bladelets. The Badegoulian is identified based on flake tools, including raclettes and transverse burins. An original industry, characterized by the production of twisted bladelets backed on the right side made from carinate cores, can be attributed to a Lower Magdalenian distinct from the Badegoulian. Finally, the Solutrean, still little excavated, is characterised by fragmented shouldered points and one fragment of a willow leaf point. The organic industry includes eyed needles, retouchers on medium-sized ungulate diaphyses and antler projectile points. The body ornaments consist of grooved and/or pierced shells, especially Dentalium, and herbivore incisors (reindeer, ibex). A few fossils, some with distant origins, were also recovered. Archaeozoological analysis shows a strong presence of reindeer, while horse is also present in various levels. The osseous material preserved in the cave will allow precise analyses of the seasons of occupation, hunting strategies, and alimentary and technical priorities

    A trade-off between classical and quantum circuit size for an attack against CSIDH

    International audienceWe propose a heuristic algorithm to solve the underlying hard problem of the CSIDH cryptosystem (and other isogeny-based cryp-tosystems using elliptic curves with endomorphism ring isomorphic to an imaginary quadratic order O). Let ∆ = Disc(O) (in CSIDH, ∆ = −4p for p the security parameter). Let 0 < α < 1/2, our algorithm requires: • A classical circuit of size 2Õ (log(|∆|) 1−α). • A quantum circuit of size 2Õ (log(|∆|) α). • Polynomial classical and quantum memory. Essentially, we propose to reduce the size of the quantum circuit below the state-of-the-art complexity 2Õ (log(|∆|) 1/2) at the cost of increasing the classical circuit-size required. The required classical circuit remains subexponential, which is a superpolynomial improvement over the classical state-of-the-art exponential solutions to these problems.     Potential impact of a nonavalent HPV vaccine on the occurrence of HPV-related diseases in France

    International audienceBackground : Human Papillomavirus (HPV) infection is known to be associated with a number of conditions including cervical, vaginal, vulvar, penile, anal neoplasias and cancers, oropharynx cancers and genitals warts (GW). Two prophylactic vaccines are currently available: a bivalent vaccine designed to prevent HPV type 16 and 18 infection and a quadrivalent vaccine targeting HPV 6, 11, 16, and 18. In France, HPV vaccination is recommended in 11-14 year-old girls with a catch-up for girls aged 15-19. The objective of this study was to assess the potential impact of an HPV 6/11/16/18/31/33/45/52/58 nonavalent vaccine on anogenital and oropharyngeal HPV-related diseases in France.Methods : HPV genotype distributions from 6 multicentric retrospective studies (EDiTH I to VI) were analyzed including 516 cases of invasive cervical cancers (ICC), 493 high-grade cervical neoplasias (CIN2/3), 397 low-grade squamous intraepithelial lesions (LSIL), 423 GW, 366 anal cancer and 314 oropharyngeal carcinomas. Low and high estimates of HPV vaccine impact were calculated as follows: low estimate: prevalence of HPV 6/11/16/18/31/33/45/52/58 genotypes alone or in association but excluding presence of another HPV type; high estimate: prevalence of HPV 6/11/16/18/31/33/45/52/58 genotypes alone or in association, possibly in presence of another HPV type.Results : Estimates of potential impact varied from 85% (low estimate) to 92% (high estimate) for ICC, 77% to 90% for CIN2/3, 26% to 56% for LSIL, 69% to 90% for GW, 81% to 93% for anal cancer, and 41% to 44% for oropharyngeal carcinomas. Compared to the quadrivalent vaccine, the proportion of additional cases potentially prevented by the nonavalent vaccine was 9.9%-15.3% for ICC, 24.7%-33.3% for CIN2/3, 12.3%-22.7% for LSIL, 2.1%-5.4% for GW, 8.5%-10.4% for anal cancer, and 0.0%-1.6% for oropharyngeal carcinoma.Conclusions : The nonavalent HPV vaccine showed significant increased potential impact compared to the HPV 6/11/16/18 quadrivalent vaccine for ICC, CIN2/3 and LSIL. Considering a 100% vaccine efficacy and high vaccine coverage, about 90% of ICC, CIN2/3, GW or anal cancer cases could be prevented by a nonavalent HPV vaccine in France

    Facteurs de risque de la tuberculose multi-résistante dans la ville de Kinshasa en République Démocratique du Congo

    Introduction: L'objectif de cette étude était de déterminer les facteurs de risque associés à la tuberculose multi résistance à Kinshasa en République Démocratique du Congo. Méthodes: Il s'agissait d'une étude cas témoins. Les cas comprenaient tous les patients tuberculeux résistants à la rifampicine et à l'isoniazide notifiés à Kinshasa de janvier 2012 à juin 2013. Les témoins étaient les patients tuberculeux traités durant la même période que les cas et qui à la fin du traitement étaient déclarés guéris. Pour cette étude, nous avons obtenu une clairance éthique. Résultats: L'échantillon était constitué de 213 participants dont 132 hommes (62%) et 81 femmes (38%). L'âge médian était de 31ans (16-73 ans). Les facteurs associés significatifs (p&lt; 0,05) à la tuberculose multi résistante étaient le non-respect des heures de prise de médicaments (0R=111) (80% chez les cas et 4% chez les témoins), l'échec au traitement (0R=20) (76% chez les cas et 13% chez les témoins) ; la notion de tuberculose multi résistante dans la famille (0R=6.4) (28% chez les cas et 6% chez les témoins); la méconnaissance de la tuberculose multi résistante (0R=3.2) (31% chez les cas et 59% chez les témoins); un séjour en prison (0R=7.6) (10% chez les cas et 1% chez les témoins) et l'interruption du traitement (0R=6.1) ( 59% chez les cas et 19% chez les témoins). Conclusion: L'émergence de la tuberculose multi résistante peut être évitée par la mise en place des stratégies de diagnostic et de traitement appropriées.Pan African Medical Journal 2016; 2

