    A laboratory micro-manufacturing method for assessing individual cheese yield

    This study aimed to propose a micro-manufacturing method for assessing individual cheese yield (CY) from dairy milk and to provide first evaluations of the method. Two water baths (WB) provided with 8 stainless containers (SC) each were used. The cheese making procedure involved coagulation of 500 ml of milk per SC previously inoculated with starter, cutting of the curd, separation of whey through drainage and pressure and weighing of the curd residue for computing CY. Spraydried milk powder and bulk milk were used to validate the procedure. For each type of milk, 5 cheese making sessions (CMS) were performed during 5 consecutive days by the same operator; for each trial 80 records (8 replicates x 2WB x 5CMS) were available. Sources of variation of CY were investigated separately for type of milk, and variance components were estimated using REML procedure for computing instrumental repeatability and reproducibility. Cheese yield averaged 11% and 13.4% for standard milk powder and bulk milk, respectively. Cheese making session significantly influenced CY, along with WB in the case of milk powder, while SC did not influence CY. Reproducible and repeatable measures of CY were obtained, indicating the method is suitable for assessing individual CY

    Monitoring of sensory attributes used in the quality payment system of Trentingrana cheese

    Abstract Trentingrana is a Protected Designation of Origin (PDO) hard cheese manufactured in the valleys of Trento province (eastern Italian Alps) by several small cooperative dairies linked in a consortium. Nine months after production, wheels are delivered to a shared facility, ripened up to 18 mo, and assessed by a panel of 8 experts for 7 sensory attributes; namely, external aspect, rind thickness, paste color, texture, odor, taste, and aroma. The evaluation takes place every 2 mo on wheels sampled within each dairy. Based on the results of the assessment, dairies receive a price premium or penalty depending on a quality index, which is the weighted sum of the scores attributed to each sampled wheel. Sensory scores and quality index of 652 wheels representing 11 dairies and 10 yr of production were analyzed using a model that included fixed effects of dairy, year, and season of production, and first-order interactions between them. The coefficients of determination ranged from 0.50 (texture) to 0.66 (aroma). All factors significantly affected the studied traits, with the exception of interactions between dairy and season of production for texture and external aspect, and between year and season of production for odor. Dairy was the most important source of variation for visually assessed traits (external aspect, rind thickness, paste color, and texture) and for quality index, whereas year of production was the most important for flavor attributes (odor, taste, and aroma). The latter traits were always highly correlated among them and with the quality index, whereas correlations among visually assessed attributes, between them and flavor attributes, and between them and the quality index were more erratic. The sensory evaluation performed by the panel of experts has proven to be a useful tool to define the quality index and address the payment system of Trentingrana cheese, but it has some limitations in correctly describing the sensory profile of cheese and identifying specific defects and possible remedies

    Field-induced p-n transition in yttria-stabilized zirconia

    Oxide ion conducting yttria-stabilised zirconia ceramics show the onset of electronic conduction under a small bias voltage. Compositions with a high yttria content undergo a transition from p-type to n-type behavior at voltages in the range 2.4 to 10 V, which also depends on oxygen partial pressure. Surface reactions have a direct influence on bulk electronic conductivities, with possible implications for voltage-induced flash phenomena and resistive switching

    Isolation and functional characterization of proinflammatory acidic phospholipase A2 from Bothrops leucurus snake venom

    AbstractIn the present study, an acidic PLA2, designated Bl-PLA2, was isolated from Bothrops leucurus snake venom through two chromatographic steps: ion-exchange on CM-Sepharose and hydrophobic chromatography on Phenyl-Sepharose. Bl-PLA2 was homogeneous on SDS-PAGE and when submitted to 2D electrophoresis the molecular mass was 15,000Da and pI was 5.4. Its N-terminal sequence revealed a high homology with other Asp49 acidic PLA2s from snake venoms. Its specific activity was 159.9U/mg and the indirect hemolytic activity was also higher than that of the crude venom. Bl-PLA2 induced low myotoxic and edema activities as compared to those of the crude venom. Moreover, the enzyme was able to induce increments in IL-12p40, TNF-α, IL-1ÎČ and IL-6 levels and no variation of IL-8 and IL-10 in human PBMC stimulated in vitro, suggesting that Bl-PLA2 induces proinflammatory cytokine production by human mononuclear cells. Bothrops leucurus venom is still not extensively explored and knowledge of its components will contribute for a better understanding of its action mechanism

