    Suitability of the nisin Z-producer lactococcus lactis subsp. lactis CBM 21 to be used as an adjunct culture for squacquerone cheese production

    This research investigated the technological and safety effects of the nisin Z producer Lactococcus lactis subsp. lactis CBM 21, tested as an adjunct culture for the making of Squacquerone cheese in a pilot-scale plant. The biocontrol agent remained at a high level throughout the cheese refrigerated storage, without having a negative influence on the viability of the conventional Streptococcus thermophilus starter. The inclusion of CBM 21 in Squacquerone cheesemaking proved to be more effective compared to the traditional one, to reduce total coliforms and Pseudomonas spp. Moreover, the novel/innovative adjunct culture tested did not negatively modify the proteolytic patterns of Squacquerone cheese, but it gave rise to products with specific volatile and texture profiles. The cheese produced with CBM 21 was more appreciated by the panelists with respect to the traditional one

    The complex microbial community of withered berries of cv. Corvina as revealed by metagenomic analysis

    Corvina is the most important of Verona\u2019s red grape varieties and forms the basis for the production of unique wines, such as Amarone and Recioto of the Valpolicella area (Italy). The distinctive features of such wines are linked to the peculiar winemaking techniques and especially to the post-harvest grape withering process, when the grapes are partially dried in attics for three/four months to concentrate sugars up to about 27% (w/v). Dehydratation increases the concentration of constituent substances in berries, determining also specific expression of grape genes. During this process, grapes are colonized with a complex microbiota to an extent which depends on environmental temperature and humidity. In this study, a whole metagenome sequencing (WMS) approach was used to analyse the microbial consortia present on Corvina berries at the end of the withering process performed in two different conditions (\u201ctraditional\u201d or \u201cfast\u201d). Representative berry samples were collected and washed to avoid grapevine DNA contamination. Bioinformatic analyses, performed on reads and scaffolds, revealed that traditionally withered berries were dominated by Clostridiales and Pseudomonadales, while the fast procedure determined the presence of Enterobacteriales and Lactobacillales. On the contrary, few consistent differences characterized the eukaryotic fraction, dominated by Aspergillus and Penicillium for both samples. Interestingly, the \u201cbinning\u201d procedure revealed 15 most abundant genomes characterizing the two conditions. These results provide insights into the microbial community of Corvina withered berries and reveal relevant variations attributable to environmental withering conditions. Further studies will be performed to determine whether the different microbial compositions could lead to significant chemical variations of the musts, with an impact on the organoleptic properties of wine. This study underlines how novel technologies, like WMS, could open novel perspectives in the knowledge and management of traditional processes as the withering process of Corvina, with an impact on the winemaking of important Italian wines

    Comparative transcriptomic analyses of Zymoseptoria tritici strains show complex lifestyle transitions and intraspecific variability in transcription profiles

    Zymoseptoria tritici causes Septoria tritici blotch (STB) on wheat. The disease interaction is characterized by clearly defined temporal phases of infection, ultimately resulting in the death of host tissue. Zymoseptoria tritici is a highly polymorphic species with significant intraspecific variation in virulence profiles. We generated a deep transcriptomic sequencing dataset spanning the entire time course of an infection using a previously uncharacterized, highly virulent Z. tritici strain isolated from a Swiss wheat field. We found that seven clusters of gene transcription profiles explained the progression of the infection. The earliest highly up-regulated genes included chloroperoxidases, which may help the fungus cope with plant defences. The onset of necrotrophy was characterized by a concerted up-regulation of proteases, plant cell wall-degrading enzymes and lipases. Functions related to nutrition and growth characterized late necrotrophy and the transition to saprotrophic growth on dead plant tissue. We found that the peak up-regulation of genes essential for mating coincided with the necrotrophic phase. We performed an intraspecies comparative transcriptomics analysis using a comparable time course infection experiment of the genome reference isolate IPO323. Major components of the fungal infection transcriptome were conserved between the two strains. However, individual small, secreted proteins, proteases and cell wall-degrading enzymes showed strongly differentiated transcriptional profiles between isolates. Our analyses illustrate that successful STB infections involve complex transcriptomic remodelling to up-regulate distinct gene functions. Heterogeneity in transcriptomes among isolates may explain some of the considerable variation in virulence and host specialization found within the species

    The influence of HAART on the efficacy and safety of pegylated interferon and ribavirin therapy for the treatment of chronic HCV infection in HIV-positive individuals

