    Riscos psicossociais da atividade docente e análise do discurso: uma investigação acerca da saúde e segurança do professor de educação básica a partir dos princípios da ergonomia

    Occupational psychosocial risks (ROP) are constituted by a set of factors that alter the worker's well-being, in relation to the work environment in which it operates. There are many complaints of psychic motivations that end up engendering pathologies at the corporal level, especially with regard to the teaching work of basic education. Usually, these professionals have multiple factors for constant health absences and frequent complaints of stress, generalized bodily pain, fatigue and depression, possibly resulting from a labor activity of constant disapproval and devaluation in today's society. The objective of the study is to identify the existence of ROP in the work environment of primary school teachers and its repercussions on the mental health of these workers. Participated in the survey 7 teachers of basic education of a municipal school in Itabira. The study was based theoretically on the ergonomic perspective and was methodologically based on the assumptions of the qualitative research, having as methodological procedures the bibliographic research and the field research. A questionnaire was used as information collection technique. The results of the questionnaire indicated an index of 100% of the sample in agreement with regard to the psychosocial risks present in the working conditions. Regarding the variable quality of life and health, the results revealed significant relationships between absence of work and health. It is remarkable a mix of psychological and sociological factors that intersect generating the sickness of the teacher.Los riesgos ocupacionales psicosociales (ROP) están constituidos por un conjunto de factores que alteran el bienestar del trabajador, con relación al ambiente de trabajo en que actúa. Son muchas las quejas de motivaciones psíquicas que acaban engendrando patologías en el nivel corporal, principalmente, en lo que concierne al trabajo docente de educación básica. Normalmente, estos profesionales poseen múltiples factores para constantes ausencias por motivos de salud y frecuentes quejas de estrés, dolores corporales generalizados, fatiga y depresión, posiblemente resultantes de una actividad laboral de constantes desaprobaciones y devaluación en la sociedad actual. El objetivo del estudio es identificar la existencia de ROP en el ambiente laboral de profesores de la enseñanza básica y sus repercusiones en la salud mental de esos trabajadores. Participaron de la investigación 7 profesores de la educación básica de una escuela municipal de Itabira. El estudio se basó teóricamente en la perspectiva ergonómica y se basó metodológicamente en los supuestos de la investigación cualitativa, teniendo como procedimientos metodológicos la investigación bibliográfica y la investigación de campo. Se utilizó como técnica de recolección de información un cuestionario. Los resultados del cuestionario apuntaron un índice del 100% de la muestra en concordancia en lo que se refiere a los riesgos psicosociales presentes en las condiciones de trabajo. Con respecto a la variable calidad de vida y salud, los resultados revelaron relaciones significativas entre la ausencia del trabajo y la salud. Normalmente, esses profissionais possuem múltiplos fatores para constantes ausências por motivo de saúde e frequentes queixas de estresse, dores corporais generalizadas, fadiga e depressão, possivelmente resultantes de uma atividade laboral de constantes desaprovações e desvalorização na sociedade atual. O objetivo do estudo é identificar a existência de ROP no ambiente laboral de professores do ensino básico e suas repercussões na saúde mental desses trabalhadores.     The Flagella of an Atypical Enteropathogenic Escherichia coli Strain Are Required for Efficient Interaction with and Stimulation of Interleukin-8 Production by Enterocytes in Vitro

    The ability of some typical enteropathogenic Escherichia coli (EPEC) strains to adhere to, invade, and increase interleukin-8 (IL-8) production in intestinal epithelial cells in vitro has been demonstrated. However, few studies regarding these aspects have been performed with atypical EPEC (aEPEC) strains, which are emerging enteropathogens in Brazil. in this study, we evaluated a selected aEPEC strain (1711-4) of serotype O51:H40, the most prevalent aEPEC serotype in Brazil, in regard to its ability to adhere to and invade Caco-2 and T84 cells and to elicit IL-8 production in Caco-2 cells. the role of flagella in aEPEC 1711-4 adhesion, invasion, and IL-8 production was investigated by performing the same experiments with an isogenic aEPEC mutant unable to produce flagellin (FliC), the flagellum protein subunit. We demonstrated that this mutant (fliC mutant) had a marked decrease in the ability to adhere to T84 cells and invade both T84 and Caco-2 cells in gentamicin protection assays and by transmission electron microscopy. in addition, the aEPEC 1711-4 fliC mutant had a reduced ability to stimulate IL-8 production by Caco-2 cells in early (3-h) but not in late (24-h) infections.     Environmental representativity in marine protected area networks over large and partly unexplored seascapes

