    Concentração de nitrito endometrial e a ocorrência de patologias uterinas em vacas

    Relacionou-se a concentração de nitrito com a ocorrência de enfermidades uterinas. Lavado intrauterino de 1 ml de PBS foi realizado em 25 peças uterinas de vacas vazias para mensuração da concentração de nitrito pela reação de Griess. Elevada concentração de nitrito exibiu relação significativa com distrofia angiomatosa (157,57±108,80 µM), endometrite (102,96±87,58 µM) e fibrose periglandular (99,15±89,75 µM); e não significativa com e hiperplasida endometrial cística (94,07±18,89 µM) e adenomiose (77,40±81,25 µM). Contudo, elevada concentração de nitrito mostrou relação significativa com adenomiose acentuada profunda pelo teste t. Os resultados indicam que a síntese de óxido nítrico está elevada nessas enfermidades.The occurrence of uterine pathologies was evaluated considering uterine nitrite concentration. The uterus of 25 non pregnant cows was washed with 1ml of PBS to measure the nitrite concentration using the Griess reaction. There was significant relationship between high nitrite concentration with angiomatosis dystrophy (157,57±108,80 µM), endometritis (102,96±87,58 µM) and periglandular fibrosis (99,15±89,75 µM). No relation was found between nitrite concentration and cystic endometrial hyperplasia (94,07±18,89 µM) and adenomyosis (77,40±81,25 µM). However, high nitrite concentration showed a significant relationship with deep accentuate adenomyosis by t test. The data suggest that nitric oxide synthesis is elevated in these illnesses

    The effects of pioglitazone, a PPARγ receptor agonist, on the abuse liability of oxycodone among nondependent opioid users

    Aims: Activation of PPARγ by pioglitazone (PIO) has shown some efficacy in attenuating addictive-like responses in laboratory animals. The ability of PIO to alter the effects of opioids in humans has not been characterized in a controlled laboratory setting. The proposed investigation sought to examine the effects of PIO on the subjective, analgesic, physiological and cognitive effects of oxycodone (OXY). Methods: During this investigation, nondependent prescription opioid abusers (N = 17 completers) were maintained for 2-3 weeks on ascending daily doses of PIO (0 mg, 15 mg, 45 mg) prior to completing a laboratory session assessing the aforementioned effects of OXY [using a within-session cumulative dosing procedure (0, 10, and 20 mg, cumulative dose = 30 mg)]. Results: OXY produced typical mu opioid agonist effects: miosis, decreased pain perception, and decreased respiratory rate. OXY also produced dose-dependent increases in positive subjective responses. Yet, ratings such as: drug "liking," "high," and "good drug effect," were not significantly altered as a function of PIO maintenance dose. Discussion: These data suggest that PIO may not be useful for reducing the abuse liability of OXY. These data were obtained with a sample of nondependent opioid users and therefore may not be applicable to dependent populations or to other opioids. Although PIO failed to alter the abuse liability of OXY, the interaction between glia and opioid receptors is not well understood so the possibility remains that medications that interact with glia in other ways may show more promise

    Efeito de diferentes formas de cultivo na ação do óxido nítrico na maturação e na integridade da membrana plasmática de complexos cumulus-oócito em bovinos

