    SMME Performance—Results from a Longitudinal Study (2003-2012)

    The small and medium enterprise sector is often purported to be a significant driving force within the modern economy. Over the last two decades various studies have concluded that creativity and innovation are influential factors that contributed significantly to job and wealth creation. The purpose of this study was firstly, to determine how SMMEs created jobs consistently over a ten year period. Secondly, the study aimed to determine customer, profit and sales growth as measures of business success. This study involved a longitudinal research design with a quantitative approach and a sample size of 117 participants. The pilot study started in 2003 with data obtained from 117 participants. 49% of these participants were still operational in 2012. 52% of these business owners prepared to provide data. The remainder of the 117 participants (represented by 51%) were untraceable and therefore, assumed to have failed. The performance parameters included the number of employees, customers, value of sales and profit. Our research clearly showed that when the economic situation is favourable, SMMEs create jobs and wealth, however, as soon as there is a downturn in the economy, both job and wealth creation is reduced

    Interventions for enhancing adherence with physiotherapy: a systematic review

    Poor adherence to treatment is commonplace and may adversely affect outcomes, efficiency and healthcare cost. The aim of this systematic review was to identify strategies to improve adherence with musculoskeletal outpatient treatment. Five suitable studies were identified which provided moderate evidence that a motivational cognitive-behavioural programme can improve attendance at exercise-based clinic sessions. There was conflicting evidence that adherence interventions increase short-term adherence with exercise. There was strong evidence that adherence strategies are not effective at improving long-term adherence with home exercise. Due to the multi-dimensional nature of non-adherence, the strategies to improve adherence with physiotherapy treatment are likely to be broad in spectrum. Combined interventions may be effective at promoting adherence with clinic appointments and exercise, though further research would be required to confirm this. Further research to increase basic understanding of the factors which act as a barrier to adherence could facilitate development of strategies to overcome non-adherence

    Interaction of intense vuv radiation with large xenon clusters

    The interaction of atomic clusters with short, intense pulses of laser light to form extremely hot, dense plasmas has attracted extensive experimental and theoretical interest. The high density of atoms within the cluster greatly enhances the atom--laser interaction, while the finite size of the cluster prevents energy from escaping the interaction region. Recent technological advances have allowed experiments to probe the laser--cluster interaction at very high photon energies, with interactions much stronger than suggested by theories for lower photon energies. We present a model of the laser--cluster interaction which uses non-perturbative R-matrix techniques to calculate inverse bremsstrahlung and photoionization cross sections for Herman-Skillman atomic potentials. We describe the evolution of the cluster under the influence of the processes of inverse bremsstrahlung heating, photoionization, collisional ionization and recombination, and expansion of the cluster. We compare charge state distribution, charge state ejection energies, and total energy absorbed with the Hamburg experiment of Wabnitz {\em et al.} [Nature {\bf 420}, 482 (2002)] and ejected electron spectra with Laarmann {\em et al.} [Phys. Rev. Lett. {\bf 95}, 063402 (2005)]

To view a copy of this license, visit http://creativecommons.org/licenses/by/4.0/The post-translational addition of C-16 long chain fatty acids to protein cysteine residues is catalysed by palmitoyl-S-acyl-transferases (PAT) and affects the affinity of a modified protein for membranes and therefore its subcellular localisation. In apicomplexan parasites this reversible protein modification regulates numerous biological processes and specifically affects cell motility, and invasion of host cells by Plasmodium falciparum merozoites and Toxoplasma gondii tachyzoites. Using inhibitor studies we show here that palmitoylation is key to transformation of zygotes into ookinetes during initial mosquito infection with P. berghei. We identify DHHC2 as a unique PAT mediating ookinete formation and morphogenesis. Essential for life cycle progression in asexual blood stage parasites and thus refractory to gene deletion analyses, we used promoter swap (ps) methodology to maintain dhhc2 expression in asexual blood stages but down regulate expression in sexual stage parasites and during post-fertilization development of the zygote. The ps mutant showed normal gamete formation, fertilisation and DNA replication to tetraploid cells, but was characterised by a complete block in post-fertilisation development and ookinete formation.     Renewable Electric Energy Integration: Quantifying the Value of Design of Markets for International Transmission Capacity