    Waning efficacy in a long-term AAV-mediated gene therapy study in the murine model of Krabbe disease

    Neonatal AAV9-gene therapy of the lysosomal enzyme galactosylceramidase (GALC) significantly ameliorates central and peripheral neuropathology, prolongs survival, and largely normalizes motor deficits in Twitcher mice. Despite these therapeutic milestones, new observations identified the presence of multiple small focal demyelinating areas in the brain after 6-8 months. These lesions are in stark contrast to the diffuse, global demyelination that affects the brain of naive Twitcher mice. Late-onset lesions exhibited lysosomal alterations with reduced expression of GALC and increased psychosine levels. Furthermore, we found that lesions were closely associated with the extravasation of plasma fibrinogen and activation of the fibrinogen-BMP-SMAD-GFAP gliotic response. Extravasation of fibrinogen correlated with tight junction disruptions of the vasculature within the lesioned areas. The lesions were surrounded by normal appearing white matter. Our study shows that the dysregulation of therapeutic GALC was likely driven by the exhaustion of therapeutic AAV episomal DNA within the lesions, paralleling the presence of proliferating oligodendrocyte progenitors and glia. We believe that this is the first demonstration of diminishing expression in vivo from an AAV gene therapy vector with detrimental effects in the brain of a lysosomal storage disease animal model. The development of this phenotype linking localized loss of GALC activity with relapsing neuropathology in the adult brain of neonatally AAV-gene therapy-treated Twitcher mice identifies and alerts to possible late-onset reductions of AAV efficacy, with implications to other genetic leukodystrophies

    Characterizing the gamma-ray long-term variability of PKS 2155-304 with H.E.S.S. and Fermi-LAT

    Studying the temporal variability of BL Lac objects at the highest energies provides unique insights into the extreme physical processes occurring in relativistic jets and in the vicinity of super-massive black holes. To this end, the long-term variability of the BL Lac object PKS 2155-304 is analyzed in the high (HE, 100 MeV 200 GeV) gamma-ray domain. Over the course of ~9 yr of H.E.S.S observations the VHE light curve in the quiescent state is consistent with a log-normal behavior. The VHE variability in this state is well described by flicker noise (power-spectral-density index {\ss}_VHE = 1.10 +0.10 -0.13) on time scales larger than one day. An analysis of 5.5 yr of HE Fermi LAT data gives consistent results ({\ss}_HE = 1.20 +0.21 -0.23, on time scales larger than 10 days) compatible with the VHE findings. The HE and VHE power spectral densities show a scale invariance across the probed time ranges. A direct linear correlation between the VHE and HE fluxes could neither be excluded nor firmly established. These long-term-variability properties are discussed and compared to the red noise behavior ({\ss} ~ 2) seen on shorter time scales during VHE-flaring states. The difference in power spectral noise behavior at VHE energies during quiescent and flaring states provides evidence that these states are influenced by different physical processes, while the compatibility of the HE and VHE long-term results is suggestive of a common physical link as it might be introduced by an underlying jet-disk connection.Comment: 11 pages, 16 figure

    Hadron Energy Reconstruction for the ATLAS Calorimetry in the Framework of the Non-parametrical Method

    This paper discusses hadron energy reconstruction for the ATLAS barrel prototype combined calorimeter (consisting of a lead-liquid argon electromagnetic part and an iron-scintillator hadronic part) in the framework of the non-parametrical method. The non-parametrical method utilizes only the known e/he/h ratios and the electron calibration constants and does not require the determination of any parameters by a minimization technique. Thus, this technique lends itself to an easy use in a first level trigger. The reconstructed mean values of the hadron energies are within ±1\pm 1% of the true values and the fractional energy resolution is [(58±3)/E+(2.5±0.3)[(58\pm3)% /\sqrt{E}+(2.5\pm0.3)%]\oplus (1.7\pm0.2)/E. The value of the e/he/h ratio obtained for the electromagnetic compartment of the combined calorimeter is 1.74±0.041.74\pm0.04 and agrees with the prediction that e/h>1.7e/h > 1.7 for this electromagnetic calorimeter. Results of a study of the longitudinal hadronic shower development are also presented. The data have been taken in the H8 beam line of the CERN SPS using pions of energies from 10 to 300 GeV.Comment: 33 pages, 13 figures, Will be published in NIM
