    Identifying novel genetic variants for brain amyloid deposition: a genome-wide association study in the Korean population

    Background: Genome-wide association studies (GWAS) have identified a number of genetic variants for Alzheimer's disease (AD). However, most GWAS were conducted in individuals of European ancestry, and non-European populations are still underrepresented in genetic discovery efforts. Here, we performed GWAS to identify single nucleotide polymorphisms (SNPs) associated with amyloid β (Aβ) positivity using a large sample of Korean population. Methods: One thousand four hundred seventy-four participants of Korean ancestry were recruited from multicenters in South Korea. Discovery dataset consisted of 1190 participants (383 with cognitively unimpaired [CU], 330 with amnestic mild cognitive impairment [aMCI], and 477 with AD dementia [ADD]) and replication dataset consisted of 284 participants (46 with CU, 167 with aMCI, and 71 with ADD). GWAS was conducted to identify SNPs associated with Aβ positivity (measured by amyloid positron emission tomography). Aβ prediction models were developed using the identified SNPs. Furthermore, bioinformatics analysis was conducted for the identified SNPs. Results: In addition to APOE, we identified nine SNPs on chromosome 7, which were associated with a decreased risk of Aβ positivity at a genome-wide suggestive level. Of these nine SNPs, four novel SNPs (rs73375428, rs2903923, rs3828947, and rs11983537) were associated with a decreased risk of Aβ positivity (p < 0.05) in the replication dataset. In a meta-analysis, two SNPs (rs7337542 and rs2903923) reached a genome-wide significant level (p < 5.0 × 10-8). Prediction performance for Aβ positivity increased when rs73375428 were incorporated (area under curve = 0.75; 95% CI = 0.74-0.76) in addition to clinical factors and APOE genotype. Cis-eQTL analysis demonstrated that the rs73375428 was associated with decreased expression levels of FGL2 in the brain. Conclusion: The novel genetic variants associated with FGL2 decreased risk of Aβ positivity in the Korean population. This finding may provide a candidate therapeutic target for AD, highlighting the importance of genetic studies in diverse populations

    Thermal Analysis of MIRIS Space Observation Camera for Verification of Passive Cooling

    We conducted thermal analyses and cooling tests of the space observation camera (SOC) of the multi-purpose infrared imaging system (MIRIS) to verify passive cooling. The thermal analyses were conducted with NX 7.0 TMG for two cases of attitude of the MIRIS: for the worst hot case and normal case. Through the thermal analyses of the flight model, it was found that even in the worst case the telescope could be cooled to less than 206°K. This is similar to the results of the passive cooling test (~200.2°K). For the normal attitude case of the analysis, on the other hand, the SOC telescope was cooled to about 160°K in 10 days. Based on the results of these analyses and the test, it was determined that the telescope of the MIRIS SOC could be successfully cooled to below 200°K with passive cooling. The SOC is, therefore, expected to have optimal performance under cooled conditions in orbit

    Thalidomide is known to have anti-inflammatory and immunomodulatory actions. However, the effect and the anti-asthmatic mechanism of thalidomide in the pathogenesis of asthmatic airways are not fully understood.This study is designed to determine the effect and the potential mechanism of thalidomide in the pathogenesis of asthmatic airways using animal model of allergic asthma.Six-week-old female BALB/C mice were sensitized with alum plus ovalbumin (OVA) and were exposed to OVA via intranasal route for 3 days for challenge. Thalidomide 200 mg/kg was given via gavage twice a day from a day before the challenge and airway hyperresponsivenss (AHR), airway inflammatory cells, and cytokines in bronchoalveolar lavage fluids (BALF) were evaluated. The expression levels of pro-inflammatory cytokines and other mediators were evaluated using ELISA, real time (RT)-qPCR, and flow cytometry. CRL-2456, alveolar macrophage cell line, was used to test the direct effect of thalidomide on the activation of macrophages in vitro.The mice with thalidomide treatment showed significantly reduced levels of allergen-induced BALF and lung inflammation, AHR, and the expression of a number of pro-inflammatory cytokines and mediators including Th2 related, IL-17 cytokines, and altered levels of allergen-specific IgG1/IgG2a. Of interesting note, thalidomide treatment significantly reduced expression levels of allergen- or Th2 cytokine-stimulated alternative activation of macrophages in vivo and in vitro.These studies highlight a potential use of thalidomide in the treatment of allergic diseases including asthma. This study further identified a novel inhibitory effect of thalidomide on alternative activation of macrophages as a potential mechanism of anti-asthmatic effect of thalidomide

    Development of the Earth Observation Camera of MIRIS

    We have designed and manufactured the Earth observation camera (EOC) of multi-purpose infrared imaging system (MIRIS). MIRIS is a main payload of the STSAT-3, which will be launched in late 2012. The main objective of the EOC is to test the operation of Korean IR technology in space, so we have designed the optical and mechanical system of the EOC to fit the IR detector system. We have assembled the flight model (FM) of EOC and performed environment tests successfully. The EOC is now ready to be integrated into the satellite system waiting for operation in space, as planned

    Conceptual Design of the NISS onboard NEXTSat-1

    The NISS onboard NEXTSat-1 is being developed by Korea astronomy and space science institute (KASI). For the study of the cosmic star formation history, the NISS performs the imaging spectroscopic observation in the near-infrared range for nearby galaxies, low background regions, star-forming regions and so on. It is designed to cover a wide field of view (2 × 2 deg) and a wide wavelength range from 0.95 to 3.8 μm by using linear variable filters. In order to reduce the thermal noise, the telescope and the infrared sensor are cooled down to 200 K and 80 K, respectively. Evading a stray light outside the field of view and making the most use of limited space, the NISS adopts the off-axis reflective optical system. The primary and the secondary mirrors, the opto-mechanical part and the mechanical structure are designed to be made of aluminum material. It reduces the degradation of optical performance due to a thermal variation. This paper presents the study on the conceptual design of the NISS
