    Susceptibilities of enterococcus faecalis biofilms to some antimicrobial medications

    Enterococcus faecalis has bEnterococcus faecalis has been suggested to be an important etiological agent in endodontic failures. The purpose of this study was to evaluate the effectiveness of chlorhexidine- or antibiotics-based medications in eliminating E. faecalis biofilms. One-day and three-day biofilms of E. faecalis were induced on cellulose nitrate membrane filters. Each biofilm-containing membrane was thoroughly covered with 1 ml of the test medications and incubated for 1 day at 37°C. Treated biofilms were then aseptically transferred to vials containing a neutralizing agent in saline solution and vortexed. Suspensions were 10-fold diluted, seeded onto Mitis salivarius agar plates, and the colony-forming units counted after 48 h of incubation. There were significant differences between the formulations tested. The association of clindamycin with metronidazole significantly reduced the number of cells in 1-day biofilms. However of all medications tested, only 2% chlorhexidine-containing medications were able to thoroughly eliminate most of both 1-day and 3-day E. faecalis biofilms

    New genetic loci link adipose and insulin biology to body fat distribution.

    Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P < 5 × 10(-8)). In total, 20 of the 49 waist-to-hip ratio adjusted for BMI loci show significant sexual dimorphism, 19 of which display a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms

    A previsão de demanda é uma atividade importante para auxiliar na determinação dos recursos necessários para a empresa. Neste artigo, a metodologia de Box-Jenkins foi utilizada para analisar dados históricos de uma empresa de assistência técnica de computadores pessoais e obter previsões do número de atendimentos. A empresa estudada apresenta três tipos de clientes diferenciados: contratos, garantia e avulsos. Como cada segmento de clientes tem suas peculiaridades, a previsão de demanda foi direcionada a cada tipo, buscando representar o comportamento de tendência e a sazonalidade por meio dos modelos de Box-Jenkins. A obtenção dos modelos mais adequados foi baseada na análise de gráficos e em testes estatísticos próprios da metodologia, os quais subsidiaram a decisão de adotar o modelo AR(1) para prever o número de atendimentos dos clientes tipo contrato, o modelo ARIMA(2,1,0) para os clientes tipo garantia e um modelo sazonal SARIMA(0,1,0)(0,1,1)12 para os clientes tipo avulsos.<br>Demand forecasting is an important tool to aid on the determination of necessary resources of a given company. In this paper, the Box-Jenkins methodology was applied to analyze historical data of a personal computer repair company and provide a forecast for the number of service calls. The company studied presents three segments of clients: contracts, warranty, and on-call. As each client has it own characteristics, in order to better represent tendency and seasonality behavior through the Box-Jenkins models, a specific forecasting model was developed for each segment. The choice of the optimum models were based into graphic analysis and statistical tests, which lead to the decision of adopting the AR(1) model to foresee the number of contract clients, the ARIMA(2,1,0) model for warranty clients and the SARIMA(0,1,0)(0,1,1)12 seasonal model for on-call clients

    Molecular Thinking for Nanoplasmonic Design

    The development of nanoplasmonics has been tremendous during the past two decades, driven in part by the improvements in colloidal synthesis of nanocrystals and manipulation of nanoparticle surface functionalities. This has granted access not only to exquisite control over the morphology of nanoparticles but also to novel multiparticle nanostructures with a variety of organizational motifs. Driven by such new possibilities, completely unforeseen plasmonic effects have been found, which let us think about applications in a variety of fields. In this Perspective, we discuss the evolution of plasmonic nanomaterials and their corresponding properties and correlations with molecular concepts that have been around for a long time. Additional thinking along these lines may lead to further expansion of nanoplasmonics and to multiple surprising discoveries in this field