    Papua New Guinean genomes reveal the complex settlement of north Sahul

    The settlement of Sahul, the lost continent of Oceania, remains one of the most ancient and debated human migrations. Modern New Guineans inherited a unique genetic diversity tracing back 50,000 years, and yet there is currently no model reconstructing their past population dynamics. We generated 58 new whole-genome sequences from Papua New Guinea, filling geographical gaps in previous sampling, specifically to address alternative scenarios of the initial migration to Sahul and the settlement of New Guinea. Here, we present the first genomic models for the settlement of northeast Sahul considering one or two migrations from Wallacea. Both models fit our data set, reinforcing the idea that ancestral groups to New Guinean and Indigenous Australians split early, potentially during their migration in Wallacea where the northern route could have been favored. The earliest period of human presence in Sahul was an era of interactions and gene flow between related but already differentiated groups, from whom all modern New Guineans, Bismarck islanders, and Indigenous Australians descend. The settlement of New Guinea was probably initiated from its southeast region, where the oldest archaeological sites have been found. This was followed by two migrations into the south and north lowlands that ultimately reached the west and east highlands. We also identify ancient gene flows between populations in New Guinea, Australia, East Indonesia, and the Bismarck Archipelago, emphasizing the fact that the anthropological landscape during the early period of Sahul settlement was highly dynamic rather than the traditional view of extensive isolation

    Assessing Zika Virus Transmission Within Households During an Outbreak in Martinique, 2015-2016.

    Since 2015, Zika virus (ZIKV) has caused large epidemics in the Americas. Households are natural targets for control interventions, but quantification of the contribution of household transmission to overall spread is needed to guide policy. We developed a modeling framework to evaluate this contribution and key epidemic features of the ZIKV epidemic in Martinique in 2015-2016 from the joint analysis of a household transmission study (n = 68 households), a study among symptomatic pregnant women (n = 281), and seroprevalence surveys of blood donors (n = 457). We estimated that the probability of mosquito-mediated within-household transmission (from an infected member to a susceptible one) was 21% (95% credible interval (CrI): 5, 51), and the overall probability of infection from outside the household (i.e., in the community) was 39% (95% CrI: 27, 50). Overall, 50% (95% CrI: 43, 58) of the population was infected, with 22% (95% CrI: 5, 46) of infections acquired in households and 40% (95% CrI: 23, 56) being asymptomatic. The probability of presenting with Zika-like symptoms due to another cause was 16% (95% CrI: 10, 23). This study characterized the contribution of household transmission in ZIKV epidemics, demonstrating the benefits of integrating multiple data sets to gain more insight into epidemic dynamics

    Meloidogyne incognita PASSE-MURAILLE (MiPM) Gene Encodes a Cell-Penetrating Protein That Interacts With the CSN5 Subunit of the COP9 Signalosome

    The pathogenicity of phytonematodes relies on secreted virulence factors to rewire host cellular pathways for the benefits of the nematode. In the root-knot nematode (RKN) Meloidogyne incognita, thousands of predicted secreted proteins have been identified and are expected to interact with host proteins at different developmental stages of the parasite. Identifying the host targets will provide compelling evidence about the biological significance and molecular function of the predicted proteins. Here, we have focused on the hub protein CSN5, the fifth subunit of the pleiotropic and eukaryotic conserved COP9 signalosome (CSN), which is a regulatory component of the ubiquitin/proteasome system. We used affinity purification-mass spectrometry (AP-MS) to generate the interaction network of CSN5 in M. incognita-infected roots. We identified the complete CSN complex and other known CSN5 interaction partners in addition to unknown plant and M. incognita proteins. Among these, we described M. incognita PASSE-MURAILLE (MiPM), a small pioneer protein predicted to contain a secretory peptide that is up-regulated mostly in the J2 parasitic stage. We confirmed the CSN5-MiPM interaction, which occurs in the nucleus, by bimolecular fluorescence complementation (BiFC). Using MiPM as bait, a GST pull-down assay coupled with MS revealed some common protein partners between CSN5 and MiPM. We further showed by in silico and microscopic analyses that the recombinant purified MiPM protein enters the cells of Arabidopsis root tips in a non-infectious context. In further detail, the supercharged N-terminal tail of MiPM (NTT-MiPM) triggers an unknown host endocytosis pathway to penetrate the cell. The functional meaning of the CSN5-MiPM interaction in the M. incognita parasitism is discussed. Moreover, we propose that the cell-penetrating properties of some M. incognita secreted proteins might be a non-negligible mechanism for cell uptake, especially during the steps preceding the sedentary parasitic phase