    The aim of the present study was to evaluate the influence of different forms of in vitro culture on the nitric oxide action in maturation and membrane integrity on bovine cumulus-oocyte complex. No significant effect was observed between different forms of culture (mineral oil vs plate; P &gt; 0.05), as much for membrane integrity as for expansion of the CC. However, it was observed that oocytes of the groups control and 10-3 M of SNP, cultivated in plate, had presented greater percentage of cell with maintenance of membrane integrity than same treatments cultivated in drop. The addition of 10-3 M of SNP showed an inhibitory effect on the expansion and membrane integrity of CC and oocytes in both, culture in drops under oil and plate (P < 0.05). The culture form did not intervene with the extrusion of the first polar corpuscle and the addition of 10-3 M of SNP inhibited this extrusion in the both systems (P < 0.05). There was a dose-response effect on the concentration of NO in the maturation medium in both types of cultivation (P < 0,05), and this was higher in the culture medium under oil, except when added 10-3 M of SNP, treatment in which there was no difference in the types of cultivation employed. (PO objetivo do presente estudo foi avaliar se diferentes formas de cultivo interferem no efeito do óxido nítrico (NO) sobre a maturação e a integridade da membrana plasmática do complexo cumulus-oócito de bovinos. Para tanto, realizou-se cultivo em gotas sob óleo mineral ou em placas de quatro poços com a adição de diferentes concentrações de nitroprussiato de sódio (SNP, doador de óxido nítrico). Não foi observada diferença (P &gt; 0,05) entre as formas de cultivo quando se avaliou a integridade de membrana plasmática e a expansão das células do cumulus (CC). Contudo, os oócitos dos grupos controle e os cultivados na presença de 10-3 M de SNP, ambos cultivados em placa, apresentaram maior porcentagem de membrana íntegra do que os mesmos tratamentos cultivados em óleo mineral (P &lt; 0,05). Observou-se que a adição de 10-3 M de SNP diminuiu o grau de expansão das CC e de integridade da membrana plasmática dos oócitos, tanto no cultivo em gota sob óleo quanto em placa, diferindo dos outros grupos (P &lt; 0,05). Semelhante à expansão, a forma de cultivo não interferiu na extrusão do primeiro corpúsculo polar, sendo que a adição de 10-3 M de SNP inibiu a extrusão em ambos os sistemas (P &lt; 0,05). Houve um efeito dose-resposta na concentração de NO no meio de maturação em ambos os tipos de cultivo (P &lt; 0,05), sendo que esta foi maior no meio de cultivo sob óleo, exceção feita quando se adicionou 10-3 M de SNP, tratamento no qual não houve diferença nos tipos de cultivo empregados. Estes dados mostram que o sistema de cultivo não interferiu na ação do NO na maturação in vitro de COC bovinos, mas interfere na integridade da membrana plasmática do oócito

    Design and operation of a fast high-granularity silicon detector system in a high-radiation environment

    Abstract We have designed, realized and operated a fast silicon detector system (50 MHz sampling frequency) to measure the angular distribution and the multiplicity of charged secondaries produced in high-energy Pb–Pb interactions, within the NA50 experiment. We present here the detector design, discuss some of the problems faced during the commissioning and report on the first results on the operation of the full system. In particular, the questions related to the operation of an integrated high-speed binary readout in a high-radiation environment (1014 particles/cm2 and about 10 Mrads) and to the radiation effects on the system during the run will be addressed

    The dependence of the anomalous J/psi suppression on the number of participant nucleons

    The observation of an anomalous J/psi suppression in Pb-Pb collisions by the NA50 Collaboration can be considered as the most striking indication for the deconfinement of quarks and gluons at SPS energies. In this Letter, we determine the J/psi suppression pattern as a function of the forward hadronic energy E-ZDC measured in a Zero Degree Calorimeter (ZDC). The direct connection between EZDC and the geometry of the collision allows us to calculate, within a Glauber approach, the precise relation between the number of participant nucleons N-part and E-ZDC. Then, we check if the experimental data can be better explained by a sudden or a smooth onset of the anomalous J/psi suppression as a function of the number of participants.     Centrality Behaviour of J/ψ\psi Production in Na50

    The J/ψ\psi production in 158 A GeV Pb-Pb interactions is studied, in the dimuon decay channel, as a function of centrality, as measured with the electromagnetic or with the very forward calorimeters. After a first sharp variation at mid centrality, both patterns continue to fall down and exhibit a curvature change at high centrality values. This trend excludes any conventional hadronic model and is in agreement with a deconfined quark-gluon phase scenario.     Blood Magnesium, and the Interaction with Calcium, on the Risk of High-Grade Prostate Cancer

