    Preparation and Characterization of Simvastatin Loaded PLGA Microparticles forTissue Engineering Applications

    Simvastatin has been reported to promote osteoblastic activity and inhibit osteoclastic activity. The successful use of simvastatin to promote in vivo bone formation depends on the local concentration, and there have been continuous efforts to find an appropriate delivery system for local delivery. Controlled drug delivery approaches based on microparticles could be a promising approach for sustained-localized delivery of simvastatin. In this study, simvastatin-loaded PLGA microparticles were prepared by using a modified single emulsion-solvent evaporation method. Uniform, spherical simvastatin loaded PLGA microparticles of size below 10μm were produced by adopting three different drug polymer ratios such as 1:40, 1:20 and 1:10 with encapsulation efficiency above 85%w/w irrespective to the drug polymer ratio and maximum simvastatin loading within PLGA microparticles was observed at drug polymer ratio of 1:10. Two stage release of simvastatin from microparticles was observed for 45 days, illustrating a controlled release. Simvastatin loaded PLGA microparticles are compatible with hFOB cells and induced in vitro bio-mineralization during 11 days treatment. These studies illustrate the feasibility of achieving local delivery of simvastatin to induce in vivo bone formation activity by suitably engrafting simvastatin loaded microparticles within porous scaffolds

    Accessing e-government: challenges for citizens and organizations

    Abstract The Internet is becoming more integral to governments and their modes of doing business and delivering services. This is creating a new imperative to address the digital divide. In Australia, as shown in this article, citizens who are the biggest users of government services are the least likely to be connected to the internet. What can be done to connect the unconnected? The article explores what has been learned from some of the Australian initiatives for connecting the unconnected to online government services. It concludes that greater attention to community-based human capital development is needed. It gives examples of factors needed for success in building socially marginalized communities' interest, enthusiasm and capacity to interact and communicate via online technologies, thereby contributing to how successful e-government can be in delivering gains in efficiency and improved services

    Building a sustainable and desirable economy-in-society-in-nature

    This report is a synthesis of ideas about what this new economy-in-society-innature could look like and how we might get there. Most of the ideas presented here are not new. The coauthors of this report have published them in various forms over the last several decades, and many others have expressed similar ideas in venues too numerous to mention. What is new is the timing and the situation. The time has come when we must make a transition. We have no choice. Our present path is clearly unsustainable. As Paul Raskin has said, Contrary to the conventional wisdom, it is business as usual that is the utopian fantasy; forging a new vision is the pragmatic necessity [10]. But we do have a choice about how to make the transition and what the new state of the world will be. We can engage in a global dialogue to envision the future we want, the theme of Rio+20, and then devise an adaptive strategy to get us there, or we can allow the current system to collapse and rebuild from a much worse starting point. We obviously argue for the former strategy. In this report, we discuss the need to focus more directly on the goal of sustainable human well-being rather than merely GDP growth. This includes protecting and restoring nature, achieving social and intergenerational fairness (including poverty alleviation), stabilizing population, and recognizing the significant nonmarket contributions to human well-being from natural and social capital. To do this, we need to develop better measures of progress that go well beyond GDP and begin to measure human well-being and its sustainability more directly

    Building a Sustainable and Desirable Economy-in-Society-in-Nature

    The world has changed dramatically. We no longer live in a world relatively empty of humans and their artifacts. We now live in the “Anthropocene,” era in a full world where humans are dramatically altering our ecological life-support system. Our traditional economic concepts and models were developed in an empty world. If we are to create sustainable prosperity, if we seek “improved human well-being and social equity, while significantly reducing environmental risks and ecological scarcities,” we are going to need a new vision of the economy and its relationship to the rest of the world that is better adapted to the new conditions we face. We are going to need an economics that respects planetary boundaries, that recognizes the dependence of human well-being on social relations and fairness, and that recognizes that the ultimate goal is real, sustainable human well-being, not merely growth of material consumption. This new economics recognizes that the economy is embedded in a society and culture that are themselves embedded in an ecological life-support system, and that the economy cannot grow forever on this finite planet. In this report, we discuss the need to focus more directly on the goal of sustainable human well-being rather than merely GDP growth.     Sonic enhancement of virtual exhibits