    <p>Abstract</p> <p>Objective</p> <p>This study was performed to investigate the impact of HAART versus no HAART and nucleoside free versus nucleoside containing HAART on the efficacy and safety of pegylated interferon and ribavirin therapy for the treatment of chronic HCV infection in HIV/HCV co-infected patients. In addition a control group of HCV mono-infected patients undergoing anti-HCV therapy was evaluated.</p> <p>Methods</p> <p>Multicenter, partially randomized, controlled clinical trial. HIV-negative and -positive patients with chronic HCV infection were treated with pegylated interferon alfa-2a and ribavirin (800 - 1200 mg/day) for 24 - 48 weeks in one of four treatment arms: HIV-negative (A), HIV-positive without HAART (B) and HIV-positive on HAART (C). Patients within arm C were randomized to receive open label either a nucleoside containing (C1) or a nucleoside free HAART (C2).</p> <p>Results</p> <p>168 patients were available for analysis. By intent-to-treat analysis similar sustained virological response rates (SVR, negative HCV-RNA 24 weeks after the end of therapy) were observed comparing HIV-negative and -positive patients (54% vs. 54%, p = 1.000). Among HIV-positive patients SVR rates were similar between patients off and on HAART (57% vs. 52%, p = 0.708). Higher SVR rates were observed in patients on a nucleoside free HAART compared to patients on a nucleoside containing HAART, though confounding could not be ruled out and in the intent-to-treat analysis the difference was not statistically significant (64% vs. 46%, p = 0.209).</p> <p>Conclusions</p> <p>Similar response rates for HCV therapy can be achieved in HIV-positive and -negative patients. Patients on nucleoside free HAART reached at least equal rates of sustained virological response compared to patients on standard HAART.</p

    Mother-offspring recognition via contact calls in cattle, Bos taurus.

    Individual recognition in gregarious species is fundamental in order to avoid misdirected parental investment. In ungulates, two very different parental care strategies have been identified: ‘hider’ offspring usually lie concealed in vegetation whereas offspring of ‘follower’ species remain with their mothers while they forage. These two strategies have been suggested to impact on mother-offspring vocal recognition, with unidirectional recognition of the mother by offspring occurring in hiders and bidirectional recognition in followers. In domestic cattle, Bos taurus, a facultative hider species, vocal communication and recognition have not been studied in detail under free-ranging conditions, where cows and calves can graze freely and where hiding behaviour can occur. We hypothesized that, as a hider species, cattle under these circumstances would display unidirectional vocal recognition. To test this hypothesis, we conducted playback experiments using mother-offspring contact calls. We found that cows were more likely to respond, by moving their ears and/or looking, turning or walking towards the loudspeaker, to calls of their own calves than to calls from other calves. Similarly, calves responded more rapidly, and were more likely to move their ears and/or look, turn or walk towards the loudspeaker, and to call back and/or meet their mothers, in response to calls from their own mothers than to calls from other females. Contrary to our predictions, our results suggest that mother-offspring vocal individual recognition is bidirectional in cattle. Additionally, mothers of younger calves tended to respond more strongly to playbacks than mothers of older calves. Therefore, mother responses to calf vocalizations are at least partially influenced by calf age

    Influence of IL28B Polymorphisms on Response to a Lower-Than-Standard Dose peg-IFN-α 2a for Genotype 3 Chronic Hepatitis C in HIV-Coinfected Patients

    Background: Data on which to base definitive recommendations on the doses and duration of therapy for genotype 3 HCV/HIV-coinfected patients are scarce. We evaluated the efficacy of a lower peginterferon-α 2a dose and a shorter duration of therapy than the current standard of care in genotype 3 HCV/HIV-coinfected patients. Methods and Findings: Pilot, open-label, single arm clinical trial which involved 58 Caucasian HCV/HIV-coinfected patients who received weekly 135 μg peginterferon-α 2a plus ribavirin 400 mg twice daily during 20 weeks after attaining undetectable viremia. The relationships between baseline patient-related variables, including IL28B genotype, plasma HCV-RNA, ribavirin dose/kg, peginterferon-α 2a and ribavirin levels with virological responses were analyzed. Only 4 patients showed lack of response and 5 patients dropped out due to adverse events related to the study medication. Overall, sustained virologic response (SVR) rates were 58.3% by intention-to-treat and 71.4% by per protocol analysis, respectively. Among patients with rapid virologic response (RVR), SVR and relapses rates were 92.6% and 7.4%, respectively. No relationships were observed between viral responses and ribavirin dose/kg, peginterferon-α 2a concentrations, ribavirin levels or rs129679860 genotype. Conclusions: Weekly 135 μg pegIFN-α 2a could be as effective as the standard 180 μg dose, with a very low incidence of severe adverse events. A 24-week treatment duration appears to be appropriate in patients achieving RVR, but extending treatment up to just 20 weeks beyond negativization of viremia is associated with a high relapse rate in those patients not achieving RVR.