    Ionized calcium (Ca) and magnesium (Mg) compete as essential messengers to regulate cell proliferation and inflammation. We hypothesized that inadequate Mg levels, perhaps relative to Ca levels (e.g. a high Ca/Mg ratio) are associated with greater prostate cancer risk.In this biomarker sub-study of the Nashville Men's Health Study (NMHS), we included 494 NMHS participants, consisting of 98 high-grade (Gleason≥7) and 100 low-grade cancer cases, 133 prostate intraepithelial neoplasia (PIN) cases, and 163 controls without cancer or PIN at biopsy. Linear and logistic regression were used to determine associations between blood Ca, Mg, and the Ca/Mg ratio across controls and case groups while adjusting for potential confounding factors.Serum Mg levels were significantly lower, while the Ca/Mg ratio was significantly higher, among high-grade cases vs. controls (p = 0.04, p = 0.01, respectively). Elevated Mg was significantly associated with a lower risk of high-grade prostate cancer (OR = 0.26 (0.09, 0.85)). An elevated Ca/Mg ratio was also associated with an increased risk of high-grade prostate cancer (OR = 2.81 (1.24, 6.36) adjusted for serum Ca and Mg). In contrast, blood Ca levels were not significantly associated with prostate cancer or PIN.Mg, Ca, or Ca/Mg levels were not associated with low-grade cancer, PIN, PSA levels, prostate volume, or BPH treatment.Low blood Mg levels and a high Ca/Mg ratio were significantly associated with high-grade prostate cancer. These findings suggest Mg affects prostate cancer risk perhaps through interacting with Ca

    Final Pre-40S Maturation Depends on the Functional Integrity of the 60S Subunit Ribosomal Protein L3

    Ribosomal protein L3 is an evolutionarily conserved protein that participates in the assembly of early pre-60S particles. We report that the rpl3[W255C] allele, which affects the affinity and function of translation elongation factors, impairs cytoplasmic maturation of 20S pre-rRNA. This was not seen for other mutations in or depletion of L3 or other 60S ribosomal proteins. Surprisingly, pre-40S particles containing 20S pre-rRNA form translation-competent 80S ribosomes, and translation inhibition partially suppresses 20S pre-rRNA accumulation. The GTP-dependent translation initiation factor Fun12 (yeast eIF5B) shows similar in vivo binding to ribosomal particles from wild-type and rpl3[W255C] cells. However, the GTPase activity of eIF5B failed to stimulate processing of 20S pre-rRNA when assayed with ribosomal particles purified from rpl3[W255C] cells. We conclude that L3 plays an important role in the function of eIF5B in stimulating 3′ end processing of 18S rRNA in the context of 80S ribosomes that have not yet engaged in translation. These findings indicate that the correct conformation of the GTPase activation region is assessed in a quality control step during maturation of cytoplasmic pre-ribosomal particles

    Cationic Amino Acid Transporter 2 Enhances Innate Immunity during Helicobacter pylori Infection

    Once acquired, Helicobacter pylori infection is lifelong due to an inadequate innate and adaptive immune response. Our previous studies indicate that interactions among the various pathways of arginine metabolism in the host are critical determinants of outcomes following infection. Cationic amino acid transporter 2 (CAT2) is essential for transport of l-arginine (L-Arg) into monocytic immune cells during H. pylori infection. Once within the cell, this amino acid is utilized by opposing pathways that lead to elaboration of either bactericidal nitric oxide (NO) produced from inducible NO synthase (iNOS), or hydrogen peroxide, which causes macrophage apoptosis, via arginase and the polyamine pathway. Because of its central role in controlling L-Arg availability in macrophages, we investigated the importance of CAT2 in vivo during H. pylori infection. CAT2−/− mice infected for 4 months exhibited decreased gastritis and increased levels of colonization compared to wild type mice. We observed suppression of gastric macrophage levels, macrophage expression of iNOS, dendritic cell activation, and expression of granulocyte-colony stimulating factor in CAT2−/− mice suggesting that CAT2 is involved in enhancing the innate immune response. In addition, cytokine expression in CAT2−/− mice was altered from an antimicrobial Th1 response to a Th2 response, indicating that the transporter has downstream effects on adaptive immunity as well. These findings demonstrate that CAT2 is an important regulator of the immune response during H. pylori infection