    Converting assemblages of marine protected areas (MPAs) into functional MPA networks requires political will, multidisciplinary information, coordinated action and time. We developed a new framework to assist planning environmental representativity in a network across the marine space of Portugal, responding to a political commitment to protect 14% of its area by 2020. An aggregate conservation value was estimated for each of the 27 habitats identified, from intertidal waters to the deep sea. This value was based on expert-judgment scoring for environmental properties and features relevant for conservation, chosen to reflect the strategic objectives of the network, thus providing an objective link between conservation commitments and habitat representativity in space. Additionally, habitats' vulnerability to existing anthropogenic pressures and sensitivity to climate change were also scored. The area coverage of each habitat in Portugal and within existing MPAs (regionally and nationally) was assigned to a scale of five orders of magnitude (from 10%) to assess rarity and existing representation. Aggregate conservation value per habitat was negatively correlated with area coverage, positively correlated with vulnerability and was not correlated with sensitivity. The proposed framework offers a multi-dimensional support tool for MPA network development, in particular regarding the prioritization of new habitats to protect, when the goal is to achieve specific targets while ensuring representativity across large areas and complex habitat mosaics.     Long‐term safety, efficacy, and quality of life in patients with juvenile idiopathic arthritis treated with intravenous abatacept for up to seven years

The efficacy and safety of abatacept in patients with juvenile idiopathic arthritis (JIA) who experienced an inadequate response to disease‐modifying antirheumatic drugs were previously established in a phase III study that included a 4‐month open‐label lead‐in period, a 6‐month double‐blind withdrawal period, and a long‐term extension (LTE) phase. The aim of this study was to present the safety, efficacy, and patient‐reported outcomes of abatacept treatment (10 mg/kg every 4 weeks) during the LTE phase, for up to 7 years of followup. Methods. Patients enrolled in the phase III trial could enter the open‐label LTE phase if they had not achieved a response to treatment at month 4 or if they had received abatacept or placebo during the double‐blind period. Results. One hundred fifty‐three (80.5%) of 190 patients entered the LTE phase, and 69 patients (36.3%) completed it. The overall incidence rate (events per 100 patient‐years) of adverse events decreased during the LTE phase (433.61 events during the short‐term phase [combined lead‐in and double‐blind periods] versus 132.39 events during the LTE phase). Similar results were observed for serious adverse events (6.82 versus 5.60), serious infections (1.13 versus 1.72), malignancies (1.12 versus 0), and autoimmune events (2.26 versus 1.18). American College of Rheumatology (ACR) Pediatric 30 (Pedi 30) responses, Pedi 70 responses, and clinically inactive disease status were maintained throughout the LTE phase in patients who continued to receive therapy. Improvements in the Child Health Questionnaire physical and psychosocial summary scores were maintained over time. Conclusion. Long‐term abatacept treatment for up to 7 years was associated with consistent safety, sustained efficacy, and quality‐of‐life benefits in patients with JIA

    Cross-scanner and cross-protocol multi-shell diffusion MRI data harmonization: algorithms and result

    Cross-scanner and cross-protocol variability of diffusion magnetic resonance imaging (dMRI) data are known to be major obstacles in multi-site clinical studies since they limit the ability to aggregate dMRI data and derived measures. Computational algorithms that harmonize the data and minimize such variability are critical to reliably combine datasets acquired from different scanners and/or protocols, thus improving the statistical power and sensitivity of multi-site studies. Different computational approaches have been proposed to harmonize diffusion MRI data or remove scanner-specific differences. To date, these methods have mostly been developed for or evaluated on single b-value diffusion MRI data. In this work, we present the evaluation results of 19 algorithms that are developed to harmonize the cross-scanner and cross-protocol variability of multi-shell diffusion MRI using a benchmark database. The proposed algorithms rely on various signal representation approaches and computational tools, such as rotational invariant spherical harmonics, deep neural networks and hybrid biophysical and statistical approaches. The benchmark database consists of data acquired from the same subjects on two scanners with different maximum gradient strength (80 and 300 ​mT/m) and with two protocols. We evaluated the performance of these algorithms for mapping multi-shell diffusion MRI data across scanners and across protocols using several state-of-the-art imaging measures. The results show that data harmonization algorithms can reduce the cross-scanner and cross-protocol variabilities to a similar level as scan-rescan variability using the same scanner and protocol. In particular, the LinearRISH algorithm based on adaptive linear mapping of rotational invariant spherical harmonics features yields the lowest variability for our data in predicting the fractional anisotropy (FA), mean diffusivity (MD), mean kurtosis (MK) and the rotationally invariant spherical harmonic (RISH) features. But other algorithms, such as DIAMOND, SHResNet, DIQT, CMResNet show further improvement in harmonizing the return-to-origin probability (RTOP).     Development of an amplicon-based sequencing approach in response to the global emergence of mpox