    Integrating large quantities of supply-driven renewable electricity generation remains a political and operational challenge. One of the main obstacles in Europe to installing at least 200 GWs of power from variable renewable sources is how to deal with the insufficient network capacity and the congestion that will result from new flow patterns. We model the current methodology for controlling congestion at international borders and compare its results, under varying penetrations of wind power, with a model that simulates an integrated European network that utilises nodal/localised marginal pricing. The nodal pricing simulations illustrate that congestion - and price - patterns vary considerably between wind scenarios and within countries, and that a nodal price regime could make fuller use of existing EU network capacity, introducing substantial operational cost savings and reducing marginal power prices in the majority of European countries.Power market design, renewable power integration, congestion management, transmission economics

    The New Zealand Kauri (Agathis Australis) Research Project: A Radiocarbon Dating Intercomparison of Younger Dryas Wood and Implications for IntCal13

    We describe here the New Zealand kauri (Agathis australis) Younger Dryas (YD) research project, which aims to undertake Δ14C analysis of ~140 decadal floating wood samples spanning the time interval ~13.1–11.7 kyr cal BP. We report 14C intercomparison measurements being undertaken by the carbon dating laboratories at University of Waikato (Wk), University of California at Irvine (UCI), and University of Oxford (OxA). The Wk, UCI, and OxA laboratories show very good agreement with an interlaboratory comparison of 12 successive decadal kauri samples (average offsets from consensus values of –7 to +4 14C yr). A University of Waikato/University of Heidelberg (HD) intercomparison involving measurement of the YD-age Swiss larch tree Ollon505, shows a HD/Wk offset of ~10–20 14C yr (HD younger), and strong evidence that the positioning of the Ollon505 series is incorrect, with a recommendation that the 14C analyses be removed from the IntCal calibration database

    Expression capable library for studies of Neisseria gonorrhoeae, version 1.0

    Background The sexually transmitted disease, gonorrhea, is a serious health problem in developed as well as in developing countries, for which treatment continues to be a challenge. The recent completion of the genome sequence of the causative agent, Neisseria gonorrhoeae, opens up an entirely new set of approaches for studying this organism and the diseases it causes. Here, we describe the initial phases of the construction of an expression-capable clone set representing the protein-coding ORFs of the gonococcal genome using a recombination-based cloning system. Results The clone set thus far includes 1672 of the 2250 predicted ORFs of the N. gonorrhoeae genome, of which 1393 (83%) are sequence-validated. Included in this set are 48 of the 61 ORFs of the gonococcal genetic island of strain MS11, not present in the sequenced genome of strain FA1090. L-arabinose-inducible glutathione-S-transferase (GST)-fusions were constructed from random clones and each was shown to express a fusion protein of the predicted size following induction, demonstrating the use of the recombination cloning system. PCR amplicons of each ORF used in the cloning reactions were spotted onto glass slides to produce DNA microarrays representing 2035 genes of the gonococcal genome. Pilot experiments indicate that these arrays are suitable for the analysis of global gene expression in gonococci. Conclusion This archived set of Gateway® entry clones will facilitate high-throughput genomic and proteomic studies of gonococcal genes using a variety of expression and analysis systems. In addition, the DNA arrays produced will allow us to generate gene expression profiles of gonococci grown in a wide variety of conditions. Together, the resources produced in this work will facilitate experiments to dissect the molecular mechanisms of gonococcal pathogenesis on a global scale, and ultimately lead to the determination of the functions of unknown genes in the genome

    Geometric reconstruction methods for electron tomography

    Electron tomography is becoming an increasingly important tool in materials science for studying the three-dimensional morphologies and chemical compositions of nanostructures. The image quality obtained by many current algorithms is seriously affected by the problems of missing wedge artefacts and nonlinear projection intensities due to diffraction effects. The former refers to the fact that data cannot be acquired over the full 180180^\circ tilt range; the latter implies that for some orientations, crystalline structures can show strong contrast changes. To overcome these problems we introduce and discuss several algorithms from the mathematical fields of geometric and discrete tomography. The algorithms incorporate geometric prior knowledge (mainly convexity and homogeneity), which also in principle considerably reduces the number of tilt angles required.