    Museums have widely embraced virtual exhibits. However, relatively little attention is paid to how sound may create a more engaging experience for audiences. To begin addressing this lacuna, we conducted an online experiment to explore how sound influences the interest level, emotional response, and engagement of individuals who view objects within a virtual exhibit. As part of this experiment, we designed a set of different soundscapes, which we presented to participants who viewed museum objects virtually. We then asked participants to report their felt affect and level of engagement with the exhibits. Our results show that soundscapes customized to exhibited objects significantly enhance audience engagement. We also found that more engaged audience members were more likely to want to learn additional information about the object(s) they viewed and to continue viewing these objects for longer periods of time. Taken together, our findings suggest that virtual museum exhibits can improve visitor engagement through forms of customized soundscape design

    A “Calvinist” Theory of Matter? Burgersdijk and Descartes on res extensa

    In the Dutch debates on Cartesianism of the 1640s, a minority believed that some Cartesian views were in fact Calvinist ones. The paper argues that, among others, a likely precursor of this position is the Aristotelian Franco Burgersdijk (1590-1635), who held a reductionist view of accidents and of the essential extension of matter on Calvinist grounds. It seems unlikely that Descartes was unaware of these views. The claim is that Descartes had two aims in his Replies to Arnauld: to show the compatibility of res extensa and the Catholic transubstantiation but also to differentiate the res extensa from some views of matter explicitly defended by some Calvinists. The association with Calvinism will be eventually used polemically against Cartesianism, for example in France. The paper finally suggests that, notwithstanding the points of conflict, the affinities between the theologically relevant theories of accidents, matter and extension ultimately facilitated the dissemination of Cartesianism among the Calvinists

    Using Automated Point Dendrometers to Analyze Tropical Treeline Stem Growth at Nevado de Colima, Mexico

    The relationship between wood growth and environmental variability at the tropical treeline of North America was investigated using automated, solar-powered sensors (a meteorological station and two dendrometer clusters) installed on Nevado de Colima, Mexico (19° 35′ N, 103° 37′ W, 3,760 m a.s.l.). Pure stands of Pinus hartwegii Lindl. (Mexican mountain pine) were targeted because of their suitability for tree-ring analysis in low-latitude, high-elevation, North American Monsoon environments. Stem size and hydroclimatic variables recorded at half-hour intervals were summarized on a daily timescale. Power outages, insect outbreaks, and sensor failures limited the analysis to non-consecutive months during 2001–2003 at one dendrometer site, and during 2002–2005 at the other. Combined data from the two sites showed that maximum radial growth rates occur in late spring (May), as soil temperature increases, and incoming short-wave radiation reaches its highest values. Early season (April–May) radial increment correlated directly with temperature, especially of the soil, and with solar radiation. Stem expansion at the start of the summer monsoon (June–July) was mostly influenced by moisture, and revealed a drought signal, while late season relationships were more varied

    Genetic association study of QT interval highlights role for calcium signaling pathways in myocardial repolarization.

    The QT interval, an electrocardiographic measure reflecting myocardial repolarization, is a heritable trait. QT prolongation is a risk factor for ventricular arrhythmias and sudden cardiac death (SCD) and could indicate the presence of the potentially lethal mendelian long-QT syndrome (LQTS). Using a genome-wide association and replication study in up to 100,000 individuals, we identified 35 common variant loci associated with QT interval that collectively explain ∼8-10% of QT-interval variation and highlight the importance of calcium regulation in myocardial repolarization. Rare variant analysis of 6 new QT interval-associated loci in 298 unrelated probands with LQTS identified coding variants not found in controls but of uncertain causality and therefore requiring validation. Several newly identified loci encode proteins that physically interact with other recognized repolarization proteins. Our integration of common variant association, expression and orthogonal protein-protein interaction screens provides new insights into cardiac electrophysiology and identifies new candidate genes for ventricular arrhythmias, LQTS and SCD