    The 2022 multicountry mpox outbreak concurrent with the ongoing Coronavirus Disease 2019 (COVID-19) pandemic further highlighted the need for genomic surveillance and rapid pathogen whole-genome sequencing. While metagenomic sequencing approaches have been used to sequence many of the early mpox infections, these methods are resource intensive and require samples with high viral DNA concentrations. Given the atypical clinical presentation of cases associated with the outbreak and uncertainty regarding viral load across both the course of infection and anatomical body sites, there was an urgent need for a more sensitive and broadly applicable sequencing approach. Highly multiplexed amplicon-based sequencing (PrimalSeq) was initially developed for sequencing of Zika virus, and later adapted as the main sequencing approach for Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). Here, we used PrimalScheme to develop a primer scheme for human monkeypox virus that can be used with many sequencing and bioinformatics pipelines implemented in public health laboratories during the COVID-19 pandemic. We sequenced clinical specimens that tested presumptively positive for human monkeypox virus with amplicon-based and metagenomic sequencing approaches. We found notably higher genome coverage across the virus genome, with minimal amplicon drop-outs, in using the amplicon-based sequencing approach, particularly in higher PCR cycle threshold (Ct) (lower DNA titer) samples. Further testing demonstrated that Ct value correlated with the number of sequencing reads and influenced the percent genome coverage. To maximize genome coverage when resources are limited, we recommend selecting samples with a PCR Ct below 31 Ct and generating 1 million sequencing reads per sample. To support national and international public health genomic surveillance efforts, we sent out primer pool aliquots to 10 laboratories across the United States, United Kingdom, Brazil, and Portugal. These public health laboratories successfully implemented the human monkeypox virus primer scheme in various amplicon sequencing workflows and with different sample types across a range of Ct values. Thus, we show that amplicon-based sequencing can provide a rapidly deployable, cost-effective, and flexible approach to pathogen whole-genome sequencing in response to newly emerging pathogens.     Tracking the international spread of SARS-CoV-2 lineages B.1.1.7 and B.1.351/501Y-V2

Using a combination of data from routine surveillance, genomic sequencing and international travel we track the international dispersal of lineages B.1.1.7 and B.1.351 (variant 501Y-V2). We account for potential biases in genomic surveillance efforts by including passenger volumes from location of where the lineage was first reported, London and South Africa respectively. Using the software tool grinch (global report investigating novel coronavirus haplotypes), we track the international spread of lineages of concern with automated daily reports, Further, we have built a custom tracking website (cov-lineages.org/global_report.html) which hosts this daily report and will continue to include novel SARS-CoV-2 lineages of concern as they are detected.Peer reviewe

    We show the distribution of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) genetic clades over time and between countries and outline potential genomic surveillance objectives. We applied three genomic nomenclature systems to all sequence data from the World Health Organization European Region available until 10 July 2020. We highlight the importance of real-time sequencing and data dissemination in a pandemic situation, compare the nomenclatures and lay a foundation for future European genomic surveillance of SARS-CoV-2

    Creation of a pandemic memory by tracing COVID-19 infections and immunity in Luxembourg (CON-VINCE).

    peer reviewed[en] BACKGROUND: During the COVID-19 pandemic swift implementation of research cohorts was key. While many studies focused exclusively on infected individuals, population based cohorts are essential for the follow-up of SARS-CoV-2 impact on public health. Here we present the CON-VINCE cohort, estimate the point and period prevalence of the SARS-CoV-2 infection, reflect on the spread within the Luxembourgish population, examine immune responses to SARS-CoV-2 infection and vaccination, and ascertain the impact of the pandemic on population psychological wellbeing at a nationwide level. METHODS: A representative sample of the adult Luxembourgish population was enrolled. The cohort was followed-up for twelve months. SARS-CoV-2 RT-qPCR and serology were conducted at each sampling visit. The surveys included detailed epidemiological, clinical, socio-economic, and psychological data. RESULTS: One thousand eight hundred sixty-five individuals were followed over seven visits (April 2020-June 2021) with the final weighted period prevalence of SARS-CoV-2 infection of 15%. The participants had similar risks of being infected regardless of their gender, age, employment status and education level. Vaccination increased the chances of IgG-S positivity in infected individuals. Depression, anxiety, loneliness and stress levels increased at a point of study when there were strict containment measures, returning to baseline afterwards. CONCLUSION: The data collected in CON-VINCE study allowed obtaining insights into the infection spread in Luxembourg, immunity build-up and the impact of the pandemic on psychological wellbeing of the population. Moreover, the study holds great translational potential, as samples stored at the biobank, together with self-reported questionnaire information, can be exploited in further research.
